Supplement of High Protein-Enriched Diet Modulates the Diversity of Gut Microbiota in WT or PD-1H-Depleted Mice

  • Supplement of high-protein food plays an important role in improving the symptoms of malnutrition and the immune capacity of the body, but the association of high-protein diet and gut microbiota remained unaddressed. Here, we systematically analyzed the internal organs and gut microbiota in C57(WT) or PD-1H-depleted (KO) mice (T cells were activated) fed with pupae or feed for six weeks. We observed that the body weight gain in the mice fed with pupae increased less significantly than that of the feed group, while the villi and small intestine lengths in the pupa group were reduced compared with that of mice given feed. However, the average body weight of the KO mice increased compared with that of the WT mice fed with pupae or feed. Pupae increased the concentration of blood glucose in WT, but not in KO mice. Moreover, in the feed group, there was no difference in the weight of the internal organs between the WT and KO mice, but in the pupae-fed group, liver weight was decreased and spleen weight was increased compared with that of KO mice. The amounts/plural/amounts of Melainabacteria, Chloroflexi, and Armatimonadetes were specifically upregulated by pupae, and this upregulation was weakened or eliminated by PD-1H depletion. Some bacteria with high abundance in the feed-fed KO mice, such as Deferribacteres, Melainabacteria, Acidobacteria, Bacteroidetes, Spirochaetes and Verrucomicrobia, were decreased in pupae-fed KO mice, and Proteobacteria and Deinococcus were specifically enriched in pupae-fed KO mice. Bacteroidetes, Firmicutes and Akkermansia were associated with weight loss in the pupae-fed group while Lachnospiraceae and Anaerobiospirillum were related glucose metabolism and energy consumption. Based on high-throughput sequencing, we discovered that some gut bacteria specifically regulated the metabolism of a high-protein diet, and PD-1H deficiency improved life quality and sustained blood glucose. Moreover, PD-1H responses to high-protein diet through modulating the type and quantity of gut bacteria. These findings provide evidence about the association among gut microbiota, T cell activation (for PD-1H depletion) and high-protein diet metabolism, have important theoretical significance for nutrition and health research.

  • 카드뮴 투여에 의해 급성중독 된 흰쥐의 간장과 신장에서의 MT합성양상을 검토하고져, 체중이 $60\pm{5g}$ 내외가 되는 Sprague-Dawley종 수컷을 이용하여, 4주간 기본식이로 사육한 후, Cd투여량 및 투여횟수에 따른 MT합성양상을 검토하였다. 즉, Cd투여량에 따른 실험에서는 체중 Kg당 0, 0.625, 1.25, 2.5, 5mg의 Cd를 24시간 간격으로 2회 복강 내에 투여하였으며, Cd투여 횟수에 따른 실험에서는 체중 Kg당 2.5mg의 Cd를 각각 0, 1, 2, 3, 4회씩 24시간 간격으로 복강내에 투여하였다. 두 실험 모두, 마지막 주사 24시간 만애 실험동물을 희생하여 혈액중의 hemoglobin과 hematocrit함량 측정과 간장 및 신장조직중에서의 MT합성양상을 관찰하였으며, 아울러 간장조직중에서의 -SH group함량을 측청하였다. 1) Cd 투여 농도에 따른 hemoglobin함량은 대조군에 비하여 체중 Kg당 2.5, 5.0mg의 Cd를 투여한 군에서 유의적으로 감소하였으나 hematocrit치는 유의적인 차이가 없었다. 또한 Cd 투여횟수를 증가시켰을 때는 모든 군이 대조군보다 감소되었으며 hermatocrit치는 3,4회 Cd 투여군에서 대조군보다 감소되었다. 2) 간종조직중의 MT합성은 2.5mg Cd/Kg of body wt.까지는 Cd투여농도에 비례하여 점진적인 증가를 하였다. 또한 Cd투여 횟수가 증가됨에 따라 간장 MT합성은 비례하여 증가하였다. 두 실험 모두에서 신장조직중의 MT합성량은 간장조직에서보다 합성량은 적었지만 그 경향은 비슷하였다. 3) 간장에 대한 신장내에서의 MT합성비율은 투여횟수가 증가함에 따라, 다소 증가하는 경향이었다. 4) Cd투여농도에 따른 간조직중의 -SH group 함량은 대조군에 비해 체중 Kg당 2.5, 5.0mg Cd투여군에서 유의적인 증가를 하였으며, Cd 투여 횟수의 증가에 따라서는 모든 군이 대조군에 비해 유의적으로 증가되었다.

유기수은 노출로 인한 흰쥐 태자골격성장 지연에 대한 마늘의 보호효과 (Protective Effects of Korean Garlic Juice against the Toxicity of Methyl Mercuric Chloride (MMC) in Relation to Fetal Ossification in Pregnant Fischer-344 Rats)

  • This study evaluates the protective effects of Korean garlic juice against the toxicity of methyl mercuric chloride (MMC) in relation to fetal ossification in pregnant Fischer 344 rats. This study has as its basis, both theory and data that neutral amino acids in garlic juice have protective effects against mercury poisoning. Pregnant rats were dosed in various combination with 20 mg MMC/kg body wt. and 0.5 or 1.0 garlic juice/kg body wt. on the 7th day of gestation. Fetuses were extracted from the dams on 20th day of gestation, and the fetal bones were stained and measured. The results obtained are as follows: 1. Fetal body weights, body lengths, and head length were significantly decreased by as much as 23.5%, 21.3%, 15.5% respectively in 20 mg/wt kg methyl mercuric chloride groups (p<0.05). But in the garlic treated groups, they were almost similar to the controls. 2. The ossification centers were significantly decreased by as much as 35.1% in pelvic phalanges, 53.5% in pectoral phalanges, 74.1% in ternebrae, 76.2% in tail in 20 mg/wt kg methyl mercuric chloride groups (p<0.05). But in the garlic treated groups, they were significantly increased by as much as $81.2{\sim}88.7%$ and $94.8{\sim}98.9%$ of controls. 3. The ossified pectoral girdles were significantly decreased by as much as 66.6% in clavicle, and in other areas $74.2{\sim}87.4%$ in the 20 mg/wt kg methyl mercuric chloride groups (p<0.05). But in the garlic treated groups, they were significantly increased by as much as $81.2{\sim}88.7%$ and $94.8{\sim}98.9%$ of the controls. 4. The ossified pelvic girdles were significantly decreased by as much as 57.1% and 56.1% in two ischium, $67.2{\sim}81.7%$ in metacarpals in the 20 mg/wt kg methyl mercuric chloride groups (p<0.05). But in the garlic treated groups, they were significantly increased by as much as $67.0{\sim}85.6%$ and $90.1{\sim}98.7%$ of the controls. 5. The ossified terenebrae were unchanged or significantly decreased; 0.0% in 5th, 54.1% in 1st, 83.9% in 2nd, 75.0% in 3rd, 72.7% in 4th, 79.8% in 6th of 20 mg/wt kg methyl mercuric chloride groups (p<0.05). But in the garlic treated groups, they were significantly increased by as much as $29.5{\sim}55.1%$ and $54.5{\sim}84.0%$ of the controls. 6. The ossified ribs were significantly decreased by as much as $8.3{\sim}18.0%$ in 20 mg/wt kg methyl mercuric chloride groups (p<0.05). But in the garlic treated groups, they were significantly increased by as much as $87.1{\sim}93.5%$ and $96.3{\sim}99.7%$ of the controls. In conclusion, Korean garlic juice significantly protected against the toxicity of MMC in relation to the fetal ossification in pregnant rats.

자기 소지의 특성에 미치는 도석첨가의 영향 (A Study on the effect of the Pottery Stone added to the Porcelain Body)

  • This study is to investigate the effect of a domestic pottery stone as a batch inclusion on the synthesis of porcelain bodies and their physical properties. The basic composition of porcelain bodies studied was consisted of 50wt% kaolin, 25wt% feldspar and 25wt% quartz. The inclusion of pottery stone was performed by partial substitution for either quartz or feldspar up to 25% of the total weight of batches. Sixteen porcelain bodied different in batch composition were prepared by firing them in the temperature range from 110$0^{\circ}C$ to 135$0^{\circ}C$ and their physical properties and microstructures were carefully examined. It was found that the inclusion of pottery stone could reudce the firing temperature for vitrification up to 5$0^{\circ}C$ but appreciably decreased the mechanical strength of sintered bodies. The most faborable result could obtain with partial substitution for both quartz and feldspar at same time up to 25% of the total batch weight.

Fumed Silica/Ceramic Wool 무기복합재의 제조 및 열적 성질 (Fabrication and Thermal Properties of Fumed Silica/Ceramic Wool Inorganic Composites)

  • Fumed Silica와 섬유상의 Ceramic Wool을 사용하여 경량의 무기복합재 샘플을 제조하기 위한 조건과 만들어진 샘플의 단열특성을 살펴보았다. 정량된 Fumed Silica 미세분말과 Ceramic Wool을 혼합한 반죽을 몰드에 넣고 상온에서 안정화시킨 후에 $150^{\circ}C$ 오븐에서 완전히 건조하여 샘플을 제작하였다. 소량의 PVA 계면접착제를 사용하지 않는 샘플에서는 Fumed Silica 조성비가 10-70wt% 사이에서 벌크밀도가 0.6-0.8 $g/cm^3$이었으며, 50wt% 이상의 샘플에서는 건조 수축으로 인한 크랙현상이 관찰되었다. 그러나 3wt%의 PVA를 사용한 샘플의 벌크밀도는 절반 정도로 크게 감소하면서도 기계적 특성과 단열성은 향상되었다. 만들어진 샘플들은 $800^{\circ}C$ 이상의 고온에서도 열크랙 없이 안정한 열적 특성을 보여주었으며, 샘플의 단열성은 Fumed Silica 조성비가 높아질수록 향상되는 것으로 나타났다. Fumed Silica 30wt%인 샘플의 열전도도는 $500^{\circ}C$에서 약 0.08 $W/m^{\circ}K$의 우수한 단열 특성을 보여 주었다.

정수 슬러지를 혼합한 계룡산 철화 분청사기 고강도 소지 개발 (Study on High- Strength Body of Chulwha Buncheong Ware from Geryong Mountain with the Addition of Purified Sludge)

  • First, a purified sludge was calcined at various temperatures viz. 800, 900, 1000, 1100 and $1200^{\circ}C$ per hour. Subsequently 100 wt% of ware from Geryong mountain was mixed with 5~25 wt% of a purified sludge. Then the ware was treated at $1250^{\circ}C$ in an electric kiln to test a bending strength. The physicochemical property of the prepared ware was characterized by XRD, Raman and SEM analysis. Among the different percentage, 25 wt% of a ware in a purified sludge calcined at $1000^{\circ}C$ showed 689 $kg/cm^2$ strength. Also the purified sludge calcined at $1000^{\circ}C$ was adequate physical properties than the other composites. Further to enhance the physical strength, 3 wt% $TiO_2$ (a mineralizer) was added into the ware and the strength was increased up to 731 $kg/cm^2$. The composites exhibit absorption and porosity rates of 0.17% and 0.39% respectively.

Measurement of Melting Temperatures of $UO_2,\;(U,Gd)O_2\;and\;(U,Er)O_2$ Fuels

  • The melting temperatures of $UO_2,\;UO_2-6wt\%Gd_{2}O_3,\;UO_2-12wt\%Gd_{2}O_3,\;UO_2-2wt\%Er_{2}O_3,\;and\;UO_2-4wt\%Er_{2}O_3$ fuels were measured. Fuel materials were loaded in a tungsten capsule of which shape met the black body condition. The melting temperature was measured by the thermal arrest method during heating of the capsule in an induction furnace. The measured melting temperature of $UO_2$ fuel was $2815{\pm}20^{\circ}C$. The solidus and liquidus temperatures of $UO_2-Gd_{2}O_3\;and\;UO_2-Er_{2}O_3$ had also been measured, and it was observed that the solidus temperatures of them were lower than the liquidus temperature by $15{\sim}25^{\circ}C$. Measured melting temperatures of $UO_2,\;UO_2-Gd_{2}O_3\;and\;UO_2-Er_{2}O_3$ fuels were as follows:

석탄회-점토계 소지의 제조 및 물성 (Manufacture and Properties of Coal Fly Ash-Clay Body)

  • Utilization of fly ash by-produced from coal fired power plants and classified as general waste became very important problem to solve in the environmental protection and recycling of waste materials. The possibility of large scale substitution of fly ash as a raw material for bricks and wet tiles was highly expected because the chemical compositions of fly ash were mostly Al2O3 and SiO2 and the properties of it were very similar with clay. Accordingly in order to investigate the substitutional limit these specimens were substituted from 0 to 100 wt% fly ash by 20wt% increment for clay. Fly ash-clay bodies were fired at 1200, 1250 and 130$0^{\circ}C$ and then their properties were measured, It was found that these specimens sintered at 125$0^{\circ}C$ had a good bending strength. Especially when these sintered bodies were added to 20, 40 and 60 wt% fly ash the bending strength of those were 201 , 205 and 191kg.cm2 respectively with the water absorption below 1%, This showed that fly ash could be substituted ab 60 wt% in this experiment.

석탄 바닥재와 점토를 이용한 인공경량골재 제조 (Manufacturing artificial lightweight aggregates using coal bottom ash and clay)

  • 화력발전소에서 발생하는 석탄 바닥재(bottom ash)와 점토를 혼합하여 성형 후, 소성하여 인공경량골재를 제조하고, 소성온도와 조성 변화에 따른 물성을 분석하였다. 바닥재는 입경이 4.75mm 이상인 입자가 13wt% 정도로 거친 분말로 압출성형을 위하여 미분쇄 공정이 필요하였다. 또한 바닥재는 미연탄소(C)를 다량 함유하고 있어 소결 시 C의 산화반응과 이에 따른 가스발생으로 소결체의 경량화를 유도하였다. 점토에 바닥재 첨가량이 증가할수록 소성 지수가 감소하였고 이에 따라 성형체의 성형성이 저하되었으나 바닥재 첨가량이 40wt% 까지의 성형체는 소성 지수 및 소성 한계값이 각각 약 10과 22로서 압출성형이 가능하였다. 바닥재가 $30{\sim}50wt%$ 첨가되고 $1150{\sim}1200^{\circ}C$ 범위에서 소결된 골재는 부피비중 $1.3{\sim}1.5$, 흡수율 $14{\sim}16%$를 나타냈고 따라서 고층빌딩이나 교량 등의 골재대체재로써의 가능성이 확인되었다.

청소년의 골밀도에 영향을 주는 요인에 관한 연구: 신체계측치, 생활습관 및 기타 환경요인 (A Study of Factors Affecting Bone Mineral Density in Korean Adolescents: Anthropometric Measurements, Life Style, and Other Environmental Factors)

  • This study was conducted to assess the bone mineral density (BMD) and the anthropometric measurements, life style, and other environmental factors affecting BMD in Korean adolescents. Subjects were 167 high school students (83 male students, 84 female students) in Seoul. BMD was measured in the lumbar spine (LS), femoral neck (FN), femoral trochanter (FT), and Ward's triangle (WT) by dual energy x-ray absorptiometry (DEXA). The questionnaire was used to assess the anthropometric measurements, life style, and other environmental factors. Bone mineral density of LS, FN, FT and WT were 0.967, 0.960, 0.795, $0.761 g/{\cal}cm^2$. The BMD of LS was not different by sex but the male students's BMD of FN, FT and WT were higher significantly than the female students (LS: 0.976 vs. 0.958, FN: 1.040 vs. 0.880, FT: 0.842 vs. 0.749, WT: 0.827 vs. $0.695 g/{\cal}cm^2$) Female students's BMD of LS, FN, WT, and FT was positively correlated with weight but male students's BMD of WT was not correlated with weight. The factors such as the life style, activity and exercise have significant influence on BMD. This study confirms that the major factor affecting BMD was body weight and the factors such as the life style, activity and exercise is related to accumulation of BMD. The classification of sexual characteristics is needed for further studies on BMD of adolescents.
