  • Plants can recognize and respond in various ways to diverse environmental stresses, including pathogenic microorganisms, salt, drought, and low temperature. Salicylic acid (SA) is one phytohormone that plays important roles in the regulation of plant growth and development. Nonexpressor of pathogenesis-related genes 1 (NPR1) was originally identified as a core protein that could function as a transcriptional co-regulator and SA receptor during systemic acquired resistance (SAR), a plant immune response that could activate PR genes after pre-exposure of a pathogen. Although the function of NPR1 in plant defense response and the role of SA hormone in the regulation of plant physiological processes have been well characterized, the biological role of NPR1 in plant abiotic stress responses is largely unknown. In this review, we will summarize and discuss the current understanding of NPR1 function in response to plant environmental stresses.

Identification and Characterization of Genes Differentially Expressed in the Resistance Reaction in Wheat Infected with Tilletia tritici, the Common Bunt Pathogen

  • The differentially virulent race T1 of common bunt (Tilletia tritici) was used to inoculate the wheat lines Neepawa (compatible) and its sib BW553 (incompatible) that are nearly isogenic for the Bt-10 resistance gene. Inoculated crown tissues were used to construct a suppression subtractive hybridization (SSH) cDNA library. Of the 1920 clones arrayed from the SSH cDNA library, approximately 10% were differentially regulated. A total of 168 differentially up-regulated and 25 down-regulated genes were identified and sequenced; 71% sequences had significant homology to genes of known function, of which 59% appeared to have roles in cellular metabolism and development, 24% in abiotic/biotic stress responses, 3% involved in transcription and signal transduction responses. Two putative resistance genes and a transcription factor were identified among the up regulated sequences. The expression of several candidate genes including a lipase, two non-specific lipid transfer proteins (ns-LTPs), and several wheat pathogenesis-related (PR)-proteins, was evaluated following 4 to 32 days post-inoculation in compatible and incompatible interactions. Results confirmed the higher overall expression of these genes in resistant BW553 compared to susceptible Neepawa, and the differential up-regulation of wheat lipase, chitinase and PR-1 proteins in the expression of the incompatible interaction.

Locating QTLs controlling overwintering seedling rate in perennial glutinous rice 89-1 (Oryza sativa L.)

  • A new cold tolerant germplasm resource named glutinous rice 89-1 (Gr89-1, Oryza sativa L.) can overwinter using axillary buds, with these buds being ratooned the following year. The overwintering seedling rate (OSR) is an important factor for evaluating cold tolerance. Many quantitative trait loci (QTLs) controlling cold tolerance at different growth stages in rice have been identified, with some of these QTLs being successfully cloned. However, no QTLs conferring to the OSR trait have been located in the perennial O. sativa L. To identify QTLs associated with OSR and to evaluate cold tolerance. 286 $F_{12}$ recombinant inbred lines (RILs) derived from a cross between the cold tolerant variety Gr89-1 and cold sensitive variety Shuhui527 (SH527) were used. A total of 198 polymorphic simple sequence repeat (SSR) markers that were distributed uniformly on 12 chromosomes were used to construct the linkage map. The gene ontology (GO) annotation of the major QTL was performed through the rice genome annotation project system. Three main-effect QTLs (qOSR2, qOSR3, and qOSR8) were detected and mapped on chromosomes 2, 3, and 8, respectively. These QTLs were located in the interval of RM14208 (35,160,202 base pairs (bp))-RM208 (35,520,147 bp), RM218 (8,375,236 bp)-RM232 (9,755,778 bp), and RM5891 (24,626,930 bp)-RM23608 (25,355,519 bp), and explained 19.6%, 9.3%, and 11.8% of the phenotypic variations, respectively. The qOSR2 QTL displayed the largest effect, with a logarithm of odds score (LOD) of 5.5. A total of 47 candidate genes on the qOSR2 locus were associated with 219 GO terms. Among these candidate genes, 11 were related to cell membrane, 7 were associated with cold stress, and 3 were involved in response to stress and biotic stimulus. OsPIP1;3 was the only one candidate gene related to stress, biotic stimulus, cold stress, and encoding a cell membrane protein. After QTL mapping, a total of three main-effect QTLs-qOSR2, qOSR3, and qOSR8-were detected on chromosomes 2, 3, and 8, respectively. Among these, qOSR2 explained the highest phenotypic variance. All the QTLs elite traits come from the cold resistance parent Gr89-1. OsPIP1;3 might be a candidate gene of qOSR2.

LC-MS Analysis According to the Combined Treatment of Paenibacillus yonginensis DCY84T and Silicon in Rice

  • We reported in our recent studies that the combined treatment of Paenibacillus yonginensis DCY84T (DCY84T) and Silicon (Si) promotes initial plant growth and increases resistance to biotic and abiotic stress. To understand the molecular background of these phenotypes, Liquid Chromatography Mass Spectrometry (LC-MS) analysis was performed, and it was confirmed that unsaturated fatty acid metabolites such as oleic acid and linoleic acid decreased in response to the combined treatment of DCY84T and Si. The stearoyl-acyl carrier protein desaturase (SACPD) introduces the cis double bond into the acyl-ACPs at C9, resulting in the production of unsaturated fatty acid. We identified OsSSI2 encoding SACPD in rice and found that the expression of OsSSI2 was reduced under DCY84T and Si treatment. Furthermore, qRT-PCR analysis revealed that the expression of OsWRKY45, which is downstream of OsSSI2, was upregulated in response to DCY84T and Si treatment. These results enable the speculation that activation of the salicylic acid (SA)-responsive gene, OsWRKY45, may contribute to enhancing biological stress resistance. Based on this, we propose a probable model for the rice defense pathway following DCY84T and Si treatment. This model retains a WRKY45-dependent but NH1(NPR1)-independent SA signaling pathway.

A WUSCHEL Homeobox Transcription Factor, OsWOX13, Enhances Drought Tolerance and Triggers Early Flowering in Rice

  • Plants have evolved strategies to cope with drought stress by maximizing physiological capacity and adjusting developmental processes such as flowering time. The WOX13 orthologous group is the most conserved among the clade of WOX homeodomain-containing proteins and is found to function in both drought stress and flower development. In this study, we isolated and characterized OsWOX13 from rice. OsWOX13 was regulated spatially in vegetative organs but temporally in flowers and seeds. Overexpression of OsWOX13 (OsWOX13-ov) in rice under the rab21 promoter resulted in drought resistance and early flowering by 7-10 days. Screening of gene expression profiles in mature leaf and panicles of OsWOX13-ov showed a broad spectrum of effects on biological processes, such as abiotic and biotic stresses, exerting a cross-talk between responses. Protein binding microarray and electrophoretic mobility shift assay analyses supported ATTGATTG as the putative cis-element binding of OsWOX13. OsDREB1A and OsDREB1F, drought stress response transcription factors, contain ATTGATTG motif(s) in their promoters and are preferentially expressed in OsWOX13-ov. In addition, Heading date 3a and OsMADS14, regulators in the flowering pathway and development, were enhanced in OsWOX13-ov. These results suggest that OsWOX13 mediates the stress response and early flowering and, thus, may be a regulator of genes involved in drought escape.

Physiology and Gene Expression Analysis of Tomato (Solanum lycopersicum L.) Exposed to Combined-Virus and Drought Stresses

  • Crop productivity can be obstructed by various biotic and abiotic stresses and thus these stresses are a threat to universal food security. The information on the use of viruses providing efficacy to plants facing growth challenges owing to stress is lacking. The role of induction of pathogen-related genes by microbes is also colossal in drought-endurance acquisition. Studies put forward the importance of viruses as sustainable means for defending plants against dual stress. A fundamental part of research focuses on a positive interplay between viruses and plants. Notably, the tomato yellow leaf curl virus (TYLCV) and tomato chlorosis virus (ToCV) possess the capacity to safeguard tomato host plants against severe drought conditions. This study aims to explore the combined effects of TYLCV, ToCV, and drought stress on two tomato cultivars, Money Maker (MK, UK) and Shalala (SH, Azerbaijan). The expression of pathogen-related four cellulose synthase gene families (CesA/Csl) which have been implicated in drought and virus resistance based on gene expression analysis, was assessed using the quantitative real-time polymerase chain reaction method. The molecular tests revealed significant upregulation of Ces-A2, Csl-D3,2, and Csl-D3,1 genes in TYLCV and ToCV-infected tomato plants. CesA/Csl genes, responsible for biosynthesis within the MK and SH tomato cultivars, play a role in defending against TYLCV and ToCV. Additionally, physiological parameters such as "relative water content," "specific leaf weight," "leaf area," and "dry biomass" were measured in dual-stressed tomatoes. Using these features, it might be possible to cultivate TYLCV-resistant plants during seasons characterized by water scarcity.

Reduction of Stress Caused by Drought and Salt in Rice (Oryza sativa L.) Crops through Applications of Selected Plant Extracts and the Physiological Response Mechanisms of Rice

  • In many areas of the world, salt damage and drought have had a negative impact on human survival due to a decrease in agricultural productivity. For instance, about 50% of agricultural land will be affected by salt damage by 2050. Biostimulants such as plant extracts can not only increase the nutrient utilization efficiency of plants, but also promote plant growth and increase resistance to abiotic or biotic stress. Therefore, the objective of this study was to determine how selected plant extracts might reduce levels of stress caused by drought and salt and to better understand the physiological response mechanisms of rice plants. In this study, we used Soybean leaves, Soybean stems and Allium tuberosum, Allium cepa, Hizikia fusiforme, and Gracilaria verrucosa extracts were used. These extracts had been used in previous studies and were found to be effective. The materials were dried in a dry oven at 50℃ for 5 days and ground using a blender. Each 50 g of materials was put in 1 L of distilled water, stirred for 24 hours, filtered using 4 layers of mirocloth, and then concentrated using a concentrator. Rice (cv. Hopumbyeo) seeds were immersed and germinated, and then sown in seedbeds filled with commercial soil. In drought experiments, three rice seedlings at 1 week after seeding was transplanted into 100 ml cups filled with commercial soils and grown until the 4-leaf stage. For this experiment, the soil weight in a cup was equalized, and water was allowed to become 100% saturated and then drained for 24 hours. Thereafter, plant extracts at 3% concentrations were applied to the soils. For NaCl treatments, rice plants at 17 days after seeding were treated with either 100 mM NaCl or plant extracts at 1%+ 100 mM NaCl combinations in the growth chamber. Leaf injury, relative water content, photosynthetic efficiency, and chlorophyll contents were measured at 3, 5, and 6 days after treatments. Shoot fresh weight of rice under drought conditions increased 28-37% in response to treatments of Soybean leaf, Soybean stem, Allium tuberosum, Allium cepa, Hizikia fusiforme, and Gracilaria verrucosa extracts at 3% when compared with control plants. Shoot fresh weight of rice subjected to 100 mM NaCl treatments also increased by 6-24% in response to Soybean leaf, Soybean stem, Allium tuberosum, Allium cepa, Hizikia fusiforme, and Gracilaria verrucosa extracts at 3% when compared with control plants. Compared to the control, rice plants treated with these six extracts and subjected to drought conditions had significantly higher relative water content, Fv/Fm, total chlorophyll and total carotenoids than control plants. With the exception of relative water contents, rice plants treated with the six extracts and subjected to salt stress (100 mM NaCl treatments) had significantly higher Fv/Fm, total chlorophyll and total carotenoids than control plants. However, the type of extract used did not produce significant difference in these parameters. Thus, all the plant extracts used in this study could mitigate drought and NaCl stresses and could also contribute substantially to sustainable crop production.

Weed Population Dynamics under Climatic Change

  • This paper provides some of the scientific background on how projected environmental conditions could affect weeds and weed management in crops. Elevated $CO_2$ levels may have positive effects on crop competitiveness with $C_4$ weeds, but these are generally outnumbered by $C_3$ species in weed populations. Moreover, higher temperatures and drought will favor $C_4$ over $C_3$ plants. The implementation of climate change adaptation technologies, such as drought-tolerant germplasm and water-saving irrigation regimes, will have consequences for crop-weed competition. Rainfed production systems are thought to be most vulnerable to the direct effects of climate change and are likely to face increased competition from $C_4$ and parasitic weeds. Biotic stress-tolerant crop cultivars to be developed for these systems should encompass weed competitiveness and parasitic-weed resistance. In irrigated systems, indirect effects will be more important and weed management strategies should be diversified to lessen dependency on herbicides and mechanical control, and be targeted to perennial rhizomatous ($C_3$) weeds. Water-saving production methods that replace a weed-suppressive floodwater layer by intermittent or continuous periods of aerobic conditions necessitate additional weed management strategies to address the inherent increases in weed competition. Thus, climatic conditions have a great effect on weed population dynamics all over the world.

A Review of Hyperspectral Imaging Analysis Techniques for Onset Crop Disease Detection, Identification and Classification

  • Recently, intensive research has been conducted to develop innovative methods for diagnosing plant diseases based on hyperspectral technologies. Hyperspectral analysis is a new subject that combines optical spectroscopy and image analysis methods, which makes it possible to simultaneously evaluate both physiological and morphological parameters. Among the physiological and morphological parameters are classifying healthy and diseased plants, assessing the severity of the disease, differentiating the types of pathogens, and identifying the symptoms of biotic stresses at early stages, including during the incubation period, when the symptoms are not visible to the human eye. Plant diseases cause significant economic losses in agriculture around the world as the symptoms of diseases usually appear when the plants are infected severely. Early detection, quantification, and identification of plant diseases are crucial for the targeted application of plant protection measures in crop production. Hence, this can be done by possible applications of hyperspectral sensors and platforms on different scales for disease diagnosis. Further, the main areas of application of hyperspectral sensors in the diagnosis of plant diseases are considered, such as detection, differentiation, and identification of diseases, estimation of disease severity, and phenotyping of disease resistance of genotypes. This review provides a deeper understanding, of basic principles and implementation of hyperspectral sensors that can measure pathogen-induced changes in plant physiology. Hence, it brings together critically assessed reports and evaluations of researchers who have adopted the use of this application. This review concluded with an overview that hyperspectral sensors, as a non-invasive system of measurement can be adopted in early detection, identification, and possible solutions to farmers as it would empower prior intervention to help moderate against decrease in yield and/or total crop loss.

소득작물 전후작용 단기성 벼 품종 "금오3호" (A New Short Growth-Duration Rice Cultivar, "Keumo 3")

  • "금오3호"는 소득작물 전 후작에 적응하는 벼를 육성하고자, 1997년 국립식량과학원 기능성작물부에서 내도복성인 농안벼에 내만식성인 신금오벼가 여교배된 $F_1$ 개체에 도열병에 저항성이고 단기성인 YR15727-B-B-B-102 계통을 교배하여 YR19124 조합을 작성 하였다. 이후 집단 및 계통육종법으로 YR19124-B-B-B-69-3 계통을 선발하고 밀양201호라는 계통명으로 지역적응성검정 시험에 공시한 결과, 그 주요특성과 수량성은 다음과 같다 1. 출수기는 평야지의 소득작물 후작 만기재배에서 8월 26일로 금오벼 보다 4일 빠른 조생종 품종이다. 2. 직립 초형이고 탈립은 약간되는 편이고, 이삭추출은 양호 하고 까락이 거의 없는 편이다. 3. 주당수수는 금오벼 보다 약간 많으며, 수당립수는 적은편이고 현미천립중은 금오벼와 비슷하였다. 4. 도정특성은 금오벼와 비슷하지만 고품질 상품에서 중요한 완전미율과 외관특성은 금오벼 보다 좋은 편이고 밥맛은 금오벼 보다 떨어 젓다. 5. 못자리 육묘기간이 50일 이상이면 본답에서 불시출수가 발생하며, 성숙기 엽노화는 금오벼 보다 빠른 편이다. 내냉성 검정에서 냉수구의 임실율이 금오벼 보다 낮았다. 6. 잎도열병 밭못자리 검정 결과 12개 시험지에서 강한 반응을 보였고, 내구저항성도 높았다. 흰잎마름병 레이스 $K_1$, $K_2$, $K_3$에는 저항성이며, $K_{3a}$에는 이병성이다. 줄무늬잎마름 병에는 저항성이며, 오갈병에는 중도 저항성이고, 검은줄오갈병에는 이병성이다. 내충성에서는 끝동매미충에 저항성이고, 벼멸구와 애멸구에는 감수성이다. 7. 지역적응시험에서 "금오3호"의 쌀 수량성은 4.48 MT/ha로 표준품종 금오벼 대비 6% 증수하였다.
