In experiment 1, dietary self-selection of the 3 macronutrients, protein, fat, and carbohydrate were examined in male rats given 3 food cups of 80% carbohydrate, 80% protein, and 70% fat diets simultaneously. All the rats showed normal growth pattern and organ weight, which means they have ability to select just right kinds and amounts of nurients in order to support their growth and development. Mean values of caloric intake, body weight gain, serum lipid values and empty carcass compositions were not significantly differ between the upper and lower quartile groups of fat proportion of empty carcass compared to the lower quartile group(LF). Same feeding design was employed in experiment 2 where the effect of mild electric stress on food selection was studied. The rats in both control and electric stress group revealed a normal growth curve and organ weights. This study was conducted to compare effects of various types of dietary carboh ydrates fed with different levels of protein on the protein utilization in weanling rats. Sixty male Sprague-Dawley rats weighing $60{\pm}1.3grams$ were adapted for 1 week with 77% starch-15% casein diet. Then the animals divided into 12 groups according to body weight and fed each experimental diet for two weeks. Carbodydrates used were glucose, starch, and sucrose and the amount of protein given were 0g, 1g, 3g, 5g casein/day. Protein portion of the diet was fed in two seperate feedings per day while nonprotein portion was fed ad libitum. It seemed that there was no significant difference in the protein utilization by using the different kinds of carbohydrate, but in P.E.R., N.P.U., weights of organs and protein and lipid in total carcass, glucose groups were tended to be slightly lower than starch and sucrose groups. The larger the amount of casein given, the higher were the value of body weight gain, F.E.R., weights of organs, total lipid in carcass and the amount of nitrogen retention. On the while, the larger the amount of casein given, the lower were the value of the intake of non-protein portion, P.E.R., N.P.U, and the percentage of nitrogen retention.
To investigate the acute toxicity of Elfvingia applanata$(P_{ers})K_{ARST}$, it was extracted with hot water. EA, the aqueous extract from the carpophores of E. applanata, was lyophilized and a dark brownish powder was obtained. The acute toxicity of EA was investigated in ICR mice of both sexes. Five groups of mice were administered orally and intraperitoneally with 375, 750, 1,500, 3,000, 6,000 mg/kg(p.o.) and 312.5, 625, 1,250, 2,500, 5,000 mg/kg (i.p.) of EA Abnormal clinical signs related to EA were not observed and no deaths occurred. $LD_{50}$ values administered orally and intraperitoneally were evaluated to be over 6,000 mg/kg and over 5,000 mg/kg, respectively. Gross finding of necropsy revealed no evidence of specific toxicity related to EA. There were no significant changes in body weights, serum biochemical values and histopathological changes compared with control group.
The effects of methidathion on humoral immune response were studied in BALB/c mice. 0.5 or 5.0 mg/kg/day methidathion were administered orally for 14 days. The parameters examined to assess apparent toxicity of methidathion included changes of body weight, relative weight of spleen, thymus, sidney and liver, and viable spllenic cell numbers. To evaluate the humoral immune resopnse, the plaque forming cell(PFC) responses sheep the red blood cells (SRBC) and the lovels of serum IgG to hen egg lysozyme (HEL) were determined. No alterations were observed in changes of body weights, relative organ weights and the numbers of viable splenocytes by exposure to any dose of methidathion. At the dose of 0.5mg/kg only PFC response was decreased, whereas both PFC response and the level of serum IgG were decreased significantly at the dose of 5.0 mg/kg. These results indicate that exposure to methidathion may cause sup[pression of humoral immune reponse in mice without overt changed in lymphoid organ weight or viability of splenocytes.
This study was carried out to investigate the supplementary effects of the rice germ oil compared with soy bean oil on blood glucose level of non-insulin dependent diabetic mice. Forty diabetic KK mice were fed two kinds of experimental diets with 20% lipid from soy bean oil as a control(CO) and rice germ oil(RG) for 8 weeks, respectively. Diet intake, body weight, organs weights and lipids levels of serum, liver and feces were measured. There was no significant difference in food and water intake, body weight gain and organs weights between experimental groups. The concentrations of fasting and random blood glucose were similar between CO and RG groups. There was no significant difference in blood glucose levels after glucose treatment during the glucose tolerance test between two groups. The levels of $HbA_{1c}$ as the index of blood glucose status, and insulin were similar in two groups. These results suggested that rice germ oil can't reduce blood glucose concentration of non-insulin dependent diabetic mice compared with soybean oil. But we need to investigate the hypoglycemic effect of rice germ oil by changing supplementary level and period.

But the concentrations of serum total cholesterol and LDL-cholesterol were lower in RG group than in CO group. The hepatic total lipid and total cholesterol levels of RG group were significantly lower than those of CO group. The contents of total lipid, triglyceride and total cholesterol excreted in feces of RG group were higher than those of CO group. These results suggested that rice germ oil can reduce serum total cholesterol and LDL-cholesterol and hepatic total lipid concentration of non-insulin dependent diabetic mice compared with soybean oil due to increasing fecal lipid excretion. But we need to investigate the hypolipidemic effect of this oil by supplementary level and period.
Artemisia iwayomogi, a member of the Compositae, is a perennial herb easily found in Korea and used as a traditional medicine to treat liver disease. AIP1, a water-soluble carbohydrate fraction from Artemisia iwayomogi, showed anti-tumor and immuno-modulating activities in animal studies. A subacute toxicological evaluation of AIP1 was performed for 4 weeks in ICR mice. After administration of AIP1 (0, 20, 100, 500 mg/kg/day), the clinical signs, mortalities, body weight changes, hematology, blood clinical biochemistry, urinalysis, organ histopathology, organ weights and gross finding were examined. The results showed that there were no significant differences in body weight changes, food intakes, water consumptions, or organ weights among different dose groups. Also we observed no death and abnormal clinical signs during the experimental period. Between the groups orally treated with AIP1 and the control group, there was no statistical significance in hematological test or serum biochemical values. Histopathological examination showed no abnormal changes in AIP1 groups. These results suggest that no observed adverse effect level (NOAEL) of the oral administration of AIP1 for 4 weeks was considered to be more than 500 mg/kg/day in mice under the condition investigated in current study.
The aim of the present study was to assess the anti-obesity effects of grape seed extract (GSE) supplement in C57BL/6J mice. Thirty mice were divided into three groups; normal diet control group (ND), high fat diet control group (HD) and high fat diet plus grape seed extract supplemented group (HD+GSE). Results were as follows: 1. GSE supplement reduced the weight gain in mice fed high fat diets; epididymal and back fat weights, were lower compared to non-supplemented HD group. 2. Blood lipid concentrations were lower in the HD+GSE group than in the HD group. Serum HDL-C concentrations were higher in the HD+GSE group compared with the other groups. 3. The concentrations of acid-insoluble acylcarnitines, (AIAC) in serum and liver were higher in the HD+GSE group than in the HD group. 4. GSE supplementation increased mRNA levels of lipolytic genes such as carnitine palmitoyltransferase-l (CPT-1) and decreased mRNA levels of lipogenic genes such as acetyl CoA carboxylase (ACC). These findings suggest that grape seed extract supplements in high fat diet might normalize body weight, epididymal and back fat weights, lipid concentrations, and carnitine levels through controlling lipid metabolism.
Objectives: The study is aimed at evaluating the possible toxicity in 90-day repeated oral administration of modified Samjung-hwan (mSJH) in Sprague-Dawley (SD) rats. This study was conducted to detect the no-observed adverse effect level (NOAEL). Methods: Modified SJH extract was administered orally in male and female SD rats at dose of 0, 1,000, 2,000, 4,000 mg/kg. Each group consisted of 10 rats of each gender. The modified SJH extract was given once a day for 90 days. We monitored the changes of mortalities, clinical signs, body weight changes, food consumption, ophthalmologic findings, urine analysis, hematology, serum biochemistry, necropsy findings, organ weights, histological markers of all animals treated with modified SJH extract during the study period. Results: There were no toxicologically significant changes in mortalities, clinical signs, body weight gains, food consumption, ophthalmologic findings, urine analysis, hematology, serum biochemistry, necropsy findings, organ weights, histological markers in any of rats tested. Conclusions: The NOAEL of the modified SJH extract in male rats and no observed effect level (NOEL) in female rats are considered 4,000 mg/kg.
Hypogonadism is a clinical syndrome that results in hormone deficiency and can be classified as 1) primary caused by the gonadal failure and 2) secondary by the hypothalamus-pituitary gland dysfunction and/or cardiometabolic complications. Recently the presence of thyroid hormone receptors in different testicular cell types was demonstrated, and thus thyroid dysfunctions would be another cause of secondary hypogonadism. Thus, we investigated the effects of perinatal hypothyroidism on hypogonadism in male Sprague-Dawley rats. Perinatal hypothyroidism was induced by daily administration of 0.05% 6-propyl-2-thiouracil (PTU) by tap water from gestation day 15, which were compared with negative control (PTU (-)) group. At postnatal day 28, hypothyroid pups were divided into 2 groups: PTU (+) group - continued PTU treatment and PTU (+/-) group - stopped PTU until postnatal day 49. Body weights, dehydrotesosterone (DHT), and testosterone levels were checked 2 and 3 weeks after grouping. Body weights were significantly decreased in PTU(+) and PTU(+/-) groups compared with PTU (-) group at postnatal day 28. 3 weeks later, PTU (+/-) group significantly gained weight compared with PTU (+) group. DHT and testosterone levels significantly decreased with PTU treatment, but increased 3 weeks after stopping PTU administration. Perinatal PTU-induced hypothyroid hypogonadism was sustained for 2 weeks after stopping PTU administration, but restored gonadal hormone levels 3 weeks after stopping PTU. These results suggest that researchers should design an experiment on hypothyroid hypogonadism based on the estimated period.
