Characterization of gender-specific bovine serum

  • Animal cell cultures generally require a nutrient-rich medium supplemented with animal serum. Adult bovine serum contains a variety of nutrients including inorganic minerals, vitamins, salts, proteins and lipids as well as growth factors that promote animal cell growth. To evaluate the potential use of gender-specific bovine serum (GSBS) for cell culture, the biochemical properties of male serum (MS), female serum (FS) and castrated-male serum (CMS) were investigated. Overall, the chemical profile of GSBS was similar to that of bovine references except for glucose, creatine kinase, lactate dehydrogenase and potassium. FS showed elevated total protein and sodium concentrations compared to MS and CMS. Proteins present in MS, FS and CMS but absent in fetal bovine serum (FBS) were selected by two-dimensional gel electrophoresis and identified by peptide mass fingerprinting. Some of the identified proteins are known to be involved in immune responses and the others have unknown physiological roles. Moreover, it was found that some proteins such as alpha-2-macroglobulin appeared to be gender-specific with higher contents in FS. Insulin and testosterone was significantly higher in MS, and $17{\beta}$-estradiol and estrone were higher in FS, as compared to the other sera. Taken together, the results indicate that each GSBS has a different ratio of components. Differences in serum constituents may affect cell cultures in a different manner and could be beneficial, depending on the specific aim of cell cultures.

  • 소의 번식효율을 증가시키기 위한 기초실험으로서 6%, 10% 및 20%의 bovine serum albumin 을 사용한 선인들의 방법과 tyrode액을 사용하여 시도한 저자의 방법으로 소의 원정액, 희석정액 및 일반시판 냉동정액으로부터 고활력정자를 분리수집하여 정자의 각종 성상과 광학현미경적 형태를 비교관찰한 결과는 다음과 같다. 1. 원정액으로부터 bovine serum albumin을 사용하여 분리한 정자는 대조군에 비하여 운동성, 운동성 정자수, 정상정자율 및 전진운동성이 현저히 높았고 정자회수율은 6%일 때 가장 높았다. 2. 원정액으로부터 bovine serum albumin을 사용하여 고활력정자를 분리한 후 냉동한 정액의 정자운동성, 정상정자율 및 전진운동성은 대조군에 비하여 현저히 높았고, 이러한 현상은 bovine serum albumin의 농도가 20%일 때에 가장 현저하게 나타났다. 3. Bovine serum albumin을 사용하여 분리한 고활력정자의 냉동전 및 냉동후의 광학현미경적인 기형율은 대조군에 비하여 현저히 낮았다. 4. Bovine serum albumin을 사용하여 분리한 고활력정자는 전자현미경으로 세포막의 확장 및 공포 형성, acrosome의 확장과 density loss의 변형율이 대조군에 비하여 낮았다. 5. 일반시판 냉동정액으로부터의 고활력정자의 분리는 bovine serum albumin을 사용할 때는 어려웠으나, tyrode액을 사용한 이 실험에서는 가능하였다. 6. 원정액, 희석정액 및 냉동정액의 고활력정자회수율은 tyrode 액을 이용하여 80분간 정치하였을때 현저히 높았다. 7. Tyrode 액을 이용하여 원정액, 희석정액 및 냉동정액으로부터 분리된 고활력정자의 운동성, 전진운동성 및 정상정자율은 대조군에 비하여 현저히 높았다. 8. Tyrode 액을 이용하여 원정액, 희석정액 및 냉동정액으로부터 분리한 고활력정자의 광학현미경적 기형율은 대조군에 비하여 현저히 낮았다.

  • The binding properties of three ginsenosides, Rb$_{1}$, Rc and Re, to bovine and human serum albumins have been examined by fluorescence probe technique. 1-anilinonphathalene-8-sulfonate (ANS) was used as the fluorescence probe. Protopanaxatriol glycoside, Re, did not quench the fluorscence of ANS to the bovine serum albumin. Competitive bindings between protopanaxadiol glycosides, Rb$_{1}$ and Rc are both 3.3 . The binding constants for Rb$_{1}$ and Rc with bovine serum albumin were 1.91 * 10$_{4}$M$_{-1}$ AND 1.04 * 10$^{[-994]}$ M$^{-1}$ , respectively. The ginsenosides, Rb$_{1}$, Rc and Re did not quench the fluorescence of ANS bound to human serum albumin.

A comparison of agglutination tests and enzyme-linked immunosorbent assay for the bovine brucellosis (소 브루셀라병 혈청 항체가 응집반응 비교 시험)

  • A total of 710 bovine serum samples which are composed 532 bovine serum samples showed negative reaction and 178 bovine serum samples showed positive reaction with tube agglutination test (TAT) from North area of Gyeong-nam, Korea were tested using all the 3 assays which are Rose-Bengal test(RBT), tube agglutination test (TAT) and enzyme-linked immunosorbent assay (ELISA, two types) and analyzed for evaluation of specificity, sensitivity, reproducibility and predictive value. In the comparison of serum antibody titer agglutination test, RBT showed almost agreement with TAT. In the comparison of TAT and two types of ELISA method, they showed difference in specificity and sensitivity about 5%. But there is no significant difference in detecting sensitivity between two types of ELISA method and TAT. In serologic tests for bovine brucellosis, the new assay ELISA would be a good candidate for serologic survey for bovine brucellosis in Korea because it is efficient in detecting many test samples quickly. But the serum agglutination tests (RBT, TAT) are more economical and easy assay for detection. In the test of comparison of antibody titer between first day of finding and 10 days after finding by TAT, there was no change in 55% (76/139) of positive cattle.

Isolation of bovine Coronavirus from calves with diarrhea (소 Coronavirus 분리에 관한 연구)

  • Fecal samples were collected from 47 calves with diarrhea and 12 clinically normal co-h-abitants, and tested for virus using MDBK cell cultures. Three cytopathic viruses were isolated from 8 fecal samples obtained from diarrheic calves. The isolated viruses were neutralized by bovine coronavirus hyperimmune serum In plaque reduction assay and were detected in the cytoplasm of MDBK cell by bovine coronavirus hyperimmune serum using immunofluorescence staining. The viruses agglutinated mouse erythrocytes only among the various animal erythrocytes tested and new isolates were identified as bovine coronavirus.

  • Attenuated varicella-zoster virus(VZV) was cultured in human embryonic lung cells. The effects of serum type and its concentration on the production of VZV were studied. Regardless of cell culture conditions, VZV yield was increased with multiplicity of infection, and the total cell concentration was decreased after virus infection. The newborn calf serum, calf serum, or horse serum was not as good as the fetal bovine serum or calf serum supplemented with iron for the propagation of VZV in the human embryonic lung cells. The yields of total VZV(cell-associated virus and cell-free virus) in the medium with calf serum containing iron were comparable to those in the medium supplemented with fetal bovine serum. Furthermore, some components of serum appear to be important for the maintenance of VZV infectivity.

Effect of Antisperm Antibodies on Capacitation and Acrosome Reaction of Bovine Spermatozoa (소의 항정자항체가 정자의 수정능획득 및 첨체반응에 미치는 영향)

  • The present study was performed to investigate the effects of caffeine and heparin on capacitation and acrosome reaction of bovine spermatozoa, effects of antisperm antibodies on acrosome reaction of bovine spermatozoa. The rates of acrosome reaction in control group, caffeine treated group, heparin treated group, caffeine-heparin complex treated group were 40.3, 54.3, 63.3, 72.3%, respectively and there were significant differences among the groups(p<0.01), especially higher in caffeine-heparin complex treated group than the others. The rates of acrosome reaction of antisperm antibodies serum supplemented groups(5, 10 and 20%) were 60.4, 48.9 and 37.1%, respectively and there were significant differences among the groups(p<0.0l), and the more increases in serum concentrations, the more decreases in acrosome reaction, but this phenomenon was not seen in fetal calf serum supplemented group and heifer serum group. When the serum concentration was 5%, the rates of acrosome reactions were significantly lower in fetal calf serum supplemented group than heifer serum group and in antisperm antibodies serum group(p<0.01), and there were no significant differences between heifer serum group and antisperm antibodies serum group(p<0.01). When the serum conecntrations were 10%, 20%, the rates of acrosome reactions were significantly lower in antisperm antibodies serum supplemented group than in fetal calf serum group and in geifer serum group(p<0.01), and there were no significant differences between fetal calf serum group and heifer serum group(p<0.01). These results indicate that caffeine-heparin complex treatment is very effective for inducing acrosome reaction of bovine spermatozoa and that antisperm antibodies block acrosome reaction.

  • Binding of nalidixic acid which is used primarily in the treatment of urinary infection and probenecid which is used as a uricosuric agent to bovine serum albumin were studied using difference spectrophotomeric method. 2-(4'-Hydroxybenzeneazo) bcnzoic acid as a spectrophotometric probe was used for measuring the binding of nalidixic acid and probenecid to bovine serum albumin. The association constants of nalidixic acid and probenecid were $1.58{\times}10^{4}M^{-1}$ and $1.70{\times}10^{4}M^{-1}$, respectively.

  • Based on the static and dynamic adsorption of Streamline DEAE, a modified tank-in-series model including particle size distribution was used to describe the adsorption performance of bovine serum albumin in an expanded bed. The calculated results indicated that the suggested model was able to simulate breakthrough curves under various conditions.

Vascular Endothelial Growth Factor Has Beneficial Effect Independent of Serum Components throughout Oocyte Maturation and Early Embryonic Development in Cattle

  • In our previous studies, we demonstrated that Vascular Endothelial Growth Factor (VEGF) enhances bovine oocyte maturation and early embryonic development in serum supplemented media. In this experiment, to determine the synergistic effect of VEGF with serum components on early embryonic development in vitro in cattle, 1 mg/ml polyvinyl-alcohol (PVA) was replaced with foetal bovine serum (FBS) in maturation and culture media. Bovine oocytes were matured in Synthetic Oviduct Fluid (SOF) supplemented with PVA, PVA+5 ng/ml of VEGF, FBS, or FBS+VEGF. Fertilized oocytes were cultured in the same conditions for 8 days. The development of embryos was examined at 48 h post- insemination and on days 6, 7 and 8. The results were analyzed using repeated measures two- factor ANOVA, in which the effects of VEGF and serum were assigned as two factors. The development rate to 4- to 8-cell embryos at 48 h was significantly higher in the PVA+VEGF group than in the PVA group (44.7% and 31.5%, respectively). However, the highest development rate to 4- to 8-cell embryos was obtained from the FBS+VEGF group (58.8%). On day 8, the blastocyst rates were higher in the PVA+VEGF (22.8%), FBS (32.1%, p<0.05) and FBS+VEGF (42.1%, p<0.05) groups than in the PVA group (17.1%). Two- factor ANOVA of the development rates indicates that VEGF had a significant effect, but had no synergistic effect with serum components on early embryonic development. The results of the present study demonstrate that VEGF improves the in vitro developmental competence of bovine oocytes and/or embryos independent of the effect of serum components.
