Establishment of Food Processing Methods for Larvae of Allomyrina dichotoma, Korean Horn Beetle (식약용으로의 장수풍뎅이 유충 전처리 방법에 대한 연구)

  • With the objective of developing a functional food source, we established optimal processing conditions for the larvae of Allomyrina dichotoma, which have been used in traditional medicine to treat hepatic disorders in Korea. Without suitable processing, the larvae are difficult to consume as a food because of their disgusting taste and smell; moreover, in this form they might be a potential microbial hazard. In this study, we investigated the effect of feeding material, sterilization, and powdering after freeze-drying on the food quality of the larvae of A. dichotoma and on cytotoxicity against Raw 264.7 cells. Three to five days feeding with the sawdust from discarded oak-trees is sufficient for the breeding process. The sawdust was sterilized by vapor for five minutes. Sterilization of the larvae at a high temperature ($115^{\circ}C$ for 5 min, 0.9 $kgf/cm^2$) is necessary to eliminate pathogenic bacteria and fungi. The results of the cytotoxicity assay showed no toxicity in the prepared extract from larvae of A. dichotoma. In addition, to prepare the larvae for human consumption, various feeds were used and the smell, color, and taste were evaluated. Our results suggested that larvae of A. dichotoma could be developed as food source when a suitable processing method is established.

Package Atmosphere and Quality as Affected by Modified Atmosphere Conditions of Persimmon (Diospyros kaki. cv. Fuyu) Fruits ('부유' 단감의 MA포장 조건에 따른 포장기체조성 및 과실의 품질 변화)

  • Persimmon (Diospyros kaki. cv. Fuyu) fruits were packaged under different conditions, and then stored at $0^{\circ}C$ for 21 weeks. The tried packages were heat-sealed bag of one fruit, heat-sealed bag of five fruits, and tie-sealed bag of five fruits, which used films of three different thickness (0.03 ㎜, 0.05 ㎜ and 0.06 ㎜). Throughout the storage, package free volume, package atmosphere and quality were measured. Package free volume decreased with time with higher rate for heat-sealed bags, in which close contact between fruit and the film was eventually reached in longer storage. However, tie-sealed bags maintained the levels of stabilized free volume. The rate of free volume decrease was faster with thinner film and larger bag size for the packs sealed by the same method. Package atmosphere covered $O_2$ concentration of $1.1{\sim}17.1%$ and $CO_2$ concentration of $1.1{\sim}8.3%$, $O_2$ concentration decreased during storage, whereas $CO_2$ increased. Thinner film package created the internal atmosphere of higher $O_2$ and lower $CO_2$ concentrations. Tie-sealed bags of 5 fruits in the films of 0.05 mm and 0.06 mm thickness maintained the equilibrated package atmosphere of $1.1{\sim}3.0%\;O_2$ and $4.0{\sim}5.5%\;CO_2$, which preserved the fruits best in terms of firmness and less physiological changes of flesh browning and peel browning. At five fruit heat-sealed bag in the films of 0.06 mm thickness with $CO_2\;8.3%$, flesh browning occurred at a high rate, whereas all treatments in the film of 0.03 mm thickness with high $O_2$ and relatively low $CO_2$ contents, flesh and peel browning rates were high.

Characteristics and breeding of a new multi-generation oyster mushroom (Pleurotus ostreatus) variety 'Dagul' (다발성 신품종 느타리 '다굴'의 육성 및 자실체 특성)

  • To develop a new variety of oyster mushroom(Pleurotus ostreatus), parental strains was selected by the method of Mon-Mon crossing between monokaryotic strains derived from ASI 2596(Suhan No.3) and ASI 2782(Black pileus mutant). The SB-73(ASI 2596-11 x 2782-8) was shown the best cultural characteristics, selected to be a new variety and named as 'Dagul'. The 'Dagul' was formed incompatibility line distinctly in the confrontation growth of parental strains Suhan No.3 and ASI 2782. The optimum temperature for mycelial growth, fruiting body development and pH arrange were $25{\sim}30^{\circ}C$, $14{\sim}17^{\circ}C$ and pH5~8, respectively. Fruiting body production per bottle was about $68.0{\pm}24.1$ g which is almost 115% quantity compared to that of other variety Suhan No.3. And also the stipe is long and individual generation is multiple. Analysis of the genetic characteristics of the new variety 'Dagul' showed different DNA bands as that of the control strains, Suhan No.3 and ASI 2782, when RAPD(Random Amplified Polymorphic DNA) primers URP7 and Rcb1 were used. This new variety 'Dagul' of oyster mushroom is characterized by multiple of individual generation and the stipe is long. We therefore expect that this new strain will increase of the income by cultivation of field.

Characteristics of Glutinous Waxy Maize Hybrids (재래종(在來種) 찰옥수수 교잡종(交雜種)의 특성비교(特性比較))

  • Characteristics of ten glutinous maize hybrids developed at the Genetics and Breeding Lab. of Department of Agronomy, College of Agriculture, Chungnam National University were compared. Inbred lines used in the development of the ten hybrids were all originated from the Korean local maize lines which were collected in 1980s and they were developed by ear-to-row method. All the inbreds used had very unique plant characteristics such as plant height, ear height, ear length, ear diameter, 100 kernel weight and kernel color. The highest inbred in plant height was Jewon and the Jewon was also highest in ear height. While the ear of Jewon inbred was slender and slimmy, the ear of Dangjin inbred was rather short and promp. 100 kernel weight of the Bosung inbred was highest and the kernel weight of Chilbo was lightest. Kernel color of Jewon, Bosung and Danyang was white and the kernel color of Yunghee and Chilbo was purple. Dangjin had yellow kernel color. A total of 11 hybrids including one check hybrid developed by the Crop Experiment Station, ADR were included in performance trial. The hybrids which showed high grain yield were Dangjin/Bosung, Sosung/Dangjin and Dangjin/Chilbo. These hybrids were about 10% higher than the check in yield capacity. Each hybrid had also unique characteristics such as plant height, ear height and flowering date.

Comparison of Weekly and Batch Management System for Sows (모돈의 주간관리와 그룹관리 비교)

  • Conventionally, many pig producers have utilized a continual sow managements system that the matings, farrowings and weanings are done weekly basis. But this transitional method is not able to cut the cycle of diseases and fully apply all-in/all-out system because of the continuous flow of sows and pigs. Conventional weekly management system is currently limiting in small farm to work efficiently both for workers and pigs. Therefore, pig producers have found novel management methods for applying all-in/all-out system, improving pig health, leading to better growth, lowering mortality and reducing medication costs nowadays. Moreover, all-in/all-out pig management system has known as a strategy for improving productivity in swine farm. The batch system is one of the best management methods to adopt all-in/all-out pig management system that prevent spreading diseases in pig and remove cycle of diseases. Batch farrowing system is a concept for providing a group of sows that delivery within a specific farrowing interval and inducing a large enough scale of piglets to fill the weaner facilities. There are different types of batch farrowing system with batch size and interval of farrowing when several factors at the swine farm are considered such as total number of sows, available facilities in the farm, and the efficiency of workforce. Sow managements such as farrowing, weaning and breeding, every 3 weeks rather than weekly, 2 or 5-week interval have advantages for workers and reproductive cycle of sows as well as pig flow. Because there are several pros and cons both in weekly and batch management system, various factors should be considered to apply the most suitable management system in each individual farm. To improve poor swine productivity in Korea compared to ED, batch system for sows will be an alternative choice which is able to prevent high incidence of diseases in swine farm such as PMWS, PRRS, PRDC and PED because all-in/all-out pig management can be also applied automatically by using this management system.

Development of Efficient Screening Methods for Melon Plants Resistant to Fusarium oxysporum f. sp. melonis (멜론 덩굴쪼김병에 대한 효율적인 저항성 검정법 개발)

  • This study was conducted to establish an efficient screening system to identify melon resistant to Fusarium oxysporum f. sp. melonis. F. oyxsporum f. sp. melonis GR was isolated from infected melon plants collected at Goryeong and identified as F. oxysporum f. sp. melonis based on morphological characteristics, molecular analyses, and host-specificity tests on cucurbits including melon, oriental melon, cucumber, and watermelon. In addition, the GR isolate was determined as race 1 based on resistance responses of melon differentials to the fungus. To select optimized medium for mass production of inoculum of F. oxysporum f. sp. melonis GR, six media were tested. The fungus produced the most spores (microconidia) in V8-juice broth. Resistance degrees to the GR isolate of 22 commercial melon cultivars and 6 rootstocks for melon plants were investigated. All tested rootstocks showed no symptoms of Fusarium wilt. Among the tested melon cultivars, only three cultivars were susceptible and the other cultivars displayed moderate to high resistance to the GR isolate. For further study, six melon cultivars (Redqueen, Summercool, Superseji, Asiapapaya, Eolukpapaya, and Asiahwanggeum) showing different degrees of resistance to the fungus were selected. The development of Fusarium wilt on the cultivars was tested according to several conditions such as plant growth stage, root wounding, dipping period of roots in spore suspension, inoculum concentration, and incubation temperature to develop the disease. On the basis of the test results, we suggest that an efficient screening method for melon plants resistant to F. oxysporum f. sp. melonis is to remove soil from roots of seven-day-old melon seedlings, to dip the seedlings without cutting in s pore s uspension of $3{\times}10^5conidia/mL$ for 30 min, to transplant the inoculated seedlings to plastic pots with horticulture nursery media, and then to cultivate the plants in a growth room at 25 to $28^{\circ}C$ for about 3 weeks with 12-hour light per day.

Precise, Real-time Measurement of the Fresh Weight of Lettuce with Growth Stage in a Plant Factory using a Nutrient Film Technique (NFT 수경재배 방식의 식물공장에서 생육단계별 실시간 작물 생체중 정밀 측정 방법)

  • The measurement of total fresh weight of plants provides an essential indicator of crop growth for monitoring production. To measure fresh weight without damaging the vegetation, image-based methods have been developed, but they have limitations. In addition, the total plant fresh weight is difficult to measure directly in hydroponic cultivation systems because of the amount of nutrient solution. This study aimed to develop a real-time, precise method to measure the total fresh weight of Romaine lettuce (Lactuca sativa L. cv. Asia Heuk Romaine) with growth stage in a plant factory using a nutrient film technique. The total weight of the channel, amount of residual nutrient solution in the channel, and fresh shoot and root weights of the plants were measured every 7 days after transplanting. The initial weight of the channel during nutrient solution supply (Wi) and its weight change per second just after the nutrient solution supply stopped were also measured. When no more draining occurred, the final weight of the channel (Ws) and the amount of residual nutrient solution in the channel were measured. The time constant (${\tau}$) was calculated by considering the transient values of Wi and Ws. The relationship of Wi, Ws, ${\tau}$, and fresh weight was quantitatively analyzed. After the nutrient solution supply stopped, the change in the channel weight exponentially decreased. The nutrient solution in the channel slowly drained as the root weight in the channel increased. Large differences were observed between the actual fresh weight of the plant and the predicted value because the channel included residual nutrient solution. These differences were difficult to predict with growth stage but a model with the time constant showed the highest accuracy. The real-time fresh weight could be calculated from Wi, Ws, and ${\tau}$ with growth stage.

Resistance Degree of Radish Cultivars to Fusarium oxysporum f. sp. raphani according to Several Conditions (발병조건에 따른 무 품종들의 시들음병에 대한 저항성 차이)

  • This study was conducted to establish the efficient screening system for resistant radish to Fusarium oxysporum f. sp. raphani. Five radish cultivars ('Myoungsan', 'Chungdu', 'Jangsaeng', 'Hannongyeorm', and 'Chungsukungjung') showing different degree of resistance to the fungus were selected. And the development of Fusarium wilt of the cultivars according to several conditions such as root wounding, dipping period of roots in spore suspension, inoculum concentration, and incubation temperature to develop the disease was tested. In distinguishing the resistance degree of the radish cultivars to the disease, non-cut roots were more effective than cut roots. And occurrence of Fusarium wilt of the radish plants increased in the proportion to increase of root-dipping period and spore concentration of the fungus. Thus, optimum conditions to differentiate susceptible and resistant cultivars to the disease were root-dipping period of 0.5 hour and spore concentration of $1{\times}10^7\;conidia{\cdot}mL^{-1}$. Disease severity of Fusarium wilt on the cultivars was changed with incubation temperature and the radish seedlings incubated at $25^{\circ}C$ represented the most difference of resistance and susceptibility to Fusarium wilt. From the above results, we suggest that the efficient screening method for resistant radish to Fusarium oxysporum f. sp. raphani would be to dip non-cut roots of fourteen-day-old radish seedlings in spore suspension of $1{\times}10^7\;conidia{\cdot}mL^{-1}$ for 0.5 hour and to transplant the inoculated plants to plastic pots with fertilized soil, and then to incubate the radish plants at a temperature of $25^{\circ}C$ for development of Fusarium wilt.

Approaches on Optimum Conditions for Agrobacterium-Mediated Transformation of Phalaenopsis (호접란의 Agrobacterium 이용 형질전환 시스템의 최적조건 구명을 위한 연구)

  • Sensitivities of PLBs of four Phalaenopsis cultivars, P. 'Taisuco Windian', P. 'Nancy Amour', P. 'Pink Twilight' and P. 'Taipei Gold' to kanamycin, spectinomycin and hygromycin at different concentrations (0, 25, 50, 100, 200, and $400mg{\cdot}L^{-1}$) were examined. Hygromycin was favorable for selecting the transformants in the genetic transformation of Phalaenopsis as PLBs of four cultivars were all dead at even $25mg{\cdot}L^{-1}$ hygromycin. Responses of PLBs of P. 'Maki Watanabe' and P. 'Brother Lawrence' to DL-phosphinothricin (PPT) were determined at different concentrations (0, 0.1. 0.25, 0.5, 1.0, 2.0, 2.5, and $5.0mg{\cdot}L^{-1}$) and $0.5mg{\cdot}L^{-1}$ PPT was thought to be suitable for selecting the transformants of Phalaenopsis. The optimum conditions for Agrobacterium cocultivation with Phalaenopsis PLBs were examined using a two-step cocultivation method in Dtps. 'City Girl' and A. tumefaciens LBA4404. In the first infection period in a 1 : 10 suspension of Agrobacterium to a VW medium, 1 hr infection showed the highest PLB survival ratio. And then, PLBs were cocultivated with a bacterial strain and a 3-day cocultivation period was better for Phalaenopsis PLBs than a prolonged period. Agrobacterium tumefaciens strains LBA4404 (pTOK233) and EHA105 (pGA643) were used to compare their efficiency on the genetic transformation of Phalaenopsis PLBs. The PLBs infected with EHA105 survived more than those infected with LBA4404 after two days in a dark condition and two weeks in light condition on a selective medium. About 1,000 PLBs for each of P. 'Maki Watanabe' and P. 'Brother Lawrence', and each bacterial strain of AGL1 (pCAMBIA3301) and LBA4404 (pTOK233) were used for the regeneration of transgenic plants. The bacterial strain AGL1 had a higher genetic transformation efficiency than LBA4404, with no significant difference between cultivars. In this study, 11 hygromycin-resistant plantlets and 32 PPT-resistant plantlets were produced, but these putative transgenic plantlets need further examinations.

Analysis of Antioxidant Activity and Serotonin Derivatives in Safflower (Carthamus tinctorius L.) Germplasm Collected from Five Countries (국외 수집 홍화 유전자원의 항산화 활성 및 세로토닌 유도체 함량 분석)

  • In order to select potential plant resources as functional materials and natural antioxidants, we evaluated antioxidant activity and serotonin derivatives of safflower germplasm collected from five countries. N-(p-Coumaroyl) serotonin (CS) and N-feruloylserotonin (FS) were analyzed by using Ultra Performance Liquid Chromatography (UPLC). Total polyphenol content (TPC) was determined by Folin-Ciocalteu method and antioxidant activities were estimated by 2,2-diphenyl-1-picryl-hydrazil (DPPH), 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) diammonium salt (ABTS), Ferric reducing antioxidant power (FRAP) and Reducing power (RP) assays. The TPC showed a range of 28.25 to $90.53{\mu}g$ gallic acid equivalent (GAE)/mg dried extract (DE). DPPH, ABTS, FRAP and RP assay were in the range of 18.76 to 93.98, 48.91 to 163.73, 3.80 to 132.29 and 26.32 to $80.08{\mu}g$ ascorbic acid equivalent (ASCE)/mg DE, respectively. Among the five countries, safflower seed collected from Iran had the highest levels of serotonin derivatives and antioxidant activities than other countries (p<0.05). CS showed high correlation with TPC, ABTS and DPPH (r=0.673,0.727,0.820), and FS showed high correlation with DPPH (r=0.740). Accessions IT321214 and IT321215 could be useful for development of new functional materials and could be used as a source of valuable natural antioxidant materials.
