Animal Cell Culture and the Production of Monoclonal Antibody(MAb) Using Biopolymer Membrane (생물고분자 막 형성을 이용한 동물세포 배양 및 단클론항체 생산)

  • Biopolymer membrane was prepared using two oppositely charged natural biopolymers. The biopolymer membrane was used for the encapsulation of two hybridoma cell lines(ATCC CRL-1606, ATCC HB-8852) to produce monoclonal antibodies. In order to reduce the down stream steps, the pre size of the membrane was controlled to retain the monoclonal antibodies in the capsules based on the diffusion experiments with standard proteins. T-flask culture showed cell densities of 8$\times$107 cells/mL and 3$\times$107 cells/mL, and MAb concentrations of 506$\mu$g/mL and 109$\mu$g/mL for encapsulated ATCC CRL-1606 and HB-8852, respectively. Two liter perfusion cultures with encapsulated ATCC HB-8852 were performed to enhance the MAb production. The MAb production of the encapsulated hybridoma increased considerably comparing to the culture using silicon tubing for oxygen transfer.

  • In order to prove physiological function of biopolymer from Bacillus coagulans CE-74, in vitro experiments simulating the passive membrane transport of gastrointestinal tract were carried out using dialysis membrane. And inhibition effect of isolated biopolymer on tyrosinase and angiotensin converting enzyme (ACE) were observed. The glucose retardation index after 30 min dialysis was 43.5% in the presence of 2% biopolymer. As the dialysis period became longer, the retarding effect toward glucose absorption decreased and the effect was close to zero after 5 hr dialysis. The bile acid retardation index after 30 min dialysis was 34% and 44.2% in the presence of 1% and 2% biopolymer, respectively. The effect decreased as the dialysis time elapsed. It was measured by arosinase inhibition activity of biopolymer that inhibition effect was 48.5% in $20\;{\mu}g/{\mu}l$. In a ACE inhibition activity, biopolymer showed inhibition activity as 97% in $10\;{\mu}g/{\mu}l$.

  • Biopolymer is a versatile material used in food processing, medicine, construction, and soil reinforcement. 𝛽-glucan is one of the biopolymers that improves the soil water content and ion adsorption in a drought or toxic metal contaminated land for plant survival. We analyzed drought stress damage reduction in sweet potatoes (Ipomoea batatas L. cv. Sodammi) by measuring the growth and major protein expression and activity under 𝛽-glucan soil amendment. The result showed that sweet potato leaf length and width were not affected by drought stress for 14 days, but sweet potatoes grown in 𝛽-glucan-amended soil showed an effect in preventing wilting caused by drought in phenotypic changes. Under drought stress, sweet potato leaves did not show any changes in electrolyte leakage, but the relative water content was higher in sweet potatoes grown in 𝛽-glucan-amended soil than in normal soil. 𝛽-glucan soil amendment increased the expression of plasma membrane (PM) H+-ATPase, but it decreased the aquaporin PIP2 (plasma membrane intrinsic protein 2) in sweet potatoes under drought stress. Moreover, water maintenance affected the PM H+-ATPase activity, which contributed to tolerance under drought stress. These results indicate that 𝛽-glucan soil amendment improves the soil water content during drought and affects the water supply in sweet potatoes. Consequently, 𝛽-glucan is a potential material for maintaining soil water contents, and analysis of the major PM proteins is one of the indicators for evaluating the biopolymer effect on plant survival under drought stress.

Cell fusion and fusants characterization of Bacillus strains producing biopolymer (Biopolymer 생산성 Bacillus 속 균주의 세포융합과 융합주의 세포특성)

  • To improve biopolymer productivity and properties of Bacillus strains, protoplast fusion was performed between biopolymer producing Bacillus subtilis K-1 and lactose utilizing Bacillus coagulans. The results were as follows; Protoplasts mixed in fusion fluid containing 33% PEG 6000, 1% PVP and 10 mM $CaCl_2$ were reacted for 5 min at $37^{\circ}C$ and then centrifused protoplasts were directly overlaid on the selective media containing $100\;{\mu}g/ml$ antibiotics and incubated for 3 days. At this conditions, the frequency of protoplast fusion was generally in the range of $4.6{\times}10^{-5}\;to\;1.8{\times}10^{-7}$ in ratio. Segregation ratio was observed between 1 to 6% indicating genetic stability of all the fusants. Fusants growth were also observed on the media contained amino acid and antibiotics as required marked materials. DNA contents of the selected fusants were 1.6 to 2.7 times more than that of parent strains. With observation by TEM microscopy, spherical protoplasts were first released from the swollen parental cells and then contracted to fuse in the process of fusion. And fused cells were observed representative vesicle. Originally, the parental cells were observed as in the morphology of thick-walled and double membrane-surrounded rod shape with TEM microscopy.

Influence of biodegradable polymer membrane on new bone formation and biodegradation of biphasic bone substitutes: an animal mandibular defect model study

  • Purpose: The purpose of this study was to evaluate the influence of biodegradable polycaprolactone membrane on new bone formation and the biodegradation of biphasic alloplastic bone substitutes using animal models. Materials and methods: In this study, bony defect was formed at the canine mandible of 8 mm in diameter, and the defects were filled with Osteon II. The experimental groups were covered with Osteoguide as barrier membrane, and the control groups were closed without membrane coverage. The proportion of new bone and residual bone graft material was measured histologically and histomorphometrically at postoperative 4 and 8 weeks. Results: At 4 weeks, the new bone proportion was similar between the groups. The proportion of remaining graft volume was 27.58 ± 6.26 and 20.01 ± 4.68% on control and experimental groups, respectively (P < 0.05). There was no significant difference between the two groups in new bone formation and the amount of residual bone graft material at 8 weeks. Conclusion: The biopolymer membrane contributes to early biodegradation of biphasic bone substitutes in the jaw defect but it does not affect the bone formation capacity of the bone graft.

Preparation and Applicaitons of Synthetic Fish Egg Capsules from Marine Polysaccharides (해양추출 다당류를 이용한 인조어란 캡슐의 제조 및 응용)

  • The biopolymer membrane could be formed using marine polysaccharides. Chiotsan and alginate were used for the formation of capsule membrane to mimic the fish eggs such as flying fish eggs and salmon eggs. The size of capsules ranging 1 to 5mm was prepared and the mechanical tests were performed to determine the mechanical similarities to natural fish eggs. The similar mechanical pattern between the synthetic capsules and natural eggs could be found. The controlling parameters for the strength of capsules were pH of the chitosan solution. This encapsulation technique can be broadly applied to medical, engineering as well as food areas.

  • This study was performed to discover the optimum operation conditions for the advanced water treatment using the ceramic membrane, introduced the first in the nation at the Y water treatment plant (WTP). The result of investigation to find the optimum operation conditions which can continue preserving the filtration performance as well as satisfying both the economics and the water quality is as follows. In the ordinary water quality condition of the Y WTP, the optimum filtration time(the backwash period), which can minimize the production of backwash waste and preserve the membrane performance was examined to be 4.0 hours on basis of institution capacity ($16,000m^3/day$). Examining the recovery rate of TMP from the chemical cleaning (CIP) discovered that the inorganic contaminants, which cause membrane fouling, such as iron, manganese, aluminum, were removed through the acidic cleaning using citric acid, whereas the membrane recovery rate was found to be low. But, on the other hand, the TMP was recovered to the initial value from the alkali cleaning using the NaOCl. Therefore, the main contaminant causing the fouling was determined to be hydrophilic organic compound( biopolymer). The membrane recovery rate is highly influenced by the temperature of the cleaning chemical. That is, the rate increased with increasing temperature.

  • Some biodegradable polymers and other materials such as hydrogels have shown the promising potential for surgical applications. Post surgical adhesion caused by the natural consequence of surgical wound healing results in repeated surgery and harmful effects. Recently, scientists have developed absorbable anti-adhesion barriers that can protect a tissue from adhesion in case they are in use; however, they are dissolved when no longer needed. Although these approaches have been attempted to fulfill the criteria for adhesion prevention, none can perfectly prevent adhesions in all situations. Overall, we developed a new method to fabricate an anti-adhesion membrane using biodegradable polymer and hydrogel. It employed a highly accurate three-dimensional positioning system with pressure-controlled syringe to deposit biopolymer solution. The pressure-activated microsyringe was equipped with fine-bore nozzles of various inner-diameters. This process allowed that inner and outer shapes could be controlled arbitrarily when it was applied to a surgical region with arbitrary shapes. In order to fulfill the properties of the ideal barriers f3r preventing postoperative adhesion, we adopted the pre-mentioned method combined with surface modification with the hydrogel coating by which anti-adhesion property was improved.
