Time-course response of epichlorohydrin on epididymal histopathology in rats

  • This research aimed to investigate the time-course effect of epichlorohydrin (ECH) on epididymal histopathology in Sprague-Dawley rats. Twenty-four male rats were randomly assigned to four groups with 6 rats in each group and were administered a single oral dose of ECH (70 mg/kg) or its vehicle. Six animals each were sacrificed on days 0 (control), 1, 2, and 7 after treatment. During the study period, clinical signs, body weights, reproductive organ weights, testicular spermatid count, epididymal sperm count, motility and morphology, and histopathology were examined. No treatmentrelated effects on body weights and reproductive organ weights were noted at any time point. On the contrary, sperm motility decreased slightly on days 1 and 2 after treatment and then decreased significantly on day 7 after treatment. The first signs of histological changes were the appearance of cell debris in the ducts and vacuolization of the epithelial cells observed in the proximal caput epididymis on day 1 after treatment. The incidences and grades of the histological changes including cell debris in the ducts, epithelial vacuolization, oligospermia, and epithelial disruption increased on day 2 and then decreased slightly on day 7 after treatment. These results show that a single oral dose of 70 mg/kg ECH to male rats results in cell debris in the ducts and vacuolization of the epithelial cells in the proximal caput epididymis, followed by reversible oligospermia, epithelial disruption, and decreased sperm motility.

Growth Performance, Relative Meat and Organ Weights, Cecal Microflora, and Blood Characteristics in Broiler Chickens Fed Diets Containing Different Nutrient Density with or without Essential Oils

  • The present study was conducted to investigate whether dietary essential oils could affect growth performance, relative organ weights, cecal microflora, immune responses and blood profiles of broiler chickens fed on diets containing different nutrient densities. A total of eight hundred-forty 1-d-old male broiler chicks were randomly allotted into twenty-eight pens (7 pens per treatment, 30 chicks per pen). There were four experimental diets containing two different nutrient densities and supplemented with or without essential oils. Experimental period lasted for 35 days. No clear interaction between nutrient density and essential oils on any of growth performance-related parameters was observed. Live body weights were affected (p<0.05) by nutrient density at 21 days and by dietary essential oils at 35 days. Essential oils significantly (p<0.05) increased daily body weight gain and feed conversion ratio during the periods of 22 to 35 and 1 to 35 days, but failed to affect feed intake during the entire experimental period. Daily weight gain at 1 to 21 days and feed intake at 1 to 21 and 1 to 35 days were significantly impaired (p<0.05) by nutrient density. There were significant treatment interactions (p<0.05) on relative weights of bursa of Fabricius and abdominal fat contents. Finally, either essential oil or nutrient density did not influence the relative percentages of breast and leg meats, the population of cecal microflora, blood parameters and antibody titers against Newcastle disease and infectious bronchitis in broiler chickens. It was concluded that dietary essential oils, independent to nutrient density, failed to stimulate feed intake, but increased growth performance in broiler chickens.

건강체중아 및 비만아의 체중조절 경험, 식습관, 운동습관 및 자아존중감 비교 (Comparison of Weight Control Behavior and Self-esteem between Healthy Weight and Obese Children)

  • The study compared eating and physical activity related behaviors and self-esteem between healthy weight and obese children by presenting 175 primary school students in Busan City and Gyeongsang province with a self-reported questionnaire and Coopersmith's self-esteem inventory. The questionnaire was composed of 25 items, weighted primarily by a Likert scale. The self-esteem inventory presented to the students comprised 25 'Yes' or 'No' response questions to different statements. The study found obese children were more likely to think they always had to control their weight (p=0.000), reportedly measuring their weights significantly more than the healthy weight children. Also the study found that obese children are significantly more likely than healthy weight children to go on a diet, however neither group were successful in losing weight as the duration of the diet in 79.5% of the total sample lasted no longer than one week. In comparison to healthy weight children, obese children reported that they consumed fewer snacks during the day, avoided snacking subsequent to an evening meal and exercised more frequently for as long as physically possible. Interestingly, we found no difference of reported self-esteem between groups, though the obese group were more likely to answer that their parents did not understand them (p=0.055) Based on these findings, we concluded that the obese children who participated in the study were more aware of their body weights than the healthy weights children. It may be necessary to investigate further the relationship between self-esteem and participants' weights while considering other variables such as personality and body image.

1,4-Dichlorobutane의 랫드 2주 반복경구투여독성시험 (2-Week repeated oral dose toxicity study of 1,4-dichlorobutane in rats)

  • Objectives: The present study investigated the potential subacute toxicity of 1,4-dichlorobutane (1,4-DCB) by a 2-week repeated oral dose in male Sprague-Dawley rats. Materials and Methods: The test chemical was administered once daily by gavage to male rats at dose levels of 0, 74, 222, 667, and 2000 mg/kg/day for 2 weeks. All rats were sacrificed at the end of treatment period. During the test period, clinical signs, mortality, body weights, food and water consumption, urinalysis, hematology, serum biochemistry, gross findings, and organ weights were examined. Results: At 2000 mg/kg/day, treatment-related clinical signs, as evidenced by hypothermia, decreased locomotor activity, piloerection, lying on side, and prone position were observed. All the rats were found dead on test day 2. At 667 mg/kg/day, polyuria, suppressed body weight gain, food consumption, and spleen and thymus weights, and increased adrenal gland and liver weights were observed.Hematological and serum biochemical investigations revealed increases in the alanine aminotransferase, alkaline phosphataseand total bilirubinand decreases in the serum $Na^+$ level, white blood cell count and lymphocyte ratio. There were no treatment-related adverse effects in the 74 and 222 mg/kg/day groups. Conclusions: In the present experimental conditions, target organs were determined to be spleen, thymus,and liver. The no-observed-adverse-effect level was considered to be 222 mg/kg/day in male rats.

Adverse Effect of Nonylphenol on the Reproductive System in F1 Male Mice: A Subchronic Low-Dose Exposure Model

  • Nonylphenols (NPs) are widely used industrial materials, and are considered as potent endocrine disrupting chemical. Present study was undertaken to clarify the effect of subchronic low-dose NP exposure to F1 generation male mice. Mice were divided into 2 groups; (1) CON, control animals and (2) NP-50 ($50{\mu}g/L$), animals were treated with NP via drinking water. NP exposures were continuously conducted from parental pre-mating period until the postnatal day (PND) 55 of F1 offsprings. Mice were sacrificed on PND 55 and the tissue weights were measured. The initial body weights (at PND 21) and terminal body weights (PND 55) of the NP-50 animals were significantly lower than those of control animals (p<0.05). NP exposure induced a significant increase in the absolute weight of the testes (p<0.05). Conversely, the NP exposure caused significant decrease in the absolute weights of the epididymis (p<0.01), prostate (p<0.05) and seminal vesicle (p<0.05). Histopathological studies revealed that NP-treated animals exerted decreased seminiferous tubule diameters, reduced luminal area, and lower number of germ cells. Also some sloughing morphologies in the tubules were observed. In the caudal epididymis, fewer mature sperms and swollen epithelial cells were found in the NP-treated group. Our results confirmed that the subchronic low-dose NP exposure altered some male parameters and induced histopathological abnormalities in testis and epididymis of F1 mice. Since the NP dose used in this study is close to the average human daily NP exposure, our results could provide practically meaningful understanding of adverse effect of EDC in human.

Dexamethasone으로 유도한 근감소 동물모델에서 상황버섯-오미자박 고상발효 열수추출물의 근감소 개선에 대한 효과 (Effect of water extract Phellinus linteus-discard Schisandra chinensis solid fermented extracts in an Animal Model of Dexamethasone-Induced Muscle Loss)

  • Objectives : In this study, it was investigated the effects of solid-phase fermentation extraction with Phellinus linteus of discarded Schisandra chinensis extract (PS) and its action mechanism on dexamethasone-induced muscle atrophy in mice. Methods : In mice, muscle atrophy model was induced by dexamethasone (5 mg/kg, I.p) once daily for 2 weeks and with PS extract administration (100 and 300 mg/kg, p.o.) as treatment groups. The changes in body weights, grip strength, Treadmill test, muscle weights, and the expression of atrophy-related genes were measured in muscle atrophy mice. The histological changes of gastrocnemius tissues were also observed by H&E staining with measurement of myofiber size. Results : The administration of PS extract increased significantly body weights, grip strength, treadmill test and muscle weights in muscle atrophy mice. PS extract administration increased significantly the area of myofibers and inhibited structural damages of muscle and increased significantly the expression of myogenin and decreased significantly the expression of MuRF1, Atrogin1 and phosphorylation of AMPK and PGC1α in muscle tissues of muscle atrophy mice. Conclusions : These results indicate that PS extract has a improvement effects on muscle atrophy with stimulation of myogenic differentiation and inhibition of mRNA degradation that could be related with the activation of AMPK and PGC1α signaling pathways in muscle. This suggests that PS extract can apply to treat muscle atrophy in clinics.

Average Direct and Maternal Genetic Effects and Heterosis Effects on Body Weight in Two Subspecies of Mice

  • Data on body weights were analyzed in the four genetic groups from all possible crosses of two subspecies of mice to estimate average direct genetic effects (ADGE), average maternal genetic effects (AMGE) and heterotic effect (HE). The genetic groups used were $CF_{{\sharp}1}$ laboratory mouse (Mus musculus domesticus), Yonakuni wild mouse (Yk, Mus musculus molossinus yonakuni) and two reciprocal $F_1$ crosses of them, CY and YC. First symbol in the reciprocal $F_1$ represent subspecies of dam. Body weight at 1 (Wk1), 3 (Wk3), 6 (Wk6) and 10 weeks of age (Wk10) were analyzed from 258 mice of the four genetic groups. The model used to evaluate body weights included main effects of genetic group and sex, and interaction effect between genetic group and sex. The ADGE and the AMGE were estimated as deviations of Yk from $CF_{{\sharp}1}$. The HE was estimated from the differences between the reciprocal $F_1$ and the midparent mean. Results of this study showed that all effects, except sex and interaction between genetic group and sex at Wk1 and Wk3, were highly significant source variation (p < 0.01). The ADGE were positive and highly significant (p < 0.01) at all ages studied for both sexes, while the AMGE were highly significant at Wk3, Wk6 and Wk10. The ADGE were larger in contributing effect on body weight differences than the AMGE. The positive value of the HE were observed at all ages for males, while for females the positive effects occured from birth through weaning.

한방초콩환이 고지방식이를 급여한 흰쥐의 체중 및 지질 변화에 미치는 영향 (Effects of Mixed Pills of Chokong (Pickled Black Soybeans) with Medicinal Herbs on Body Weight Gain and Lipid Profiles in Rats Fed High-fat Diet)

  • Objectives : In this study we investigated the antiobese effects of mixed pills of pickled black soybeans with herbs(herbal Chokong pill, hereafter HCKP) in rats fed high-fat diet. It was evaluated by measuring the changes of body weight, adipose tissues weight and lipid profiles in serum. Methods : Black soybeans were pickled in vinegar for 15 days to prepare Chokong, at room temperature. For treatment group, HCKP was prepared, which five kinds of medicinal herbs(Rhynchosia nulubilis, Glycyrrhiza uralensis, Zizyphus vulgaris, Atractylodes macrocephala K, Astragalus membranaceus and Cornus officinalis) were added to dried Chokong. Four groups of male Sprague-Dawley rats were fed different diets during 9 weeks: normal diet containing 5%(w/w) com oil, high-fat diet containing 10%(w/w) lard plus 5%(w/w) corn oil (HF), high-fat diet supplemented with 1%(T1) and 5%(T5) HCKP powder, respectively. Results : The T5 group had markedly lower body weight gain and weights of epididymal adipose tissue when compared with HF group. There were significant differences in visceral adipose tissue weights, serum total cholesterol and triglyceride concentrations between the HF and T5 group. Then, the efficacy of powered HCKP on body weight and lipid profiles change in rats fed high-fat diet were induced dose dependantly. Conclusion : These results suggest that the possibility of HCKP, as an antiobese functional formula, by suppression of body weight gain and improved lipid profiles.

GROWTH AND COMPOSITION OF THE OMANI DHOFARI CATTLE 1. BODY WEIGHT GROWTH AND CARCASS COMPOSITION

  • A study was conducted which demonstrated that Dhofari cattle responded well to intensive management systems in terms of growth rates and carcass yields. Twenty-four Omani Dhofari bulls and steers (12 of each) were reared from birth until slaughter at 110, 160 and 210 kg body weight (for of each sex at each slaughter weight). Calves were fed ad libitum a diet of concentrate (16.5% CP) and Rhodesgrass hay (8.8% CP). Bulls and steers reached the predetermined slaughter weights of 110, 160 and 210 kg at 154 and 164; 219 and 233; 273 and 310 days of age, respectively. Respective mean pre and post-weaning daily body weight gains for bulls and steers were 581 and 530; 796 and 706 g averaging 645 and 596 g over 36 weeks. At 210 kg, the heaviest weight of the study, dressing-out percentage (DO) was 54.5 and 56.9 yielding carcasses of 115.9 and 118.5 kg which contained 60.4 and 61.5% muscle; 11.6 and 11.4% bone and 24.5 and 22.9% fat for bulls and steers, respectively. Proportion of bone in the carcass decreased, that of fat increased, whereas that of muscle remained unchanged between slaughter weights of 110 to 210 kg. That resulted in increased muscle : bone and decreased muscle : fat ratios. At 160 kg body weight, bulls had less fat and more muscle and bone than steers but there were no sex differences in carcass composition at 210 kg slaughter weight.

  • 본(本) 실험(實驗)은 thyroxine 및 propylthiouracil의 투여(投與)에 의(依)한 갑상선(甲狀腺)의 기능변화(機能變化)와 아울러 체성장(體成長)에 미치는 영향(影響)을 구명(究明)코저 실시(實施)하였는 바, 25일령(日齡)의 암 흰쥐를 갑상선척출군(甲狀腺剔出群), propylthiouracil 처리군(處理群), Thyroxine 처리군(處理群) 및 정상대조군(正常對照群)의 사개군(四個群)으로 나누어 배치(配置)하고 각군(各群)을 처리후(處理後) 시간경과(時間經過)에 따라 도살(屠殺)하여 갑상선(甲狀腺)의 중량측정(重量測定) 및 조직학적(組織學的) 검색(檢索)과 아울러 체중(體重)의 변화(變化)를 조사(調査)하였다. 1. 갑상선(甲狀腺)의 중량(重量)은 모든 관찰시간(觀察時間)에서 비교군간(比較群間)에 유의성(有意性)이 인정(認定)되었는데 PTU 처리군(處理群)은 정상대조군(正常對照群)에 비(比)하여 높은 중량(重量)을 나타냈고, Thyroxine 처리군(處理群)은 낮은 값을 나타냈다. 2. 갑상선(甲狀腺)의 조직상(組織像)은 PTU 처리군(處理群)에서는 처리(處理) 1주후(週後)부터 여포상피세포(濾胞上皮細胞)가 증식(增殖) 비대(肥大)를 동반(同伴)하는 원주화현상(圓柱化現象)을 나타내기 시작하여 실험기간(實驗期間)이 경과(經過)될 수록 더욱 진행(進行)되는 조직소견(組織所見)을 보였으며, Thyroxine 처리군(處理群)에서는 여포상피세포(濾胞上皮細胞)가 도평화(屠平化)되는 퇴행성(退行性) 변화(變化)가 인지(認知)되었다. 3. 체중(體重)은 처리(處理) 2주후(週後)부터 비교군간(比較群間)에 유의성(有意性)이 인정(認定)되는 변화(變化)를 보였는데 PTU 처리군(處理群)과 갑상선척출군(甲狀腺剔出群)은 정상대조군(正常對照群)에 비(比)하여 낮은 값을 나타냈고 Thyroxine 처리군(處理群)은 높은 체중치(體重値)를 보였다. 그러나, 다중검정결과(多重檢定結果) PTU 처리군(處理群)과 갑상선척출군(甲狀腺剔出群) 상호간(相互間)에는 유의성(有意性)이 인정(認定)되지 않았다.
