Antioxidant and Anti-Inflammatory Effects of Kamisipjeondaebotang in RAW 264.7 Cells (가미십전대보탕의 RAW 264.7 세포에서 항산화 및 항염증 효과)

  • As Kamisipjeondaebotang (KSD) extract is an herbal ingredient, safety is very important due to possible cell poisoning or heavy metal toxicity to organs when administered to humans or animals. Accordingly, this study examined the antioxidant and anti-inflammatory effects of KSD extract to confirm its medicinal safety by using RAW 264.7 cells after heavy metal screening, functional index test of the liver and kidney, and cell survival rate test. Heavy metals were not found in KSD extracts or were less than standard amounts. Liver function indices such as aspartate aminotransferase and alanine aminotransferase revealed low values and kidney function indices such as creatinine and blood urea nitrogen were not significantly different from the normal group. This proved the safety to the human. RAW 264.7 cells showed no poisoning compared to the control group in terms of survival rate. Regarding the antioxidant effect of KSD extract, 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity and 2,2'-azino-bis(3-ethylbenzothiazo-line-6-sulphonic acid) radical scavenging activity increased at concentrations over $10{\mu}g/mL$. The anti-inflammatory effect of KSD extract significantly decreased based on the amount of nitric oxide at concentrations of 10 and $100{\mu}g/mL$ compared to the control group. Expression of interleukin (IL)-$1{\beta}$ and IL-6 decreased in a concentration-dependent manner. There was no significant difference in tumor necrosis factor-${\alpha}$ level. Based on the results, KSD can be regarded as a safe antioxidant with anti-inflammatory effects for fracture treatment.

Effect of Dendropanax morbifera Pharmacopuncture on Sleep-related Hormones, Cognition in Psychosocial Stress-induced Rats (황칠(黃漆) 약침이 정신 스트레스를 받은 백서의 수면 관련 호르몬, 인지에 미치는 영향)

  • Objectives: The purpose of this study was to check the therapeutic effect of the Dendropanax morbifera Pharmacopuncture for insomnia and cognition caused by psychosocial stress. Methods: We separated Wistar rats (older than age 10 weeks, 300 g) into normal, control, positive control (Zolpidem administered, PC), and Dendropanax morbifera Pharmacopuncture experimental groups (DPA). Psychosocial stress such as circadian rhythm change, clammy straw, predator stress, restraint stress, noise bursts, and flashing lights were applied to the control, PC, DPA groups. The Dendropanax morbifera Pharmacopuncture group was given 0.5×, 1× and 2× amount concentration of Dendropanax morbifera Pharmacopuncture, respectively. The levels of Melatonin, Serotonin, Corticosterone, GABA, BDNF and CREB were measured, and the Y-maze test, weight and blood test were performed. Results: In all of the DPA groups, the Melatonin level showed no noticeable difference. In the DPA-2x group, Serotonin increased significantly. In all the experimental groups, Corticosterone decreased significantly and GABA showed increasing tendency. The DPA-1x and DPA-2x groups showed remarkable increase in BDNF and DPA-0.5x and the DPA-2x groups showed significant increase in CREB. The DPA-2x group showed remarkable increase in the Alternation behavior category of the Y-maze test. In all of the experimental groups the weight change showed increasing tendency, whereas no noticeable differences were found among all experimental groups regarding AST, ALT, BUN, Creatinine and CBC. Conclusions: Dendropanax morbifera Pharmacopuncture increases levels of serotonin and GABA, decreases corticosterone, increases levels of BDNF and CREB, and increases the ratio of alternation behavior in the Y-maze test. Thus, I suggest that Dendropanax morbifera Pharmacopuncture has the effect of treating insomnia caused by psychosocial stress, activating the brain, and improving cognition.

A Study of Carry Over Contamination in Chematology (이월오염에 대한 연구)

  • Carry over contamination has been reduced in some systems by flushing the internal and external surfaces of the sample probe with copious amount of diluent. It between specimens should be kept as small as possible. A built-in, continuous-flow wash reservoir, which allows the simultaneous washing of the interior and exterior of the syringe needles, addresses this issue. In addition, residual contamination can further be prevented through the use of efficient needle rinsing procedures. In discrete systems with disposable reaction vessels and measuring cuvets, any carry over is entirely caused by the pipetting system. In analyzers with reuseable cuvets or flow cells, carry over may arise at every point through which high samples pass sequentially. Therefore, disposable sample probe tips can eliminate both the contamination of one sample by another inside the probe and the carry over of in specimen into the specimen in the cup. The results of the applicative carry over experiment studied on 21 items for total protein (TP), albumin (ALB), total bilirubin (TB), alkaline phosphatase (ALP), aspratate aminotranferase (AST), alanine aminotranferase (ALT), gamma glutamyl transferase (GGT), creatinine kinase (CK), lactic dehydrogenase (LD), creatnine (CRE), blood urea nitrogen (BUN), uric acid (UA), total cholesterol (TC), triglyceride (TG), glucose (GLU), amylase (AMY), calcium (CA), inorganic phosphorus (IP), sodium (Na), potassium (K), chloride (CL) tests in chematology were as follows. Evaluation of process performance less than 1% in all tests was very good, but a percentage of ALB, TP, TB, ALP, CRE, UA, TC, GLU, AMY, IP, K, Na, and CL was 0%, implying no carry over. Other tests were ALT(-0.08%), GGT(-0.09%), CK(0.08%), LD(0.06%), BUN(0.12%), TG (-0.06%), and CA(0.89%).

Effect of New Remedies Mainly Comprised of Hovenia dulcis Thunb on Alcohol Degradation and Liver Protection in Sprague Dawley Male Rats (헛개나무 열매를 주성분으로 제조한 새로운 처방이 알코올 분해 및 간 손상에 미치는 영향)

  • We investigated whether two-kinds of mixture (NHDT-1 and NHDT-2) mainly composed of Hovenia dulcis Thunb had beneficial actions for alcohol and acetaldehyde degradation in acute alcohol treatment and liver protection from fatty liver induced by chronic alcohol administration. In acute alcohol degradation experiment, serum alcohol and acetaldehyde concentrations exhibited lower 1, 3 and 5 hours after taking 3 g ethanol per kg body weight in NHDT-1 treated rats, but not NHDT-2 including ginseng. On the contrast to the acute effect on alcohol degradation, the long-term alcohol administration revealed that NHDT-2, not NHDT-1, protected the increase in serum concentration of aspartate aminotransferase, alanine aminotrasferase and ${\gamma}-triglyceride$ metabolism similar to the rats not consuming alcohol, leading to decreased triglyceride accumulation in blood and liver. In liver morphological study, NHDT-1 preserved the regular hepatocyte morphology, decreased fat accumulation and reduced sinusoidal leukocyte infiltration in hepatocytes. In conclusion, NHDT-1 plays an important role in alcohol and acetaldehyde degradation without protecting liver damage while NHDT-2 works as hepatocyte protector from alcohol mediated damage.

Observational Study of the Treatment of 116 Cases of High Risk Group for Dementia and Depression in Multiple Korean Medicine Clinic (치매 및 우울증 고위험군 116례의 한의 치료에 대한 관찰 연구)

  • In order to reduce the social and economic costs due to the increase in dementia patients, which is a representative senile disease in an aging society, it will be important to prevent dementia and to detect and treat early in high-risk groups. This study reported the results of treatment for the elderly with high risk of dementia and depression who received Korean medicine (KM) treatment in 9 KM clinics. Medical charts were surveyed on 116 patients with high risk of dementia and depression who received KM treatment at 9 KM clinics in Gangseo-gu from September 1, 2020 to December 31, 2020. The majority of the patients were female(76.72%), the average age was 71.66±7.18 years old, and the average education level was 8.96±3.91 years. The average treatment period was 61.47±10.30 days, the average number of treatments was 15.38±1.06 times, and both acupuncture and herbal medicine were administered. After receiving KM treatment, cognitive assessment scores such as MMSE-DS and MoCA-K, depression-related GDSSF-K score, dementia knowledge, attitude, and preventive behavior, blood debilitation scale, and GQOL-D score were improved compared to before. There was no specific adverse reaction except that one patient with elevated AST and ALT levels was observed. This study showed improvement in cognitive function, depression, dementia-related scale, and quality of life in high-risk groups for dementia and depression over 60 years old who received KM treatment. More systematic and large-scale planned clinical studies will be needed.

Effects of hematological parameters and plasma components of olive flounder, Paralichthys olivaceus by acute nitrite exposure according to water temperature (수온별 아질산 급성 노출에 따른 넙치, Paralichthys olivaceus의 혈액학적 성상 및 혈장성분의 영향)

  • Olive flounder (Paralichthys olivaceus) (Weight 110.9±17.1 g, length 22.3±1.2 cm) were exposed to waterborne nitrite at 0, 30, 60, 120, 240, 480 and 960 mg NO2-/L according to water temperature at 20℃ and 25℃ for 96 hours. The lethal concentration 50 (LC50) of olive flounder, P. olivaceus exposed to waterborne nitrite was 513.87 mg NO2-/L at 20℃ and 208.35 mg NO2-/L at 25℃, which means a significant difference in LC50 by the water temperature. Hemoglobin and hematocrit were significantly decreased by waterborne nitrite exposure. The inorganic component, plasma calcium, was significantly decreased, and the organic components such as plasma glucose and cholesterol were significantly decreased showing a similar tendency with calcium. In enzymatic components, the AST and ALP were also significantly decreased by nitrite exposure. The results of this study indicate that exposure to nitrite can affect the survival and hematological physiology of P. olivaceus, and the effect of exposure to nitrite had a significant effect on nitrite toxicity depending on the water temperature.

Hepatotoxicity and nephrotoxicity of saponin-enriched extract of Asparagus cochinchinensis in ICR mice

  • The inhibitory effects of Asparagus cochinchinensis against inflammatory response induced by lipopolysaccharide (LPS), substance P and phthalic anhydride (PA) treatment were recently reported for some cell lines and animal models. To evaluate the hepatotoxicity and nephrotoxicity of A. cochinchinensis toward the livers and kidneys of ICR mice, alterations in related markers including body weight, organ weight, urine composition, liver pathology and kidney pathology were analyzed in male and female ICR mice after oral administration of 150, 300 and 600 mg/kg body weight/day saponin-enriched extract of A. cochinchinensis (SEAC) for 14 days. The saponin, total flavonoid and total phenol levels were found to be 57.2, 88.5 and 102.1 mg/g in SEAC, respectively, and the scavenging activity of SEAC gradually increased in a dose-dependent manner. Moreover, body and organ weight, clinical phenotypes, urine parameters and mice mortality did not differ between the vehicle and SEAC treated group. Furthermore, no significant alterations were measured in alkaline phosphatase (ALP), alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH), blood urea nitrogen (BUN) and the serum creatinine (Cr) in the SEAC treated group relative to the vehicle treated group. Moreover, the specific pathological features induced by most toxic compounds were not observed upon liver and kidney histological analysis. Overall, the results of the present study suggest that SEAC does not induce any specific toxicity in the livers and kidneys of male and female ICR mice at doses of 600 mg/kg body weight/day.

Effects of Steaming Process on Liriopis Tuber to Antioxidant Activities and Hyperlipidemia Induced Rats. (맥문동(麥門冬)의 증숙(蒸熟)에 따른 항산화 효능 및 고지혈증 유발 흰쥐에 대한 효능 연구)

  • Objectives : This study is aimed to compare the changes in Antioxidative capacity of Liriopis Tuber by steaming process and to compare the effects in hyperlipidemia induced rats fed high cholesterol diet between Simvastatin and Liriopis Tuber by steaming process. Methods : The SD rats were divided into six groups: normal diet (Nor), high cholesterol diet (Veh), high cholesterol diet plus Simvastatin 5 mg/kg (Sim), high cholesterol diet plus LT0 extract 200 mg/kg (LT0), high cholesterol diet plus LT6 extract 200 mg/kg (LT6) and high cholesterol diet plus LT9 extract 200 mg/kg (LT9). We compared the total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL), low density lipoprotein cholesterol (LDL) contents and reactive oxygen species (ROS) from each serums. Protein expression in liver tissues related to antioxidant and cholesterol was analyzed. Results : The Antioxidant activity of Liriopis Tuber increased by steaming process. In vivo, TC, TG, LDL-c, atherogenic index (AI) and cardiac risk factor (CRF) decreased and HDL-c increased with increasing steaming frequency. aspartate aminotransferase (AST), alanine aminotransferase (ALT), creatinine, and blood urea nitrogen (BUN) decreased with increasing steaming frequency. ROS decreased only in LT9, and SOD, catalase and glutathione peroxidase (GPx) increased with increasing steaming frequency. phospho-AMP-activated protein kinase (p-AMPK) increased and sterol regulatory element-binding protein 2 (SREBP-2), Phospho-Acetyl-CoA Carboxylase (p-ACC) and HMG-CoA reductase (HMGCR) decreased with increasing steaming frequency. Liver staining showed a decrease in hepatic fat accumulation of LT9. LT9 showed significant results in all experiments. Conclusions : LT9 showed significance of anti-lipid effect and improved fatty liver of hyperlipemia induced rats fed on high cholesterol diet, In conclusion, LP9 can be effectively used for the treatment of hyperlipidemia.

Effect of Red Ginseng with Processed Sulfur Extracts on Serum Lipids Concentration and Metabolic Variables in Diabetic Rats (홍삼의 법제유황 처리가 당뇨쥐의 혈중지질 및 대사지표물질에 미치는 영향)

  • Objectives : The purpose of this study was to evaluate effects of processed sulfur with red ginseng on streptozotocin(STZ) induced diabetic rats for expansion of processed sulfur internal application. Methods : We prepared red ginseng with non processed sulfur extracts(GS), red ginseng with processed sulfur I extracts(GPS I) and red ginseng with processed sulfur II extracts(GPS II). In the present study, we examined about contents of crdue saponin, antioxidant activity, $\alpha$-glucosidase inhibitory activity, and effects of STZ induced diabetic rats. Results : Contents of crude saponin increased by processed sulfur, and GPS II was shown highest contents in crude saponin and sulfur compared with another groups. Electron donating ability of GPS II was shown highest activity compared with GS and GPS I, SOD-like activity showed same tendency as electron donating ability at 1 $mg/m\ell$ concentration. Inhibitory activity of $\alpha$-glucosidase was approximately same level in acarbose and GPS II. Blood glucose level of GPS II group was decreased 18.34% compared with DC(diabetes control) group and maintained stability range in glucose level. but GS and GPS I showed high level compared to GPS II. Serum triglycerides concentration also showed lowest level in GPS II. The activity of ALT, AST and ALP was shown high level in diabetic induced groups, and lowest level in GPS II. Creatinine was shown non-significantly difference in each groups and GPS II was shown lowest level in BUN. Conclusions : These results suggested that processed sulfur with red ginseng have improvement effects on diabetes and internal application of processed sulfur with red ginseng have no specific toxicity in liver and kidney.

Development of electrochemical biosensor for determination of galactose (4갈락토오즈 측정을 위한 전기화학적 바이오센서 개발)

  • In principle, the blood galactose level may be determined conveniently with a strip-type biosensor similar to that for glucose. In this study, we describe the development of a disposable galactose biosensor strip for point-of-care testing. The sensor strip is constructed with screen-printed carbon paste electrode (SPCE) and sample amount (< $100{\mu}L$). The developed strip the galactose level in less than 90 s using bienzymatic system of galactose oxidase (GAO) and horseradish peroxidase (HRP). The effects of pH, mediator (1,1-ferrocenedimethanol) concentration, ratio of enzymes, and applied potential were determined preliminarily with glassy carbon electrodes, and optimized further with the strip-type electrodes. The sensor exhibits linear response in the range of $0{\sim}400{\mu}M$ ($r^2$ = 0.997, S/N = 3). Since a low working potential, in principle, the fabricated disposable galactose biosensor has -100 mV (vs. Ag/AgCl), it is applied for the detection of galactose, interfering responses from common interferents such as ascorbic acid, uric acid and acetaminophen could be minimized. The sensor has been used to determine the total galactose level in standard samples with satisfactory reproducibility (CV = 5 %).
