Studies of the actions of Aconiti tuber butanol fraction on the mechanical and electrical properties of isolated rabbit atrium (부자(附子) Butanol fraction의 강심작용(强心作用)에 관한 연구(硏究))

  • In Chinese medicine, it is said that Aconiti tuber has cardiotonic, diuretic and analgesic effects. Kim et al reported that alkaloid free part of Aconiti tuber, $CHCI_3$ insoluble fraction, showed inotropic effect on isolated frog heart and inotropic effect is potenciated by n-butanol fractionation. To investigate the effect of Aconiti tuber butanol fraction on the mechanical and electrical properties of heart, change of active tension, excitability and refractory period of isolated rabbit atrium in the presence of butanol fraction were measured and the comparison with that of ouabain and quinidine was done. The observed results are as follows. 1. $5{\times}10^{-4}g/ml$ concentration of Aconiti tuber butanol fraction showed approximately same effect with therapeutic concentration of ouabain on the increment of contractile force, and the effect of $2{\times}10^{-3}g/ml$ was greater than that of $1{\times}10^{-5}g/ml$ of ouabain. 2. Acceleration of rate of contractile force increment in the presence of Aconiti tuber butanol fraction was greater than in ouabain, and the time to maximum tension was shorter in Aconiti tuber butanol fraction than in ouabain. 3. The excitability of isolated atrium was slightly increased at low concentration of Aconiti tuber butanol fraction, while decreased at higher concentration. 4. Aconiti tuber butanol fraction slightly prolonged refractory period of isolated right atrium at the concentration of $2{\times}10^{-3}g/ml$.

  • This study focuses on preparation of PVDF/PDMS composite membranes to effectively separate butanol from water-butanol mixture using pervaporation. We prepared various composite membranes by changing PVDF concentration of support layer and PDMS cross-linking condition of active layer. Pervaporation performance was tested by measuring butanol flux and separation factor with various cases of butanol concentration, temperature, and flow rate of feed. As results, performance of our novel PVDF/PDMS membranes surpasses that of PVDF/POMS membrane, manufactured by GKSS (Germany), in term of butanol flux, permeate concentration, and separation factor.

  • Combustion characteristics of a 1-butanol gel fuel were studied in atmospheric pressure condition. The butanol gel fuel was manufactured by adding hydroxypropyl-methyl cellulose (HPMC) as a gellant and the effect of the gellant concentration was observed. The combustion process of a single butanol gel droplet was divided into 3 stages including droplet heating, microexplosion, and gellant combustion. The flame was distorted compared to butanol + water mixture because of micro-explosion during the combustion. Increase of gellant concentration delayed the droplet ignition, but the combustion rate was improved due to the mass ejection during the micro-explosion.

Removal Characteristics of Ethyl Acetate and 2-Butanol by a Biofilter Packed with Jeju Scoria

  • The removal characteristics of ethyl acetate and 2-butanol were investigated in a bench-scale down-flow biofilter packed with Jeju scoria medium. Various inlet concentrations and gas flow rates were tested. The adaptation times of microorganisms to the change of the influent concentration of ethyl acetate and 2-butanol gas were found to be about 3 days. At the inlet concentration of 300 ppmv and empty bed contact time (EBCT) of 15 see, the removal efficiencies of the biofilter for ethyl acetate and 2-butanol were above $99.9\%$. The maximum removal capacity of the biofilter for ethyl acetate was $316-318\;g/m^3/h$ and that for 2-butanol was $245-251\;g/m^3/h$. Overall, the removal capacity of the biofilter for ethyl acetate was $50-70\;g/m^3/h$ larger than that for 2-butanol. During the operation of 65 days, the pressure drop through the biofilter column was maintained below $13\;mmH_{2}O/m$. Although the pH in the drain water decreased from 7.2 to 5.0, the pH drop did not affect the removal of the gases. From the above results, the biofilter using Jeju scoria as a packing material seemed to very effectively treat waste gases such as ethyl acetate and 2-butanol.

The investigation on the Ignition Delay of n-heptane/n-butanol Blend Fuel using a Rapid Compression Machine at Low Temperature Combustion Regime (저온연소조건에서 급속압축기를 이용한 n-heptane/n-butanol 혼합연료의 착화지연에 관한 연구)

  • This study presents both experimental and numerical investigation of ignition characteristics of n-heptane and n-butanol mixture. The $O_2$ concentration was fixed to 9-10% to make high exhaust gas recirculation(EGR) rate condition. Experiments were performed using a rapid compression machine. In addition, a numerical study of the ignition delay time was performed using CHEMKIN codes to validate experimental results and predict chemical species after combustion process. The results showed that the ignition delay time increased with increasing n-butanol ratio and the reactivity decreased by low $O_2$ concentration.

Effects of Chenopodium album Linne on Gastritis and Gastric Cancer Cell Growth

  • In our previous study, we investigated Chenopodium album Linne (CAL) ethanol extract and its fractions on anti-gastritic actions using the HCl/ethanol and indomethacin induced gastric lesion model and Helicobacter pylori (H. pylori). Based on the results, butanol fraction was most effective among fractions obtained from CAL. This study aims to elucidate the mechanisms of butanol fraction, and betaine as a constituent of the butanol fraction, on gastritis and anti-gastric cancer cell growth. First, we examined antioxidant properties using hydrogen peroxide and superoxide radical, and we found that butanol fraction and betaine may be good antioxidants. Second, cytotoxicity was assessed by measuring cell viability and 4,6-diamidino-2-phenylinodole dihydrochloride (DAPI) staining of human gastric cancer cells (AGS cells). We also examined the relationship between the cytotoxicity and intracellular $Ca^{2+}$ signaling mechanism. The butanol fraction demonstrated cell viability 71.49% at the concentration of 100 ${\mu}g/ml$ and increased intracellular $Ca^{2+}$ concentration in a dose dependent manner. Finally, we observed the mucus content as a defensive factor and gastric secretion as an aggressive factor, and found that the mucus content noticeably increased when treated with butanol fraction and betaine and gastric secretion decreased when treated with betaine in vivo study. From these results, we suggest that CAL butanol fraction and betaine may have protective effects on gastritis.

Pervaporation of n-Butanol/water Mixture through Organophilic ZSM-5 Zeolite Membrane (소수성 ZSM-5 제올라이트 분리막을 이용한 n-부탄올/물 혼합물의 투과증발)

  • Organophilic ZSM-5 membrane was synthesized on the inside of a porous stainless steel support by a hydrothermal secondary growth with seed crystals. They are used to separate n-butanol from its aqueous solution. Pervaporation characteristics such as a permeation flux and a separation factor are investigated as a function of the feed concentration and the operating temperature. The concentration of n-butanol was changed from 0.001 mole fraction to 0.015 mole fraction with an interval of 0.005 mole fraction; the operating temperature was controlled to be 25C, $35^{\circ}C$ and $45^{\circ}C$. When the operating temperature was $45^{\circ}C$, the flux of n-butanol significantly increased from 2 to $27g/m^2/hr$ as the mole fraction of n-butanol in the feed side increased from 0.001 to 0.015. Consequently, the concentration of n-butanol in the permeate substantially increased from 0.0016 to 0.052 mole fraction. When the feed concentration was 0.015, the flux of n-butanol significantly increased from 13 to $27g/m^2/hr$ as the operating temperature increased from $25^{\circ}C$ to $45^{\circ}C$. As a result, the concentration of n-butanol in the permeate also increased from 0.045 to 0.052 mole fraction.

The effect of Aconiti tuber butanol fraction on the rabbit heart microsomal $Na^+-K^+$-activated ATPase activity (부자 Butanol Fraction이 가토 심장근 Microsomal $Na^+-K^+$-activated ATPase 활성도에 미치는 영향)

  • Aconiti tuber butanol fraction shows positive inotropic effect on the isolated atrium of rabbit heart. To investigate the mechanism, the effect on microsomal ATPase activity of rabbit heart is observed. The microsomal fraction which contains the $Na^+$- and $K^+$-activated ATPase in the presence of $Mg^{++}$ is isolated from the left ventricle of rabbit heart. The microsomal ATPase activity is maximally stimulated at $Na^+$ and $K^+$ concentration of 100 mM and 10 mM respectively. Microsomal $Na^+-K^+$-activated ATPase is inhibited by ouabain and Aconiti tuber butanol fraction. Ouabain and Aconiti tuber butanol fraction depress $Na^+$-stimulation on microsomal ATPase activity, and the inhibitory effects are not completely reversed at $Na^+$ concentration of 300 mM. Also, $K^+$-stimulation on microsomal ATPase activity is inhibited by ouabin and Aconiti tuber butanol fraction and the inhibitions are not compeletely reversed at $K^+$ concentration of 30 mM. It is, therefore, suggested that the inhibitory effect of Aconiti tuber butanol fraction on the microsomal ATPase activity may contribute to leading to the positive inotropic effect.

Pervaporation of Butanol from their Aqueous Solution using a PDMS-Zeolite Composite Membrane (PDMS-Zeolite 복합막을 이용한 부탄올 투과증발)

  • Pervaporation is known to be a low energy consumption process since it needs only an electric power to maintain the permeate side in vacuum. Also, the pervaporation is an environmentally clean technology because it does not use the third material such as an entrainer for either an azeotropic distillation or an extractive distillation. In this study, Silicalite-1 particles are hydrothermally synthesized and polydimethylsiloxane(PDMS)-zeolite composite membranes are prepared with a mixture of synthesized Silicalite-1 particles and PDMS-polymer. They are used to separate n-butanol from its aqueous solution. Pervaporation characteristics such as a permeation flux and a separation factor are investigated as a function of the feed concentration and the weight % of Silicalite-1 particles in the membrane. A 1,000 $cm^3$ aqueous solution containing butanol of low mole fraction such as order of 0.001 was used as a feed to the membrane cell while the pressure of the permeation side was kept about 0.2~0.3 torr. When the butanol concentration in the feed solution was 0.015 mole fraction, the flux of n-butanol significantly increased from 14.5 g/ $m^2$/hr to 186.3 g/$m^2$/hr as the Silicalite-1 content increased from 0 wt% to 10 wt%, indicating that the Silicalite-1 molecular sieve improved the membrane permselectivity from 4.8 to 11.8 due to its unique crystalline microporous structure and its strong hydrophobicity. Consequently, the concentration of n-butanol in the permeate substantially increased from 0.07 to 0.15 mole fraction. This composite membrane could be potentially appliable for separation of n-butanol from insitu fermentation broth where n-butanol is produced at a fairly low concentration of 0.015 mole fraction.

Modeling and Analysis of Extractive Butanol Fermentation with Pervaporation (투과증발을 이용한 부탄올 추출발효 시스템의 모델링과 분석)

  • Results from experiments and mathematical modeling were compared for pervaporative butanol fermentation. The developed model includes expressions to predict characteristics of butanol fermentation, such as, microbial growth, solvent (butanol, acetone, and ethanol) formation and organic acid (acetate and butyrate) production. Butanol diffusivity was 1.15${\times}$10(sup)-7 ㎡/hr at 1.5 L/min-tubing of air flow rate using a pervaporative module. The model correlated well with experimental results (cell growth, glucose consumption and concentrations of solvents and organic acids) for batch fermentation with and without pervaporation. Larger surface area and thinner module tubing resulted in an increased glucose consumption and a decreased residual butanol concentration. Optimum membrane area and thickness were 0.34 ㎡ and 120 $\mu\textrm{m}$, respectively.
