Physicochemical Properties of a Giant Embryo Mutant Induced by T-DNA Insertion in Rice

  • This study was conducted to determine the physicochemical properties of a giant embryo rice 'P47JB-4-B-5-B' derived from the cross between 'P47', a mutant of 'Hwayoung' induced by T-DNA insertion, and 'Junam'. The grain appearance and chemical components of the embryo were analyzed and compared with a donor cultivar, 'Hwayoung'. The proportion of embryo weight to grain weight of 'P47 JB-4-B-5-B' was 2.2 times heavier (6.7%) than that (3.1%) of 'Hwayoung'. Total free amino acid content (75.81 mg/100 g) of 'P47JB-4-B-5-B' was 2.1 times higher than that of 'Hwayoung'. The GABA content in brown rice was 14.06 mg/100 g in 'P47JB-4-B-5-B' and 6.8 mg/100 g in 'Hwayoung'. Especially, the GABA content in brown rice of 'P47JB-4-B-5-B' remarkably increased (about 33 times from 1.48 mg to 44.81 mg/100 g) 2 days after germination. Continuous frequency distributions and transgressive segregation in embryo length and width were observed in the $F_2$ population of the cross between 'P47' and 'Cheongcheong', indicating that the giant embryo was controlled by quantitative trait loci. However, embryo length and width demonstrated high broad sense heritability, implying that giant embryonic traits could be selected in earlier generations in comparison with other quantitative traits.

Genetic Analysis of the Ability of Callus Formation and Plant Regeneration in Seed Culture of Rice (벼의 종자배양에서 캘러스 형성과 식물체 재분화 능력의 유전)

  • This study was conducted to determine the inheritance of the ability of callus formation and plant regeneration in seed cultures of rice. The culturabilities of three Japonica rices,'Chucheongbyeo', 'Nagdongbyeo', and 'Daeribbyeo 1', were higher than those of Tongil type cultivars, 'Milyang 23' and 'Samgangbyeo 'The frequency for callus growth in F₂ populations of the three crosses, 'Milyang 23/Chucheongbyeo', 'Milyang 23/Daeribbyeo 1', and' Samgangbyeo/Nagdongbyeo', revealed a nearly normal distribution. The broad-sense heritability estimated from the ability of callus formation in the crosses were ranged from 83.8% to 90.1%. The frequency distribution of plant regenerability in F₂ population of 'Milyang 23/Daeribbyeo 1' showed a continuous variation. But the segregation mode of plant regenerability from seed-derived callus in the F₂ progenies of 'Milyang 23/Chucheongbyeo' and 'Samgangbyeo/Nagdongbyeo' appeared to fit the expected 3 : 1 ratio for the high and low regenerability. These results suggest that the high plant regenerability of 'Chucheongbyeo' and 'Nagdongbyeo' was regulated by a single dominant gene.

  • Narrow and broad sense heritabilities for the characters ranged from 0.1618 to 0.6914 and from 0.7494 to 0.9357, respectively, in F$_1$ hybrids of $5{\times}5$ full diallel crosses. Plant height and days to flowering had the highest heritabilities and yield and quality the lowest. Partial dominance was exhibited by plant height, number of leaves, leaf area per plant and days to flowering. Over dorminance was detected by leaf width and yield of cured leaf. Leaf length and quality would be considered being controlled by a complete dominance effect.

Variation of Anther and Pistil Length in Rice (수도의 약장, 자약장의 변이)

  • The variations of the anther and pistil length of some rice cultivars at the different planting density and fertility levels were tested and their inheritance mode was studied. The anther length of a spikelet on a secondary panicle branch was longer than the one of a spikelet on a primary panicle branch. In the cultivar Z97B, both the anther and the pistil length were increased slightly along the increased plant-ing spacings, But, in the cultivar IR43 no general tendency was observed. The fertilizer levels applied n the field did not affect both the anther and pistil length of a given cultivar. In a cross of IR56/IR8, the anther length showed continuous variation with longer anther being recessive in the F2 population. While, the pistil length showed a continuous variation with longer pistil being over-dominance. In all of the crosses which IR56 involved, the general and specific combining abilities were high for both the anther and pistil length. The broad sense heritability for anther length was 0.46, and for pistil length was 0.88. The correlation coefficients of anther length and pistil length were 0.33 for phenotypic and 0.44 for geno-typic.

Varietal Difference and Inheritance of Plant Regenerability in Anther Culture of Rice (벼 악배양 효율의 품종간 차이와 식물체 재분화 능력의 유전)

  • This study was carried out to identify varietal difference and the inheritance of the ability of plant regeneration in anther culture of rice. The anthers of 33 Japonica, 11 Indica/Japonica, and 3 Indica rice cultivars were culture on $N_6-Y$_1$$ medium with 1 mg/L NAA and 2 mg/L kinetin. The remarkable variability in plant regeneration were observed among the genotypes, ranging from 0.0% to 19.4% Most of Japonica rices were resulted to have better culturability than that of Indica/Japonica and Indica type cultivars. Newly developed Japonica rices, 'Ilmibyeo' and 'Hangnambyeo' showed to have regenerability with the frequency of 19.4% and 18.1% respectively. The segregation mode for callus formation and plant regenerationin anther culture of F$_2$ population of 'Milyang 23/Chucheongbyeo' showed a continuous variation. The variation of plant regeneration frequency in anther culture of F$_2$ plants ranged from 0% to 33.3% with a mean of 6.3% The broad-sense heritability estimated from the ability of plant regeneration was considerably high (82.7%).

Genetic Variation of Flower Production in Breeding Seedling Seed Orchards of Quercus acuta and Q. glauca

  • This study was conducted to test the significant difference of fertility variation among families and to select superior families for acorn production in the breeding seedling seed orchards (BSSOs) of Quercus acuta and Quercus glauca. The seed orchards were located in Jeju island and established by seedlings raised from selected parents for genetic testing in 2006. In the spring of 2021, the numbers of female and male flower were counted from 5 to 10 individuals per family in the BSSOs. To test statistical significance of which parameter is not satisfied through the normality test, we used a nonparametric analysis. Correlation analysis was performed to quantify the association between female and male flower production. As the results, the significant difference of flower production among families was found in both seed orchards. The averages of female flower production were 65.3 and 181.9 in Q. acuta and Q. glauca. The positive Spearman's rank correlation was existed between male and female flower production. Broad-sense heritability on female and male flower production were 0.191 and 0.147 in Q. acuta, and 0.285 and 0.068 in Q. glauca, respectively. Sexual asymmetry (e.g., maleness index) between female and male, and contribution variation among families (e.g., parental balance) were analyzed to find reasonable alternatives in the management of seed orchards. Effective population size of seed crops was predicted as a concept of status number. Loss of gene diversity (accumulation of group coancestry) would not be alarming in the BSSOs. Our results would be helpful to select breeding materials for establishing new seed orchards and to supply genetically improved seeds of evergreen oaks, which is one of the backbones of the strategy of carbon sink in the 2050 Carbon Neutrality of Korea Forest Service.

Studies on the selection in soybean breeding. -II. Additional data on heritability, genotypic correlation and selection index- (대두육종에 있어서의 선발에 관한 실험적연구 -속보 : 유전력ㆍ유전상관, 그리고 선발지수의 재검토-)

  • The experimental studies were intended to clarify the effects of selection, and also aimed at estimating the heritabilities, the genotypic correlations among some agronomic characters, and at calculating the selection index on some selective characters for the selection of desirable lines, under different climatic conditions. Finally practical implications of these studies, especially on the selection index, were discussed. Twenty-two varieties, determinate growing habit type, were selected at random from the 138 soybean varieties cultivated the year before, were grown in a randomized block design with three replicates at Chinju, Korea, under May and June sowing conditions. The method of estimating heritabilities for the eleven agronomic characters-flowering date, maturity date, stem length, branch numbers per plant, stem diameter, plant weight, pod numbers per plant, grain numbers per plant and 100 grain weight, shown in Table 3, was the variance components procedures in a replicated trial for the varieties. The analysis of covariance was used to obtain the genotypic correlations and phenotypic correlations among the eight characters, and the selection indexes for some agronomic characters were calculated by Robinson's method. The results are summarized as follows: Heritabilities : The experiment on the genotype-environment interaction revealed that in almost all of the characters investigated the interaction was too large to be neglected and materially affected the estimates of various genotypic parameters. The variation in heritability due to the change of environments was larger in the characters of low heritability than in those of high heritability. Heritability values of flowering date, fruiting period (days from flowering to maturity), stem length and 100 grain weight were the highest in both environments, those of yield(grain weight) and other characters were showed the lower values(Table 3). These heritability values showed a decreasing trend with the delayed sowing in the experiments. Further, all calculated heritability values were higher than anticipated. This was expected since these values, which were the broad sense heritability, contain the variance due to dominance and epistasisf in addition to the additive genetic variance. Genotypic correlations : Genotypic correlations were slightly higher than the corresponding phenotypic correlations in both environments, but the variation in values due to the change of environment appeared between grain weight and some other characters, especially an increase between grain weight and flowering date, and the total growing period(Table 6). Genotypic correlations between grain weight and other characters indicated that high seed yield was genetically correlated with late flowering, late maturity, and the other five characters namely branch numbers per plant, stem diameter, plant weight, pod numbers per plant and grain numbers per plant, but not with 100 grain weight of soybeans. Pod numbers and grain numbers per plant were more closely correlated with seed yields than with other characters. Selection index : For the comparison and the use of selection indexes in the selection, two kinds of selection indexes were calculated, the former was called selection index A and the later selection index B as shown in Table 7. Selection index A was calculated by the values of grain weight per plant as the character of yield(character Y), but the other, selection index B, was calculated by the values of pod numbers per plant, instead of grain weight per plant, as the character of yield'(character Y'). These results suggest that selection index technique is useful in soybean breeding. In reality, however, as the selection index varies with population and environment, it must be calculated in each population to which selection is applied and in each environment in which the population is located. In spite of the expected usefulness of selection index technique in soybean breeding, unsolved problems such as the expense, time and labor involved in calculating the selection index remain. For these reasons and from these experimental studies, it was recognized that in the breeding of self-fertilized soybean plants the selection for yield should be based on a more simple selection index such as selection index B of these experiments rather than on the complex selection index such as selection index A. Furthermore, it was realized that the selection index for the selection should be calculated on the basis of the data of some 3-4 agronomic characters-maturity date(X$_1$), branch numbers per plant(X$_2$), stem diameter(X$_3$) and pod numbers per plant etc. It must be noted that it should be successful in selection to select for maturity date(X$_1$) which has high heritability, and the selection index should be calculated easily on the basis of the data of branch numbers per plant(X$_2$), stem diameter(X$_3$) and pod numbers per plant, directly after the harvest before drying and threshing. These characters should be very useful agronomic characters in the selection of Korean soybeans, determinate growing habit type, as they could be measured or counted easily thus saving time and expense in the duration from harvest to drying and threshing, and are affected more in soybean yields than the other agronomic characters.

Correlation and Combining Ability related to the Growth Characteristics in F1 Hybrids by Diallel Cross of Viola tricolor (팬지의 일대 잡종에서 생육관련 형질의 상관관계 및 조합능력)

  • This study was conducted to determine correlations and combining abilities of leaf lengths, leaf widths, petiole lengths, number of branches, number of leaves, leaf areas, fresh weights, dry weights and number of flowers in F1 crosses made with the partial seven-parent diallel cross in Viola tricolor. Leaf lengths and leaf widths showed highly positive correlation with petiole length and negative correlation with number of branches, number of leaves and number of flowers. There was positive correlation between the number of leaves and flowers as well as between leaf area and fresh weight. Mean squares of general combining ability(GCA) and specific combining ability(SCA) were highly significant for all the parameters. Variance component values of SCA were greater than those of GCA for all the parameters except leaf length, implying preponderance of non-additive gene actions for these characters. The lines C and G for leaf lengths and widths, the lines A and F for number of leaves, the lines A, B and G for leaf areas, and the lines B and F for number of flowers showed relatively high GCA effects. The crosses of A×B and B×D exhibited high SCA effects on increasing leaf lengths, leaf widths, number of leaves, fresh weights and number of flowers. And also the crosses of B×E and D×G exhibited high SCA effects on decreasing leaf lengths, leaf areas and fresh weights as well as increasing number of flowers. The broad sense heritabilities of most characters were high compared with the narrow sense one. Those of leaf length, leaf width, petiole length and number of leaves were high in both the broad and narrow sense heritabilities.

  • Lipoxygenase activity of soybean seeds of approximately 507 genotypes as well as its inheritance and selection efficiency in early breeding generation was measured in the Department of Agronomy, Seoul National University to facilitate breeding for low lipoxygenase activity of soybean. Average seed lipoxygenase activity of 507 cultivars and strains was 350 unit (unit: $\Delta$0.01 /min. /mg at 234nm) and ranged from 50 to 670 unit. There was no difference in mean lipoxygenase activity according to apparent seed characters such as seed coat and embryo color. But early mature soybean genotypes had fairly low lipoxygenase activity. Lipoxygenase activity was inherited quantitatively, in which additive effect was greater than dominant one and proportion of gene with positive effects was similar to that with negative ones. Estimated narrow- and broad-sense heritabilities were 0.78 and 0.86 for lipoxygenase activity, respectively. Heritability measured from selection in early breeding lines for high or low lipoxygenase activity was 64~76% or 54~62%, respectively. And selection for high lipoxygenase activity increased by 29.7~44.7%, whereas that for low ones decreased by 21.8~27.3%, respectively, when compared to random population. Clear effect in selecting of lipoxygenase activity was present in early generation.

  • Seven rice varieties and $42F_1$ hybrids of $7{\times}7$ diallel cross were used in genetic analysis for grain size, weight and shape. Effects of average heterosis for grain length and shape showed significantly negative values. GCA, SCA and RCA variances were significant for all characters and also additive variance was more important than dominance variance for grain length, width, thickness and weight. GCA effects of BG 2 and Arborio were highly positive for grain size and weight, and those of BG 2 and Nagdongbyeo were highly negative for grain shape. Partial dominance was observed for all characters. The direction of dominance was negative for grain length, width and shape, and ambidirections for grain thickness and weight. The effective gene number was two for grain length and shape. Both narrow and broad sense heritability were high in all characters.
