Effects of Arbuscular Mycorrhizal Fungi and Soil Conditions on Crop Plant Growth

  • We investigated the effects on various crops of inoculation with species of arbuscular mycorrhizal fungi (AMF) in soils from different sources and selected AMF species suitable for domestic environment-friendly farming. Effects on plants varied with the AMF species used. In carrot, Scutellospora heterogama, Acaulospora longula, and Funneliformis mosseae had a positive effect on growth of the host, whereas AMF had only weak effects on the growth of red pepper and leek. AMF inoculation had positive effects on the growth of carrot and sorghum. The results of this study indicate the nature of the relationship between soil, plants, and AMF; this study therefore has important implications for the future use of AMF in environment-friendly agriculture.

Unreported Endophytic Fungi Isolated from the Leaves of Evergreen Woody Plants in Korea: Muyocopron lithocarpi and Didymocyrtis cladoniicola

  • In this study, endophytic fungi were isolated from the leaves of evergreen woody plants in Hansando Island in South Korea. The fungal isolates were identified based on morphological characteristics and phylogenetic analysis of the DNA sequences of the internal transcribed spacer and the large subunit regions of rDNA. It was confirmed that two endophytic fungal species, Muyocopron lithocarpi and Didymocyrtis cladoniicola, isolated in this study have not previously been reported in South Korea.

  • We isolated endophytic fungi from the roots of Quercus spp. in Korea. The isolates were identified based on morphological and molecular characteristics from the nucleotide sequences of the internal transcribed spacer and large subunit of rDNA regions. We confirmed that the four species we isolated - Oidiodendron citrinum, Acremonium crotocinigenum, Mammaria echinobotryoides, and Pleotrichocladium opacum - have not been recorded previously in Korea. Herein, we present their morphological characteristics and the result of phylogenetic analysis.

  • Endophytic fungi were isolated from needle leaves of two conifer species in Korea. The fungal isolates were identified based on their morphological characteristics and phylogenetic analyses of their internal transcribed spacer and large-subunit rDNA regions. Three species of endophytic fungi, namely Celosporium laricicola, Neocatenulostroma germanicum, and Phaeophleospora eucalypticola were the first records in Korea. In this study, we reported the morphological and phylogenetic characteristics of these fungi.

Human Intersectin 2 (ITSN2) binds to Eps8 protein and enhances its degradation

  • Participates in actin remodeling through Rac and receptor endocytosis via Rab5. Here, we used yeast two-hybrid system with Eps8 as bait to screen a human brain cDNA library. ITSN2 was identified as the novel binding factor of Eps8. The interaction between ITSN2 and Eps8 was demonstrated by the in vivo co-immunoprecipitation and colocalization assays and the in vitro GST pull-down assays. Furthermore, we mapped the interaction domains to the region between amino acids 260-306 of Eps8 and the coiled-coil domain of ITSN2. In addition, protein stability assays and immunofluorescence analysis showed ITSN2 overexpression induced the degradation of Eps8 proteins, which was markedly alleviated with the lysosome inhibitor NH4Cl treatment. Taken together, our results suggested ITSN2 interacts with Eps8 and stimulates the degradation of Eps8 proteins.

Exosomes derived from microRNA-584 transfected mesenchymal stem cells: novel alternative therapeutic vehicles for cancer therapy

  • Exosomes are small membranous vesicles which contain abundant RNA molecules, and are transferred from releasing cells to uptaking cells. MicroRNA (miRNA) is one of the transferred molecules affecting the adopted cells, including glioma cells. We hypothesized that mesenchymal stem cells (MSCs) can secrete exosomes loading miRNA and have important effects on the progress of gliomas. To determine these effects by treating exosomal miRNA in culture media of miRNA mimic transfected MSCs, we assessed the in vitro cell proliferation and invasion capabilities, and the expression level of relative proteins associated with cell apoptosis, growth and migration. For animal studies, the mice injected with U87 cells were exposed to exosomes derived from miRNA-584-5p transfected MSCs, to confirm the influence of exosomal miRNA on the progress of glioma. Based on our results, we propose a new targeted cancer therapy wherein exosomes derived from miRNA transfected MSCs could be used to modulate tumor progress as the anticancer vehicles.
