The Evaluation of Antithrombotic and Fibrinolytic Activities of Nattokinase from Bacillus subtilis Natto (Bacillus subtilis Natto가 생산하는 Nattokinase의 항혈전 및 피브린 용해능 효능평가)

  • We previously reported that Ultra nattokinase$^{(R)}$ showed high fibrinolytic activity and revealed antithrombotic effect in rat blood plasma based on its ability to suppress collagen-induced platelet aggregation. This research was carried out to verify the clot lysing activity and blood flow enhancing effects of Ultra nattokinase$^{(R)}$ via monitoring and comparing the antithrombotic effects in rat artery between oral administration of Ultra nattokinase$^{(R)}$ and maltodextrin. SD rats were fed with 1.11 mg/kg of Ultra nattokinase$^{(R)}$ for 4 weeks. The effect on arterial thrombosis was then evaluated using an antithrombotic model after induction by $FeCl_3$. Detected fibrinolytic activity was proportional to the content of Ultra nattokinase$^{(R)}$ and statistical extents of the antithrombotic activity was enhanced strongly twice rather than control group. The PT and the aPTT, however, showed only a small difference between two groups. The results suggest that Ultra nattokinase$^{(R)}$ can effectively treat thromboembolism and enhance blood flow, and that Ultra nattokinase$^{(R)}$ can also prevent venous occlusion by aiding clot lysis.

Evaluation of blood clot, platelet-rich plasma, and platelet-rich fibrin-mediated regenerative endodontic procedures in teeth with periapical pathology: a CBCT study

  • Objectives: This study compared the clinical and radiological outcomes of regenerative endodontic procedures (REPs) using blood clots (BCs), platelet-rich plasma (PRP), and platelet-rich fibrin (PRF) through intraoral periapical radiography (IOPAR) and cone-beam computed tomography (CBCT). Materials and Methods: Forty-five single-rooted necrotic teeth with periapical pathology were randomly allocated to receive BC, PRP, or PRF as an individual scaffold. Outcomes were evaluated in 35 teeth in 23 patients with a follow-up period of 12-24 months through qualitative IOPAR scoring and quantitative CBCT measurements. Healing of periapical lesions and in immature teeth, changes in the apical foramen diameter (AFD), root wall thickness (RWT), and root length (RL) were assessed. A p value less than 0.05 was considered to indicate statistical significance. Results: All teeth were asymptomatic except 1 in the PRP group. Periapical lesion healing was seen in all except 2 teeth in the BC group and 3 in the PRP group. Both IOPAR and CBCT revealed no significant differences in bone healing or changes in AFD, RWT, and RL among the 3 groups. A positive pulp sensibility response to the cold test was seen in 2 teeth in the BC group, but none to the electric pulp test. Intracanal calcification (ICC) was evident in more teeth in the BC group than in the PRP and PRF groups, and was also significantly higher in immature teeth. Conclusions: Our results revealed that BC, PRP, and PRF have similar potential as scaffolds in REPs, and ICC may be a concern for long-term outcomes.

Evaluation of Antithrombosis and Antioxidant Activities of the Ethanol Extract of Different Parts of Hibiscus cannabinus L. cv. 'Jangdae' (케나프 장대 품종의 부위별 에탄올 추출물의 항혈전 및 항산화 활성)

  • Kenaf (Hibiscus cannabinus L.), one of the four major fiber crops, is attracting attention for its efficient CO2-absorbing ability and versatility for producing daily supplies, including textiles. In Korea, a new cultivar 'Jangdae' was established in 2013. The ease of cultivation and seed gathering of 'Jangdae' has led to its nationwide cultivation. However, evaluation of the bioactivities of the different parts of kenaf, and especially the 'Jangdae' cultivar, remains rudimentary. In this study, the antithrombosis and antioxidant activities of extracts prepared from different parts of the 'Jangdae' cultivar were evaluated by determining their effects on blood clot formation. Extracts prepared from seeds (HC-SD), seedpods (HC-SP), leaves (HC-L), stems (HC-S), and roots (HC-R) of the 'Jangdae' cultivar strongly inhibited blood clot formation. In particular, the HC-SD, HC-SP, and HC-S extracts showed strong inhibition against the coagulation factors prothrombin, and thrombin. The HC-SP extract showed strong antioxidant activities, such as scavenging ability against DPPH anion, ABTS cation, nitrite, and reducing power. Since blood clot formation is closely related to oxidative stress, the HC-SP extract could be developed as a novel anticoagulation and antioxidant resource. This is the first report of the antithrombosis activities of different parts of H. cannabinus L. cv. 'Jangdae'.

Experimental of Cerebral Vasospasm and Measure the Mean Blood Flow Velocity in the Middle Cerebral Artery

  • To determine the appropriate concentration of papaverine hydrochloride(PPV) for therapeutic intraarterial infusion against cerebral vasospasm and to measure the mean blood flow velocity of the middle cerebral artery in rabbits. Vasospasm was induced in the experimental groups (3 days after infusion; group 1, n=3, 7 days after infusion; group 2, n=3) and a control group (n=l) by placing a blood clot in the subarachnoid space around the top of the internal carotid siphon. PPV (5 mg/kg) was infused into the internal carotid artery. The vascular diameters of the internal carotid artery (ICA) and middle cerebral artery (MCA) were measured on angiograms before and after infusion. The mean blood flow velocity in the MCA was measured on transcranial doppler sonograms before and 24 hours after infusion. After fixation, the MCA was dissected out, stained, and examined microscopically. After PPV infusion in both groups, vascular dilatation of about $20\%$ was seen. The mean increase in blood flow velocity in the group $1(30\%)$ was smaller than in the group $2(70\%)$. The mean blood flow velocity in the MCA decreased by about $30\%$ in both groups, but increased again after 24 hours nearly to the level before PPV infusion. PPV infusion may be more effective in early stages of vasospasm when vascular walls have fewer histologic changes.

  • The purpose of this study was to the observe effects of the various bleaching agents on blood clot. Absorbent papers(N9.5) were coated with O type blood and were dried in shadow for 72 hours. Bleaching agents used were 5% NaOCl 30% $H_2O_2$, saturated solutions of $NaBO_3$ $4H_2O$, $Ca(OCl)_2$ and $Na_2O_2$ at $20^{\circ}C$. After the specimens, blood cloted papers were dipped in 10CC of solution for 1 minute, 5 minutes, 30 minutes, 2 hours and 24 hours, specimens were washed with distilled water and dried. Hue Value/Chroma contrived by Munsell was measured in each specimen. The following results were obtained. 1. Superoxol changes 2.5YR 4.5/2 into N9.5 (white) after 24 hours. 2. All bleaching agents except superoxol change YR into Y.

  • This 58-year-old woman was transferred from a local hospital due to symptoms of acute headache and decreased consciousness. Computed tomography revealed a subarachnoid hemorrhage with blood clot in prepontine cistern. On the first day in the hospital, diagnostic cerebral angiography revealed a basilar tip aneurysm. We performed basilar artery to bilateral posterior cerebral artery[PCA] stent placement to reconstruct the basilar artery apex.

  • Thromboelastography (TEG) analysis consists of ${\alpha}$, G, K, MA, and R types of values and tests the effectiveness of blood clotting, which can be assessed for platelet function, clotting strength, and fibrinolysis. Canine heartworm diseases caused by Dirofilaria immitis, a vascular nematode, can lead to hyperfibrinolysis and hypercoagulation. In this study, G and MA values showed a statistically significant decrease over time after treatment of heartworm disease. Additionally, the ${\alpha}-value$ showed a high correlation with G, K, MA, and R values. The G value showed a high correlation with K and MA values, while The K value showed a high correlation with MA and R values. This study clearly found a gradual decrease in G and MA values in dogs with heartworm disease over time, both before and after treatment. This suggests that the clot formation time is longer and that the intensity of clot formation is lowered and may improve the risk of thromboembolism in dogs with heartworm disease.

Relationship between sonorheometry parameters and laboratory values in a critical care setting in Italy: a retrospective cohort study

  • Purpose: This preliminary retrospective cohort study analyzed the relationship between the parameters provided by sonorheometry device Quantra and the coagulation values obtained from standard venous blood samples in patients admitted in intensive care unit (ICU). Methods: We reviewed medical charts of 13 ICU adult patients in whom at least one coagulation study with Quantra was performed. The relationship between Quantra and laboratory data was analyzed with the Spearman rank correlation coefficient (rho). The 95% confidence interval (CI) was computed. A P-value <0.05 was considered statistically significant. Results: We collected 28 data pairs. Statistically significant moderate correlations were found for the following parameters: clot time (CT) and activated partial thromboplastin time (rho=0.516; 95% CI, 0.123-0.904; P=0.009; clot stiffness (CS) and the international normalized ratio (INR; rho=0.418; 95% CI, 0.042-0.787; P=0.039); INR and platelet contribution to CS (rho=0.459; 95% CI, 0.077-0.836; P=0.022); platelet count and platelet contribution to CS (PCS; rho=0.498; 95% CI, 0.166-0.825; P=0.008); and fibrinogen and fibrinogen contribution to CS (FCS; rho=0.620; 95% CI, 0.081-0.881; P=0.001). Conclusions: Quantra can provide useful information regarding coagulation status, showing modest correlations with the parameters obtained from laboratory tests. During diffuse bleeding, CT and FCS values can guide the proper administration of clotting factors and fibrinogens. However, the correlation of INR with CS and PCS can cause misinterpretation. Further studies are needed to clarify the relationship between Quantra parameters and laboratory tests in the critical care setting and the role of sonorheometry in guiding targeted therapies and improving outcomes.

  • A 3-month-old, female Persian cat was presented with sclera mass. The mass was detected after cat's squeals during the playing with a dog before 2 days. On ophthalmic examination, a $2{\times}5mm$ dark purple colored raised mass was observed 1 mm behind the limbus in the right eye. Anterior chamber was filled with blood and epithelial defect was detected at 2 o'clock in the right central cornea. After general anesthesia, resection of the protruded iris was performed with iris scissors and bipolar cautery. Ruptured sclera and conjunctiva were closed with simple interrupted and simple continuous suture, respectively. Also, irrigation and aspiration (I/A) of the anterior chamber was performed to remove blood clot with bimanual I/A handpiece in the right eye. Four months after the surgery, the right eye was recovered completely and menace response was positive in spite of the retinal lesions.

  • In this experiment, artificial blood vessel was fabricated from electro-spun PU/PCL. To assist with endothelial growth, PU/PCL surface was coated with the RGD peptide. To prevent a clot of blood, anti-thrombus agent was loaded to the fibrous mat and values were reflected through FT-IR data. In vitro study, SEM and MTT data showed that the component was of excellent biocompatibility and cell proliferation. In in vivo study, the artificial blood vessel was implanted in a dog's artery. The results of the CT scan, ultrasound and H&E staining showed that artificial blood vessel was excellent for artery replacement applications.
