Diversity and Distribution of Bulb-associated fungi of Fritillaria Cirrhosae Bulbus Source Plants used in Traditional Chinese Medicine

  • Diversity and community composition of bulb-associated fungi of Fritillaria Cirrhosae Bulbus source plants, which are used in traditional chinese medicine, in the eastern Himalaya-Hengduan Mountains, southwestern China, were estimated based on the internal transcribed spacer rDNA sequence analysis, using host plant species, geographic area, and plant phenology as variables. A total of 1,486 fungal sequences assigned to 251 operational taxonomical units (OTUs) were obtained from the bulbs. Fungal OTUs comprised 96.41% Ascomycotina, 3.52% Basidiomycotina, and 0.07% Zygomycotina. Sordariomycetes, Hypocreales, and Nectriaceae were the most frequent fungal lineages at each taxonomic rank. Fusarium, Ilyonectria, Tetracladium, Leptodontidium, and Tomentella were the top OTU-rich genera. Fusarium sp. 03, Ilyonectria rufa, Fusarium sp. 08, Ilyonectria sp. 03, and Leptodontidium orchidicola 03 represented the most frequent OTUs. Fusarium spp. were the most frequent general taxa. The distribution of fungal community exhibited preferences for host plant species, geographic area, and plant phenology. These findings are the foundation of our research on culturing and active metabolites of bulb-associated fungi of Fritillaria Cirrhosae Bulbus source plants.

Identification of Pathogens Associated with Bulb Rot of Lily during Storage and Effects of Bulb Disinfection on Development of Lily Bulb Rot (백합 저장중 구근부패에 관여하는 병원균의 동정과 종구 소독효과)

  • Several pathogens associated with bulb rot of lilies in storage house were identified with respect to rot types. Rot patterns were grouped into four different types; brown rot of bottoms, brown rot of shoots, water-soaked rot and blue mold. Brown rot of bottoms was the highest in frequency with 72.5%, and brown rot of shoots the least with 23.0%. Dominant pathogens were differed with rot patterns, brown rot of bottoms by Fusarium oxysporum, blue mold and brown rot of shoots by Penicillium brevicompactum and P. fellutanum. In wound-inoculation tests, Penicillium and Fusarium isolates caused severe rot on the bulbs. Bulb disinfection before storage by captan showed the most prominent control value of 95.2% followed by thiophanate-methyl with 85.6%.

Preconditioning for Cryopreservation of in vitro Grown Bulblets of Lily using Droplet-Vitrification

  • This study was conducted to improve and supplement the system of cryopreservation for adventitious bulbs induced by tissue cultured bulb-scales of lily (Lilium spp.) cvs. 'Milky way'. The explants, bulblets and bulb-scale-bulblets, were treated to low temperature (4℃) for 7 days prior to the pre-culture. The adventitious bulbs were pre-cultured in Murashige and Skoog (MS) liquid medium supplemented with sucrose (0.3 and 0.7M). The pre-cultured adventitious bulbs were treated to loading solution (LS1 or LS2, C4 or C6) containing 35% of PVS3 (LS1, C4) or 40% of PVS3 (LS2, C6) for 40 min and exposed to dehydration solution (PVS3, B1) containing 50% glycerol and 50% sucrose for 60 min at 25℃. The adventitious bulbs were moved onto droplets containing 3 µl PVS3 on sterilized aluminum foils, and then soaked into liquid nitrogen (LN) for 60 min. The result of highest regrowth rate as 65.7% was obtained in cold treatment (4℃), osmoprotected with LS1 solution, and cultured in PCM3 medium by using bulb-scale-bulblet for cryopreservation. This result shows that droplet-vitrification could be used as a promising method for long-term storage of lily genetic resource.

The Effect of The Planting Time of Mother bulb and Mulching Materials for The Seed Production of Onion (Allium cepa L.) (양파 채종을 위한 모구의 재식시기와 멀칭재료가 채종량에 미치는 영향)

  • This study examines the effects of the planting time of mother bulb and mulching materials for the seed production of onion on the amount of seed production, and the following results are obtained : Growth, bolting rate, flowering rate, floret numbers per scape and fertilization rate are best in the case of the seed production under transparent vinyl mulching between Sept. 10 and Sept. 25, and seed yield per plant and seed yield per 10a are increased in the case of the seed production under transparent vinyl mulching between Sept. 10 and Sept. 25, so it is found that it is appropriate to plant the onion mother bulb under the transparent vinyl mulching between Sept. 10 and Sept. 25 in the southern areas of Korea.

Enhancing in vitro Growth of Bulbs for Mass Propagation of Lily Germplasm

  • Plants regenerated from in vitro cultures carry chromosomal variations, especially in long-term culture. Reducing the duration of plant tissue culture is one of the ways to reduce genetic and epigenetic changes. In this study, we reduced the duration of long-term culture and repeat subculture using small bulblets derived from bulb scales in two lily cultivars. The adventitious bulblets derived from bulb-scale tissue were cultured on three different media containing Murashige and Skoog (MS) basal medium supplemented with 1 g/L Charcoal, MS medium containing 0.3 mg/L IAA and 0.4 mg/L BA hormone with or without Charcoal, respectively. About seven weeks later, the number of newly propagated multiple shoots in the two media, A and B media, showed little differentiation. Compared to both media, the number of propagated multiple shoots increased 5-fold in MS medium containing 0.3 mg/L IAA and 0.4 mg/L BA hormone without Charcoal (C medium). The number of propagated multiple shoots ranged from 5 to 6 and 4 to 6 with an average of 5 in TropicalPink and GreenStar cultivars, respectively. The flow cytometric measurements indicated no variation in the ploidy level between control and in vitro propagated plants.

Pyruvic Acid Content according to Different Portions in Onion(Allium cepa L.) (양파의 부위에 따른 pyruvic acid 함량)

  • By analysis of pyruvic acid and sugar levels in bulbs, leaf sheaths, and leaves, and with regard to phyllotaxis, it was shown that the lower portion of each plant component had the greatest pyruvic acid content, the upper portion less, and the middle portion the lowest. When bulb scales were examined, pyruvic acid content was lowest in the second bulb scale from the papery scale. Pyruvic acid level increased toward the inner portion of the bulb. Pyruvic acid levels in leaf sheaths and leaves were greater than those in the bulb during the early stages of bulb growth. However, at harvest, the pyruvic acid content of bulbs increased slightly, whereas those of leaf sheaths and leaves decreased markedly. Tissue sugar contents were similar at the beginning of bulb growth. However, bulb sugar content increased greatly as the bulb grew, and sugar contents of leaf sheaths and leaves decreased. With respect to phyllotaxis, outer older leaves had the lowest total sugar and pyruvic acid contents. Young leaves showed a tendency to have less pyruvic acid and sugar compared with mature leaves.

  • To obtain information on the status of vegetative and reproductive growth of Fritillariae rhizoma-bulb were sown in 5 levels-weight (1. 5 gram, 3gram. .5gram, 7gram, and 9gram). Results observed were as follows; A clear plus correlations were shown between the germination status, plant height and other agronomic characteristics and the weight of rhizoma-bulb. As the weight of rhizoma-bulb was increasing, also the number of flower and yields were increased.

  • The zone of greenhouse in Korea increase on a scale yearly. Particularly, greenhouse take up a important value on the agriculture economy. Greenhouse of scale, material, heating system, and drain is progressing to industrialization. The dry bulb temperature, humidity, photosynthesis and so forth are necessary to maintain environment control of greenhouse. The dry bulb temperature among them greatly affects growing of a plant. The purpose of this study is to analyzed the indoor environment control system and the characteristic of dry bulb temperature distribution on a large scale greenhouse in summer season.
