  • High molecular weight dextran (39% alcohol, v/v), less soluble dextran, eluted from this column between T 500 and T 2000, a commercial linear dextran, Soluble dextran (45% alcohol, v/v) eluted at between T 70 and T 150 dextran. The molecular weight average of total dextran (50% alcohol, v/v) was between 150,000 to 500,000. A few oligosaccharides were detected from hydrolyzates of less soluble dextran. The hydrolyzates of soluble dextran were a family of DP 1 to 6 isomaltooligosaccharides. Compounds greater than DP 4 were branched isomaltooligosaacharides.

A New Sponge, Antho (Acarnia) seogwipoensis (Poecilosclerida: Microcionidae) from Korea

  • A new marine sponge, Antho (Acarnia) seogwipoensis n. sp., of the family Microcionidae, was collected from Seogwipo-si, Jeju-do, Korea, about 100 m in depth using a gill net on 1969. The genus Antho Gray, 1867 including Demospongiae, Poecilosclerida, Microcionidae, is a large group of sponges. About 100 species in Antho were reported from worldwide. The genus Antho contains five subgenera: Antho, Acarnia, Isopenectya, Jia, and Plocamia. Among them, about 30 species in Acarnia were described in world sponge. A new sponge's body shape is branching, size up to 124 mm wide, 213 mm high, 3-8 mm thick in branch and 7-9 mm thick in stalk. Antho (Acarnia) seogwipoensis n. sp. is similar to A. (A.) novizelanicum Ridley and Duncan, 1881 based on their spicules type and skeletal structure, but differs in the spicules dimension and growth form. This new species is branched growth form and have three kinds of toxa.

Penicillifer diparietisporus: a New Record from Field Soil in Korea

  • A fungus was isolated from field soil collected from Daegu, Korea. The colony of the isolated fungus showed short, branched, and light to dark yellow pigments with hyaline, yellowish red to orange brown aerial mycelia. In addition, the fungus produced solitary to aggregated perithecia, ovoid to pyriform, short neck, and asci as well as biseriately arranged ascospores. Phylogenetic analysis using the internal transcribed spacer region and translation elongation factor $1-{\alpha}$ sequences and morphological characteristics identified the isolated fungus as Penicillifer diparietisporus, which belongs to the family Nectriaceae. To our knowledge, this is the first report of Penicillifer diparietisporus in Korea.

  • We describe a hidden Markov model, HMMTIR, for general protein sequence based on the I-sites library of sequence-structure motifs. Unlike the linear HMMs used to model individual protein families, HMMSTR has a highly branched topology and captures recurrent local features of protein sequences and structures that transcend protein family boundaries. The model extends the I-sites library by describing the adjacencies of different sequence-structure motifs as observed in the database, and achieves a great reduction in parameters by representing overlapping motifs in a much more compact form. The HMM attributes a considerably higher probability to coding sequence than does an equivalent dipeptide model, predicts secondary structure with an accuracy of 74.6% and backbone torsion angles better than any previously reported method, and predicts the structural context of beta strands and turns with an accuracy that should be useful for tertiary structure prediction. HMMSTR has been incorporated into a public, fully-automated protein structure prediction server.

First Record of the Giant Grouper Epinephelus lanceolatus (Perciformes: Serranidae: Epinephelinae) from Jeju Island, South Korea

  • A single specimen of Epinephelus lanceolatus, belonging to Epinephelinae of the family Serranidae, was newly collected from Jeju Island, South Korea, in December 2009. Epinephelus lanceolatus is characterized by anterior lateral-line scales with branched tubules, cycloid scales, irregular white spots, and fins with numerous small black spots. We propose a Korean name, "Dae-wang-ba-ri' for this species.

  • One specimen of Chirolophis saitone, belonging to the family Stichaeidae, was collected by a bottom trawl in the southern area of the Yellow Sea. The present specimen was characterized by 52 dorsal fin spines, 38 anal fin spines, a caudal fin with four vertical dark bands and an orbital region with three pairs of branched cirri. We propose a new Korean name, "Jak-eun-goe-do-ra-chi', for C. saitone.

  • Five unrecorded species (Oedipodium griffithianum, Myurella tenerrima, Trachypus bicolor, Coscinodon humilis, Meteorium miquelianum subsp. atrovariegatum) with one unrecorded family and two unrecorded genera, were reported as new to the Korean moss flora. The monotypic family Oedipodiaceae consisting of a single genus and single species was distinguished from Splachnaceae by small plants that were 10 mm in length simple or sometimes branched, obovate-spathulate to ${\pm}$ orbicular, costa ending below the apex and entire margins except for the lower long-ciliate portion The species was first found at the top of the Mt. Seorak at an altitude of 1708 m. The two genera, Myurella and Coscinodon were found in the Korean Peninsula. Myurella tenerrima (Theliaceae) was found around the top of Jung-bong on Mt. Jiri. It was mixed with other mosses in the crevices of rocks in the alpine regions. The species is similar to M. sibirica, yet it can be distinguished by the position of papilla in the median laminal cells. Coscinodon humilis (Grimmiaceae) were found on the ridge of Mt. Gaya. C. humilis has a variety of hyaline apex according to leaf position and forms a capsule so it can be distinguished by family. Trachypus bicolor and Meteorium miquelianum subsp. atrovariegatum are unrecorded species. T. bicolor (Trachypodaceae) were found on the ridge of Mt. Gaya. T. bicolor is similar to T. humilis but distinguished by the costal lengths of the leaves. M. miquelianum subsp. atrovariegatum (Meteoriaceae) was found in Gageo-do. This species was distinguished by the plant form and morphology of stem leaves in the same genus.

Mannan-binding lectin of the sea cucumbers Stichopus japonicus has common antigenic determinants with human serum mannan-binding lectin

  • The host defense system or immune system of all modern animals has their roots in very ancient organisms. After analyzing literature data concerning properties of invertebrates and vertebrates lectins we suggest that mechanism of mannans recognition may exist in marine invertebrates, as a universal mechanism for homeostasis maintenance and host defense, and mannan-binding lectins family of vertebrates has ancient precursor, as was shown for another S-type lectins family. We carried out the screening of mannan-binding type lectin among different species of echinoderms inhabiting in Piter the Grate Bay, the sea of Japan. As a result, the C-type lectins (SJL-32) specific for high mannose glycans was isolated from the coelomic plasma of the sea cucumbers Stichopus japonicus by ion-exchange chromatography on a DEAE-Toyopearl 650M, affinity chromatography on a mannan-Sepharose 6B and gel filtration on a Sephacryl S-200. SJL-32 is homodimer with molecular mass about 32 kDa on SDS-PAGE under non-reducing conditions. Protein part of the lectin has high conteins Asn, Glu, Ser. Hemagglutination of trypsin-treated O blood group human erythrocytes by SJL-32 was competitively inhibited by high-branched -D-mannan composed of -1,2 and -1,6 linked D-mannopyranose residues. In contrast, a variety of mono-, oligo-, and polysaccharides composed of residues of galactose and fucose showed absence or little inhibitory activities. The lectin activity strong depends on Ca2+ concentration, temperature and pH. Monospecific polyclonal antibodies were obtained to the lectin. As was shown by ELISA assay, antibodies to SJL-32 cross-reacted with human serum mannan-binding lectin. This data allows making conclusion about common antigenic determinants and structural homology of both lectins. In our opinion, SJL-32 belongs to evolutionary high conservative mannan-binding lectins (MBLs) family and takes part in the host defense against pathogenic microorganisms.

Antioxidant potential of aerial part of Asclepias curassavica. Linn (Family-Asclepiadaceae)

  • Asclepias curassavica. Linn, an erect, simple (or) much branched perennial herb with a somewhat woody base, belonging to the family Asclepiadaceae. It has been reported to have multiple pharmacological effect of anti-inflammatory, cardiotonic, anticancer, anthelmintic and to treat piles and gonorrhoea. It is to be expected that several activities might be related to a possible antioxidant action from this plant. The hydro alcoholic extract of Asclepias curassavica was tested in vitro for its antioxidant activities, such as DPPH radical, nitric oxide radical, superoxide anion radical, lipid peroxidation assay, hydroxyl radical, reducing power, and total phenol content. The extract exhibited scavenging potential with $IC_{50}$ value of $8.7\;{\mu}g/ml,\;198.4\;{\mu}g/ml\;and\;21.7\;{\mu}g/ml$ for DPPH, nitric oxide and superoxide anion radicals. The values were found to higher than those of Vitamin-C, rutin, and curcumin, as standards. The extract showed 50% protection at the dose of $134.2\;{\mu}g/ml\;and\;41.4\;{\mu}g/ml$ in lipid peroxidation as well as deoxyribose degradation, those values more to that of standard, vitamin E $(IC50\;values,\;119.2\;{\mu}g/ml\;and\;32.5\;{\mu}g/ml,\;respectively)$. The reducing power of the extract depends on the concentration and amount of extract. Since a significant amount of polyphenol could be detected by the equivalent to $0.0495\;{\mu}g$ of pyrocatechol from 1 mg of extract. It can be concluded that hydro alcoholic extract of aerial part of Asclepias curassavica could be considered as potent antioxidant, which makes it suitable for the prevention of human disease.

한국산 개구리류의 방광에 기생하는 Gorgoderid 흡충류의 분류 (Gorgoderid trematodes (Digenea: Gorgoderidae) from the urinary bladder of frogs in Korea)

  • Gorgoderidae에 속하는 흡충류는 어류, 양서류 및 파충류 등의 방광에 기생하며 우리 나라의 양서류에서는 Gonodera japonica 1종만이 기록되어 있다가 유와 이(1983)에 의해 Corgoderina bombinae 1종이 신종으로 보고된 바 있다. 본 연구에서는 1989년부터 1994년에 걸쳐 전국 각지에서 채집한 양서류를 대상으로 그들의 방광에 기생하는 윤충류를 조사한 결과 Gorgoder japonica 및 Conoderinc bombinae 2종을 동정하였으며. 분류된 2종에 대한 형태학적인 특징을 기술하고 이들의 분류학적 위치에 대해 상세히 논하였다 G. japonica가 G. cygnoides와 명확히 구별되는 점은 난황선이 깊은 열개에 의해 뚜렷한 여러 개의 엽을 형성하는 점이었다. G. bombinoe는 기존의 종들과 비교한 결과 뚜렷한 종으로서 확인되었으며, 이 종에 대한 국명을 "무당개구리방광흡충"으로 제안한다.
