삼정환(三精九)의 항우울 효과에 대한 실험적 연구 (Experimental Study on the Antidepressant Effect of Sam-Jeong-Hwan)

  • Objective: The purpose of this study was to investigate the protective effects of Sam-Jeong-Hwan(SJH) on the animal model of depression induced immobilization stress. Method: The subject were divided into 4 groups(l. normal 2. saline solution administered during immobilization stress treatment 3. SJH of 100mg/kg administered 4. BKJ of 400mg/kg administered). Immobilization stress was treated for 1 hours on day. During 2 days of immobilization stress treatment, they were executed forced swimming test, passive avoidance test, elevated plus maze test. Corticosterone and ACTH in blood were measured. Results: In forced swimming test, SJH of 400mg/kg group showed decreased immobilization. In passive avoidance test, SJH of 400mg/kg group showed increased learning execution. In EPM test, SJH of 400mg/kg group showed decreased anxiety. In locomotor activity test, SJH groups showed significantly increased locomotor activity. Stress group showed significantly increase in serum level of corticosterone, SJH of 400mg/kg group showed decreased serum level of corticosterone. Stress group showed significantly increase in serum level of ACTH, SJH of 400mg/kg group showed decreased serum level of ACTH. Conclusion: These results suggest that Sam-Jeong-Hwan(SJH) is effective in the treatment of depression.

Anti-depressant Effect of Chilbokum under the Forced Swimming Test in Rats

  • Objectives : The aim of this study was to evaluate the anti-depressant effect of Chilbokum on rats under the forced swimming test (FST) Methods : The rats were treated with the herbal extract, Chilbokum. In order to induce depression-like symptoms, the FST was conducted afterwards. The immobility time was measured during a 5-min experimental session. The alterations of the 5-HT level in the hypothalamus and hippocampus and the change of corticosterone level in the blood induced by FST were determined in the rats. Results : The results were as follows: 1. The immobility time during 5 min of FST in the drug administration group showed significant decreases compared with the control group (p<0.05). 2. The FST+Chilbokum group had significantly increased 5-HT levels of the hypothalamus and hippocampus, compared with the control group (p<0.05, respectively). 3. The FST+Chilbokum group had significantly decreased corticosterone levels, compared with the control group (p<0.05). Conclusions : These results demonstrate that the reduced immobility time by Chilbokum may be mediated by the increase in 5-HT level in the hypothalamus and hippocampus, suggesting that Chilbokum has a potential therapeutic efficacy for human depression.

Stress related activities of Sun-ginseng in SD Rats and ICR Mice

  • The main aim of this study was to investigate stress related activities of Sun-ginseng extract as a candidate for anti-stress-related functional supplement by comparing its effect to those of red ginseng, which is also known to alleviate stress. Normal group was not exposed to stress while the control group was exposed to stress. Rats were orally administered once a day with 200 mg red ginseng (RG) extract, 100 or 200 mg Sun-ginseng (SG) extract/kg body weight. Mice were given water containing 400 mg red ginseng extract, 200 or 400 mg SG/100 mL potable water. Rats were given supplements for 5 days without stress, and 5 days with restraint and electroshock stress. After final stress, stress-related behavioral changes of experimental animals were recorded and levels of blood corticosterone were measured. Mice were given supplements for 5 days through drinking water, and then fatigue related motor activity were recorded. SG-supplementation partially blocked stress effect on locomotion and elevated plus maze test in rats, and also partially blocked stress-induced behavioral changes such as freezing, burrowing, smelling, facewashing, grooming and rearing behavior in rats. SG-supplementation decreased blood corticosterone level which is increased by stress in rats. Effects of SG may not be modulated by GABAnergic nervous system. SG-supplementation prolonged swimming time and staying time on the wire and rotarod wheel in mice. These results suggest that SG partially protects living organisms from stress attack in some cases and thus has the potential to be used as a functional food to alleviate stress response.

  • Gardenia Jasminoides(GJ) is traditionally used for treatment of hepatic disease, insomnia, anxiety, and inflammatory disease. The aim of this study is to examine effects of GJ extract in response to stress. Animals of the normal group were not exposed to any stress and the control group were exposed to stress. The rats of the Ginseng and GJ supplementary group were orally administered once a day with 100mg of red ginseng extract, 100mg of GJ extract/kg body weight. The mice were given water containing 200mg of red ginseng extract, 200mg of GJ extract/100ml potable water. Animals were given supplements for 7 days without stress, and then were given supplements for 5 days with restraint and electroshock stress. After loading final stress, we examined stress related behavioral changes of experimental animals and measured the levels of blood corticosterone. GJ-supplementation partially blocked the stress effect on locomotion and elevated plus maze test in rats, and also partially blocked stress-induced behavioral changes such as freezing, burrowing, face-washing, smelling and rearing behavior in rats. The effect was almost equipotent to Ginseng's effect. GJ-supplementation didn't influence on fatigue related behavior or physical stress resistance. GJ-supplementation decreased the levels of blood corticosterone which is increased by stress in rats. These results suggest that GJ protects partially the living organism from stress attack and it has the potential to be used as a functional material to alleviate stress response.

조도가 육계 병아리의 생산성, 혈액성상 및 면역 수준에 미치는 영향 (Effects of Light Intensity on the Growth Performance, Blood Parameter and Immune Status of Broiler Chicks)

  • 본 연구는 육계 병아리 시기(0~7일령)에 조명의 조도 차이에 따른 생산성, 혈액 조성 및 스트레스 수준에 미치는 영향을 대하여 조사하였다. 육계(Ross 308) 초생추(48.0±0.2 g) 수컷 240수를 조도에 따라 3처리구(20, 30 및 50 lx)로 나누어, 처리구 당 4반복, 반복 당 20수를 완전임의 배치하였다. 7일령에 체중, 사료요구율, 가슴육과 간의 무게, 소장 길이를 측정하였으며, 혈액을 채취하여 혈구, 혈청 생화학 조성, corticosterone, IL-6 및 TNF-α을 분석하였다. 체중, 증체량, 가슴근육 및 간 무게는 20 lx 처리구보다 50 lx 처리구에서 유의적으로 높게 나타났으나(P<0.05), 사료 섭취량, 사료 요구량, 소장 길이는 유의적인 차이를 보이지 않았다. 혈구 분석 결과, 백혈구, 적혈구, 혈소판 조성, 혈청 생화학 성분(glucose 및 IP 제외)은 처리구 사이에서 유의적인 차이가 보이지 않았다. Corticosterone과 IL-6은 20 lx 처리구에서 높은 농도를 나타내었다(P<0.05). 결론적으로, 육계 병아리 시기에 조도를 50 lx로 처리하였을 때 증체량 및 가슴근육 비율이 증가하였으며, corticosterone 및 IL-6는 낮은 함량을 나타내었다. 따라서 병아리 시기의 높은 조도는 생산 능력, 스트레스 및 면역 상태를 개선하는 것으로 사료된다.

LC-MS-based metabolomic analysis of serum and livers from red ginseng-fed rats

  • Serum and liver metabolites in rats fed red ginseng (RG) were analyzed by ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry. The mass data were analyzed by partial least squares-discriminant analysis (PLS-DA) to discriminate between control and RG groups and identify metabolites contributing to this discrimination. The RG group was clearly separated from the control group on PLS-DA scores plot for serum samples, but not liver samples. The major metabolites contributing to the discrimination included lipid metabolites (lysophosphatidylcholine, acyl-carnitine, and sphingosine), isoleucine, nicotinamide, and corticosterone in the serum; the blood levels of all but isoleucine were reduced by RG administration. Not all metabolites were positively correlated with the health benefits of RG. However, the blood levels of lysophosphatidylcholine, which stimulate various diseases, and long-chain acylcarnitines and corticosterone, which activate the stress response, were reduced by RG, suggesting long-term RG might relieve stress and prevent physiological and biological problems.

Anti-stress Effects of Kimchi

  • The anti-stress effects of kimchi were studied in the Sprague-Dawley rats dosed with kimchi. The rats in the stress groups were subjected to immobilization stress for 2 hr/day for 5 days. At the end of the experimental period, daily average food intake and body weight (BW) gain had been reduced in the stress group compared to the control group. Daily average food intake was significantly increased in the stress-kimchi diet group compared to the stress-only group. The weights of the thymus and spleen were decreased by immobilization stress, but this reduction was partially suppressed by the addition of kimchi. The weights of the adrenal gland and epididymal adipose tissue were increased in the stress group, but ingestion of kimchi completely and partially suppressed these stress-induced changes, respectively. Serum levels of total cholesterol and triglyceride, and plasma levels of corticosterone were increased in the stress group, but at control levels in the stress-kimchi diet group.

Lactobacillus mucosae and Bifidobacterium longum Synergistically Alleviate Immobilization Stress-Induced Anxiety/Depression in Mice by Suppressing Gut Dysbiosis

  • We isolated Lactobacillus mucosae NK41 and Bifidobacterium longum NK46 from human feces, which induced BDNF expression in corticosterone-stimulated SH-SY5Y cells, and examined their anti-depressive effects in mice. NK41, NK46, and their (1:1) mixture significantly mitigated immobilization stress (IS)-induced anxiety-like/depressive behaviors, hippocampal $NF-{\kappa}B$ activation, BDNF expression, $Iba1^+$ cell population, and blood corticosterone, $TNF-{\alpha}$, IL-6, and lipopolysaccharide levels. Furthermore, they inhibited colitis marker $NF-{\kappa}B$ activation, and $TNF-{\alpha}$ expression in mice with IS-induced anxiety/depression. They additionally suppressed gut Proteobacteria and Bacteroidetes populations and bacterial lipopolysaccharide production. These findings suggest that NK41 and NK46 may alleviate anxiety/depression and colitis by suppressing gut dysbiosis.

케이지 사육시 사육밀도가 산란계의 깃털손상도, 계란품질, 혈액성상 및 스트레스 호르몬 농도에 미치는 영향 (Effect of Stocking Density on the Feather Condition, Egg Quality, Blood Parameters and Corticosterone Concentration of Laying Hens in Conventional Cage)

  • 본 연구는 케이지 사육 시 사육밀도가 산란계의 깃털 손상도, 계란품질, 혈액성상 및 스트레스 호르몬 농도에 미치는 영향을 구명하기 위하여 수행되었다. 산란계(Hy-Line Brown) 700수를 공시하여 배터리 케이지에 750 ㎠/수 및 500 ㎠/수 사육밀도 처리구로 50 반복을 두어 32주부터 60주까지 총 28주간 실시하였으며, 사료와 물은 자유 급이 하였다. 사육밀도가 증가함에 따라 주령이 증가할수록 깃털손상도가 증가하였으며, 등과 꼬리 부위의 손상이 유의적으로 증가하였다. 계란품질(난중, Haugh unit, 난각 강도 등)은 사육밀도에 따른 처리구 간 유의적 차이는 나타나지 않았다. 또한, 혈구 성분 및 혈액 생화학 지표는 사육밀도에 의한 영향을 받지 않는 것으로 나타났다. 그러나 시험종료 시점의 혈액과 깃털 내 스트레스 호르몬인 corticosterone 농도를 분석한 결과, 500 ㎠/수 사육밀도에서 사육 시 750 ㎠보다 높게 나타났다. 결론적으로 케이지에서 500 ㎠/수 사육밀도 사육은 산란계의 깃털손상도를 증가시켜 부정적인 영향을 미치는 것으로 보이며, 향후 산란계의 행동과 관련된 추가 연구가 필요할 것으로 판단된다.

일반농장과 동물복지농장 닭의 혈액 내 gas, 생화학인자, 호르몬 농도 및 근육 내 스트레스 반응인자 비교 (Comparison of Blood Gas, Biochemical Factors, and Hormone Concentrations, and Muscle Stress Response Factors of Chickens in General Farms and Animal Welfare Farms)

  • 본 연구는 육계에서 동물복지농장의 스트레스 저감효과를 과학적인 지표를 통해 확인하고자 시행되었다. 농장 내 gas 농도를 비교한 결과 일반농장에서 육계에 부정적인 영향을 미칠 수 있는 CO2와 NH3의 농도가 높았다. PCO2는 일반농장에서, PO2는 동물복지농장에서 유의하게 높았다. 일반농장의 육계에서 혈중 CREA, GLU, AST, CK가 유의적으로 높았으며, TP, GLOB는 유의하게 낮았다. 또한 혈중 corticosterone과 norepinephrine이 일반농장에서 유의하게 높은 값을 나타내었다. 이러한 결과를 통해 동물복지농장에 비해 일반농장 육계의 스트레스 수치가 더 높은 것으로 판단되며, 동물복지농장이 스트레스가 감소된 사양 환경을 제공하고 있다는 것을 의미한다.
