Development and Field-evaluation of Automatic Spreader for Bedding Materials in Duck Houses (오리사 바닥 깔짚자동살포장치 개발 및 실증)

  • The automatic-spreader of bedding materials was developed to reduce labor cost, and to achieve successful biosecurity in duck houses. Algorithm of the device was designed to realize a concept of the automatic unmanned operation including entire processes such as loading and spreading of the bedding materials. From the field measurement, the relationship between the expected water content reduction and weight of bedding materials per unit area according to the operation condition of the devices was induced. In the case of the measurement of particulate matters during the process of spreading works, the results of using both conventional manual-spreader and automatic-spreader were exceeded the allowable limit of inhalable and respirable dust, respectively. But, workers using automatic-spreader could be free from harmful aero-condition because they did not stay inside the facility during the spreading works. In addition, from the Duck hepatitis virus PCR test, influence on pulmonary tissue of the duck was not found. It could be expected that the development of the automatic-spreader of bedding materials for duck house can contribute to the advancement of duck breeding facilities.

Development of a Genetic Map of Chili Pepper Using Single Nucleotide Polymorphism Markers Generated from Next Generation Resequencing of Parents (양친의 대량 염기서열 해독을 통해 개발된 SNP 분자표지를 이용한 고추 유전자지도 작성)

  • Molecular markers, as an efficient selection tool, have been and is being used for practical breeding program in chili pepper (Capsicum annuum L.). Recently, a lot of researches on inheritance and genetic analysis for quantitative traits including capsaicinoids, carotenoids, and sugar content in pepper are being performed worldwide. It has been also reported that QTL mapping is a necessary tool to develop molecular markers associated with the quantitative traits. In this study, we suggested a new method to construct a pepper genetic map using SNP (HRM) markers generated from NGS resequencing of female and male parents. Plant materials were C. annuum 'NB1' (female parent), C. chinense 'Jolokia' (male parent), and their $F_2$ population consisting of 94 progenies. Sequences of 4.6 Gbp and 6.2 Gbp were obtained from NGS resequencing of 'NB1' and 'Jolokia', respectively. Totally, 4.29 million SNPs between 'NB1' and 'Jolokia' were detected and the 1.76 million SNPs were clearly identified. Among them, total 145 SNP (HRM) primer pairs covering pepper genetic map were selected, and the 116 SNP (HRM) markers of them were located on this map. Total distance of the map, which consisted of 12 linkage groups and matched with basic chromosome numbers of pepper, was 1,167.9 cM. According to the mapping result, we concluded that our mapping method was suitable to construct a pepper genetic map fast and accurately. In addition, the genetic map could be directly used for QTL analysis of traits different between both parents.

Evaluation and classification of selected rice varieties for salinity tolerance at seedling stage

  • To identify the new source of breeding materials for rice salt tolerance, the salinity tolerance of thirty-four varieties was evaluated under 0.5% saline condition at seedling stage. The salinity score showed highly significant correlations to dry weight and dead leaf ratio. The tested varieties were classified into three groups by visual score, reduction ratio of dry weight, and dead leaf ratio. Eighteen varieties were classified as the highly tolerant group (salinity scores of 1.3-3.7), seven varieties were fallen into the tolerant group (salinity scores of 4.2-5.8), and others were susceptible (salinity scores of 6.7-9.0). In highly tolerant group, most indica varieties including Getu, Dikwee and Kuatic Putic, didn't exsert a panicle under the Korean climate. But six varieties, Xiangcho V, Annapuruna, HP 3319-2wx-6-3-1, Giza 175, and GZ 2447-S-17, GZ 4255-6-3 were suitable to the Korean climate, and their heading date (6-16, August) and culm length (65-78㎝) were similar to the Korean varieties. Accordingly, these varieties can be utilized as crossing materials for the salt tolerance in japonica rice.

  • Eggshell quality is one of the most important factors that influence hatchability. The porosity and overall quality of eggshell have a very significant effect on exchange of vital gas (carbon dioxide and oxygen) between the developing embryo and the air during incubation. Thin-shelled eggs showed the greater weight loss than do thick-shelled eggs. causing the chick to have difficulty in hatching. Thin-shelled eggs also have a much greater chance of being cracked during handling. On the other hand. thick-shelled eggs showed the higher hatchability as a result of greater fertility and lower intermediate and late embryonic mortalities. Even a small percentage improvement in the eggshell quality could result in significant saving to the breeder industry in an increasingly competitive environment. Many factors including nutrition. management practices, environmental conditions and breeding are known to be related to eggshell quality. In this review brief accounts of each factor associated with eggshell quality and hatchability were provided. We conducted three experiments to investigate the effects of dietary Ca. vitamin D3 metabolite and some bioactive materials on eggshell quality and hatching egg production. The results of our studies showed that relatively high levels of dietary Ca in combination with 25-hydroxycholecalciferol had beneficial effects on improving eggshell quality and reproductive performances in aged egg-type breeder hens. It was also suggested that the proper use of some feed additives such as isoflavon and chitosan might provide means of improving eggshell quality and reproductive performances in aged egg-type breeder hens.

Growth Characteristics and Seed Yield of Medicinal Soybeans Collected in Korea

  • Seeds of medicinal soybean [Glycine max(L.) Merill] are characterized by a black seed coat, white stripe at hilum border, yellow cotyledon, and very small seed weight. Production of this medicinal soybean has recently increased as a consumption increased. The objective of this study was to evaluate the growth characteristics and seed yield of collected medicinal soybeans and to obtain basic information on production practices and breeding materials. The collected medicinal soybean lines were cultivated at three locations for two years. Twenty-seven lines were planted at the Research Farm, Korea University, Namyangju city, on May 23, and at the Research Farm, Kyungpook National University, Taegu, on May 20, 1995. In 1996 field experiments, forty-four lines were planted on May 25, at Research Farm, Korea University, and twenty-seven lines among those were planted on June 7 at the Research Farm of National Yeongnam Agricultural Experiment Station, Milyang city. The investigated lines had purple or white flower. Flowering and maturing dates were similar or later than those of the control cultivars. Branch number was greater for the investigated lines. One hundred-seed weight of the lines ranged from 8.5 to 15.0 g. Mean seed yields ranged from 1.54 to 2.89 MT/ha. Nine lines of the investigated medicinal soybeans showed higher yield capacity than the control cultivars. Further research should be done on the improvement of the production system and breeding program of medicinal soybeans.

Identification and Quantitative Determination of Glucosinolates in Brassica napus cv. Hanakkori

  • The objective of this study was to identify and quantify glucosinolates (GSLs) in Brassica napus cv. Hanakkori and its parents and to evaluate its potential bitter taste. 'Hanakkori' materials were cultivated with commercial chemical nutrients (20 kg/ha, N-P-K: 16-10-10) at the field. GSLs were isolated by means of extraction with 70%(v/v) boiling methanol (MeOH) followed by desulfation from those plants by reversed-phase high performance liquid chromatography (HPLC) and identified by electronic spray ionization-mass spectrometry (ESI-MS) analysis. In 'Hanakkori', 11 GSLs were identified as progoitrin, glucoraphanin, glucoalyssin, gluconapoleiferin, gluconapin, 1-methylpropyl, glucobrassicanapin, glucobrassicin, 4-methoxyglucobrassicin, gluconasturtiin, and neoglucobrassicin. The total GSL contents were 109 and 36.1 mmol/kg dry weights (d.w.) for the seeds and edible parts, respectively. The major GSLs (>5 mmol/kg d.w.) in the seeds were progoitrin (78.8), gluconapin (10.7), and glucobrassicanapin (7.81), whereas they in the edible parts were progoitrin (16.1) and glucobrassicanapin (8.58). In addition, the bitter taste in the edible parts was presumably related with the presence of progoitrin (>45% to the total GSL).

Inhibition of Metarhizium anisopliae infection of Protaetia brevitarsis seluensis larvae using several effective microorganisms

  • The purpose of this study was to determine the best method for minimizing the occurrence of Metarhizium anisopliae infection of Protaetia brevitarsis seluensis during mass breeding on agricultural farms. There is a high demand for the use of P. b. seluensis larvae in animal feed and as food for humans. However, mass breeding results in the entomopathogenic fungal (usually M. anisopliae) infection of P. b. seluensis. A mixture of microorganisms (Bacillus subtilis, Lactobacillus plantarum, and Saccharomyces cerevisiae) delayed fungal infection by M. anisopliae, which infected fewer P. b. seluensis when the microorganism mixture was added to sawdust as feed for P. b. seluensis. When sawdust with the effective microorganisms (EM) was given to P. b. seluensis for 30 d, their mortality rate was approximately 35 % less than that of the control group, which was fed sawdust without the EM. In addition, the growth of M. anisopliae on agar media spread with each bacterium as inhibited by up to 80 % more than those spread with 4 % sodium hypochlorite, which is a harmless fungal inhibitor generally used in agricultural farms for disinfection.

  • Agriculture is the most primitive civilized Activities of mankind but also the propellant of civilization development. Because it is the most basic material goods source of mankind. Among these materials rice is one of the most important part of these, we call them the substance of survival. From the beginning of the agricultural activities to the present we have experienced three industrial revolutions and are experiencing the Fourth Industrial Revolution. With the development of science and technology makes the efficiency of agricultural production is higher and higher, but compared with the original we are facing the same problem: natural disasters; pests and diseases; now also face the depletion of resources, environmental degradation and other issues. Therefore, improve and cultivate new crop varieties to make it better resistance and more production for better develop modern agriculture. It's very helpful for human social development. And also it is the responsibility and task of modern molecular breeding. In this study, I used bacterial leaf blight to find a better resistance gene to improve the resistance of rice. Frist Cultivate k3 of bacterial leaf blight, than inoculation by leaf clipping method (Kauffman,1973) in CNDH and SNDH population at 40days after rice transplanting. Check the lesion length by inoculation plants at 14days after inoculation, and record data for QTL analysis program. Than I get 4 intervals in 3 different chromosomal regions. I found these defense genes in the 4 intervals. So I used NCBI Justbio, Rapdb, etc. to finding these genes in physical map, than design primer for map base cloning. At last these defense genes will be employed in further research for introduction of the gene to the parental plant and rice breeding for solving food crisis.

  • To develop neohaplonts for Ganoderma breeding, protoplasts were isolated from dikaryotic mycelium and regenerated. Selection rate of neohaplonts varied between ASI7074, ASI7091, ASI7094, ASI7100 and ASI7115, showing 5.24% on the average. Auxotropic mutants from Ganoderma monokarions were recovered by UV irradiation on protoplasts. Protoplast survival rates were 1.9% ASI 7074, 0.17% ASI 7091, and zero percent ASI 7100 using 300 second irradiation. Four auxotrophic strains were recovered from 1,536 colonies screened that will be further utilized for protoplast fusion and transformation.

Genetic Analysis on Some Quantitative Characters in Tobacco (Nicotiana tabacum L.) Breeding IV. Changes of Genetic Parameter according to different Cultivated Systems and Generations. (연초(Nicotiana tabacum L.)육종을 위한 제형질의 통계유전학적 연구 IV. 재배법 및 세대에 따른 유전적 Parameter의 변동)

  • The study was conducted to obtain the genetic informations for some useful characters in tobacco breeding. The eight parents and a set of 28 crosses of F1's and F2's were used as materials, and planted on two different cultivated systems, i. e., oriental's and burley systems, during 1986-1988 at taegu Experiment Station, Korea Ginseng & Tobacco Research Institute. The observed characters were six agronomic characters which were plant height, number of leaves per plant, leaf length, leaf width, days to flowering, yield and five chemical components, nicotine, total nitrogen, protein nitrogen, total volatile base and pet, ether extract. The results obtaining are summarized as follows: 1. The higher heritabilities were found for days to flowering yield and nicotine in both generations, but values for number of leaves per plant, leaf length, total nitrogen and protein nitrogen were lower than other characters. 2. Genotypic correlation coefficients among all pairs of characters were slightly higher than the corresponding phenotypic correlations in F1's and F2's on two different cultivated systems. The relationship between leaf length and leaf width was the positive correlation, but that between number of leaves per plant and leaf width was negative. 3. From the genotypic correlations between yield and other characters, a conclusion that the yield was highly correlated with plant height, leaf length, leaf width and days to flowering was given. 4. Quality was positively correlated with number of leaves per plant and nicotine, but negatively with the other agronomic characters and chemical components.
