수온과 먹이생물의 농도 변화에 따른 재첩의 여과율 변동 (The Influence of Water Temperature and Food Concentration on the Filtration Rates of the Asiatic Clam, Corbicula fluminea)

  • This study was undertaken with the intent to describe the influence of water temperature and food concentration on the filtration rates of the Asiatic clam, Corbicula fluminea. The clams were collected at Lake Geumho near Yeongsan river, during March 2001. Food organism, Scenedesmus sp. (KMCC FC-34), was indoor-cultured in f/2 medium, and was used to measure the filtration rate of the clams. Filtration rate of the clams was measured by indirect method. Cell concentrations of food organisms were determined by direct counting cells using the hemacytometer under the light microscope. The filtration rate of the clams increased with water temperature up to circa $25^{\circ}C$. Above this temperature, the filtration rate decreased rapidly. The minimal filtration rate of the clams was recorded at $5^{\circ}C$. Thermal coefficient, $Q_10$ values at low temperature range were much higher than those at high temperature range. These results indicate the asiatic clam is more sensitive in cold water like most of marine bivalves. There was a strong reversed correlation between filtration rate and food concentration. Filtration rate of the clams was reduced as food concentration was increased.

  • Heavy metal level and distribution in sediments and bivalves from the tidal flats along the mid-western coast, Korea, were investigated. The averages of heavy metal concentrations in sediments from 5 sampling sites ranged from 86.35-187.77mg/kg for Mn, 13.25-17.73mg/kg for Zn, 6.80-11.85mg/kg for Cr, 4.28-6.25mg/kg for Ni, 1.85-3.09mg/kg for Cu, 0.004-0.020mg/kg for Cd and 5.70-10.41mg/kg for Pb, respectively. Among the mussels of the limited body size(8.5-11cm), heavy metal concentration both in the flesh and in the whole soft parts were not strongly related to the body size. It was shown that, in S. strictus, Cu moves fast into the flesh after absorption, but Pb and Fe move very slowly. Overally, the heavy metal concentration in the flesh tends to increase with that in the whole soft parts. The concentration factors(heavy metal concentration in S. strictus/heavy metal concentration in sediment) showed that, of the examined metals, Cd is the most cumulative in the body of S. strictus, followed by Zn and Cu, while Mn, Cr, Ni and Pb are not cumulative.

Variation in Physiological Energetics of the Ark Shell Scapharca broughtonii (Bivalvia: Arcidae) from Gamak Bay, South Coast of Korea

  • This study presents physiological rates of respiration and excretion, clearance rate, and assimilation efficiency of the ark shell Scapharca broughtonii, determined during 2007 from specimens collected in Gamak Bay on the south coast of Korea. Physiological parameters were measured monthly under static, laboratory controlled conditions with ambient conditions, and measurements were performed seasonally in order to estimate scope for growth and its probable sources of variation. Temperature directly influenced respiration and excretion. Clearance rates showed a tendency to be low during May-August, which is a period of gametogenesis. Assimilation efficiency was not significantly different seasonally and was independent of the concentration of chlorophyll a. The scope for growth was negative during high-temperature months (July-August), reflecting the high temperature and low clearance rate, and had its highest positive values during spring and autumn. The energy budget or growth potential of bivalves has been applied to other economically important species. Data on the physiological parameters and scope for growth of S. broughtonii obtained in this study will be used to assess the carrying capacity for ark shell cultivation.

한국 해산 기회성 연체동물의 분포양상 (Distribution Patterns of Opportunistic Molluscan Species in Korean Waters)

  • 본 연구는 해양환경이 크게 교란을 받고 있는 4개 저서환경을 선택하여 연체동물 중 어떠한 종이 기회종의 특성을 보이는지를 조사하였다. 기회종의 특성을 보인 5종의 이매패류에 대한 분포특성을 파악하고자 하였다. 이매패류인 애기반투명조개 (Theora fragilis), 대양조개(Macoma incongrua), 종밋(Musculista senhousia) 등이 빈 산소 현상을 보이는 반폐쇄성 연안역이나 유기물 축적이 많은 퇴적물을 가진 항만해역에서 우점하여 서식하고 있었으며, 소형 개체인 말발굽조개 (Thyasira tokunagai)와 쇄개량조개 (Raetellops pulchella)는 외해역의 폐기물 해양투기장에서 우점하는 종으로 나타났다.

Corbicula fluminea (Bivalvia: Corbiculidae): a possible second molluscan intermediate host of Echinostoma cinetorchis (Trematoda: Echinostomatidae) in Korea

  • More than 1,500 clams of Corbicula fluminea, the most favorable food source of freshwater bivalves in Korea, were collected from 5 localities to examine cercarial and metacercarial infection with Echinostoma cinetorchis. Although 3 clams infected with suspicious E. cinetorchis metacercariae out of 200 specimens collected at Kangjin, Chollanam-do were detected, no cercarial and metacercarial infections with E. cinetorchis were observed in field-collected Corbicula specimens. In the susceptibility experiments with laboratory-reared clams, those infected with miracidia of E. cinetorchis did not release their cercariae up to 60 days after infection. To confirm the identity of second intermediate host of E. cinetorchis experimentally, a total of 30 clams were exposed to the cercariae from Segmentina hemisphaerula that had been infected with miracidia of E. cinetorchis. The clams were susceptible to cercariae of E. cinetorchis with an infection rate of 93.3%. Metacercariae from clams taken more than 7 days after cercarial exposure were fed to rats (S/D strain), and adult worms of E. cinetorchis, characterized by 37-38 collar spines on the head crown, were recovered from the ileocecal regions. This is the first report of C. fluminea as a possible second intermediate host of E. cinetorchis.

한국연안 진주담치와 굴의 중금속농도에 관한 연구 (Study on the Heavy Metal Concentration in Mussels and Oysters from the Korean Coastal Waters)

  • 1985-1989년간 한국연안의 서로 다른 서식지로부터 채집된 진주담치 Mytilus edulis와 굴 Crassostrea gigas중의 중금속 농도를 알아보았다. 두이매패류에서 가장 높은 중금속농도는 조사점 1, 2, 5, 23 그리고 25에서 발견되었던 반면 가장 낮은 중금속농도는 진해만 주위의 조사점에서 발견되었다. 5개년간 굴과 진주담치의 평균중금속농도비는 Cu 16:1, Cd 1.63:1, Pb 1.36:1, Zn 7.61:1 그리고 Hg 2.08:1로서 굴이 진주담치보다 Cu, Zn, Hg에서 보다 효과적인 생물농축자로 나타났다. 진주담치의 크기별 분포는 크기가 클수록 중금속농도도 증가하였다.

Quantification of Reproductive Output of the Butter Clam, Saxidomus purpuratus(Sowerby, 1852) Using Enzyme-Linked Immunosorbent Assay (ELISA)

  • An immunological method was developed in this study to quantify reproductive output of the female butter clam, Saxidomus purpuratus. A clam egg-specific polyclonal antibody was developed using the purified butter clam egg as an antigen. An indirect Enzyme-Linked Immunosorbent Assay (ELISA) was used in quantitative measurement of the eggs. Size of the butter clam eggs ranged from $70.81{\pm}7.52{\mu}m$ in histology or $88.56{\pm}11.31{\mu}m$ in intact eggs. The predominant egg constituent was protein (37.44%), followed by lipids (11.40%) and carbohydrates (9.68%). The SDS-PAGE showed that the egg proteins are composed of several peptides of molecular weights consisting of 247, 200, 99, 91, 54 and 47 kDa. ELISA indicated that the clams collected from Geoje Island in May 2002 produced 8.2 to 26.8% of their body weight as eggs or 9,307,309 to 31,156,333 with a mean of 16,931,893 eggs per individual clam. The results of this study thus suggest that indirect ELISA using rabbit anti-clam egg IgG as a primary antibody is a rapid, affordable and sensitive method to assess reproductive output of 5. purpuratus and possibly other bivalves using a small amount of eggs.

유세포 분석기를 이용한 바지락(Ruditapes philippinarum)의 면역력 측정 (Flow Cytometric Assessment of Immune Parameters of the Manila Clam (Ruditapes philippinarum))

  • Immmune parameters of the Manila clam Ruditapes philippinarum collected from four tidal flats, Nudong, Gonam, Hwangdo and Bangpo on Anmyeon-do, Korea were optimized and evaluated at the single cell level using flow-cytometry Hemocytes were withdrawn from the sinus of each clam, and total hemocyte counts (THC), phagocytosis rate, hemocyte mortality (HM) and DNA damage of hemocyte were analyzed. The highest hemocyte counts was recorded from the clams collected from Gonam, followed by Hwangdo, Nudong and Bangpo (P<0.001). Phagocytosis rate and hemocyte mortality of Gonam and Nudong clams were significantly higher than those of clams from Hwangdo and Bangpo (P<0.001). DNA damage in the clams from Nudong was higher twice than that of clams from Gonam (P<0.05). We suggest that the flow-cytometry has a high potential for evaluation of immunity of marine bivalves.

바지락독에 관한 연구 (THE SHELLFISH POISON IN TAPES PHILIPPINARUM LISCHKE)

  • Incidents of poisoning caused by edible marine bivalve, Tapes philippinarum in Kojedo, Korea were reported in March 1968 and 1969. The results of the present investigation revealed that the incidents were caused by hemorrhagic diathesis and liver injuries, as those caused by acute yellow atrophic liver. The minimal lethal dose of the liver extract of the bivalve varied with seasons. The dose was 0.02ml in March and increased to 0.25ml in April-May 1969. After lune the extract showed no toxicity. Phthalein reaction of smashed liver solution of the bivalve showed a great variation with seasons. In March the values of $P_1$(crude solution) and $P_2$ (boiled one) were 0.1 and 10, at its strongest toxicity. In May those of $P_1\;and\;P_2$ were 1 and 1,000. However the mouse died after intraperitoneal administration of 0.25ml of the liver extract at the $P_2$ value of 1,000. For this reason, determination of the toxicity is difficult by judging from the phthalein reaction alone. The bivalves collected from Pusan, Kaduk, Koje, Hansan, Yeosu, Mokpo and Kunsan were found nontoxic during April-August 1969.

Oocyte Degeneration Associated with Follicle Cells in Female Mactra chinensis (Bivalvia: Mactridae)

  • Ultrastructural studies of oocyte degeneration in the oocyte, and the functions of follicle cells during oocyte degeneration are described to clarify the reproductive mechanism on oocyte degeneration of Mactra chinensis using cytological methods. Commonly, the follicle cells are attached to the oocyte. Follicle cells play an important role in oocyte degeneration. In particular, the functions of follicle cells during oocyte degeneration are associated with phagocytosis and the intracellular digestion of products. In this study, morphologically similar degenerated phagosomes (various lysosomes), which were observed in the degenerated oocytes, appeared in the follicle cells. After the spawning of the oocytes, the follicle cells were involved in oocyte degeneration through phagocytosis by phagolysosomes. Therefore, it can be assumed that follicle cells reabsorb phagosomes from degenerated oocytes. In this study, the presence of lipid granules, which occurred from degenerating yolk granules, gradually increased in degenerating oocytes. The function of follicle cells can accumulate reserves of lipid granules and glycogen in the cytoplasm, which can be employed by the vitellogenic oocyte. Based on observations of follicle cells attached to degenerating oocytes after spawning, the follicle cells of this species are involved in the lysosomal induction of oocyte degeneration for the reabsorption of phagosomes (phagolysosomes) in the cytoplasm for nutrient storage, as seen in other bivalves.
