Path Matching Algorithm for Bridges Puzzle (가교 퍼즐에 관한 경로 매칭 알고리즘)

  • The problem of the bridges(Hasjiwokakero, Hasi) puzzle, which connects the bridge(edge) required by the island(vertex) without crossing the horizontal and vertical straight bridges except for the diagonal to form a connected network, is a barren ground for research without any related research. For this problem, there is no algorithm that presents a generalized exponential time brute-force or branch-and-bound method. This paper obtained the initial solution of the lattice graph by drawing a grid without diagonal lines for a given BP, removing unnecessary edges, and supplementing essential bridges. Next, through insufficient island pair path matching, the method of adding insufficient edges to the route and deleting the crossed surplus edges(bridges) was adopted. Applying the proposed algorithm to 24 benchmarking experimental data showed that accurate solutions can be obtained for all problems.

The Effect of Stress Among Middle School Students and the Effect of Motive on Their Addiction to the Internet (중학생의 스트레스와 인터넷 이용동기가 인터넷 중독에 미치는 영향)

  • The following research aimed to determine the effect of stress among middle school students on their addiction to the Internet. This research's target was a group of male students who had a high probability of getting addicted to the Internet while playing c/t games. The study distributed 357 questionnaires and used 340 copies, which meant discarding 17 copies that were considered inadequate. The research results are as follows: First, there appeared subordinate factors in the stress suffered by the students. These included stress from their families, from conflicts with their teachers, from the living environment, current schoolwork and future course in college, insecurity over their physical appearance, bullying from other students, and relationships with friends. Among these factors, stress caused by conflicts with teachers and family was the most frequently cited, while stress from their friends was the least cited. The motive in using the Internet was found to be bound with several factors. These include: a form of diversion, a way to communicate with others, a means in coping with loneliness, a source of news and information, a form of passing away time, a kind of habit, and others. Among these motives, passing away time and indulging a habit were cited the most, followed by news and information search, and a form of diversion. Second, as a subordinate factor in Internet addiction, the following were cited: formation of tolerance, health issues, occurrence of problems related to daily life, satisfaction or a pleasant sensation, withdrawal, cover-up on the use of the Internet, and formation of virtual interpersonal relationships, and others. Among these, the formation of tolerance came out the highest, followed by health issues, daily life, and problems related to daily life. Third, in terms of the effects of stress on the motive in using the Internet, the research found that the more the students felt stressed out by conflicts with their teachers and family, the more they tended to use the Internet to communicate with others, to cope with loneliness, to obtain newsI and information, to passawaytime, and to indulge a habit Also, the more they felt stressed out by the living environment, the more they tended to use the Internet to communicate with others, to cope with loneliness, and use news and information. The more they felt stressed out by their schoolwork and future course in college, they tended to use the Internet as a form of diversion and to secure news and information. The more they felt stressed out by their insecurity over their physical appearance and being victimized by bullies, the more they tended to use the Internet to cope with loneliness. Fourth, as for the effect of several variables on student addiction to the Internet, the study found that the more students felt stressed out by their living environment, by schoolwork and future course in college, by their physical appearance, and bullying from other students, the more they used the Internet as a form of diversion, a communication tool, and as a means of passing away time or indulging a habit. The study came up with the finding that the more the students used the computer and the Internet, the probability of their getting addicted to the Internet got higher.

Modulation of Cellulalr Quinone Reductase Inducibility by Roasting Treatment and Acid Hydrolysis of Perilla (들깨의 볶음처리와 산가수분해에 의한 세포모델계 Quinone Reductase 활성유도능의 변화)

  • Increased activities of phase 2 enzymes including quinone reductase(QR) have been reported to be associated with protection of animals from neoplastic, mutagenic, and other toxic effects of many carcinogens. In previous study, we found that methanol extract of roasted and defatted perilla meal induced the activity of quinone reductase, an anticarcinogenic marker enzyme, in murine hepalc1c7 cells. Current study showed that unroasted perilla had a limited QR-inducing activity, suggesting that roasting cause the generation of active component(s). Thus we hypothesized that QR inducer in perilla might be covalently linked to sugar moiety and released during roasting process. Methanol extract of defatted raw perilla was subject to acid treatment in order to hydrolyze the potential sugar moiety. Prolonged hydrolysis of methanol extract of defatted raw perilla at $98{\sim}100^{\circ}C$ increased the ability to induce cytosolic QR activity of hepalclc7 cells. Furthermore roasting at 180 and $200^{\circ}C$ resulted in significant induction of QR activity. The result strongly support the idea that QR inducer(s) is present in bound form in raw perilla and released during roasting. Cellular QR activity was induced proportionately with the increase of concentration of methanol extract of roasted perilla. The induction of QR by defatted perilla was also examined in the cytosols of liver, small intestine, stomach, lung and kidney of male ICR mice. Induction patterns showed specificity with respect to target tissue and roasting of perilla. Unroasted perilla meal (defatted) significantly induced QR in liver and lung, while roasted perilla meal induced QR in liver and stomach. The observation that raw perilla showed similar QR induction patterns to roasted perilla is consistent with our proposal that QR inducer(s) is present in bound form and released by physical and chemical treatments as digestive or microbial enzymes could release the inducers from inactive glycoside forms in gastrointestinal tract of mice. In conclusion, perilla could exert protective effect against chemically induced carcinogenesis by inducing phase 2 enzymes in biological systems regardless of chemical and physical process such as roasting.

  • When the spermine, which is one of the polyamines containing cation in vivo, binds to DNA, it can increase the stability of DNA. At the same time, it can cause B-form to Z-form transformations of DNA. However, because we cannot determine the binding geometry of the spermine to DNA by using spectroscopic methods, nobody can show the accurate binding mechanism of a DNA-spermine complex. Thus, we used DAPI as a spectroscopic probe of spermine, which binding geometry was well known. At the result of base selective binding geometry of spermine to synthetic DNA, the concentration of spermine gets higher, it grows the hydrophobic environment of DAPI which bound the minor groove of adenine-thymine base pair. Simultaneously, spermine seems to bridge the backbones around the minor groove of $poly[d(A-T)_2]$. So that, the intensity of fluorescence spectrum of that shows sudden increasement. In guanine-cytocine base pair, $poly[d(G-C)_2]$, we can suppose that spermine bind to the major groove of that, shoving out the DAPI which is partially intercalated between the base pocket across the major groove of it. In both cases, spermine doesn't show the base selectivity against to DNA.

Changes of Polyamine Content and Phytoavailability in Lactuca sativa cultivated in Cadmium and Arsenic Treated Soil (카드뮴과 비소처리가 상추의 polyamine함량 및 유효도에 미치는 영향)

  • Absorption of Cadmium(Cd) and Arsenic(As) by lettuce following to the change of polyamine content in lettuce were examined to find the effect of these elements on lettuce growth and on the basic resistance mechanism. As the contents of Cd and As were increased in soil, the contents of these elements were significantly increased in lettuce but lettuce growth was decreased. Sequential fractionation experiment of the Cd and As treated soil was shown that exchangeable, dilute acid extractable, and organically bound form, which were more mobile and phyto-available, of Cd were increased by 5.6, 42.9, and 56.7% during a growing season whereas 17.6, 25.0, and 24.1% were increased in case of As, respectively. Specially, the Cd content of leaves and roots in lettuce was positively correlated with exchangeable, dilute acid extractable, and residual form of Cd in the treated soil. However, the As content of roots in lettuce was positively correlated with all chemical forms of As in the treated soil. The contents of putrescine and cadaverine, a kind of polyamine, were also increased in lettuce as both elements were increased in soil.

  • 호르몬의 면역학적(免疫學的) 측정법(測定法)은 내분비학적(內分泌學的)인 입장(立場)에서 가장 중요(重要)한 연구방법(硏究方法)의 하나이며, 또한 핵의학적(核醫學的) 입장(立場)에서도 방사성(放射性) 동위원소(同位元素)를 이용(利用)한 성체관내(誠體管內) 측정(測定)의 하나로 중요(重要)한 位置를 차지하고 있다. 이 방법(方法)의 일반원리(一般原理)는 방사성(放射性) 동위원소(同位元素)로 표지(標識)된 호르몬이 특정항체(特定抗體)에 대(對)하여 가역적(加逆的)으로 반응(反應)하고, 표지(標識)된 항원-항체(抗原-抗體) 복합체(複合體)를 형성(形成)하는데 근거(根據)를 두고 있다. 단일(單一) system내(內)에서 표지(標識)되지 않은 호르몬이 표지(標識)된 호르몬과 면역학적(免疫學的)으로, 특정항체(特定抗體)에 대(對)해 경합적(競合的)으로 작용(作用)하기 때문에, 실제적(實際的)으로는 일정량(一定量)의 표지(標識)된 호르몬과 일정량(一定量)의 항체(抗體)가 있는 조건(條件)하에서 미지성료(未知誠料)와 알려진 양(量)의 호르몬을 가진 표준(標準) 호르몬을 동시(同時)에 처리(處理)하여, 표준곡선(標準曲線)을 얻고, 여기에서 미지성료(未知誠料)의 흐르몬 양(量)을 추정(推定)하게 된다. 이때 경합적(競合的)으로 결합(結合)한 표지(標識)호르몬(bound form)과 결합(結合)하지 못한 표지(標識)호르몬(free form)을 분리(分離)하는 것이 필요(必要)하며, 전기영동법(電氣泳動法), 이중항체법(二重抗體法), solid phase법(法), 염석법(鹽析法) 및 효소(酵素)에 의한 단백소산법(消散法) 등이 이 목적(目的)을 위해 개발(開發)되어 있다. 이 중(中)에서 전기영동법(電氣泳動法)은 변성(變性)된 호르몬 및 유리(遊離)된 방사성옥소(放射性沃素)를 알 수 있는 특장(特長)이 있고, 이중항체법(二重抗體法) 등은 다수(多數)의 성료(誠料)를 처리하거나, 시간(時間)을 단축(短縮)시키는 이점(利點)이 있다 호르몬에 방사성(放射性) 동위원소(同位元素)를 부착(附着)시키는 데에는 $^{131}I$$^{125}I$가 주(主)로 쓰이나, 각기 장단점(長短點)이 있으며, 높은 방사성(放射性) 활성도(活性度)를 얻기 위해 chloramine-T 및 Na-metabisulfite를 사용하는 부착방법(附着方法)이 주(主)로 사용된다. 미량의 호르몬을 실제 측정(測定)하기 때문에 표지(標識)호르몬의 비방사능도(比放射能度)가 높을 필요성(必要性)이 있으며, 이 경우에는 방사능(放射能)에 의(依)한 흐르몬의 변성(變性)이 큰 문제점으로 등장하게 된다.

Studies on Scintillation Proximity Assay for the mesurement of alpha-hCG (Alpha-hCG 측정을 위한 섬광 근접 측정법 (Scintillation Proximity Assay)에 관한 연구)

  • Purpose: Scintillation Proximity Assay (SPA) does not require the physical separation of receptor bound form from free form. SPA was applied to the study of interaction of human chorionic gonadotropin (hCG) and $anti-{\alpha}$ hCG in serum. Materials and methods: $Anti-{\alpha}$ hCG was biotinylated for the binding to streptavidin. The assay was based on the simple competitive binding method between $[^{125}I]hCG$ and the hCG in sample serum, with $anti-{\alpha}$ hCG-coated beads. Aliquots of biotinylated $anti-{\alpha}$ hCG were dispensed into scintillation vials containing $100{\mu}{\ell}\;[^125}I]hCG\;and\;200{\mu}{\ell}$ of either a standard concentration of hCG for preparation of standard curve or unknown sample, and incubated for 20 min. at room temperature. Then $20{\mu}{\ell}$ streptavidin-coated beads were added to vials, and finally incubated for 10 min at room temperature. Values for unknown samples were then calculated from the standard curve. Results: Optimal background counts were certificated using varied radioactivity of radionuclides. Appropriate standard curve was obtained from SPA method successively, and the concentration of hCG from unknown serum was determined by standard curve. The result from SPA assay was similar to that of RIA. Conclusion: This observation confirms that SPA method could be useful for clinical diagnosis.

Contribution of a Low-Barrier Hydrogen Bond to Catalysis Is Not Significant in Ketosteroid Isomerase

  • Low-barrier hydrogen bonds (LBHBs) have been proposed to have important influences on the enormous reaction rate increases achieved by many enzymes. ${\Delta}^5$-3-ketosteroi isomerase (KSI) catalyzes the allylic isomerization of ${\Delta}^5$-3-ketosteroid to its conjugated ${\Delta}^4$-isomers at a rate that approache the diffusion limit. Tyr14, a catalytic residue of KSI, has been hypothesized to form an LBHB with the oxyanion of a dienolate steroid intermediate generated during the catalysis. The unusual chemical shift of a proton at 16.8 ppm in the nuclear magnetic resonance spectrum has been attributed to an LBHB between Tyr14 $O{\eta}$ and C3-O of equilenin an intermediate analogue, in the active site of D38N KSI. This shift in the spectrum was not observed in Y30F/Y55F/D38N and Y30F/Y55F/Y115F/D38N mutant KSIs when each mutant was complexed with equilenin, suggesting that Tyr14 could not form LBHB with the intermediate analogue in these mutant KSIs. The crystal structure of Y30F/Y55F/Y115F/D38N-equilenin complex revealed that the distance between Tyr14 $O{\eta}$ and C3-O of the bound steroi was within a direct hydrogen bond. The conversion of LBHB to an ordinary hydrogen bond in the mutant KSI reduced the binding affinity for the steroid inhibitors by a factor of 8.1-11. In addition, the absence of LBHB reduced the catalytic activity by only a factor of 1.7-2. These results suggest that the amount of stabilization energy of the reaction intermediate provided by LBHB is small compared with that provided by an ordinary hydrogen bond in KSI.

Ultrastructure of Oocytes During Oogenesis and Oocyte Degeneration Associated with Follicle Cells in Female Sinonovacula constricta(BIVALVIA: PHARIDAE) in Western Korea

  • The ultrastructure of oocytes during oogenesis and oocyte degeneration associated with follicle cells in female Sinonovacula constricta(Lamarck, 1818) were investigated by electron microscope observations. Ovarian follicles are surrounded by a matrix of vesicular connective tissue cells(VCT cells). VCT cells contain large quantities of glycogen particles and several lipid droplets in their cytoplasm. It is suggested that VCT cells act as a source of nutrients for vitellogenesis during oogenesis. In early vitellogenic oocytes, several coated vesicles, which appear at the basal region of the oocyte, lead to the formation of membrane-bound vesicles via endocytosis. The uptake of nutritive materials in coated vesicles formed by endocytosis appears through the formation of coated pits on the oolemma during vitellogenesis. During the late stage of oogenesis, yolk precursors(yolk granules), mitochondria and lipid droplets are present in the cytoplasm of late vitellogenic oocytes. In particular, proteinaceous yolk granules containing several different components are intermingles and form immature yolk granules. In the mature oocyte, small immature yolk granules are intermingled and form large mature yolk granules. Vitellogenesis occurs through a process of autosynthesis, involving combined activity of the Golgi complex, mitochondria and rough endoplasmic reticulum in the cytoplasm of vitellogenic oocytes. The process of heterosynthesis is where extraovarian precursors are incorporated into oocytes by endocytosis at the basal region of early vitellogenic oocytes before the formation of the vitelline coat. Follicle cells appear to play an important role in vitellogenesis and oocyte degeneration. The functions of attached follicle cells to the oocyte during oocyte degeneration are phagocytosis and digestion of phagosomes originating from oocyte degeneration. After digestion of phagosomes, it is assumed that the function of follicle cells can permit a transfer of yolk precursors necessary for vitellogenesis and allows for the accumulation of glycogen and lipid during oocyte degeneration, which can be employed by vitellogenic oocytes. Follicle cells of S. constricta may possess a lysosomal system for induction of oocyte breakdown and might resorb phagosomes in the cytoplasm for nutrient accumulation during oocyte degeneration.

The Crystal and Molecular Structure of Sulfadiazine (Sulfadiazine의 結晶 및 分子構造)

  • Sulfadiazine, $C_{10}H_{10}N_4O_2S$, forms monoclinic crystals of space group $P21}c$ from a mixture of acetone and ethanol with $a=13.71{\pm}0.04,\;b=5.84{\pm}0.03,\;c=15.11{\pm}0.05{\AA},\;{\beta}=115.0{\pm}0.3^{\circ}$, and four molecules per cell. Three dimensional photographic data were collected with $CuK\alpha$ radiation. The structure was determined using Patterson and Fourier synthesis methods and refined by block diagonal least-squares methods with isotropic thermal parameter for all non-hydrogen atoms. The final R value was 0.15 for the 1517 observed independent reflections. The dihedral angle between the planes through the benzene ring and the pyrimidine ring is $76^{\circ}$. The conformational angle formed by the projection of the S-C(5) bond with that of N(1)-C(1) where the projection is taken along the S-N(1) bond is $77^{\circ}$. The imino nitrogen atom, N(1), and pyrimidine nitrogen atom, N(3), form intermolecular $N-H{\cdots}N$ hydrogen bond between the molecules related by center of symmetry. Amino nitrogen atom, N(4), forms two intermolecular $N-H{\cdots}O$ hydrogen bonds, with O(1) and O(2) atoms of different molecules separated by b. A two dimensional network of hydrogen bonds form infinite molecular sheets parallel to the (100) plane. Adjacent sheets are bound together by van der Waals forces.
