INFLUENCE OF SLAUGHTER WEIGHT, SIRE, CONCENTRATE FEEDING AND MUSCLE ON THE PHYSICAL AND CHEMICAL CHARACTERISTICS IN JAPANESE BLACK BEEF

  • Seventeen Japanese Black steers were used to evaluate the influence of slaughter weight (550 kg, 600 kg or 650 kg), sire (two sires), concentrate feeding (barley or corn) and muscle (six muscles) on the following characteristics: pH value, cooking loss, shear value, moisture and crude fat contents. Crude fat contents were higher, and moisture content and shear values were lower in muscles from the large slaughter weight group 650 kg than those from other slaughter weight groups. Cooking loss, shear value, moisture and crude fat contents differed between sires. Corn feeding increased crude fat content in muscle compared to barley feeding. Muscles containing a large amount of intramuscular fat showed lower shear values and less cooking loss than those containing a small amount of intramuscular fat.

Growth Characteristics and Ginsenoside Contents of 4 Years Old Korean Ginseng (Panax ginseng C.A. Meyer) by Shade Materials and Green Manure Crops (해가림자재 종류와 녹비작물 재배에 따른 4년생 인삼의 생육과 진세노사이드 함량)

  • This research carried out to figure out the effect of the green manure crop cultivated at a preparation field and the shading net on the growth, development, and quality of ginseng. Followings are results obtained from the research. Leaf width of ginseng under the shading net of a two-layered blue and two-layered black polythylene net (TBTBPN) was good at rye and hairy vetch cultured group. Leaf length of ginseng under the shading net of a three-layered blue and one-layered black polyethylene net (TBOBPN) was good at barley and hairy vetch cultured group. Meanwhile, leaf width was good at hairy vetch cultured group. Leaf length of ginseng under a blue polyethylene sheet (BPS) was good at a barley and barley + hairy vetch cultured group, but stem length was shorter compare to other shading net cultivations. Root weight of ginseng was good under the shading net of a two-layered blue and two-layered black polyethylene net (TBTBPN) at a rye and hairy vetch cultured group, and was good under the shading net of a three-layered blue and onelayered black polyethylene net (TBOBPN) at a barley + hairy vetch cultured group, but there was no significant difference under blackout screen according to manure crop varieties. Ratio of rusty root was 10.2% at the barley cultured group under the shading net of a two-layered blue and two-layered black polyethylene net (TBTBPN), and was 23.1% at hairy vetch cultured group under shading net of a three-layered blue and one-layered black polyethylene net (TBOBPN). Ratio of rusty root was the lowest at a rye cultured group regardless the shading nets. Content of the ginsenoside was the highest at the rye cultured group under the shading net of two-layered blue and two-layered black polyethylene net (TBTBPN), was the highest at the barley cultured group under the shading net of a three-layered blue and one-layered black polyethylene net (TBOBPN), and was the highest at the rye cultured group under the blackout screen.

Effects of Feeding Total Mixed Rations Containing Different Winter Forage Crop Silages on Feed Intake, Nutrient Digestibility and Blood Characteristics in Korean Black Goats (동계사료작물 사일리지를 첨가한 TMR 급여가 흑염소의 사료섭취량, 영양소 소화율 및 혈액성상에 미치는 영향)

  • This study was conducted to determine the feed value of total mixed rations (TMR) containing different winter forage crop silages on feed intakes, daily gain, nutrient digestibility, and nitrogen retention in Korean black goats. A total of 12 male goats were used in this study and each goat was housed in individual metabolism crates. The treatments were four diets, consisting of whole crop barley silage (T1), rape silage (T2), rye silage (T3), and Italian ryegrass silage (T4). The experiment was designed as a $4{\times}4$ Latin square arrangement in three replicates. Daily intakes for dietary dry matter (DM), organic matter (OM), crude protein (CP) per head were highest in rape silage treatment, and lowest in whole barley silage (p<0.05). Digestible intakes for CP and acid detergent fiber were highest in rape silage treatment. The daily gain of rape, rye, and Italian ryegrass silages were significantly (p<0.05) higher than that of whole crop barley silage. Digestibility of DM. CP, fiber were highest in whole crop barley silage, and lowest in rape silage (p<0.05). Nitrogen (N) intake and N retention were significantly highest in rape silage and lowest in whole crop barley silage (p<0.05). The results showed that, among different winter forage crop silages, rape silage incorporation into TMR had the best performances indicating increases of feed intakes and N retention in Korean black goats.

Antioxidant Content and Activity in Methanolic Extracts from Colored Barley (유색보리 Methanol 추출물의 항산화 성분 및 항산화 활성)

  • We examined the antioxidant content and activity in methanolic extracts from split polished barley (Spb), pearled black barley (Pbb), hulled yellow barley (Hyb), hulled purple barley (Hpb), and hulled black barley (Hbb). The extraction yields of Spb, Pbb, Hyb, Hpb, and Hbb were 2.85, 3.62, 4.62, 4.41, and 6.58%, respectively. The polyphenolic and flavonoid contents were 57.93 and 24.02 for Spb, 64.01 and 27.92 for Pbb, 122.88 and 36.38 for Hyb, 134.94 and 36.51 for Hpb, and 163.43 and 39.70 mg/100 g for Hbb, respectively. The antioxidant activity of hulled barley was significantly higher than that of split or pearled barley. The results of this study show that the antioxidant activity in barley bran has significant health benefits.

PREDICTING MALTING QUALITY IN WHOLE GRAIN MALT COMPARED TO WHOLE GRAIN BARLEY BY NEAR INFRARED SPECTROSCOPY

  • Predicting quality traits using near infrared (NIR) spectroscopy on whole grain samples has gained wide acceptance as a non-destructive, rapid and cost effective technique. Barley breeding programs throughout southern Australia currently use this technology as a tool for selecting malting quality lines. For the past 3 years whole grain barley calibrations have been developed at VIDA to predict malting quality traits in the early generation selections of the breeding program. More recently calibrations for whole grain malt have been developed and introduced to aid in selecting malted samples at the mid-generation stage for more complex malting quality traits. Using the same population set, barley and malt calibrations were developed to predict hot water extracts (EBC and IoB), diastatic power, free $\alpha$-amino nitrogen, soluble protein, wort $\beta$-glucan and $\beta$-glucanase. The correlation coefficients between NIR predicted values and laboratory methods for malt were all highly significant ($R^2$ > 0.84), whereas the correlation coefficients for the barley calibrations were lower ($R^2$ > 0.57) but still significant. The magnitude of the error in predicting hot water extract, diastatic power and wort $\beta$-glucan using whole grain malt was reduced by 50% when compared with predicting the same trait using whole grain barley. This can be explained by the complex nature of attempting to develop calibrations on whole grain barley utilizing malt data. During malting, the composition of barley is modified by the action of enzymes throughout the steeping and germination stages and by heating during the kilning stage. Predicting malting quality on whole grain malt is a more reliable alternative to predicting whole grain barley, although there is the added expense of micro-malting the samples. The ability to apply barley and malt calibrations to different generations is an advantage to a barley breeding program that requires thousands of samples to be assessed each year.

Waxy Black Barley (Hordeum vulgare L.) Cultivar, 'Heuksoojeongchal', with High Yield and Low Amylose Content (아밀로스 함량이 낮은 흑색 다수성 찰보리 '흑수정찰')

  • A new black barley variety, "Heuksoojeongchal," was developed as a cross between "Keunalbori 1" and "Masankwamaek/Mortoni" at the National Institute of Crop Science, RDA in 2014. Heuksoojeongchal had a longer culm length (89 cm) and a later average heading date and maturity than Seodunchalbori. It also had a spike length of 5.0 cm, $543spikes/m^2$, and 54 grains/spike. The 1000-grain weight of Heuksoojeongchal (36.0 g) was heavier than that of Seodunchalbori. Heuksoojeongchal showed resistance to barley yellow mosaic virus (BaYMV). The yield potential of Heuksoojeongchal was approximately 7% higher than that of Seodunchalbori upland, but the average yield potential was similar in upland and paddy fields. Among the quality characteristics, the ${\beta}-glucan$ content (6.7%) was higher and the amylose content (5.5%) was lower than those of Seodunchalbori. The whiteness of the black barley was low, and its absorption rate and spreadability were similar to those of Seodunchalbori.

Rapid and Visual Detection of Barley Yellow Dwarf Virus by Reverse Transcription Recombinase Polymerase Amplification with Lateral Flow Strips

  • Barley yellow dwarf virus (BYDV) has been a major viral pathogen causing significant losses of cereal crops including oats worldwide. It spreads naturally through aphids, and a rapid, specific, and reliable diagnostic method is imperative for disease monitoring and management. Here, we established a rapid and reliable method for isothermal reverse transcription recombinase polymerase amplification (RT-RPA) combined with a lateral flow strips (LFS) assay for the detection of BYDV-infected oat samples based on the conserved sequences of the BYDV coat protein gene. Specific primers and a probe for RT-RPA reacted and optimally incubated at 42℃ for 10 min, and the end-labeled amplification products were visualized on LFS within 10 min. The RT-RPA-LFS assay showed no cross-reactivity with other major cereal viruses, including barley mild mosaic virus, barley yellow mosaic virus, and rice black streaked dwarf virus, indicating high specificity of the assay. The sensitivity of the RT-RPA-LFS assay was similar to that of reverse transcription polymerase chain reaction, and it was successfully validated to detect BYDV in oat samples from six different regions and in individual aphids. These results confirm the outstanding potential of the RT-RPA-LFS assay for rapid detection of BYDV.

Antioxidant activity of black rice and grains (흑미 및 곡류 추출물의 항산화 효과)

  • In this study, the comparison of antioxidative effect of the methanol (MeOH) extracts from grains were investigated in vitro radical scavenging system. Ten grains (black rice, rice, barley, wheat, millet, sorghum, glutinous millet, buckwheat, phellines linteus rice and brown rice) were extracted with MeOH. Among the MeOH extracts of grains, sorghum and black rice showed effective scavenging activities of 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical. The $IC_{50}$ values of sorghum and black rice were $47.4{\mu}g/mL$ and $50.6{\mu}g/mL$, respectively. In addition, black rice also exerted the strongest activities on hydroxyl radical (${\cdot}OH$) scavenging activity. Furthermore, the MeOH extracts of black rice showed effective and dose dependant scavenging activities of DPPH radical and ${\cdot}OH$. These results indicated that black rice showed strong free radical scavenging activity. It suggested that black rice could be a promising natural antioxidant against free radical-induced oxidative damage.

Anticariogenic Effects of Different Types of Tea (음용 차의 항우식 효과)

  • The aim of this study was to investigate the effects of green tea, black tea, barley tea and roasted corn tea used in South Korea to make tea on the formation of Streptococcus mutans biofilm. The aqueous samples of 4 types of tea were extracted from commercial tea bags using cold water ($7^{\circ}C$) or hot water ($72^{\circ}C$). S. mutans UA 159 was introduced into 96-well plates which contained the tea samples and 1% sucrose media. Crystal violet staining was used to assess the effects of teas on S. mutans biofilm formation. In both groups of green tea and black tea, the biofilm significantly decreased with the solution of $50{\mu}L$ aqueous sample in $200{\mu}L$ media and $100{\mu}L$ aqueous sample in $100{\mu}L$ media (p < 0.05). S. mutans biofilm with $25{\mu}L$ of barley tea or roasted corn tea aqueous sample in hot water ($72^{\circ}C$) for 10 minutes showed significant decrease compared of green tea or black tea (p < 0.05). In this study, green tea and black tea suppress the formation of S. mutans biofilm.

Comparison of antioxidant activities of pearled and wholegrain barley harvested in Jeju (도정 유무에 따른 제주산 보리의 항산화 활성 비교)

  • Barley contains health-beneficial compounds, such as phenolic acids, flavonoids, lignans, and tocopherols. The antioxidant activities of pearled and wholegrain hull-less, premature, and black barley harvested in Jeju were investigated by measuring total phenolic content (TPC), total flavonoid content (TFC), phenolic compounds, tocopherols (T), and tocotrienols (T3), along with their in vitro antioxidant activities. Consequently, TPC and TFC in wholegrain barley groups were higher than those in pearled barley. Gallic acid (1.55-2.98 ㎍/g) and protocatechuic acid (0.67-2.84 ㎍/g) were the predominant phenolic compounds in barley. Total T and T3 concentrations of wholegrain barley groups were significantly higher than those of pearled barley (p<0.05). Except for the metal chelating effect and reducing power, the in vitro antioxidant activities of wholegrain barley were significantly higher than those of pearled barley. These results indicate that wholegrain Jeju barley can be used as a natural antioxidant source in the food industry.
