홍화 추출물이 치주인대세포, 조골세포 활성도에 미치는 영향 (The biologic effects of safflower(Carthamus tinctorius $Linn\acute{e}$) extract and Dipsasi Radix extract on periodontal ligament cells and osteoblastic cells)

  • Safflower(Carthamus tinctorius $Linn\acute{e}$ has been traditionally used for the treatment of blood stasis, and Dipsasi Radix has been used as a drug for fracture in Chinese medicine. The purpose of present study was to examine the biologic effects of safflower extract and Disasi radix extracts on the periodontal. ligament cells and osteoblastic cells and on the wound healing of rat calvarial defect. The ethanolic extract of safflower blossom, safflower seed and Dipsasi Radix(125, 250, and 500 ${\mu}g/ml$) were prepared as test group, and PDGF-BB(lOng/ml) and unsafonifiable fraction of Zea Mays L.(125, 250, and 500 ${\mu}g/ml$) were employed as positive control. The effects of each agents on the growth and survival, ALPase activity, expression of PDGF-BB receptor, chemotactic response of PDL cell and ATCC human osteosarcoma MG63 cells in vitro were examined. The tissue regenerative effect of each extracts was evaluated by histomorphometric measuring of newly formed bone on the 8mm defect in rat calvaria after oral administration of 3 different dosages groups : 0.02, 0.1 and 0.35g/kg, per day. It was also employed the same dosages of unsaponifiable fraction of Zea Mays L. as positive controls. Safflower blossom extract, safflower seed extract, and Dipsasi Radix extract stimulate the cellular activity of MG63 cells in concentration range of $125-500{\mu}g/ml$, and safflower bolssom extract and safflower seed extract stimulate also the cellular activity of periodontal ligament cells in concentration range of $250-500{\mu}g/ml$. In activity of ALPase, $250-500{\mu}g/ml$ of safflower blossom extracts showed significant stimulating effects on MG63 cells, and the same concentration range of safflower seed extracts showed significant effect on periodontal ligament cells. In the recovery on PDGF-BB receptor expression which was depressed by $IL-1{\beta}$, $125-250{\mu}g/ml$ of safflower blossom extracts and $250-500{\mu}g/ml$ of safflower seed extracts showed significant increasing effect on MG63 cells, and $500{\mu}g/ml$ of safflower blossom extract and $250-500{\mu}g/ml$ of safflower seed extracts showed significant effect on periodontal ligament cells. In chemotactic response, among all tested group, safflower seed extracts only were chemotactic to MG63 cells and periodontal ligament cells in concentration range of $125-500{\mu}g/ml$. Also in the view of bone regeneration in rat calvarial defect model, the only group that was orally administrated 0.35g/kg, day of safflower seed extract showed significant new bone formation. These results suggested that safflower extracts might have a potential possibilities as an useful drug for adjunct to treatment for regeneration of periodontal defect.

  • Objective: Although the effect of Ulmus davidiana Planch (UD) extracts on collagen-induced-arthritis (CIA) and bone metabolism has been studies, research on its effect on human immunomodulatory activity is further a due. The objective of the present study was to investigate the immunomodulatory activity of UD on cellular and humoral immunity. Methods : Oral administration of the ethanolic and water extracts of UD, at doses of 20, 100 and 200 mg/kg in mice, dose dependently potentiated the delayed type hypersensitivity reaction induced both by sheep red blood cells (SRBC) and oxazolone. Results : It significantly enhanced the production of circulating antibody titre in response to SRBC in mice. Extracts of UD failed to show any effect on macrophage phagocytosis. Chronic administration of UD extracts significantly ameliorated the total white blood cell count and also restored the myelosuppressive effects induced by cyclophosphamide. Conclusion : The present investigation reveals that UD extracts possesses immunomodulatory activity.

SAM p6 mouse에서 자하거 주사제의 골다공증 개선에 대한 연구 (Effects on osteoporosis of Senescence Accelerated Mice (SAM) P6 by Injection of Placenta extracts ($Beuronel^{(R)}$))

  • Objectives : The purpose of this study was to examine the effects of the Hominis placenta extracts on improvement of senile osteoporosis (Type 2) in SAM P6. In traditional Oriental medicine, Placenta (Jahageo) is indicated for insufficiency of blood and tidal fever due to steaming bone disorder. Methods : At 10 weeks after birth, Placenta ex. was given 100 mL, 200 ml/kg/day, i.p. 3 times a week for 4 weeks in SAM P6 mice. And then we measured complete blood cells, blood urea nitrogen, creatinine, inorganic phosphate and total iron. In addition, we tested bone mineral density (BMD) using the soft X-ray. Results : As a result, in SAM P6, injection of Placenta ex. (200 mL) increased in RBC, HB and PLT, in comparison with control group. It was also found that the inorganic phosphate levels increased significantly in the injection groups of the Placenta ex. from that of the control group, but blood urea nitrogen was no significant. Moreover, Placenta ex. were showed a trend of increase in bone mineral density (BMD) of the vertebrae lumbales and tibia in P6 mice. Conclusions : These findings suggest that Placenta ex. are effective in preventing bone loss in SAMP6.

당귀천궁복합물이 대퇴골 골절 동물모델에서 골 유합에 미치는 영향 (Effects of Extracts from Cnidium officinale and Angelica sinensis on Bone Fusion in Mice with Femoral Fracture)

  • Objectives The purpose of this study is to evaluate the fracture healing effect of extracts from Cnidium officinale and Angelica sinensis (CO/AS) in mice with femoral fracture. Methods C57BL/6 mice were randomly divided into normal, control (phospate-bufferd saline), positive control (tramadol), CO/AS extract 40 mg/kg and 80 mg/kg. By using Collier's method, all groups except normal group went through femoral fracture. Aspartate aminotransferase (AST), alanine transferase (ALT), lactate dehydrogenase (LDH), blood urea nitrogen (BUN) and creatinine were measured to evaluate the safety of CO/AS. Hematoxylin & eosin, Safranin O staining, x-ray, tensile and compressive force were conducted to assess the effect of CO/AS on fracture. Results The liver function test showed AST, ALT and LDH in CO/AS at 14th and 28th days were not significantly different compared with control group. The renal function test showed BUN in CO/AS at 14th days and BUN and creatinine in CO/AS at 28th days were significantly decreased compared with control group. The morphological & histological analysis and x-ray showed that CO/AS promoted cartilage and callus formation process compared with control group. The tensile and compressive forces test showed tensile in CO/AS 40 mg/kg and tensile & compressive forces in CO/AS 80 mg/kg were significantly increased compared with control group. Conclusions CO/AS extract showed the possibility that it promotes early fracture union and increases bone tensile and compressive strength, while does not have hepatotoxicity. In conclusion, CO/AS has a potential to promote healing of bone fracture and this study warranted the clinical usage of CO/AS at bone fracture.

Extracts from the Red Algae Gracilaria vermiculophylla have Antioxidant Effects in Human Bone Marrow Mesenchymal Stem Cells

  • The red algae Gracilaria vermiculophylla is widely spread around seaside areas across the globe, and has been used as a food resource in Southeast Asian countries. Previous studies have shown that Gracilaria red algae extracts have beneficial antihypercholesterolemic, antioxidant, anti-inflammatory, and antimicrobial effects. In this study, we investigated the antioxidant effects of Gracilaria vermiculophylla extracts (GV-Ex) on human bone marrow mesenchymal stem cells (hBM-MSCs). The acetone and DMSO/ethanol solvents of the tested GV contain higher total flavonoid and polyphenolic contents that can strongly scavenge reactive oxygen species (ROS). Pre-treatment with GV-Ex protected hBM-MSCs against oxidative stress induced by hydrogen peroxide treatment. The protective effects of GV-Ex treatment were confirmed by MTT assay. The elevated levels of ROS in hBM-MSCs caused by hydrogen peroxide induced oxidative stress were significantly decreased by GV extract treatment. The levels of the antioxidant proteins superoxide dismutase 1 (SOD1), superoxide dismutase 2 (SOD2), and catalase (CAT) were also restored or protected by GV-Ex treatment, suggesting that GV extracts moderate excess ROS levels and prevent cells from oxidative damage.

Oral Administration of Mice with Cell Extracts of Recombinant Lactococcus lactis IL1403 Expressing Mouse Receptor Activator of NF-kB Ligand (RANKL)

  • Receptor activator of NF-kB ligand (RANKL) is known to play a major role in bone metabolism and the immune system, and its recombinant form has been expressed in bacterial systems for research since the last two decades. However, most of these recombinant forms are used after purification or directly using living cells. Here, there were cell extracts of recombinant Lactococcus lactis expressing mouse RANKL (mRANKL) used to evaluate its biological activity in mice. Mice were divided into three groups that were fed phosphate-buffered saline (PBS), wild-type L. lactis IL1403 (WT_CE), and recombinant L. lactis expressing mRANKL (mRANKL_CE). The small intestinal transcriptome and fecal microbiome were then profiled. The biological activity of mRANKL_CE was confirmed by studying RANK-RANKL signaling in vitro and in vivo. For small intestinal transcriptome, differentially expressed genes (DEGs) were identified in the mRANKL_CE group, and no DEGs were found in the WT_CE group. In the PBS vs. mRANKL_CE gene enrichment analysis, upregulated genes were enriched for heat shock protein binding, regulation of bone resorption, and calcium ion binding. In the gut microbiome analysis, there were no critical changes among the three groups. However, Lactobacillus and Sphingomonas were more abundant in the mRANKL_CE group than in the other two groups. Our results indicate that cell extracts of mRANKL_CE can play an effective role without a significant impact on the intestine. This strategy may be useful for the development of protein drugs.

Stewartia pseudocamellia and Torilis japonica Extracts Inhibit RANKL-induced Osteoclastogenesis in RAW 264.7 Cells

  • Osteoporosis is a disease that causes the weakening of bone by increasing porosity, which often results in fractures. Osteoporosis treatment measures include the use of Bisphosphonates and estrogen. However, these treatments cannot be used in the long term as these treatments have adverse side effects. Therefore, there is a need to identify better and safer treatment options. For this, 63 plant extracts were screened and among them, six extracts showed high anti-osteoclastic activity with low cytotoxicity. Of these six extracts, three extracts, Cudrania tricuspidata (P371), Ulmus davidiana var. japonica (P401), and Torilis japonica (P411), showed more than 50 percent osteoclast inhibition. While the remaining, Stewartia pseudocamellia extracts I and II (P370, P397) and Cuscuta chinensis (P418), showed moderate or between 40-50 percent osteoclast inhibition. Among all the extracts, Torilis japonica (P411) showed the highest inhibitory action against osteoclast development. Torilis japonica (P411) primary components include Kaempferol, Quercetin, and Luteolin, all proven to inhibit osteoclastogenesis. Stewartia pseudocamellia extracts I and II (P370 and P397) showed moderate or 44% osteoclast inhibition. Stewartia pseudocamellia extract II (P397) enhanced the growth of RAW 264.7 cells by 19%. Torilis japonica (P411) and Stewartia pseudocamellia extract II (P397) suppressed the expression of osteoclast-specific genes in RANKL-induced osteoclastogenesis in RAW 246.7 cells. Torilis japonica (P411) extracts even increased osteoblast-specific RUNX2 gene expression. This results provide that six extracts could be used as a potential treatment option for osteoporosis disease with the extracts of Torilis japonica (P411) and Stewartia pseudocamellia (P397) as an ideal candidates. However, the combination of the extract with higher osteoclastic inhibition and less toxic effects with further analysis should be recommended.

CT 영상에서 스네이크 알고리즘과 파티클 필터를 이용한 뼈 영역 추출 방법 (A Bone Region Extraction Method based on Snake Algorithm and Particle Filter in CT image)

  • 본 논문에서는 CT 영상에서 스네이크 알고리즘과 파티클 필터를 이용한 뼈 영역 추출 방법을 제안한다. 스네이크 알고리즘을 이용하여 뼈 외곽선을 추출하고, 이 외곽선을 따라서 파티클 필터를 움직여 가면서 뼈 영역을 추출한다. 뼈 외곽선 주위에 다른 뼈가 근접해 있는 경우에 스네이크 알고리즘이 뼈 외곽선을 완벽하게 추출하지 못할 수 있는데, 이때에 파티클 필터가 이러한 오류를 보완해가면서 뼈 영역을 추출한다. 본 논문에서 제안한 방법을 기존의 형태학적 처리를 이용한 방법과 비교한 결과, 추출하고자 하는 뼈 영역 주위에 다른 뼈가 근접해 있지 않은 경우에는 비슷한 결과를 보였지만, 다른 뼈가 근접해 있는 경우에는 본 논문에서 제안한 방법의 정확도가 높음을 알 수 있었다.

쇄양(鎖陽)과 두중(杜仲)이 골다공증 유발 후 흰쥐의 골밀도와 체중에 미치는 영향 (The Efficacy of Cynomorii Herba and Eucommiae Cortex on Treatment of Osteoporosis in Ovariectomized Rats)

  • Objectives : The present study had been undertaken to investigate the effects of Cynomorii Herba and Eucommiae Cortex on treatment of osteoporosis in ovariectomized rats. Methods : In this experiment, the rats of experimental groups were ovariectomized. Rats of medicinal groups were administered by water extracts of Cynomorii Herba or Eucommiae Cortex. The levels of bone mineral density and body weight were measured. Results : The levels of spinal bone mineral density was significantly increased in comparison with OVX group at 8 weeks after medication in Cynomorii Herba group. The levels of femoral and fibula-tibial bone mineral density were not significantly increased in comparison with OVX group after medication in Cynomorii Herba and Eucommiae Cortex group. The levels of body weight were not significantly decreased in comparison with OVX group after medication in Cynomorii Herba and Eucommiae Cortex group. Conclusions : Reviewing these experimetal results, it appears that Cynomorii Herba have efficacy on treatment of osteoporosis. And further study should be conducted to illustrate in depth the curing and prevention of osteoporosis.

Methods : In this experiment, the rats were ovariectomized. Rats were administered by water extracts of Trigonellae Semen. The levels of bone mineral density and body weight were measured. Results : The levels of spinal bone mineral density was significantly increased in comparison with OVX group at 4 and 8 weeks after medication in Trigonellae Semen group. The levels of femoral and fibula-tibial bone mineral density were significantly increased in comparison with OVX group at 4 weeks after medication in Trigonellae Semen group. The levels of body weight was significantly increased in comparison with OVX group at 4 weeks after medication in Trigonellae Semen group. Conclusion : Reviewing these experimetal results, it appears that Trigonellae Semen have efficacy on treatment of osteeoporosis. And further study should be conducted to illustrate in depth the curing and prevention of osteoporosis.
