고지방 식이 섭취로 유발된 비만 쥐의 시상하부 BDNF발현과 렙틴 mRNA 발현, 혈청 렙틴과 항비만과의 관계에 대한 운동트레이닝의 효과 (Effects of Exercise Training on the Relationship with Brain-derived Neurotrophic Factor Expression and Leptin mRNA Expression in Hypothalamus, Serum Leptin, and Anti-obesity in High-fat Diet-induced Obese Rats)

  • 본 연구의 목적은 고지방식으로 유도된 비만이 에너지 항상성 조절인자인 신경영양인자와 섭식조절 호르몬인 렙틴 mRNA 발현에 어떠한 관련성이 있는지를 규명하는 것과 비만 쥐의 시상하부 BDNF 발현과 렙틴 리셉터 mRNA 발현, 혈청 렙틴과 항비만과의 관계에 대한 운동트레이닝과 정상식이로의 전환에 대한 효과를 확인하고자 하였다. 그 결과 8주 후 비만 대조군(HF)에 비해 운동군(HFT), 정상식이군(HFND), 운동 및 정상식이 트레이닝군(HFNDT)에서 각각 유의한 체중 및 혈중 TC 감소를 보였다(P<0.05). 혈중 TG에서는 HF군에 비해 HFNDT군에서 유의한 감소를 보였다(P<0.05). 혈중 렙틴은 HF군에 비해 HFT군에서 유의한 감소를(P<0.05), HFT군에 비해 HFND군에서 유의한 감소를 보였으며(P<0.05), HFND군에 비해 HFNDT군에서 유의한 감소를 나타냈다(P<0.05). 시상하부 BDNF 단백질 발현은 HF군에 비해 모든 군에서 유의한 증가를 보였다(P<0.05). 시상하부 렙틴 리셉터 mRNA는 HF군에 비해 HFT와 HFND군에서 약간의 감소를, HFNDT군에서는 증가를 나타내었다. 이상의 결과는 고지방식이로 유도된 비만쥐에 대한 운동트레이닝은 항비만 효과가 있는 것으로 나타났으며, 운동 또는 정상식이의 병행은 식욕과 체중조절에 최적의 비만예방 및 개선효과가 있을 것으로 사료된다.

DEPRESSION: CELLULAR AND PHYSIOLOGICAL CONSEQUENCES OF STRESS (ANTIDEPRESSANT EFFECT OF SEROTONIN N-ACETYLTRANSFERASE INHIBITOR)

  • Melatonin is secreted during the hours of darkness and is thought to influence the circadian and seasonal timing of a variety of physiological processes. Serotonin N-acetyltransferase (AA-NAT) which is found to be expressed in pineal gland, retina, and various tissues, catalyses the conversion of serotonin to N-acetylserotonin and is known as the rate-limiting enzyme in the biosynthetic pathway of melatonin. The compounds that modulate the activity of AA-NAT can be used to treat serotonin-and melatonin-related diseases such as insomnia, depression and seasonal affective disorders (SAD). Several assay methods have been developed by which to measure AA-NAT activity. We have also developed a simple, rapid and sensitive AA-NAT assay method that takes advantage of differences in the organic solubilities between acetyl CoA and N-acetyltryptamine. We screened modulators of AA-NAT activity from the water extracts of the medicinal plants. We found MNP1005 which strongly inhibited the activity of AA-NAT ($IC_{50}$=2.2$\mu$M). Enzyme inhibitory kinetic studies revealed that MNP1005 exhibited a noncompetitive inhibition toward tryptamine. The antidepressant effect of MNP1005 was investigated on behavioral despair test so called forced swimming test (FST). MNP1005 significantly increased swimming behavior by reducing immobility with treatment of 10 mg/kg when compared to the vehicle-treated control group (P < 0.05). This suggests that MNP1005 possesses antidepressant activity. The influence of chronic MNP1005 treatment on the expression of brain-derived neurotrophic factor (BDNF) was examined by in situ hybridization and Northern blot. Chronic treatment of MNP1005 blocked the downregulation of BDNF mRNA in the frontal cortex and other cortex regions in response to restraint stress.

Protective effect of Capsosiphon fulvescens on oxidative stress-stimulated neurodegenerative dysfunction of PC12 cells and zebrafish larva models

  • Reactive oxygen species (ROS) at high concentrations induce oxidative stress, an imbalanced redox state that is a prevalent cause of neurodegenerative disorders. This study aimed to investigate the protective effect of Capsosiphon fulvescens (CF) extract on oxidative stress-induced impairment of cognitive function in models of neurodegenerative diseases. CF was extracted with subcritical water and several solvents and H2O2 (0.25 mM) or aluminum chloride (AlCl3; 25 µM) as an inducer of ROS was treated in PC12 neuronal cells and zebrafish larvae. All statistical analyses were performed using one-way analysis of variance and Dunnett's test using GraphPad Prism. H2O2 and AlCl3 were found to significantly induce ROS production in PC12 neuronal cells and zebrafish larvae. In addition, they strongly affected intracellular Ca2+ levels, antioxidant enzyme activity, brain-derived neurotrophic factor (BDNF) and tropomyosin receptor kinase B (TrkB) signaling, acetylcholinesterase (AChE) activity, and hallmarks of Alzheimer's disease. However, treatment of H2O2-induced PC12 cells or AlCl3-induced zebrafish larvae with CF subcritical water extract at 90℃ and CF water extract effectively regulated excessive ROS production, intracellular Ca2+ levels, and mRNA expression of superoxide dismutase, glutathione peroxide, glycogen synthase kinase-3 beta, β-amyloid, tau, AChE, BDNF, and TrkB. Our study suggested that CF extracts can be a potential source of nutraceuticals that can improve the impairment of cognitive function and synaptic plasticity by regulating ROS generation in neurodegenerative diseases.

Chronic Administration of Baicalein Decreases Depression-Like Behavior Induced by Repeated Restraint Stress in Rats

  • Baicalein (BA), a plant-derived active flavonoid present in the root of Scutellaria baicalensis, has been widely used for the treatment of stress-related neuropsychiatric disorders including depression. Previous studies have demonstrated that repeated restraint stress disrupts the activity of the hypothalamic-pituitary-adrenal (HPA) axis, resulting in depression. The behavioral and neurochemical basis of the BA effect on depression remain unclear. The present study used the forced swimming test (FST) and changes in brain neurotransmitter levels to confirm the impact of BA on repeated restraint stress-induced behavioral and neurochemical changes in rats. Male rats received 10, 20, or 40 mg/kg BA (i.p.) 30 min prior to daily exposure to repeated restraint stress (2 h/day) for 14 days. Activation of the HPA axis in response to repeated restraint stress was confirmed by measuring serum corticosterone levels and the expression of corticotrophin-releasing factor in the hypothalamus. Daily BA administration significantly decreased the duration of immobility in the FST, increased sucrose consumption, and restored the stress-related decreases in dopamine concentrations in the hippocampus to near normal levels. BA significantly inhibited the stress-induced decrease in neuronal tyrosine hydroxylase immunoreactivity in the ventral tegmental area and the expression of brain-derived neurotrophic factor (BDNF) mRNA in the hippocampus. Taken together, these findings indicate that administration of BA prior to the repeated restraint stress significantly improves helpless behaviors and depressive symptoms, possibly by preventing the decrease in dopamine and BDNF expression. Thus, BA may be a useful agent for the treatment or alleviation of the complex symptoms associated with depression.

스코폴라민으로 유도된 기억력 손상에 대한 복신의 보호 효과 및 작용기전 연구 (Effect of Poria Cocos on the Scopolamine-induced Memory Impairment and Its Underlying Molecular Mechanism)

  • This study was performed to investigate the memory enhancing effect of Poria cocos Wolf (Hoelen cum radix) against scopolamine-induced amnesia in Sprague-Dawley (SD) rats. To induce amnesia, scopolamine (0.75 mg/kg) was intraperitonically injected into SD rats 30 min before starting behavior tests. We have conducted Morris water-maze and Y-maze tests to monitor learning and memory functions. Poria cocos effectively reversed scopolamine-induced memory impairment in SD rats which was represented by an improvement of mean escape latency in water-maze test and spontaneous alterations in Y-maze test. To elucidate possible molecule mechanism, we have measured mRNA as well as protein expression of acetylcholine esterase (AchE), choline acetyltransferase (ChAT), muscarinic acetylcholine receptor (mAchR), and brain-derived neurotrophic factor (BDNF) using RT-PCR and Western blot analysis, respectively. Poria cocos increased mRNA levels of ChAT and mAchR in rat hippocampus compared with those in the scopolamine-injected amnesic group. In addition, protein expression of ChAT and BDNF was also elevated by Poria cocos intake. Furthermore, as an upstrem regulator, the activation of cAMP response element-binding protein (CREB) was assessed by immunohistochemistry. In this immunohistochemical analysis, the phosphorylation of CREB (p-CREB) was reduced by scopolamine injection, which was restored back to control levels by administration of Poria cocos. These results suggest that Poria cocos may improve memory and cognitive deficit in amnesia and have therapeutic potentials through up-regulation of ChAT, mAchR, and BDNF, which seemed to be mediated by activation of CREB.

Hippocampus-dependent cognitive enhancement induced by systemic gintonin administration

  • Background: A number of neurological and neurodegenerative diseases share impaired cognition as a common symptom. Therefore, the development of clinically applicable therapies to enhance cognition has yielded significant interest. Previously, we have shown that activation of lysophosphatidic acid receptors (LPARs) via gintonin application potentiates synaptic transmission by the blockade of $K^+$ channels in the mature hippocampus. However, whether gintonin may exert any beneficial impact directly on cognition at the neural circuitry level and the behavioral level has not been investigated. Methods: In the current study, we took advantage of gintonin, a novel LPAR agonist, to investigate the effect of gintonin-mediated LPAR activation on cognitive performances. Hippocampus-dependent fear memory test, synaptic plasticity in the hippocampal brain slices, and quantitative analysis on synaptic plasticity-related proteins were used. Results: Daily oral administration of gintonin for 1 wk significantly improved fear memory retention in the contextual fear-conditioning test in mice.We also found that oral administration of gintonin for 1 wk increased the expression of learning and memory-related proteins such as phosphorylated cyclic adenosine monophosphate-response element binding (CREB) protein and brain-derived neurotrophic factor (BDNF). In addition, prolonged gintonin administration enhanced long-term potentiation in the hippocampus. Conclusion: Our observations suggest that the systemic gintonin administration could successfully improve contextual memory formation at the molecular and synaptic levels as well as the behavioral level. Therefore, oral administration of gintonin may serve as an effective noninvasive, nonsurgical method of enhancing cognitive functions.

반복 스트레스에 의한 흰쥐 해마조직내 신경전구세포의 생성과 brain-derived neurotrophic factor (BDNF) mRNA 발현 변동에 미치는 고려홍삼 사포닌의 반복 투여 효과 (Effects of Korea Red Ginseng Total Saponin on Repeated Unpredictable Stress-induced Changes of Proliferation of Neural Progenitor Cells and BDNF mRNA Expression in Adult Rat Hippocampus)

  • 본 연구 결과를 통하여 홍삼 성분인 고려홍삼 사포닌을 반복 투여시 흰쥐 해마 SGZ 부위의 신경전구세포 생성이 유의하게 증가되었으며, 이와 같은 경향은 반복 스트레스에 노출되어도 유지되었다. 또한 스트레스를 가하지 않은 흰쥐에서 고려홍삼 사포닌 반복 투여시 해마 CA3와 CA1 부위에서 BDNF mRNA의 발현이 증가되었으나, 반복 스트레스를 가한 흰쥐의 CA3와 CA1부위에서 BDNF mRNA의 감소를 차단하지는 못하였다. 따라서 고려홍삼 사포닌 반복 처치에 의한 해마 신경전구세포의 생성에 BDNF 보다는 다른 요인이 관여할 가능성이 클 것으로 추정된다.

강도별 유산소운동이 중년여성의 적혈구생성인자, BDNF와 인지기능, 작업기억에 미치는 영향 (Influence of Erythropoiesis Factors, BDNF, Cognitive Function and Working Memory by Intensity Aerobic Exercise in Middle Aged Women)

  • 본 연구는 강도별 규칙적인 유산소운동이 중년여성의 적혈구생성인자, BDNF와 인지기능, 작업기억에 어떠한 영향을 미치는지 규명하기 위해 실시하였다. 경기도 Y시 소재 G구에 거주하는 40~60대 중년여성을 대상으로 통제집단(n=9, non-exercise), 중강도 유산소운동집단(n=10, 50%VO2max, 200 kcal), 고강도 유산소운동집단(n=9, 70%VO2max, 200 kcal)으로 분류하여 12주간 주 3회의 빈도로 집단의 특성에 맞게 운동처치를 실시하여 운동전과 6주, 12주 후 채혈과 인지기능 및 작업기억검사를 통해 적혈구생성인자, BDNF, 인지기능 및 작업기억을 측정하였다. 모든 자료는 SPSS Ver. 15.0를 통해 집단, 시기, 상호교호작용의 비교를 위해 반복측정분산분석, 일변량분석 및 사후검증(LSD)을 실시하였다. 그 결과 적혈구생성인자, BDNF, 인지기능, 작업기억에서 시기에 따른 유의한 차이가 나타나 일변량분석을 통해 집단 내 시기에 따른 분석을 실시한 결과 중강도 유산소운동은 적혈구 수, 헤마토크릿, BDNF와 작업기억에서 유의하게 증가하였고(p<.05), (p<.01), (p<.05), (p<.05), 고강도 유산소운동은 적혈구 수와 작업기억에서 유의하게 증가하였다(p<.01)(p<.001). 또한 집단 간 비교 결과 헤마토크릿은 중강도 유산소운동집단과 고강도 유산소운동집단이 통제집단에 비해 유의하게 높았고(p<.01), 헤모글로빈은 중강도 유산소운동집단이 통제집단에 비해 유의하게 높은 것으로 나타났다(p<.05). 따라서 본 연구의 결과를 비추어볼 때 중년여성의 적혈구생성인자, BDNF, 인지기능 및 작업기억에 12주의 중강도 이상의 유산소운동이 긍정적인 영향을 미치는 것을 알 수 있었다.

조현병 환자의 신경과학적 근거에 대한 고찰 (Review of the Neuroscientific Evidences for the People With Schizophrenia)

  • 본 연구는 조현병의 신경과학적 연구에 대한 흐름을 알아봄으로써 임상가들에게 조현병 환자 치료의 토대를 제공하고, 보다 효과적인 치료를 선정하는데 도움이 되고자 하였다. 현재까지 조현병 환자의 신경과학적 연구는 매우 활발히 이루어지고 있는데, 본 고찰에서는 조현병의 발생에 대한 연구와 조현병으로 인한 신경해부학적 기능 이상에 초점을 두고 살펴보았다. 조현병의 발생과 관련된 연구는 신경생화학적 이상, 신경발달적 모델, 뇌단백물질, 뇌유래신경영양인자에 대한 근거들을 다루었으며, 조현병의 신경해부학적 기능 이상에 대해서는 주로 신경영상학적 특징들에 근거하였다. 결론적으로, 조현병은 광범위한 기능적 손실을 가져오는 정신과적 질환으로 이제까지 원인과 치료에 대한 많은 연구들이 실행되고 있지만 아직 명백한 결과를 가져오지는 못하고 있다. 그러나 이제까지 밝혀진 많은 근거들은 임상가들이 조현병 환자를 치료하고 재활함에 있어서 충분한 도움이 될 것으로 생각된다. 앞으로도 조현병의 원인 규명과 치료에 대한 다양한 신경과학적 연구가 진행될 것으로 보인다.

국소 허혈성 뇌손상 흰쥐 모델에서 환경강화 조건 하 수정된 건측억제유도 운동치료가 앞다리 운동기능 증진에 미치는 영향 (Effects of Improved Forelimb Sensorimotor Function on the Modified CIMT Applied Under the influence of Environmental Enrichment in a Focal Ischemic Brain Injury Rat Model)

  • Environmental Enrichment (EE) alone is not capable of enhancing the fine digit and the forelimb functions. Therefore, we applied modified constraint-induced movement therapy (mCIMT) under the influence of EE to assess its effect on promoting improved forelimb sensorimotor functions. Focal ischemic brain injury was produced in Sprague-Dawley rats (60 rats, $250{\pm}50$ g) through middle cerebral artery occlusion (MCAO). Before MCAO induction, all rats were trained in modified limb placing tests and reaching tasks for 1 week. Then they were randomly divided into three groups: Group I: application of standard environment (SE) after MCAO induction (n=20), Group II: application of EE after MCAO induction (n=20), Group III: MCAO+EE, mCIMT and task-oriented training that was initiated at 10th day after MCAO induction (n=20). We also applied mCIMT (between 9 AM and 5 PM/daily) which included restraining the forelimb ipsilateral to the lesion using the 'Jones & Schallert' method. We assessed the change of modified limb placing, single pellet reaching test and the immunoreactivity of BDNF by immunohistochemistry (pre, 1st, 5th, 10th and 20th day). Group I showed no improved outcome, whereas group II and III significantly improved on the use of the forelimb and the immunoreactivity. The qualitative analysis of the skilled reaching test, of group III showed the greatest improvement in the fine digit and the forelimb function. These results suggest that EE combined with mCIMT is more functional in promoting enhanced fine digit and forelimb functional movements.
