꾸지뽕나무 근피의 항당뇨병 효과 (Hypoglycemic Effect of Cudrania tricuspidata Root Bark)

  • The present study was undertaken to elucidate the hypoglycemic effect and inhibitory effect of Cudrania tricuspidata root bark on aldose reductase activity. C. tricuspidata MeOH ext. 1,000 mg/kg showed a significant blood glucose lowering effect on alloxan-induced hyperglycemic rats and increasing body weight. C. tricuspidata MeOH ext. showed a potent inhibitory effect on bovine lens aldose reductase.

당뇨쥐에서 다시마와 Metformin의 급여가 지질과산화물과 항산화영양소 함량에 미치는 영향 (Effect of Sea Tangle and Metformin on Lipid Peroxide and Antioxidants Levels in Diabetic Rats)

  • The present study was conducted to investigate te effect of sea tangle and hypoglycemic agent(Metformin) on lipid peroxidation and antioxidants levels in normal and diabetic rats. Male Sprague-Dawley rats were fed AIN-76 based experimental diets containing 5% (w/w) cellulose or 15%(w/w) sea tangle for 3 weeks, and then rats of diabetic groups were rendered diabetic by intramuscular injection of streptozotocin(STZ, 45mg/kg BW). After induction of diabetes Metformin(350mg/kg BW) was given once a day using a feeding tube for 5 days. Blood glucose level in diabetic rats fed sea tangle was significantly lower than that of diabetic rats fed cellulose. Metformin feeding resulted in further lowering blood glucose. Plasma and liver microsomal levels of lipid peroxides were increased significantly in diabetic rats as compared to normal rats, and the plasma level tended to be decreased by sea tangle feeding. Plasma level of retinol was reduced by STZ treatment, but it was increased by Metformin feeding in diabetic rats fed sea tangle. The liver contents of retinyl plamitate were reduced in diabetic rats but recovered by sea tangle feeding to some extent. Liver contents of total vitamin A were increased significantly by sea tangle in diabetic rats. Although difference in either plasma or liver level of $\alpha$-tocopherol by diet and STZ treatment was not significant, $\alpha$-tocopherol levels were the highest in the group with simultaneous feeding of sea tangle and metformin. Liver contents of zinc and copper were not influenced by either STZ treatment or sea tangle feeding. The present study indicates that the lowering blood glucose level could be attained by simultaneous trial of sea tangle diet and hypoglycemic agent and the increased oxidative stress caused by STZz treatment could be relieved by sea tangle feeding.

김천산 자두가 Streptozotocin 당뇨 유발 쥐에 미치는 영향 (Effect of Plums Produced in Gimcheon on Metabolism in Streptozotocin-induced Diabetic Rats)

  • This study examined the effects of plums produced in Gimcheon area on the glucose and lipid metabolism in streptozotocin-induced diabetic rats. Male Sprague-Dawley diabetic rats were divided into four groups: control, diabetic control, Diabetes-low plum (containing 10% plum powder), and Diabetes-high plum (containing 20% plum powder). The animals were fed isocaloric experimental diets based on an AIN-76 diet for 6 weeks. Feed efficiency ratio (FER) of the diabetic groups were significantly lower than that of the control. On the other hand, among the diabetic groups, the FER of the high plum intake group was higher than that of the diabetic control. The liver weight per 100 g body weight of each group was similar but the liver weights tended to decrease as the amount of plum intake was increased. Kidney weight per 100 g body weight of the plum intake groups were significantly different compared to that of the diabetic control. The supplementation of plums lowered the fasting blood glucose level of the diabetic groups and improved the glucose tolerance, thereby lowering the glycosylated hemoglobin index. In addition, the supplementation of plum was lowered the blood total cholesterol concentration and increased the HDL-C/TC (%) significantly, thereby lowering the atherosclerotic index (AI) and hepatic peroxide level. A steady diet of plums produced in Gimcheon may be effective in controlling the blood glucose level and preventing chronic diabetes mellitus.

보건소 중식실습교육이 제 2 형 당뇨병환자의 혈당개선에 미치는 영향 (Group Lunch Visits at the Public Health Center Improve Glycemic Control in Older Adults with Type 2 Diabetes Mellitus)

  • This study was carried out to investigate the effect of nutrition education program for diabetic patients on the glycemic control at the public health center. The study subjects, aged 61.7 $\pm$ 9.4 years, were 93 sex-and age-matched patients with type 2 diabetes mellitus. They were divided into three groups: nutrition education & diet practice group (EDG), nutrition education-only group (EG), and the control group (CG). Height, weight, and the postprandial 2 hour blood glucose (PP2) were measured at baseline, and 4, 6 and 8 week after the diabetic nutrition education program. At baseline there were no differences in height, weight, and blood glucose levels among the three groups. Nutrition education programs, especially that with group lunch practice sessions were found to be effective in lowering the blood glucose levels in patients with NIDDM patients. At 4 week blood glucose levels were decreased by 40.6% and 19.6% in EDG and EG, respectively, which was further dropped by 50.2% and 35.1% at 8 week, as compared to the CG group. For the EDG group, the total energy intake, which was 162.3% of the prescription before the diet counselling session, was decreased to 113.6% of the prescription after the lunch visit, with most decrease coming from the reduction in carbohydrate and fat intake. Multiple stepwise regression analysis revealed that the total energy intake explained 47.9% and 57% of blood glucose changes for men and women, respectively, and that percent energy intake from protein explained 15.8% for women. These results demonstrate that the public health center nutrition education programs for diabetic patients, especially that with group lunch practice sessions are very effective for the glycemic control in patients with diabetes mellitus.

db/db 마우스에서 상백피탕의 혈당강하 활성 및 기전연구 (Blood Glucose Lowering Activity and Mechanism of Sangbackpitang (SBPT) in db/db Mouse)

  • Antidiabetic activity and mechanism of Sangbackpitang (SBPT) was examined in db/db mice, which is a spontaneously hyperglycemic, hyperinsulinemic and obese animal model. SBPT and acarbose were administered orally for 4 weeks. Fasting and non-fasting serum glucose, glycated hemoglobin and triglyceride were all reduced when compared between db/db control group and SBPT treated group. At 12th week after birth, SBPT increased an insulin secretion although statistic significance was not seen. Total activities of sucrase, maltase and lactase in SBPT treated group were all decreased when compared to db/db control. On the other hand, sucrase and maltase activities in acarbose treated groups were increased. Effect of SBPT on mRNA expression of glucose transporter(GLUT-4) was also examined. Quantitation of glucose transporter was performed by RT-PCR and in vitro transcription with co-amplification of rat-action gene as an internal standard. Muscular GLUT-4 mRNA expression in SBPT treated group was increased significantly. These results may suggest that SBPT lowered blood glucose ascribing to inhibition of glycosidase-catalyzed reaction and upregulation of muscular GLUT-4 mRNA expression.

수종의 한약재가 Streptozotocin으로 유발된 고혈당 생쥐에 미치는 영향 (The Effects of Several Herbal Medicine Concentrated Solution on the Hyperglycemic Mice Induced with Streptozotocin)

  • Objective : This study has been carried out to understand the effect of several herbal medicine concentrated solution on the hyperglycemic mice Induced with streptozotocin(STZ). Methods : The 60 mg/kg of STZ injection into mice twice by 24 h interval and then 120 mg/kg of STZ injection again 3 days after the earlier injection. Control group was subjected to natural recovery, however, treated groups were fed 0.2 ml of several herbal medicine-concentrated solution (PA (x2, several herbal medicine-concentrated solution 1 group); PB (x4 several herbal medicine-concentrated solution 2 group) daily for 6 weeks. Result : The weight of PA was higher than that of control, but weight of PB was lower than control. The blood level of control increased continuously, reaching to 350mg/dL after 6 weeks, however, PA and PB showed a fast reduction of blood glucose. In blood glucose tolerance test, PA and PB showed better resistance than control. The GOT level in significantly(p<0.05) decreased in PA and PB compared with control group. The BUN and creatinine levels are significantly(p<0.01) decreased in PA compared with control group. Feeding of several herbal medicine-concentrated solution in a concentration of PA had an efficient effect on regeneration or recovery of Langerhans islet and ${\beta}-cell$ damaged by STZ. More Langerhans islet and high insulin-immunohistochemical resistance were observed in PA compared with control, but they were higher in PB than in PA. The number of Langerhans islet ${\beta}-cell$ and Langerhans islet. Conclusions : The result from the six weeks of observation demonstrates that the several herbal medicine concentrated solution have a positive effect of lowering the level of blood sugar and they increased insulin concentration. They have an effect for recovery of pancreas tissue and recovery of kidney, liver function from a diabetes mellitus.

제2형 당뇨병 환자에서 혈당강하제와 보중익기탕 병용 투여가 혈당 변화에 미치는 영향 및 안전성 연구 (A Retrospective Study of the Safety and Effect of Co-administration of Glucose-lowering Medication and Bojungikgi-tang on Blood Glucose Level in Patients with Type 2 Diabetes Mellitus)

  • Objective: This study aimed to assess the safety and effect on glucose level of Bojungikgi-tang in patients with type 2 diabetes mellitus. Methods: To review patients' clinical characteristics and laboratory tests retrospectively, we investigated 15 hospitalized patients with type 2 diabetes mellitus who took Bojungikgi-tang at Kyung Hee University Korean Medical Hospital for at least one day between January 2012 and December 2022. The blood glucose levels, including fasting blood sugar (FBS), 2-hour postprandial glucose (PP2) levels, and glycated hemoglobin level, were collected to determine the effect of the Bojungikgi-tang on blood sugar changes. Furthermore, to evaluate the safety of Bojungikgi-tang, hepatic function and renal function tests were implemented, including aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, γ-glutamyltransferase, blood urea nitrogen, creatinine, and Modification of Diet in Renal Disease estimated glomerular filtration rate. Results: When Bojungikgi-tang and a standard treatment for diabetes were administered for patients with type 2 diabetes, it was confirmed that there were no statistically significant changes in FBS and PP2 levels in the analysis of each medication taken. There was no significant difference in the safety profile after taking Bojungikgi-tang. Conclusions: The combined administration of Bojungikgi-tang with standard hypoglycemic medication for patients with type 2 diabetes may not affect blood glucose levels and safety.

직장인의 만성질환 위험 요소별로 구성된 영양 교육의 효과 비교 연구 (Effects of Workplace Nutrition Education Program Tailored for the Individual Chronic Disease Risks)

  • The incidence of chronic disease is continuously increasing in Korea. Especially, office workers have higher risk of chronic disease because of their dietary habit and lifestyle. The study aimed to investigate the effect of tailored nutrition counseling on improving chronic disease risk factors. Ninety-nine male workers (age $46.9{\pm}7.0$ yrs) volunteered for 12 weeks of nutrition program containing dietary intake and physical activity adjustment. Five individualized programs were performed with the main theme of weight loss (WL, n = 16), blood pressure lowering (BL, n = 34), normalizing blood glucose (GL, n = 21), lipid lowering (LL, n = 13) and reducing MS risk factors (ML, n = 15). Anthropometric data, blood-pressure, self-reported questionnaire, blood profiles were measured before and after 12weeks of nutrition education. The education program included 5 times of 1:1 interview. Compared to 0 week, anthropometric data (weight, BMI, fat, visceral fat, waist, SBP, DBP) were significantly decreased after 12 weeks (p < 0.001). Fasting blood glucose and total cholesterol were decreased (p < 0.05). The primary outcomes with individually tailored programs showed to be more effective than one general nutrition program.

Heat Stability and Glucose-Lowering Effect of 1-Deoxynojirimycin from Silkworm (Bombyx mori) extract Powder

  • Silkworm powder, which contains 1-deoxynojirimycin (DNJ), is a promising complementary and alternative medicine (CAM) in Korea. Silkworm powder was produced from Yeonnokjam pupae at d 3 of the 5th instar at the National Academy of Agricultural Science. The powder was derivatized with 9-fluorenylmethyl chloroformate (FMOC-Cl), and the DNJ-FMOC content was measured by HPLC. We investigated the content of 1-DNJ in the silkworm powder and its glucose-lowering effect when it was treated at different temperatures. The content of 1-DNJ was the lowest at $150^{\circ}C$, while it was constant at other temperatures. The silkworm extract powder was orally administered to diabetic mice (20 mg/kg/d) for 4 wk. Water intake did not significantly change when compared with the control group (T0). The blood glucose levels significantly decreased when mice where administered silkworm powder treated at $60^{\circ}C$ (T60) compared to the control group, but no difference was observed between the groups T100 and T150. Moreover, the blood levels of TG significantly decreased compared with the control group. Based on these results, we surmise that the properties of the silkworm extract powder were stable upon heating at $100^{\circ}C$ but not at $150^{\circ}C$.

Inhibition of GLUT-1 Expressed in Xenopus laevis Oocytes by Acetoxyscirpendiol of Paecilomyces tenuipes

  • Paecilomyces tenuipes, a caterpillar fungus, contains many health-promoting ingredients. Recent reports indicate that consumption of P. tenuipes helps reducing blood sugar content for diabetes. Mechanism for reduction in the circulatory sugar content, however, still remains least understood. Methanolic extraction of P. tenuipes (MPT) was prepared and acetoxyscirpendiol (ASD) was subsequently purified limn MPT. Glucose transporter-1 (GLUT-1) was expressed in the Xenopus oocytes and the effect of MPT or ASD on the expressed GLUT-1 was analyzed according to the uptake of 2-dideoxy-D-glucose (2-DOG). MPT was shown to inhibit GLUT-1 activity significant1y compared to the non-treated control. In the presence of ASD and its derivatives, GLUT-1 activity was greatly inhibited in a dose-dependent manner. Among ASD and its derivatives, AS-1 showed most significant inhibition. Taken together, these results strongly indicate that ASD in P. tenuipes may serve as a functional substance in lowering blood sugar in the circulatory system. ASD and its derivatives can be utilized as inhibitors of GLUT-1.
