Morphological characteristics and genetic diversity of Calanthe species native to Korea (한국 자생 새우난초의 형태적 특성 및 유전적 다양성)

  • This study was conducted to research the morphological characteristics and analyze the genetic diversity by using RAPD in Calanthe species native to Korea. Nine samples were selected by flower color and 19 morphological characteristics. In the length and width of leaf, dorsal sepal, the lateral sepal, the petal, the central lip, and the lateral lip, C. discolor was the shortest and narrowest, respectively, but C. sieboldii was the longest and the widest, respectively. The flower stalk length was the shortest in C. discolor, and the longest in C. sieboldii. Three variants were the intermediate between C. discolor and C. sieboldii in the above morphological characteristics, but spur length was the longest in C. discolor, the shortest in C. sieboldii, and intermediate in the variants. The ovary length in C. discolor was shortest and C. sieboldii and variants were similar with each other. The flower color of C. discolor were brownish red, the value of CIE Lab was between 40 and 50. The flower color of C. sieboldii was yellowish, the value of CIE Lab was between 110 and 130. And variants had various colors between 50 to 70 in the value of CIE Lab. By analyzing multiple band patterns of PCR products, 154 bands were selected as polymorphic RAPD markers. The analysis of genetic similarity of Calanther species using RAPD showed that C. discolor and C. sieboldii are more distant from each other than variants, and these results demonstrated that genetic position of variants located between C. discolor and C. sieboldii.

Investigation of Co- and Pr-doped yttria-stabilized cubic zirconia (YSZ) single crystal grown by skull melting method (스컬용융법에 의해 성장시킨 Co와 Pr이 첨가된 이트리아안정화큐빅지르코니아(YSZ) 단결정의 연구)

  • Co-(0.7 wt%) and Pr-(2.0, 3.5 or 5.0 wt%) doped cubic zirconia ($ZrO_2:Y_2O_3=50:50wt%$) single crystals grown by a skull melting method were heat-treated in $N_2$ at $1150^{\circ}C$ for 5 hrs. The brown colored as-grown single crystals were changed into either dark brownish green, greenish blue and light green color after the heat treatment. Before and after the heat treatment, the YSZ (yttria-stabilized zirconia) single crystals were cut for wafer form (${\phi}7.5mm{\times}t3mm$). The optical and structural properties were examined by UV-VIS spectrophotometer and X-ray diffraction. Absorption by $Co^{2+}$(${\fallingdotseq}589nm$: ${\Gamma}_8[^4A_2(^4F)]{\rightarrow}{\Gamma}_8+{\Gamma}_7[^4T_1(^4F)]$, ${\fallingdotseq}610nm$: ${\Gamma}_8[^4A_2(^4F)]{\rightarrow}{\Gamma}_8[^4T_1(^4F)]$], ${\fallingdotseq}661nm$: ${\Gamma}_8[^4A_2(^4F)]{\rightarrow}{\Gamma}_6[^4T_1(^4F)]$]) and $Pr^{3+}$(${\fallingdotseq}450nm$: ${^3}H{_4}-{^3}P{_2}$, ${\fallingdotseq}473nm$: ${^3}H{_4}{\rightarrow}{^3}P{_1}$, ${\fallingdotseq}484nm$: ${^3}H{_4}{\rightarrow}{^3}P{_0}$), change of ionization energy and lattice parameter were confirmed.

Gemological and Minearlogical Properties of the Red Garnet Stones (적색 석류석 보석의 보석.광물학적 특징)

  • Chemical composition, crystal structure, refractive index, specific gravity, color, and luster were studied fur pyrope-almandine series garnets. The main coloring agents determining the reddish or brownish garnets were also investigated. It was also examined if there is any relationship between mineralogical properties with respect to the various chemical compositions in the solid solution, in the hope to figure out the existing classification values of R.I. and S.G. using gem- testing facilities to distinguish pyrope from almadine. It was found that 17 out of the 24 specimens belong to pyrope and the rest almandine. R.I. of pyrope goes up to 1.77 and that of almandine is higher than the value.5.5. of pyrope reaches to 3.88 and that of almandine is greater than the value of pyrope. X-ray diffraction data revealed that pyrope-almandine garnets are isometric with space group Ia3d, and also show that the variation of cell parameters are not significant enough to parallel with the chemical compositions of the series. R.I. and S.G. increase with FeO content. Fe and Mn are most responsible to the red-purple and orange coloration of the specimens, respectively. Both zircon and rutile crystals are most common inclusions in the reddish stones.

Comparison of Growth Characteristics and Ginsenosides Content of 6-Year-Old Ginseng (Panax ginseng C. A. Meyer) by Drainage Class in Paddy Field (논토양에서 배수등급별 6년근 인삼의 생육특성 및 진세노사이드 함량 비교)

  • To develop the practical cultivation for paddy field, we investigated the properties of paddy soil, growth characteristics and ginsenoside content of 6-year-old ginseng, Cheonpung variety between poor drainage class (PDC) and imperfect drainage class (IDC). Groundwater level in PDC showed monthly small changes of 20~30 cm, while IDC showed monthly great changes of 28~71 cm depending on rainfall. Soil moisture content in PDC and IDC was 17.2%, 22.5%, respectively. Air temperature in IDC was lower than $0.3^{\circ}C$, while soil temperature was higher than $0.8^{\circ}C$ compare to PDC, respectively. Main soil color of PDC was grayish olive, while IDC was brownish olive. PDC showed yellowish mottles only at underground of 20~40 cm, while IDC showed that at underground of 30~90 cm. IDC showed lower pH, EC, potassium, calcium and magnesium content, but higher organic matter, phosphate, and iron content than that of PDC, respectively. All of EC, organic matter, potassium, calcium, and magnesium content were decreased, but iron content was increased at the subsoil layers of PDC. All of EC, organic matter, phosphorus, and potassium content were decreased, but calcium and magnesium content were increased at the subsoil layers of IDC. Root yield in IDC was more increased by 33% than that of PDC. The moisture content and rusty ratio of ginseng root in IDC were lower than that of PDC. Ginsenoside content in IDC was higher than that of PDC because the ratio of lateral and fine root showing relatively high content of ginsenoside was higher in IDC than that of PDC.

Cultural characteristics of Inonotus obliquus isolated from Betula costata at Mt. Jumbong in Korea (점봉산 거제수나무에서 분리한 차가버섯의 배양특성)

  • The sterile form of Inonotus obliquus is used for preparation of the medicine befungin that is effective in the treatment of gastritis, gastric ulcers, and several tumors. The fungus is known to be produced mainly on the stems of Betula platyphylla var. japonica that grows at high altitudinal (above 1,100 m) region in Korea. But, we found the mushroom on the stem of Betula costata at Mt. Jumbong in Korea. We isolated a pure culture of the fungus from the stem of B. costata by use of potato dextrose agar (PDA) medium with streptomycin. We could isolate the fungus from plant's tissue filled with hyphae, but not from other parts. The spore collected from the sclerotia showed $6.0{\sim}10.0{\times}4.5{\sim}6.0{\mu}m$ in diameter, and the hypha was $2.5{\sim}5.0{\mu}m$ in thickness. The colony showed irregular features and scattered yellow color at the center as the culture ages. We could find brownish setae at the yellow region of colony at 20 days of culture, and the size ranged $4{\sim}6{\times}100{\sim}420{\mu}m$. The oatmeal agar (OA) provided best growth for I. obliquus among five media (CDA, CMA, MA, OA and PDA). Optimum temperature ranged $25{\sim}30^{\circ}C$, and optimum pH was relatively alkaline with the range of pH 8.0~9.5.

Sclerotinia Rot on Basil Caused by Sclerotinia sclerotiorum in Korea (Sclerotinia sclerotiorum에 의한 바질 균핵병)

  • During growing season of 2011 to 2013, Sclerotinia rot symptoms consistently have been observed on basil in Yesan-gun, Chungcheongnam-do in Korea. The typical symptom formed initially brownish spot on leaf and stem, and then advancing margins, wilting the whole plant and blighting, eventually died. On the surface of diseased lesions was observed cottony, white, dense mat of mycelial growth, and sclerotia ($30-100{\mu}m$ diameter) formed on stem and leaf. Morphological and cultural characteristic on potato dextrose agar, color of colony was white and colorless chocolate, sclerotium of irregular shape of the oval was black and $5-50{\mu}m$ diameter in size. In pathogenicity test, necrosis and wilt of the inoculated stem were observed in all plants and the pathogen was reisolated from stems. On the basis of mycological characteristics, pathogenicity, and internal transcribed spacer rDNA sequence analysis, this fungus was identified as Sclerotinia sclerotiorum. This is the first report of Sclerotinia rot on basil caused by S. sclerotiorum in Korea.

Be Age-dating of Marine Sediments from NE Pacific (북동태평양 해저퇴적물의 Be 연대측정)

  • In order to investigate absolute age of marine sediment in the KR5 region, northeastern Pacific, we measured the Be isotope ($^{10}Be$ and $^{9}Be$) of box core (BC08-02-13) sediment with depth. Core sediment is divided into three sedimentary facies (Facies I, Facies II, FaciesIII). Facies I mainly consists of brown to dark brown (10YR4/3) homogeneous mud with high water content. Facies II shows brownish yellow (10YR6/6) color. The unconformity is recognized at the boundary between Facies I and Facies II, Facies III consists of very dark brown (10YR2/2) mud. Many bioturbated burrows are observed at FaciesII and FaciesIII. Based on Be dating results, FaciesIII and Facies II had been deposited before 3.7 Ma and 2.3 Ma, respectively. After 2.3 Ma, the upper part of Facies II was eroded due to the change of sedimenary environment. Facies I has been deposited since 1.8 Ma. The unconformity located at the boundary between Facies I and Facies II may be driven by the envrionmental change due to the migration of Intertropical Convergence Zone.

  • Brewing method and quality of 15 samples of traditional Kochuzang in Junrabook-do area were investigated. Average combination ratio of glutinous rice: Meju (soybean Koji): red pepper powder was 2 : 0.7 : 1.1. In order to improve the taste of Kochuzang, some amount of natural soysauce was added in the Kochuzang at early stage of fermentation. Average content of sodium chloride was 7.32%; amino nitrogen, 123.3%; total sugar, 31.9%; reducing sugar, 19.4%; ethyl alcohol, 1.13% and the activity of acid protease was 0.07; liquefying amylase, 9.67 and saccharogenic amylase was 15.82. Five samples were selected as excellent Kochuzang through sensory evaluation. These samples were similarly strong in hot and sweet taste with alcoholic flavor, glossy surface and brownish red color.

The orange, yellowish brown and brown colored as-grown single crystals were changed into either brownish red, yellow and green color after the heat treatment. Different Responses of Zoysiagrass (Zoysia spp.) Ecotypes against Puccinia zoysiae Causing Rust Disease in Field (포장에서 녹병 병원균인 Puccinia zoysiae에 대한 한국잔디의 종별 상이한 반응)

  • An obligate plant pathogenic fungus Puccinia zoysiae Dietel, the causal agent of rust disease, which is implicated in the damage of zoysiagrass (Zoysia spp.) in Korea. The fungus is one of the important pathogens, which attacks and colonizes susceptible zoysiagrass. Zoysia rust disease was observed in Jangseong, Hoengseong and Hapcheon in 2014 and 2015, Korea. The typical symptoms of the disease appeared first as small white spots on the leaf and stem of zoysiagrass plants, and turn brownish, dark brown spores revealed on the lesions. Uredospores were light brown in color, ellipsoid or spherical in shape and $22.0{\sim}25.0{\times}15.0{\sim}17.5{\mu}m$ in size. On the basis of the morphological characteristics, the fungus was identified as P. zoysiae. Pathogenicity of the fungus was proved by artificial inoculation on Z. japonica. The rust disease firstly appeared approx. early or middle June and then increased middle of October in Korea. In field, Z. matrella ecotypes were more resistant than Z. japonica ecotypes against the pathogen. Disease response against the pathogen was different among ecotypes of each species, respectively.
