Effect of Kamicheonggan-tang on Pre-hepatocarcinogenesis Induced by N-nitrosomorpholine (가미청간탕이 N-nitrosomorpholine으로 유도된 전암성 간병변에 미치는 영향)

  • The purpose of this study was to evaluate the protective effect of Kamicheonggan-tang(KCGT) on pre-hepatocarcinogenesis induced by N-nitrosomorpholine. The studied using blood chemistry, lipidperoxidation, antioxidant, immunohistochemistry and morphological change. The results were obtained as follows. In the pre-hepatocarcinogenesis induced by NMP, serum AST, ALT, ALP and total bilirubin were not changed in NMP and NK treated group after 1 st week, but desreased in NK treated group after 4th week as compared with NMP treated 4th week group. The content of GSH was similary to in NK treated groups as compares with data of normal group. The content of MDA was increased in NK treated group after 1st and 4th week, and more increased in the NMP treated group than those of their NK treated group. The immunohistochemically, stain of GST-p, positive lesions of KCGT were significantly decreased than those of NMP treated group. The histopathologically, fat changes, nucleotic changes, oval cell and inflammatory cells in periportal were observed in NMP treated fater 4th week, but those were significantly decreased from 4th week in the NK treated group. And the enlarged nucleus was not changed in KCGT treated group, but increased in NMP treated group after 1st and 4th week. The ultrastructurally, nucleotic changes, glycogen degeneration, lipid droplet and rER fragmentation were observed in NMP treated group after 4th week, but those changes were significantly decreased from 4th week in the NK treated group. These results suggested that KCGT extracts has protective effect on prehepatocarcinogenesis by NMP, might be usefully applied for clinical treatment of hapatic disease and also it was necessary to do more studies about its mechanisms.

A study on the effect on obesity and lipid metabolism in liver hypofunction animal-experimental model induced by Acetaminophen(AAP) injection (Acetaminophen(AAP)으로 유발한 간기능 저하 동물 모델에서의 비만 및 지질대사에 대한 영향 평가)

  • Objectives: The objective of this research is to develop new animal-experimental model for Sasang Constitutional Medicine, especially for partial Taeyangin(one of four constitution which has good pulmonary function and poor hepatic function) by AAP intraperitoneal injection, and to estimate from the viewpoint of obesity and lipid metabolism. Methods: The C57bl/6J mice was divided into 4 groups ; Normal group, AAP group, High-Fat-Diet(HFD) group, and HFD+AAP group. 200mg AAP was injected intraperitoneally to the AAP group twice a week for six weeks, and HFD group was fed with 60%-High-fat Diet for six weeks. HFD+AAP group got both AAP injection and 60%-High-fat Diet at the same time for the same period. In this period, We measured the weight and Food Efficiency Ratio(FER, %) once a week. After six weeks, We conducted the blood chemical test from the groups, and extracted the fat tissue to measure weight. Results & conclusion: In the liver function test, two AAP groups had higher AST and ALP, and normal LDH. The blood level of creatinine from all groups were normal. The rate in weight was lesser by 7.8% in HFD+AAP group, and had lesser FER than HFD group. Also They had lesser Total cholesterol and LDL cholesterol, and had more HDL cholesterol than HFD group. HFD+AAP group hadmore glucose in serum and lesser Insulin-like Growth Factor 1(IGF-1) than HFD group.

A Clinical Study of Shi Ho Cheong Gan-San on Blood Heat Pattern Atopic Dermatitis: a randomized, double-blind clinical trial (혈열형 아토피피부염에 대한 시호청간산의 유효성과 안전성 연구)

  • 목적 : 본 임상시험을 통하여 혈열(血熱)형으로 변증된 아토피피부염의 한약 치료 후 임상 효능과 안전성을 관찰함으로써 아토피피부염에 대한 한약치료의 유용성을 평가하고자 한다. 방법 : 본 임상시험은 무작위배정, 이중맹검, 양성대조군, 평행 설계로 진행되었다. 자의에 의해 임상시험 동의서에 서명한 대상자 중 선정기준 및 제외기준에 부합된 36명의 대상자들에게 시험약과 양성대조약 과립제를 1일 3회(5.0g* 3회/ 1일) 4주간 복용하도록 하였다. 36명 중 31명의 환자가 4주간의 치료를 종료하였다(시호청간산 복용군: n = 16, 소풍산 복용군: n =15). 아토피피부염 증상을 평가하기 위해 Scoring atopic dermatitis (SCORAD) index와 Eczemaarea and severity index (EASI) 를 사용하여 시험 시작 전과 4주 후 시험 종료일에 환자들의 피부소견을 평가하였다. 아토피 피부염에 대한 한약치료의 안정성을 평가하기 위하여 치료 전후 혈중 AST, ALT, BUN, creatinine 변화를 검토하여 한약치료의 간/신독성 여부에 대해 조사하였다. 결과 및 결론: 두 군 모두에서 유의하게 SCORAD 점수와 EASI 점수가 감소하였으나 두 군 간의 차이는 유의하지 않았다. 부종/구진, 찰상 항목의 점수에는 두 군 간에 유의한 차이가 관찰되었다. 31명의 환자에게서 한약 치료 후 간장과 신장에 미치는 독성이 발견되지 않았으며 한약투여와 관련된 이상반응은 보고되지 않았다. 위의 결과로부터 시호청간산과 소풍산이 혈열형 아토피피부염에 대한 효과적이고 안전한 치료 방법이 될 수 있을 것으로 생각된다.

Expression and Clinical Significance of Myeloid Derived Suppressor Cells in Chronic Hepatitis B Patients

  • We here document discovery of expression profile of myeloid derived suppressor cells (MDSCs) in chronic hepatitis B (CHB) patients and changes in the course of disease. The study population was composed of 75 outpatient HBV cases and 15 healthy control cases. Peripheral blood samples were collected for separation of mononuclear cells. Levels of MDSCs labeled with Lin-DR-CD11b+CD33+ obtained from peripheral blood mononuclear cells (PBMC), were revealed to have significant differences between the CHB and other groups. They were 0.414% for health control cases and 0.226% for CHB cases (Z=-2.356, p=0.0189). It also observed that the group of HBeAg positive cases had significant difference in MDSCs/PBMC median ($X^2=11.877$, p=0.003), compared with group of HBeAg negative cases and the healthy control group. It suggested considerable MDSCs might be involved in HBeAg immune tolerance. In addition, negative correlations between MDSCs/PBMC and parameters of ALT, AST and TBil, while positive correlation between MDSCs/PBMC and ALB parameter were found. Multiple comparisons between the four phases and health control phase again, there was a statistically sifnificant difference ($X^2=17.198$, p=0.002). Taken together, these findings may provide a new immunotherapy strategy for reduced the expression levels of MDSCs in CHB patients, through induction of an autoimmune response to virus removal.

Hotwater Extract of Hovenia dulcis Peduncle Improves Exercise Performance and Anti-fatigue Effect in Mice (헛개나무 열매 열수추출물 투여에 의한 생쥐의 지구력 운동 수행 능력 향상 및 피로개선 효과)

  • The aim of this study was to evaluate the effect of hot water extract of peduncle obtained from Hovenia dulcis Thunb (HD) which is commercially developed for the protective effect on the alcoholic hepatotoxicity, on the endurance capacity for weight loaded forced swimming mice. The swimming times to exhaustion in mice fed 100 and 200 mg/kg HD for 2 weeks were prolonged 3.6 and 3.7 fold, and for 4 weeks 1.9 and 2.7 fold compared with each vehicle control ($42.8{\pm}20.5$ min and $67.7{\pm}47.8$ min, for 2 and 4 weeks), respectively. Blood biochemical parameters for ALT, AST, creatinine and BUN were not significantly different between from HD fed or control mice. Although HD fed mice swam over 2 fold longer time than vehicle control mice at 4 weeks, blood biomarkers of physical fatigue such as glucose, triglyceride and free fatty acid, lactate were not significant different and even tended to ameliorate. Hepatic lipid peroxidation and SOD activity did not significantly change in HD fed- and vehicle control exhausted swimming mice at 2 or 4 weeks. However, catalase activity in HD-fed mice was significantly increased in a dose-dependent manner compared with vehicle control mice. The present study indicates that HD improved physical fatigue and exercise performance in mice. Therefore, it has a potential for the pharmacological effect of anti-fatigue.

Effects of Starvation on Growth and Physiological Response in Cultured Catfish, Silurus asotus (기아에 따른 양식 메기, Silurus asotus의 성장 및 생리적 반응)

  • An experiment was conducted for 80 days to determine the effect of feeding and starvation on the survival, growth, gonad-somatic index (GSI), hepato-somatic index (HSI), viscera index (LSI), the morphometric changes and hematological parameter in a cultured catfish, Silurus asotus. The survival and growth of the starved group was lower than those of the fed group during the experiment. Starvation resulted in retardation of growth, which provides examples of the reduction of final body weight, growth rate of body weight, specific growth rate and condition factor, whereas the fed group continued to grow and remained in good condition. Blood analysis (cortisol, glucose, aspartate aminotransferase, alanine aminotransferase, red blood cell and hemoglobin) showed significant differences between the fed and starved groups at the end of the experiment. These results suggest that all nutritional parameters used for starvation and feeding in this study appear to be a useful index of nutritional status in the cultured catfish.

Prognostic Significance of Basic Laboratory Methods in Non-Small-Cell-Lung Cancer

  • Background: In our study, the LDH, albumin, hemoglobin, neutrophile, thrombocyte, lymphocyte counts and prognostic significance of neutrophile-lymphocyte and thrombocyte-lymphocyte ratios in NSCLC derived from these counts obtained during regular examinations of patients were examined. Materials and Methods: Histopathologically diagnosed non-small-cell-lung cancer patients between 2008 and 2010 were included in the study. Before the treatment, full blood count including routine lymphocyte count, blood biochemistry examinations including liver (AST, ALT, total protein, Albumin), LDH and kidney (BUN, Cre) function tests were performed. Results: A total of 156 patients, 76 of whom (48.7%) were female and 80 of whom (51.3%) were male were included. Mean hemoglobin level was determined as 12. Overall survival was found to be significantly dependent on whether patients were anemic or not (p: 0.005). Mean LDH level was determined as 233.4. There was nosurvival difference between patients with and without high LDH (p: 0.532). In patients where NLR showed systemic inflammatory response, overall survival was 10.8 months whereas this duration was 19.6 months in patients where the systemic inflammatory response was negative (p: 0.012). In patients where TLR showed systemic inflammatory response, overall survival was 13.6 months whereas this duration was 21.9 months in patients where the systemic inflammatory response was negative (p: 0.04). Conclusions: Molecular methods have been changing rapidly in today's world and they manage the treatment besides defining the prognosis of patients. However, easily accessible and cheap laboratory parameters should be considered in the prognosis of patients besides these new methods.

Protective Effect of Panax ginseng on TCDD-induced Liver Toxicity in Male Guinea Pigs

  • This study was carried out to investigate the preventive and therapeutic effect of Panax ginseng water extract (PG-WE) on the toxicity induced by 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), one of the most notorious toxic environmental pollutants belonging to the group of polyhalogenated aromatic hydrocarbons. Normal control (NC) group guinea pigs (180~200 g) received vehicle and saline, and TCDD-treated (TT) group was given TCDD and saline. P100 and P200 group animals received PG-WE for 28 days since 1 week before TCDD exposure at daily doses of 100 mg/kg b.w. and/or 200 mg/kg b.w., respectively. C100 and C200 group received PG-WE for 14 days starting 1 week after TCDD-exposure. Toxicity was induced by a single intraperitoneal injection of TCDD (1 $\mu\textrm{g}$/kg b.w.). Abnormal increase in AST and ALT activities in TT group was significantly improved by the administration of PC-WE. Microscopically, there were mild to moderate swelling of hepatocytes, hyperchromatism of individual cells, acidophilic cytoplasm and cytoplasm vacuolation of some hepatocytes, slight to moderate variations of staining density, occasional single cell necrosis, variable size and shape of some hepatocytes, small groups of degenerating hepatocytes surrounded by mononuclear cells, dilated sinusoids of centrilobular zone and some loss of lobular architecture in TT group liver. From these results, we could find the protective and therapeutic role of PG-WE in TCDD-induced liver toxicity by examining the blood chemical parameters and histopathological observation.

Effects of the Integrated Obesity Management Program, which Includes Exercise Therapy, Psychotherapy and Dietary Treatment, on the Body Composition and Blood Composition of Obese Patients Worked in the Industry (운동치료, 심리치료 및 식이요법이 포함된 통합비만관리프로그램이 산업체에 근무하는 비만 환자의 체성분 및 혈액성분에 미치는 영향)

  • Purpose : The purpose of this study was to find out how changes in body shape of obese patients could be affected by the combined management of diet and psychotherapy along with exercise therapy to control energy imbalance, the cause of obesity. Methods : In this study, the aerobic exercise program was conducted on 12 industrial workers of "D" company for a total of eight weeks for obese patients with a high body mass index (25 kg/㎡ or higher). The experimental group did not apply the program three times a week. The body type analysis was carried out using the body analyzer (Inbody 770, Inbody, Korea) and blood test and psychological test were performed after 8 weeks of exercise before and after exercise. Results : The results of the group showed a change in weight loss of 6.03 kg (p=.000) on average, which is nearly 10 % of the change in weight. It was also shown that the BMI (body mass index) decreased by 1.76 kg/㎡ (p=.000). Abdominal fat (AO) is also 0.14 % (p=.000), waist circumference (WC, 12.72 cm, p=.000), internal fat (VF, 25.12 %, p=.000), liver function levels AST (5.47 U/L, p=.04), ALT (13.64 U/L, p=.000), total cholesterol, pTC -14.22. Conclusion : Based on the results of this study, the obesity management program will be able to maximize the effects of obesity control if exercise programs and psychotherapy are combined.

Effects of Extracts from Mixed Culture with Tricholoma Matsutake Mycelium and Cordyceps Militaris Mycelium on Blood Glucose in Streptozotocin-Induced Diabetic Rats (송이버섯과 동충하초 균사체 혼합배양 추출물의 투여가 Streptozotocin으로 유발한 당뇨 쥐의 혈당에 미치는 영향)

  • This study was performed to investigate the influence of extracts from mixed culture with Tricholoma matsutake mycelium and Cordyceps militaris mycelium on hypoglycemia in streptozotocin-induced diabetic rats. Experimental animals were divided into 7 groups : normal control group(NC), diabetes control group (DC), positive control group(PC), non-fermented OCM(oriental medicine & cereal medium) extracts group (UM), crude polysaccharide of non-fermented OCM extracts group (UME), fermented OCM extracts group (UF), crude polysaccharide of fermented OCM extracts group (UFE). NC, DC groups were orally administered saline, PC group was orally administered acarbose. UM, UME, UF, UFE groups were orally administered each extract once a day for 14days. Blood glucose level was lower in the all administering OCM extract groups (UM, UME, UF, UFE) than in the diabetes group(p<0.05), and specially UF, UFE groups were similar to tendency of PC group. ALT, ALP activity in OCM groups were not significantly lowered than PC group(p<0.05). AST activity was not different with PC group. The results of this study show that extracts from mixed culture with Tricholoma matsutake mycelium and Cordyceps militaris mycelium may have a beneficial effect on the hypoglycemia.
