채혈바늘에 의한 2차 감염 방지기능을 갖는 랜싯과 전동식 채혈기의 개발 (Development of the Electric Lancing Device and the Lancet to Prevent the Secondary Contamination by Needlestick Injury)

  • 본 연구는 채혈바늘에 의해 발생하는 2차 감염을 방지하기 위한 안전 랜싯(Lancet)을 설계 개발하고, 연속하여 채혈하는 경우에 쉽게 그리고 편리하게 사용할 수 있도록 무선 전동방식 채혈기의 설계 제작을 목적으로 한다. 안전랜싯은 채혈침과 보호캡이 일체형으로 설계되어 채혈 순간을 제외하고는 채혈침이 항상 보호캡 속에 있도록 하여 채혈바늘에 의한 사고를 방지하도록 하였다. 전동 채혈기는 충전건전지를 전원으로 하고 솔레노이드의 작동으로 랜싯을 타격하여 채혈하도록 설계되었으며, 연발타격 또는 단발타격의 기능을 선택하도록 설계 제작되었다. 또한 타격봉에 의한 충격력과 사용된 배터리의 수명 등이 측정되었다. 이 채혈기는 한방병의원의 사혈기로도 사용되도록 제작되었다.

연 재생공장 인접 근로자들의 연 폭로정도에 관한 조사 (Exposed level of workers in the factory next to a lead recycling factory)

  • 부산 시내 모 공단에 위치한 폐밧데리로 연을 재생하는 산업장에 인접한 산업장에 근무하는 근로자들의 연 폭로 정도를 알기 위하여 연 폭로원에 인접한 작업장 A의 39명과 연 폭로원에서 8.5km 떨어진 곳에 위치한 동일한 원료, 공정, 자동차 부품을 생산하는 작업장 B의 62명을 연구대상자로 선정하고 표준화된 설문지 조사와 혈중 연 농도를 분석, 비교하였다. 또한 두 산업장의 세 작업장 기중 연 농도를 분석, 비교하였다. 그 결과는 다음과 같다. 1. 산업장 A의 평균연령 (38.7세)이 산업장 B(29.5세) 보다 유의하게 높았고, 근무연수의 차이는 없었다. 2. 평균 기중 연 농도는 산업장 A $2.6{\mu}g/m^3$, 산업장 B $1.2{\mu}g/m^3$로 산업장 A가 유의하게 높았다(P<0.05). 또한 산업장 A의 평균 기중 연 농도는 실내작업장, 실외작업장, 사무실 순으로 연 폭로원과 거리가 가까울수록 높았고 세 작업장 사이에 유의한 농도차가 있었다(P<0.05). 3. 평균 혈중 연 농도는 산업장 A $14.9{\mu}g/dl$, 산업장 B $12.2{\mu}g/dl$로 산업장 A가 유의하게 높았다(P<0.001). 또한 작업장 A의 세 작업 분야별 평균 혈중 연 농도 사이에는 유의한 통계적 차이(P<0.05)가 있었지만 작업장 B는 없었고 작업장 A의 평균 혈중 연 농도 순은 기중 연 농도 순과 연 폭로원과 떨어진 거리 순과 일치하였다. 4. 혈중 연 농도와 연령, 근무연수 사이의 상관관계는 산업장 A의 경우 상관이 낮았고, 산업장 B에서는 연령과 유의한 상관(r=0.382, P=0.0023)을 보였고, 연령을 보정하여도 두 산업장간 평균 혈중 연 농도 차는 유지되었다(P<0.001). 산업장 A에서 보인 작업장소간, Part간 농도의 차이는 연 폭로원에 인접한 산업장의 경우 연 폭로원의 작업환경과 밀접한 관계가 있음을 시사한다. 따라서 유해산업장의 작업환경으로 인하여 발생하는 인접산업장 근로자들의 유해물질에 대한 환경적 폭로는 해당 유해산업장의 환경개선 노력에 의하여 적절히 예방될 수 있을 것이므로 유해산업장의 작업환경에 대한 보다 체계적이고 철저한 감시가 필요할 것으로 생각된다.

생물의약품 제조공정에서 Bovine Parvovirus 정량 검출을 위한 Real-Time PCR (Real-Time PCR for Quantitative Detection of Bovine Parvovirus during Manufacture of Biologics)

  • 소의 혈액, 세포, 조직, 기관 등은 생물의약품과 조직공학제제, 세포치료제의 원료로 널리 사용되고 있다. 소유래 물질을 원료로 사용한 제제의 경우 소유래 원료 물질에 다양한 바이러스가 오염된 사례가 있기 때문에 바이러스 안전성 검증이 필수적이다. BPV는 소에게 가장 흔하게 감염되는 바이러스 중의 하나이다. 소유래 물질을 원료로 하는 생물의약품, 조직공학제제, 세포치료제 등에서 BPV 안전성을 확보하기 위해, 원료물질, 제조공정, 완제품에서 BPV를 정량적으로 검출하고, 제조공정에서 BPV 제거 검증을 위한 시험법으로 활용이 가능한 BPV real-time PCR 시험법을 확립하였다. BPV에 특이적인 primer를 선별하였으며, 형광염료 SYBR Green I을 사용하여 BPV DNA 정량 검출 시험법을 최적화하였다. 세포배양법에 의한 감염역가와 비교한 결과 real-time PCR 민감도는 $1.3{\times}10^{-1}\;TCID_{50}/mL$이었다. 확립된 시험법의 신뢰성(reliability)을 보증하기 위해 시험법 검증을 실시한 결과 특이성(specificity)과 재현성 (reproducibility)이 우수함을 확인하였다. 확립된 real-time PCR을 생물의약품 제조공정 검증에 적용할 수 있는지 확인하기 위하여 인위적으로 BPV를 오염시킨 CHO 세포주와 소유래 콜라겐에서 BPV 검출 시험을 실시하였다. BPV를 감염시킨 CHO 세포에서 세포변병효과를 관찰할 수 없었지만, 세포와 세포배양 상청액에서 BPV를 정량적으로 검출할 수 있었다. 소유래 콜라겐에서도 $1.3{\times}10^0\;TCID_{50}/mL$까지 정량적으로 검출할 수 있었다.

소고기의 유통 단계별 병원성 미생물 오염도에 관한 연구 (A study on the contamination level of pathogenic microorganisms in beef distribution stages)

  • Contamination levels of pathogenic microorganisms in 145 cases of beef, which were distributed in Gwangju province, had been investigated in each distributed stage and also monitored by general bacterial count and E coli count index. General bacterial count of beef from the slaughterhouse was 10$^4$cfu/g less than the level of promotion(10 cfu/$\textrm{cm}^2$) and E coli count index was also under the level of 10$^2$cfu/$\textrm{cm}^2$ recommended level of the ministry of agriculture and forestry. Pathogenic microorganisms were detected from 23.2% of samples in the consumption stage, 12.5% in the slaughtering stage and 5.6% in the transporting and processing stage. Staphylococcus aureus was isolated in the largest number and its ratio was 9.0%, listeria monocytogenes 5.5% and salmonella spp 1.4%. There were no samples that bacteria had been detected dually. E coli O157:H7 and campylobacter jejuni were not isolated. In raw and chilled beef, isolation rate of pathogenic microorganisms were 13.3% and 16.5% each. Especially in raw beef, L monocytogenes was. isolated in 3 samples among 30 cases (10%) and S aureus in one sample (3.3%). According to a scale of meat store, isolation rates of pathogenic microorganisms were different. It was 28.6% in the small-scale meat store and 16.7% in the large-scale meat store each. Four cases (16.7%) of S aureus were isolated in the large-scale meat store and seven cases (20.0%) of L monocytogenes and 2 cases (5.7%) of salmonella spp were isolated in the small-scale meat store. S aureus was isolated in two places among 10 feeding facilities of the elementary school. This result shows that the sanitation of elementary school feeding facilities is so poor and more careful policy consideration is needed. Eleven strains of S aureus isolated showed ${\beta}$-hemolysis on blood agar, 1 strain ${\alpha}$-hemolysis, and 1 strain ${\gamma}$-hemolysis. Isolated strains of L monocytogenes were reconfirmed in 560 bp by PCR. Conclusively, these results show that the sanitary condition in the stages of slaughtering, transportation-processing and consumption influences the degree of pathogenic microorganisms contamination in beef severely It is necessary to apply thoroughly hazard analysis critical control point in a process of beef distribution and also to develop rapid test methods for microorganism diagnosis. This effort is very important for the supply of safe and clean meat from farm to table and helpful for the improvement of public health.

THE EFFECTS OF SURFACE CONTAMINATION ON THE SHEAR BOND STRENGTH OF COMPOMER

  • The lastest concepts in bonding are "total etch", in which both enamel and dentin are etched with an acid to remove the smear layers, and "wet dentin" in which the dentin is not blown dry but left moist before application of the bonding primer. Ideally, the application of a bonding agent to tooth structure should be insensitive to minor contamination from oral fluids. Clinically contaminations such as saliva, gingival fluid, blood and handpiece lubricant are often encountered by dentists during preparation of a restoration. The aim of this study was to evaluate the effect of contamination by hem-ostatic agents on shear bond strength of compomer restorations. One hundred and ten extracted human maxillary and mandibular molar teeth were collected. The teeth were cleaned from soft tissue remnant and debris and stored in physiologic solution until they were used. Small flat area on dentin of the buccal surface were wet ground serially with 400, 800 and 1200 abrasive paper on automatic polishing machine. The teeth were randomly divided into 11 groups. Each group was conditioned as follows: Group 1 : Dentin surface was not etched and not contaminated by hemostatic agents. Group2 : Dentin surface was not etched but was contaminated by Astringedent (Ultradent product Inc., Utah, U.S.A.). Group3 : Dentin surface was not etched but was contaminated by Bosmin (Jeil Phann, Korea.). Group4 : Dentin surface was not etched but was contaminated by Epri-dent (Epr Industries, NJ, U.S.A.). Group5: Dentin surface was etched and not contaminated by hemostatic agents. Group 6 : Dentin surface was etched and contaminated by Astringedent. Group7 : Dentin surface was etched and contaminated by Bosmin. Group8 : Dentin surface was etched and contaminated by Epri-dent. Group9 : Dentin surface was contaminated by Astringedent. The contaminated surface was rinsed by water and dried by compressed air. Group10 : Dentin surface was contaminated by Bosmin. The contaminated surface was rinsed by water aud dried by compresfed air. Group 11 : Dentin surface was contaminated by Epri-dent. The contaminated surface was rinsed by water and dried by compresfed air. After surface conditioning, F2000 was applicated on the conditoned dentin surface. The teeth were thermocycled in distilled water at $5^{\circ}C\;and\;55^{\circ}C$ for 1000 cycles. The samples were placed on the binder with the bonded compomer-dentin interface parallel to the lmife-edge shearing rod of the Universal testing machine(Zwick 020, Germany) running at a cross head speed of 1.0mmimin. There were no significant differences in shear bond strength between groups 1 and group 3 and 4, but group 2 showed significant decrease in shear bond strength compared with group 1. There were no significant differences in shear bond strength between group 5 and group 7 and 8, but group 6 showed significant decrease in shear bond strength compared with group 5. There were no significant differences in shear bond strength between group 5 and group 9, 10 and 11.

도축되는 한우 혈액에서 회수한 혈청의 생물학적 안전성 분석 (Assessing Biological Safety of the Hanwoo Serum Obtained During Slaughtering Process)

  • 본 연구에서는 성체 한우의 도축과정 중에 채혈된 혈액으로부터 분리 정제한 혈청의 생물학적 안전성을 검증하기 위하여 세균, 마이코플라즈마 및 바이러스의 검출과 동정을 실시하였다. 검사를 실시한 대부분의 혈청에서 전염성 병원체가 검출되지 않았고 두 개의 혈청에서 토양 유래의 것으로 추정되는 Bacillus thuringiensis가 검출 되었다. 본 연구의 결과는 국내에서 도축되는 한우는 생물학적 안전성을 담보할 수 있는 최소한의 기본적인 안전장치를 통해 도축된다는 사실을 보여주며 한우의 도축과정에서 부산물로 얻을 수 있는 혈액을 안전한 생물학적 제제로 개발할 수 있다는 가능성을 보여주는 것으로 생각된다.

다제내성균 환경표면 오염도 및 소독 효과 평가 (Evaluation of environmental surface contamination and disinfection effects on multidrug-resistant organism)

  • 본 연구는 다제내성균 환자가 사용한 병실 환경에 대한 환경관리 평가 방법 중 미생물 배양검사와 Adenosin Triphosphate Bioluminescence 방법을 이용하여 오염도를 파악 한 후 환경소독제를 이용하여 소독효과를 평가하고자 시도되었다. 환경표면은 다제내성균으로 격리된 환자병실을 대상으로 하였다. 검체채취는 인퓨전펌프, IV 폴대, 상두대, 침대난간, 키보드 혈압기 커프 5곳을 실시하였고 소독 전, 소독 직후, 소독 후 5분 후 ATP와 미생물배양검사를 실시하였다. 연구결과 Infusion pump의 환경표면이 소독·후 오염도가 통계적으로 유의하게 감소하였다. 또한 IV 폴대, 침상난간, 키보드는 소독 전후 균 검출이 감소하였다. 즉 정기적인 환경표면소독은 환자에게 감염으로부터 안전한 환경을 제공할 수 있다. 따라서 향후 다기관을 대상으로 소독제의 지속성 등을 평가하여 환경표면 소독방법, 소독주기 등의 지침을 마련하는 것이 필요하다.

Brucella 감영농장에서 감염경로의 역학적 연구 (Epidemiological study for infection route of brucellosis in a infected dairy farms)

  • A dairy farm that has been suffered continuously(more than 2 years) from brucellosis in Korea in spite of repeated legal test-and-slaughter was investigated the main source of infection in the farm. All cattle(22 milking cows, 44 heifers, 60 calves, 8 bull), dogs(3 mixed breed), feces from wild birds(3 samples), drinking water(3 sites), and soil in the paddocks(14 sites) inside the farm were examined with serological and/or bacteriological methods including specific DNA detection with PCR method. Brucella spp in the milk and blood were detected in 12/22 and 5/22 milking cows, respectively, although all of them were negative with conventional tube agglutination test. The number of serologically positive heifer was 15(15/44), but the isolation of Brucella spp was succeeded in the only 11(11/15) of them. Brucella were detected in vagina 1(1/11) and nasal(3/12) excretion in serologically positive heifers. All the three dogs were serologically positive, and Brucella spp were isolated from their blood. However, Brucella spp were not detected in the drinking water, soil in the paddocks, nor the feces of wild birds. The results suggest that milking cow secrete Brucella spp through milk, genital tract and nasal cavity, which are the major source of infection in this farm, The main infection route of Brucella spp is contact to contact with Brucella spp excreting animals rather than environmental contamination. The animals, living together with infected cow such as dogs, are the readily susceptible and are required to be examined for Brucella spp.

오적산을 투여한 흰쥐의 혈액중 금속농도 비교에 관한 연구 - 용량-반응관계와 기전을 중심으로 - (Comparison of Blood Metal Concentration in Ohjeok-san(Wuji-san) Treated Rats - Dose-Response Relationship and Mechanism -)

  • Objectives: In recent years, extensive focus has been laid on adulteration of herbal medicine with heavy metals. This may be mainly due to soil contamination by environmental pollution. The objective of this study is to identify the contents of various heavy metals in the blood from Ohjeok-san (Wuji-san) Decoction (OD) treated-rats. Methods: For this study, 13 kinds of metals including essential and heavy metals, i.e. A1, As, Cd, Co, Cr, Cu, Fe, Hg, Mn, Ni, Pb, Se and Zn were analyzed by a slight modification of EP A methods and the following results are obtained. Results: 1. There was no significant difference between the OD-treated groups and control group in liver, kidney, bone, brain and weight, especially no significant difference at the 5th and 10th days in weight and the amount of food intake. 2. The amount of each metal analyzed in the blood were as follows; A1: 2.3~3.07 mg/l, As: 2.90~3.66 mg/l, Cd: 0~0.001 mg/l, Co: 0~0.01 mg/l, Cr: 0.40~043 mg/l, Cu: 0.93~1.88 mg/l, Fe: 414.35~464.46 mg/l, Hg: 0.01 mg/l, Mn: 0.10~0.17 mg/l, Ni: 0.01 mg/l, Pb: 0.03~012 mg/l, Se: 0.73 mg/l, Zn: 3.41~4.13 mg/l by groups, respectively. In control and experimental group, Experimental I and other experimental II, III, IV, and V groups, there were no significant differences. 3. The amount of non-toxic metals (A1, Co, Cu, Fe, Mn, Se, Zn) were $64.1{\pm}7.71{\;}mg/l$ in the control group and 60.70~67.58 mg/l in the experimental groups I, II, III, IV and V. The amount of Toxic metals (As, Cd, Cr, Ag, Pb) were $0.68{\pm}0.21{\;}mg/l$ in the control group and 0.57 ~ 0.66mg/l in the experimental groups. The total amount of metals were 32.35 mg/l in the control group and 30.48~34.12 mg/l in the test groups I, II, III, IV and V, respectively. Conclusions: There was no significant difference of metal concentration in the blood from the OD-treated-rats compared to those of the control group even if higher dosage (1~8 times the dosage for a person) of OD was administered. This may be mainly due to a decoction treatment which contains only supernatants filtered from the herb-mass after boiling. This indicates the legal limitation for metal concentration in herbal medicine must be applied according to different treatment methods of herbal medicine.

Non-Conventional Concentrates in Temperate Asian-Australasian Countries - Review -

  • The huge amount of demand for feedgrains from this region could not possibly be met by producing countries from the other regions. In order to fulfill this increasing demand for conventional raw materials, an alternative for the conventional raw materials produced in the Asia and Pacific region is becoming increasingly more important. A potential alternative is concentrates or non-conventional concentrates produced locally in relative abundance in this region. These feedstuffs include feed grains, by-products from the milling, sugar industries, brewing and distilling industries. Vegetable, citrus, and animal by-products from abattoir, feather meal and blood meal are also possibilities. In addition to more widespread use of unconventional feed sources, the following approach is recommended to improve utilization and performance. These include establishing the nutritive value of non-conventional feeds, quality control to minimize variability, proper storage and processing to assure the nutritive value and prevent mycotoxin contamination, properly balance amino acids with protein sources, supplementation with synthetic amino acids and the use of enzymes to increase digestibility. Currently, practical applications for these resources in feed formulation are negligible despite the potential. The socio-economic aspects will dominate the use of these non-conventional concentrates. In the future, the feed industry will resolve the problems in using locally available raw feed materials.
