The water extract of Seongpungtang(SPT) has commonly been used for treatment of ischemic brain damage in Oriental traditional medicine. However, little is known about the mechanism by which the water extract of SPT rescues brain cells from ischemic damage. To elucidate the protective mechanism of ischemic induced cytotoxicity, the regulation of Lipopolysaccharide (LPS) and PMA (phobol-12-myristate-13-acetate) induced iNOS expression in microglial cells was investigated. LPS and PMA treatment for 48 hr in microglial cells markedly induced nitric oxide (NO), but treatment of the cells with the water extract of SPT decreased nitrite formation. In addition, LPS and PMA treatment for 48 hr induced severe cell death in microglial cells. However treatment of the cells with the water extract of SPT did not induce significant changes compared to the control cells. Furthermore, NO production was markedly decreased by treatment of nuclear factor kappa B(NF-kB) inhibitor, pyrrolidine dithiocarbamate(PDTC). According to the above results, it is suggested that the protective effects of the water extract of SPT against ischemic brain damage may be mediated by regulation of iNOS during ischemic condition.
The present study was carried out to determine the possible use of cTn-I in the cardiac myofibrillar architecture, as a potential target for in vivo radioimmunodetection of cardiac damage in a brain death pig model. Radioiodiantion of the anti-cTn-I 5F4 McAb was carried out by lactoperoxidase method. the percentage iodine incorporation achieved was 70-75%. The radioiodinated McAbs were purified on Sephadex G-25 column and characterised by Paper chromatography, Phast Gel electrophoresis and electroimmunoblotting. Radioiodinated anticTn-I 5F4 McAbs were employed alongside Pyrophosphate($Tc_{99m}$-PPi$) and $Thallium^{201}$ chloride($TI^{201}$) in 24 landrace pigs (brain-dead=18 & sham-operated=6). The percentage cardiac uptake of the radiolabelled antibody injected dose was significantly higher in the brain dead animals(0.196%) as compared to that of sham-operated animals (0.11%). Specific in vivo localization of radiolabelled McAbs in the infarcted cardiac tissue was confirmed by computer-aided reconstruction of 3-D images of the isolated heart. The preliminary results of the study revealed preferential uptake of radiolabelled antibody at the site of myocyte damage resulting from artificially induced brain death.
Objectives : This research was performed to investigate effect of Samul-tang-gamibang against focal cerebral ischemic damage after middle cerebral artery occlusion(MCAO). Methods : This research was used rats which were against focal cerebral ischemic damage by MCAO. It was used Zea Longa's theory and Belayev's methods to give rise to focal cerebral ischemic damage by MCAO. After 7days later, we drew out the brain and then had frozen and dyeing it and we had taken a picture to measure of the damaged area in each brain section. We determined the Neurological Index and tested the Foot-fault test and Roatated test to appraise the fall of motion ability result from cerebral ischemic damage. Results : The results of the experiment are as follows. 1. Samul-tang-gamibang reduced infarct size of sample group compared to control group at 7 day after MCAO. 2. Samul-tang-gamibang reduced infarct volume of sample group compared to control group at 7 day after MCAO. 3. Samul-tang-gamibang reduced foot-fault index of sample group compared to control group at 5,7 day after MCAO. Conclusions : Samul-tang-gamibang has protective effects against ischemic brain damage and had significant reduced infarct size and infarct volume of Rt-MCAO.
Background: Herbal medicines are popular approaches to capably prevent and treat obesity and its related diseases. Excessive exposure to dietary lipids causes oxidative stress and inflammation, which possibly induces cellular senescence and contribute the damaging effects in brain. The potential roles of selective enhanced ginsenoside in regulating high fat diet (HFD)-induced brain damage remain unknown. Methods: The protection function of Ginsenoside F1-enhanced mixture (SGB121) was evaluated by in vivo and in vitro experiments. Human primary astrocytes and SH-SY5Y cells were treated with palmitic acid conjugated Bovine Serum Albumin, and the effects of SGB121 were determined by MTT and lipid uptake assays. For in vivo tests, C57BL/6J mice were fed with high fat diet for 3 months with or without SGB121 administration. Thereafter, immunohistochemistry, western blot, PCR and ELISA assays were conducted with brain tissues. Results and conclusion: SGB121 selectively suppressed HFD-induced oxidative stress and cellular senescence in brain, and reduced subsequent inflammation responses manifested by abrogated secretion of IL-6, IL-1β and TNFα via NF-κB signaling pathway. Interestingly, SGB121 protects against HFD-induced damage by improving mitophagy and endoplasmic reticulum-stress associated autophagy flux and inhibiting apoptosis. In addition, SGB121 regulates lipid uptake and accumulation by FATP4 and PPARα. SGB121 significantly abates excessively phosphorylated tau protein in the cortex and GFAP activation in corpus callosum. Together, our results suggest that SGB121 is able to favor the resistance of brain to HFD-induced damage, therefore provide explicit evidence of the potential to be a functional food.

Purpose: This study evaluated the extent of damage due to hypothermia in the mature and immature brain. Methods: Hippocampal tissue cultures at 7 and 14 days in vitro (DIV) were used to represent the immature and mature brain, respectively. The cultures were exposed at $25^{\circ}C$ for 0, 10, 30, and 60 minutes (n=30 in each subgroup). Propidium iodide fluorescent images were captured 24 and 48 hours after hypothermic injury. Damaged areas of the cornu ammonis 1 (CA1), CA3, and dentate gyrus (DG) were measured using image analysis. Results: At 7 DIV, the tissues exposed to cold injury for 60 minutes showed increased damage in CA1 (P<0.001) and CA3 (P=0.005) compared to the control group at 48 hours. Increased damage to DG was observed at 24 (P=0.008) and 48 hours (P=0.011). The 14 DIV tissues did not demonstrate any significant differences compared with the control group, except for the tissues exposed for 30 minutes in which DG showed less damage at 48 hours than the control group (P=0.048). In tissues at 7 DIV, CA1 (P=0.040) and DG (P=0.013) showed differences in the duration of cold exposure. Conclusion: The immature brain is more vulnerable to hypothermic injury than the mature brain.
Objectives: This study was designed to assess neuroprotective effects of herb medicine against Alzhheimer's disease related brain damage in organotypic hippocampal slice culture. Methods: We induced dementia related brain damage in organotypic hippocampal slices by $\beta$-amyloid. Those slices were treated with herb medicines - Hwangryeonhaedoktang, Sopungsoongiwon. Using by PI staining, the extents of cell death were assessed. After that, we selected the best effective one among those herb medicines and the major components of that medicine were studied to reveal neuroprotective effects and related proteins by using PI stating. Results: In PI staining, Sopungsoongiwon is the best effective herb medicine between Hwangryeonhaedoktang and Sopungsoongiwon. Notopterygii Rhizoma, Corni Fructus, Areca Catechu, Aurantii Fructus Immaturus, Plantaginis Semen is the best effective one among the components of Sopungsoongiwon. Conclusions: We suggested that purgative effect would be the best effetive medicine on dementia related brain damage between clearing heat and toxic materials.
The purpose of this study was to analyze the presence of ipsilesional movement deficit, with segmental performance in each proximal or distal upper extremity. The visuoperceptual complex task of the ipsilesional upper extremity was investigated in patients with unilateral brain damage and a control group of healthy sex-age-matched controls. Tracking movements were tested in the proximal and distal upper extremities. Movements were measured by the accuracy index, which was normalized to each subject's own range of motion and took into account any differences between subjects in the excursion of the tracking target. The findings revealed that stroke patients experienced difficulties with tracking movement of both proximal and distal segments in the upper extremities on the so-called "non-affected side", irrespectively of the extent of patient's age, time since onset, or severity of contralateral upper extremity. Therefore, the unilateral brain damage affected ipsilateral motor function of the proximal and distal upper limbs in the performance of complex motor tasks, requiring central processing and the higher order cognitive function in the integrity of both hemispheres.
This study focuses on the concept of multi-scale wireless sensor networks for damage detection in civil infrastructure systems by first over viewing the general network philosophy and attributes in the areas of data acquisition, data reduction, assessment and decision making. The data acquisition aspect includes a scalable wireless sensor network acquiring acceleration and strain data, triggered using a Restricted Input Network Activation scheme (RINAS) that extends network lifetime and reduces the size of the requisite undamaged reference pool. Major emphasis is given in this study to data reduction and assessment aspects that enable a decentralized approach operating within the hardware and power constraints of wireless sensor networks to avoid issues associated with packet loss, synchronization and latency. After over viewing various models for data reduction, the concept of a data-driven Bivariate Regressive Adaptive INdex (BRAIN) for damage detection is presented. Subsequent examples using experimental and simulated data verify two major hypotheses related to the BRAIN concept: (i) data-driven damage metrics are more robust and reliable than their counterparts and (ii) the use of heterogeneous sensing enhances overall detection capability of such data-driven damage metrics.
There is a correct way to avoid any sequale in the central motor area during neurosurgery procedures. A clear way to find the circumference of the central sulcus, central motor, and sensory areas by giving cortical electrical stimulation to the central motor area immediate after surgery is proposed. Looking at patients who underwent brain surgery September 2009 to July 2013, the central sulcus and speech areas around the central area of the brain was investigated, using the practices of either a localized brain map check or a direct cortical electrical stimulation test. Brain maps localized around the surgical site through functional movement or speech areas were identified. Accurate tests done during surgery without damage to motor neurons or after surgery were conducted smoothly. Although successful brain map test localization can be accomplished, there are some factors that can interfere. The following phenomena can reverse the phase: (1) the first sensory / motor in the case of patients severe nerve damage; (2) placement of the electrode on top of the vessel; (3) presence of a brain tumor near the brain cortex; (4) use of anesthesia if patient cooperation is difficult; and (5) location of the electrode position and stimulus is inappropriate.
