Effect of Broussonetia papyrifera L. (paper mulberry) silage on dry matter intake, milk composition, antioxidant capacity and milk fatty acid profile in dairy cows

  • Objective: This study was carried out to investigate the possible application of Broussonetia papyrifera (B. papyrifera) silage as a functional feeding stuff in dairy cattle. Methods: Seventy-two Holstein cows were divided into four groups randomly and allocated to 6 pens with 3 individuals in each group and fed the original total mixed ratio (TMR) in the dairy farm or the new TMR with 5%, 10%, and 15% B. papyrifera silage, separately. Feed intake were recorded, milk and blood samples were collected, and milk composition, blood metabolites and milk fatty acids composition were measure at the end of the experiment. Results: Dry matter intake of cows decreased when they fed on diet with B. papyrifera, but no differences were observed in body condition score, milk yield, milk protein and lactose, feed efficiency and serum metabolites between groups. Both 10% or 15% of B. papyrifera silage in the diet significantly increased the immunoglobulin A (IgA) and IgG in serum, 15% of B. papyrifera silage increased the content of serum catalase, superoxide dismutase, total antioxidant capacity, and decreased the content of 8-hydroxy-2'-deoxyguanosine. Furthermore, 10% or 15% of B. papyrifera silage resulted in a significant decrease in the milk somatic cell count, and increased the polyunsaturated fatty acids content in the milk. Conclusion: The diets with 10% to 15% of B. papyrifera silage might enhance the immune and antioxidant function of dairy cows and increase the polyunstaturated fatty acid concentration in the milk.

Effects of concentrate level and chromium-methionine supplementation on the performance, nutrient digestibility, rumen fermentation, blood metabolites, and meat quality of Tan lambs

  • Objective: This study was conducted to evaluate the effects of concentrate level and chromium-methionine (Cr-Met) supplementation on the growth performance, carcass characteristics, meat quality, and fatty acid composition of Tan lambs. Methods: Sixty male Tan lambs (21±1.23 kg body weight) fed a finishing diet (concentrate-to-forage ratio: 35:65 [LC group] or 55:45 [HC group]) with daily Cr-Met supplementation (0, 0.75, or 1.50 g) were used in a completely randomized design with a 2×3 factorial arrangement of treatments. Results: Lambs from the HC group had higher average daily gain, dry matter (DM) digestibility, dressing percentages, leg proportions, intramuscular fat (IMF) contents, and saturated fatty acid levels, but lower feed conversion ratios, globulin (GLB) and total protein (TP) concentrations, shear force, and monounsaturated fatty acid (MUFA) levels (all p<0.05). Cr-Met supplementation increased the DM digestibility, GLB and TP concentrations, rack and loin percentages, and cooking loss, but decreased the IMF contents and leg proportions (all p<0.05). Cr-Met supplementation at 0.75 g/d increased the conjugated linoleic acid (CLA) content in both the HC and LC groups (p<0.01). Significant interactions between the concentrate level and Cr-Met dosage were observed for MUFA (p<0.01) and polyunsaturated fatty acid (PUFA) (p<0.01) levels. Meat from the lambs fed an unsupplemented LC diet presented the highest PUFA and MUFA levels (p<0.01). However, the MUFA and PUFA levels decreased significantly with increasing Cr-Met supplementation levels in the LC group (p<0.01), whereas the opposite trend was seen in the HC group. Conclusion: The HC diet improved the growth performance of Tan lambs, increased their profitability by increasing leg and rack joint proportions, and improved meat quality by promoting an IMF content that was more visibly acceptable to consumers. Cr-Met supplementation at 0.75 g/d in a HC diet was the best choice and may be economically beneficial.

Hypolipidemic and Antithrombotic Effects of Increasing Intake of Linolenic Acid Derived from Perilla Oil in Rats

  • This study investigated the hypolipidemic and antithrombotic effects of linolenic acid derived from Korean perilla oil. The experimental rats(male, Sprague-Dawley) were divided into 5 groups using a Randomized Complete Block Design and fed one of the five following diets : DO*/O#. D4/O, D4/4, D4/8, or D4/20(D*/# represents the ratio of linoleic to linoenic acid as the percentage of total dietary energy intake) for 4 or 8 months. Bleeding time and whole blood clotting time were determined and the composition of serum and platelet lipids analyzed. Comparisons from the DO/O to the D4/20 group showed that serum lipids (total lipid, triglyceride, total cholesterol, and HDL-cholesterol) gradually decreased with increasing linolenic acid intake - the hypolipidemic effect. The composition of platelet fatty acids[the ratio of eicosapentaenoic acid(EPA)/arachidonci aci(AA)] increased gradually with increasing linolenic acid intake. Higher linolenic acid intake increased bleeding time and whole blood clotting time, and decreased malondialdehyde(MDA) production in the platelets, though no significant differences. These results suggest that linolenic acid derived from perilla oil appears to suppress the conversion of linoleic acid to AA and the EPA transformed from linolenic acid appears to suppress the conversion of AA to TXA2. Since TXA2 is a platelet-aggregating and vasoconstricting agent, the redulction of TXA2 released by platelets with increasing intake of perilla oil containing a lot of linolenic acid confers an antithrombotic effect.

산란계의 혈액 및 계란지질에 관한 고수준의 미세캡슐화 이눌린 급여효과 (Effect of Feeding the High Levels of Microcapsulated Inulin on Egg and Blood Lipid Profile in Laying Hens)

  • 본 연구는 산란계 사료 내 돼지감자(Helianthus tuberosus L.)로부터 제조한 미세캡술화 이눌린(microcapsulated inulin, MI)의 고수준 첨가가 혈액지질, 계란콜레스테롤 및 지방산조성에 미치는 영향을 조사하기 위해 실시하였다. 25주령 갈색산란계를 이용하여 4주 동안 실험사료를 급여하였으며, 실험처리구는 MI 무첨가 대조군, 800 ppm, 900 ppm 및 1,000 ppm로 구분하였다. 혈액 중성지방, 총콜레스테롤 및 계란 콜레스테롤은 MI 무첨가군과 비교할 때 첨가군에서 유의하게 감소하였다. 계란 포화지방산 조성은 MI무첨가군과 비교할 때 첨가군에서 낮았으나 불포화지방산은 그 반대로 유의하게 높았다. 본 연구결과는 미세캡슐화 이눌린을 산란계 사료 내 첨가해주면 혈액지질 감소에 의해 계란품질을 향상시킬 수 있음을 시사해준다.

산초유의 지방산 조성과 마우스 혈청 및 간장의 지방산 조성변화 (Fatty Acid Composition of Serum and Liver in Mice and Sancho(Zanthoxylum schinifolium) Seed Oil)

  • 산초유 전지질 분획에서 주요 구성지방산 조성은 eicosenoic acid 30.88%, linoleic acid 23.4%, oleic acid 19.94%, palmitic acid 10.52% 순으로 나타났으며, n-9계열 일가불포화지방산이 약 60% 이상을 차지하였다. 또한 산초유의 각 지질 분획의 구성지방산 조성에서는, triglyceride 분획에서 eicosenoic acid가 41.16%로 가장 높았고, 나머지 분획들에서는 oleic acid가 34.32~53.58%로 높게 나타났다. 마우스를 24시간 절식시킨 상태에서 산초유를 마리당 500 mg씩 강제 경구투여 한 후 혈청 및 간장 전지질의 지방산 조성을 투여전, 투여 3시간 및 6시간 후의 경시적인 변화를 관찰한 결과, 혈청 전지질 중의 eicosenoic acid가 투여 전에는 존재가 확인되지 않았으나, 투여 3시간 및 6시간 후에는 경시적으로 유의적인 증가를 보였다. 간장에서도 산초유 투여 6시간 후에 eicosenoic acid의 유의적인 증가가 나타났다. 따라서, 혈청 및 간장 전지질 중의 eicosenoic acid의 증가는 산초 원료유의 섭취 영향과 지방질 대사과정에서 oleic acid로부터의 전환이 시사되어진다. 이상의 결과로부터, 산초유에는 일반 종실유와는 달리 n-9 계열 일가불포화지방산인 eicosenoic acid와 oleic acid가 많이 함유되어 있으며, 마우스의 혈청 및 간장 전지질 중의 지방산 조성의 변화는 경구투여한 원료유 지방산 섭취의 영향을 받는 것으로 시사되어졌다.

곶감 부산물의 급여가 육계의 생산성 및 계육의 지방산 조성에 미치는 영향 (Effect of Dietary Dried Persimmon By-product on Broiler Performance and Fatty Acid Contents in Chicken Meat)

  • 본 실험은 곶감 부산물의 사료적 가치를 구명하기 위하여 첨가수준(0, 1, 3, $5\%$)에 따라 육계의 생산성, 계육의 지방산 조성 및 혈중 콜레스테롤 등에 미치는 영향을 고찰하기 위하여 5주 동안사양 시험하였다. 전기 3주간 사료내 조단백질은 $21.5\%$, ME는 3,100kcal/kg 수준으로 공급하였고, 후기에는 조단백질 $19\%$, ME 3,100kcal/kg 수준으로 급여하였다. 대조구(무첨가구), 곶감 부산물 $1\%$ 첨가구(T1), $3\%$ 첨가구(T2), $5\%$ 첨가구(T3)로 처리구당 4반복, 반복당 10수로 총 160수를 공시하였다. 그 결과 증체량은 전기사료 급여시에 대조구보다 곶감 부산물을 급여한 처리구에서 높은 경향이었으나, 후기 사료 급여시는 대조구와 T1 및 T2에서 T3보다 유의적으로 높게 나타났다. 사료 효율은 처리구간에 유의성이 없었다. 곶감 부산물의 급여 수준에 따른 계육의 일반 성분 중 수분과 조회분은 유의성이 없으며, 조지방은 증가하고 단백질은 감소하는 경향이었다(P<0.05). 총 콜레스테롤, HDL-C, 중성지방은 처리구에서 높았고, LDL-C은 처리구보다 대조구에서 높았다(P<0.05). 계육의 주된 지방산은 oleic acid, palmitic acid, linoleic acid, stearic acid 순이었다. 곶감 부산물의 급여량이 증가함에 따라 stearic acid, linoleic acid는 감소하였으나 oleic acid의 함량은 증가하였다(P<0.05). 이러한 실험 결과로 곶감부산물은 육계사료에 $3\%$수준까지 급여하더라도 생산성에 영향을 미치지 않을 것으로 사료된다.

  • In this study, Artemisia capillaris, which has been used in oriental medicine and folks remedy, was investigated to characterize the nutritional composition (protein, lipid, ash, fiber, amino acids, fatty acids, Na, K, P, Mg, Ca and Fe) and effective substance (scopoletin). Artemisia capillaris has often been cited in medical literature for its medicinal effects. The Korean Food and Drug Administration allowed Artemisia capillaris as a food stuff and indicated as In-Jin-Sook. From the view point of the subjects who eat In-Jin-Sook products, most of them ingested it for a certain pharmacological efficacy rather than as a beverage or a food. Therefore, we need to develop functional foods using In-Jin-Sook. Artemisia capillaris was superior in protein, crude fiber, amino acid and fatty acid compositions to wild greens according to Korean Nutritional Composition Table. Also scopoletin, blown for blood vessel relaxation and hypotensive effect, was detected at the level of 326.13 $\mu\textrm{g}$/g. These results demonstrated that Artemisia capillaris has both pharmaceutical effects and balanced nutritional contents. Artemisia capillaris has sufficient values to use as a food stuff for functional foods.

Changes of the Blood Composition of Periparturient Cows in Relation to Time of Day

  • In order to determine the appropriate sampling time for blood metabolites of periparturient cows, the changes of the blood composition in relation to time of day were evaluated in sixteen multiparous Holstein cows at 1 wk prepartum, 1 and 6 d postpartum. Blood samples were collected at 08:30, 10:00, 15:30 and 17:00 h in each sampling day, and the sampling times at 08:30 and 15:30 h were prior to feeding. The rectal temperature of cows increased gradually from 08:30, to 17:00 h, but blood Hct and Hb decreased constantly. Plasma non-esterified fatty acid (NEFA) concentration at 08:30 h was two-fold higher than those at 10:00, 15:30 and 17:00 h from 1 wk prepartum to 6 d postpartum, and the value was maximum at 1 d postpartum. The highest plasma urea-N was observed at 10:00 h from 1 wk prepartum to 6 d postpartum. Plasma glucose and total protein were not affected by sampling time. The data indicated that blood samples of periparturient cows should be collected before morning feeding for the diagnosis of energy status, because plasma NEFA was the highest before morning feeding.

된장으로 부터 fibrin 용해 세균의 분리에 관한 연구 (The study on isolation of fibrinolytic bacteria from soybean paste)

  • fibrin 단백질을 분해할 수 있는 효소를 분비하는 균을 된장으로부터 분리하여 동정하였다. 된장으로부터 protein 분해능이 있는 균주를 분리하고 분리한 균주들 중에서 혈전용해능이 가장 우수한 균주인 KDO-13을 선발하였다. 이 균주는 호기성으로 포자를 형성하는 Gram(+)균 이었으며 gas chromatography에 의한 세포의 지방산 분석은 $C_{15:0}$ anteiso fatty acid가 47.7%, $C_{15:0}$ iso fatty acid가 13.5% 이었다. 이 균주는 생리, 생화학적 특성 및 세포지방산 분석을 통하여 57.7%의 similarity를 갖는 Bacillus atrophaeus로 잠정 동정되었다. Bacillus atrophaeus KDO-13의 fibrinolytic enzyme 생성을 위한 최적 배양 온도와 pH는 각각 $37^{\circ}$와 6.0 이었다.

Comparison of overfed Xupu and Landes geese in performance, fatty acid composition, enzymes and gene expression related to lipid metabolism

  • Objective: The aim of this study was to compare overfeeding performance, fatty acid composition, blood chemistry, enzymes and genes expression overfed Xupu and Landes geese. Methods: Sixty male Xupu geese (80 d) and Landes geese (80 d) were selected. After a period of one-week of pre-overfeeding, Xupu and Landes geese were overfed three meals of 550 and 350 g/d, respectively, of a high-carbohydrate diet in the first week of the overfeeding period. The next week, geese were given four meals of 1,200 and 850 g/d, respectively, over 8 to 14 d. Finally, geese were given five meals of 1,600 and 1,350 g/d, respectively, for the last two weeks. Results: After overfeeding for 28 d: Compared with Landes geese, Xupu geese liver weight and liver-to-body weight ratio decreased (p<0.05), while final weight, slaughter weight, total weight gain, abdominal fat weight, and feed-to-liver weight ratio increased (p<0.05). The levels of elaidic acid (C18:1t9), oleic acid (C18:1n-9), eicosenoic acid, and arachidonic acid in the liver of Xupu geese significantly increased (p<0.05), and the levels of myristic acid and stearic acid significantly decreased (p<0.05), while methyleicosanoate acid significantly increased (p<0.05). Xupu geese had higher plasma concentrations of triglyceride and very low density lipoprotein cholesterol (p<0.05), and decreased activities of alanine aminotransferase, aspartate aminotransferase, and lipase (LPS) (p<0.05). Landes geese had higher LPS activity (p<0.05), but lower cholinesterase activity (p<0.05) when compared with Xupu geese. The mRNA expression levels of fatty acid dehydrogenase (FADS) gene, elongase of long-chain fatty acid 1 (ELOVL1) gene, ELOVL5, and acyl-Co A: cholesterol acyltransferase 2 (ACAT2) gene were significantly upregulated (p<0.05) in Landes goose when compared with Xupu geese. Conclusion: This study demonstrates that the liver production performance of Landes geese was better than that of Xupu geese to some extent, which may be closely related to LPS activity, as well as the expression of FADS, ELOVL1, ELOVL5, and ACAT2.
