Origin-related study of genetic diversity and heteroplasmy of Mongolian sheep (Ovis aries) using mitochondrial DNA

  • Food and agricultural production sector, especially livestock production is vital for Mongolia's economic and social development. Domestic sheep play key roles for Mongolians, providing food (meat, milk) and raw materials (wool, sheepskin), but genetic diversity, origin of sheep populations in Mongolia have not been well studied. Studies of population genetic diversity is important research field in conservation and restoration of animal breeds and genetic resources. Therefore, this study aimed to investigate genetic characteristics and estimate origin through the analysis of mitochondrial DNA control region D-loop and Cytochrome b of Mongolian indigenous sheep (Mongolian native, Orkhon and Altanbulag) and one Europe sheep (Suffolk). As a result of there were found, 220 SNPs (Single nucleotide polymorphism) in the D-loop region, 28 SNPs in the Cytochrome B region, furthermore, 77 Haplotypes. The nucleotide diversity was only found in D-loop region (n = 0.0184). Phylogenetic analysis showed that 3 (A, B, and C) of 5 haplogroups of sheep have been identified in our research. Haplogroup C was only found in Mongolian indigenous sheep. Haplogroup D and E were not observed. As a result of haplogroups, haplogroup A was dominant (n = 46 of 94 sheeps), followed by haplogroup B (n = 36) and haplogroup C (n = 12). Sequence analysis showed that T deletion, insertion and heteroplasmy in D-loop region occurred at a high rate in Mongolian indigenous sheep population (T insertion = 47, T deletion = 83). The heteroplasmy, which has never been found in Mongolian sheep, has been newly discovered in this study. As a result, the Mongolian sheep varieties, which mainly derived from Asia, were in hybridization with European sheep varieties.

  • This study has been divided into two parts, 1) literature review and 2) a case study. Based on literature review and case studies found in Korea and abroad, this study aimed at classifying wildlife passages and establishing their development procedures in order to establish a theoretical ground for the development of wildlife passage as a way to link habitats that have been fragmented by the construction of roads and other developments. The results of the literature review are as follows : 1) Impact of road construction on surrounding ecological environment is found in various aspects. Along with damage of habitats as direct impact, decrease of biodiversity and simplification of species due to limited breeding and movement are cited as possible indirect impact. 2) In order to lessen the impact generated from fragmented habitats, various types of wildlife passage are being developed in Europe and are showing positive results. 3) In Korea, interests on animal passages have been rising recently. The ministry of Environment and local authorities are establishing development plans for the passages. However, practical materials on the usefulness of adoped methods are not available.4) In order to create an effective wildlife passage, decisions have to be made first on basic factors of planning and designing such as ecosystem rehabilitation methods and surveys on wildlife movement routes and habitat environment. Based on this, in a planning and designing stage, the location and type of a passage, size and measures to attract animals and prevent a collision need to be finalized. This should be followed by a series of process of maintenance, management and monitoring after the development. Based on the results of a theoretical study to develop an wildlife passage, Part 2 of this study deals with the results of an experiment through a specific planning and design that includes a process of ecological surveys, site selection, type selection, ecosystem infrastructure, planting design and construction details on an actual study area and presents them as a model project.

Benefit Analysis and Profit Business of Fishery Sector about Tidal Power Plant in Garolim Bay (가로림만 조력발전 사업관련 수산부문 수익사업 도출과 편익 분석)

  • While promoting a variety of projects by using sea space, there has been a lot of social controversy among the stakeholders based on the results of the analysis. In particular, the benefits of fisheries sector were over-estimated and occurred a lot of opposition from fishermen and environment groups. In order to this solve, we have been studied focussed such as excavation of new projects, the method and result of the estimated benefits for fisheries sector. With regard to the tidal power plant of Garolim, we came up with five kinds of problems such as doing not reflect of the cost of the initial investment, over unit production, over unit price, excessive selection of breed, issues of cost of production in benefits study of fisheries sector derived previously. In order to these problems, we have derived new profitable projects based on the initial materials such as improvement of raising breed, discharge project of fingerlings and a small sea ranch project. As a result, previous study was analyzed in 30.2 billion won(1993), 54.8billion won(2009), but this study was estimated to 26.4 billion won and differences of benefits amount were occurred. In a previous study was calculated benefits by simple method, but this study came up with new projects for fisheries sector and because we estimated benefits considering new project. The method for benefits calculations for fisheries sector was not attempted in previous studies and in this study, we approached for the first time Also, in this study suggested new method to increase incomes fishermen such as development of suitable ranch breeding for the marine environment, ranch license(limited license), development of fisheries resources and the operation of fisheries facility. In the future, we hope to be helpful for the same field of study.

IL-1B (C+3954T) Gene Polymorphism and Susceptibility to Gastric Cancer in the Iranian Population

  • Background: Gastric cancer as one of the most important diseases affecting health in all worldwide. Current studies have confirmed associations of cytokine gene polymorphisms with the risk of gastric cancer development. The current research aimed to assess the association of IL-1B+3954 genotypes with the risk of gastric cancer in the Iranian population. Materials and Methods: This case-control study covered 49 gastric cancer patients compared to 53 cancer free individuals as a control group. Genomic-DNA extraction was carried out from bioptic samples of patients and peripheral blood of healthy volunteers. Polymorphism of IL-1B +3954 genotypes were analysed with a polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. Results: The frequencies of IL-1B +3954 A1A1, A1A2 and A2A2 genotypes in healthy individuals were 26.4, 66 and 7.6 %, respectively. However, in gastric cancer patients, A1A1, A1A2 and A2A2 with 4.1, 51 and 44.9% were observed (p<0.05). Conclusions: The findings of our results show a positive association between the IL-1B+3954 genotype distribution and the risk of gastric cancer disease in the Iranian population.

  • 1. In order to investigate Physiologic races of Puccinia gramininis f. sp. tritici as basic materials for tile breeding of resistant varieties to stem rust of wheat, this experiment was carried out during 1971 and 1973. 2. Twenty seven isolates of the fungus were identified as follows: two isolates of race 11, eight . isolates of race 17, and 21, four isolates of race 34, one each of race 37 and 56, respectively The remaining three isolates identified were not identical with the key issued by Stakman, et at in 1962. Therefore, the three new races were tentatively numbered $K_3,\;K_4\; and\; K_4,$ in addition to $K_l\;and\;K_2$ described in 1961. 3. Therefore, attempts to produce resistant varieties to stem rust of wheat should be accompanied by intensive investigation of Physiologic races of p. graminis f. sp. tritici in the future.

Morphological and Molecular Characterization of Thamnocalamus falconeri Hook f. ex. Munro

  • The economy of India and so also of many Asian countries depends on bamboos and their uses are not only in domestic items but also in rural housing and raw materials to several industries and germplasm characterization is an important link between the conservation and utilization of plant genetic resources. Classical taxonomic studies of the bamboos are based on floral morphology and growth habit, which can cause problems in identification due to erratic flowering coupled with different biotic agencies and environmental factors. Identification and genetic relationships among accessions of Thamnocalamus falconeri were investigated using morphology and random amplified polymorphic DNAs (RAPD) technique. Analysis started by using 51 vegetative characters and forty two 10-mer primers that allowed us to distinguish different genotypes hailing from different eco- zones of Garhwal Himalayas (India). The selected primers (12) were used for identification and for establishing a profiling system to estimate genetic diversity. A total of 79.33% polymorphism was estimated by using 12 selected primers. The genetic similar analysis was conducted based on binary digits i.e. presence (1) or absence (0) of bands, which revealed a wide range of variability among the species whereas genetic relatedness was quite high based on vegetative characters. Cluster analysis clearly showed two major clusters for both of the markers viz. morphology and RAPD belonging to 10 accessions of T. falconeri. Two major clusters were further divided into minor clusters. Cluster based on RAPD marker showed grouping of accessions of closed locality whereas analogy was reported for vegetative traits. The RAPD technique has the potential for use in species identification and genetic relationships studies of bamboo for breeding program.

Varietal Variation in Germinative Energy, Capacity and Water Sensitivity for Malting Barley (맥주보리에서 발아세, 발아율 및 수감수성의 품종간 차이)

  • Forty-nine malting barley varieties were tested at 3 and 8 weeks after harvest, stored in natural glasshouse so as to investigate varietal variation of germinative energy(GE), capacity(GC) and water sensitivity(WS). The differences among GE, GC, promptness index(PI), and WS on paper at two moisture levels (4$m\ell$ and 8$m\ell$ tests) were great at 3 weeks after harvest, but small at 8 weeks after harvest. Excess water depressed the germination of dormant grains. The GE of whole varieties were excellent, but WS of only two varieties, CI 12062 and CI 15631 were bad at 8 weeks after harvest. So in the case of broad germplasm to be used for crossing materials, it is suggested that WS will be tested to screen good quality malting lines. WS was highly significantly correlated with GE, GC, and PI of 8 ml test, but was not with three factors of 4 $m\ell$ test. GE and WS were various with storage period, varieties and grain plumpness, indicated that breeding good grain-filling varieties and improving production practice for good grain-filling would be studied in the future.

Selection Index and Genetic Advance on Quantitative Characters of Sesame (참깨의 양적형질에 대한 선발지수 및 유전진전)

  • The studies were intended to clarify the effects of selection in sesame breeding. The 82 cultivars of sesame were used as the materials, and 14 quantitative characters were measured on individual plant basis. Selection indexes and genetic advances were calculated by Robinson's methods. In genetic advance values, combinations of days to flowering + number of capsule per plant, days to flowering + length of stem with capsule + number of capsule per plant and days to flowering + number of nodes with capsule were higher than those of other combinations. The highest genetic advance was the combination of all characters, but unreasonable problems such as the expence, time and labor involved in calculating the selection index are remained. For these reasons, it was realized that the selection index for selection should calculated on the basis of the data of 2-3 useful characters, i.e., days to flowering, length of stem with capsule and number of capsule per plant.

  • Resistance to various diseases of Korean native varieties were investigated in field. The results obtained are summarized as follows: 1) The varieties Ubangtchio and Osib-eubthio were moderately resistant to black shank, but Suantchio, Usultchio and Gwangtchio were slightly resistant. Dixie Bright 101, $H_2$ and Bright Yellow 4 were more resistant to black shank than any of the Korean native varieties. 2) Mokgitchio, Ubangtchio and Osib-eubtchio were moderately resistant to black root rot, but Useultchio Suantchio and Hoetchio were susceptible than Others. 3) Muktchio, Hoetchio and Ubangtchio were slightly resistant to wild fire. 4) Osib-ebthio was highly resistant to brown spot, but Ubangtchio, Hyangtchio and Mokgitchio were moderately resistant and Useultchio was susceptible to brown spot, but it was resistant than Bright ellow 4 or Bernhart 1000-1. 5) Hyangtchio was slightly resistant to CMV-Y. According to these results, except Virus disease, Ubangtchio was resistant to all of the above mentioned diseases. Osib-eubtchio, Hoetchio, Hyangtchio and Suantchio showed proferable disease resistant and these might be well utilized as breeding materials.

Growth Characteristics·Fresh and Dry Weight of the Corn Inbred Lines with Multi-Tiller and Ear using Forage Crop (사료용 다얼·다수성 옥수수의 생육특성·생체 및 건물 수량)

  • This study was carried out to find a new corn cultivar with multiple tillers that is suitable for forage. Materials were developed at the Corn Breeding Laboratory of Chungnam National University. A total of 3,650 accessions were collected domestically and from other countries, including China. Among them, 16 CNU inbreds, tillering and non-tillering, were compared to each other in terms of stem heigh, number of tillers, fresh weight, and dry weight per plant. Of these surveyed traits, the stem heigh at tasseling stage was 140~190 cm and number of tillers per plant ranged from two to four. Fresh weight and dry weight of these lines were higher in the tillered than in the non-tillered sample.
