Neotypification of Veronica pusanensis (Scrophulariaceae)

  • A type specimen is the specimen originally used to name a new species; thus, it is one of the most significant materials for taxonomic study. In our study on the inventory and management of endemic species in Korea, we checked the type materials for Pseudolysimachion pusanensis (Y. Lee) Y. Lee, combination name Veronica pusanensis Y. Lee, recorded as deposited at the herbaria of the Ewha Womans University and the National Institute of Biological Resources, and failed to find any specimens. Thus, we concluded that all type specimens of V. pusanensis have since been misplaced. According to articles 9.11 and 9.13 of the International Code of Nomenclature for algae, fungi, and plants, it is necessary to select a neotype if the holotype is missing and no other original material exists. Therefore, we designate the neotype maintained in the herbarium of the National Institute of Biological Resources of the Republic of Korea.

A Basic Study on the Variation of Temperature Characteristics for Attenuation Coefficient and Sound Velocity in Biological Tissues

  • This study is concerned with the temperature dependence characteristics of ultrasound parameters in biological tissues, which are basic on the noninvasive deep body temperature estimation. Used parameters are ultrasonic attenuation coefficient and sound velocity In order to accomplishment our purpose, several signal processing methods were used. Attenua4iorl coefficient was estimated by spectral difference method and sound velocity was estimated by P-P method. And we also examined these methods through a series of IN VITRO experi mentis that used tissue-mimicking phantom samples and biological tissue samples. In order to imitate the biological soft tissue two kinds of phantom samples are used, one is agar phantom sample which is composed of agar, graphite, N-propyl alcohol and distilled water, and the other is fat phantom sample which is composed of pure animal fat. And the ultrasound transmission mode and reflection mode experiments are performed on the pig's spleen, kidney and fat. As a result, it is found that the temperature characteristics are uniform in case of phan- tom samples but not in biological tissues because of complicate wave propagation within them. Consequently, the possibility of temperature measurement using ultrasound on biological tissue is confirmed and its results may contribute to the establishment of reference values of internal temperature measurement of biological tissues.

Three New Records of Mortierella Species Isolated from Crop Field Soil in Korea

  • Three new fungal species of the genus Mortierella, Mortierella zychae, Mortierella ambigua, and Mortierella indohii, have been reported in Korea. The fungi were encountered during a study on the fungal community of soil samples collected from different locations in Korea. The species were identified based on molecular and morphological analyses. This study presents detailed descriptions of the morphological observations and molecular phylogenetic analysis of these three fungi. All three species were found to be sensitive to triphenyltetrazolium chloride staining. M. zychae demonstrated the highest intensity of mycelial staining, indicating that this species has the highest potential to produce arachidonic acid of the three species. The staining results indicated that the newly recorded species could potentially be useful for arachidonic acid production.

Melanogenesis Inhibitory Activity of Epicatechin-3-O-Gallate Isolated from Polygonum amphibium L.

  • This study aimed to investigate the melanogenesis inhibitory activity of epicatechin-3-O-gallate (ECG) isolated from Polygonum amphibium L. ECG was isolated from the ethanol extract of P. amphibium L, and its chemical structure was determined using spectroscopic methods such as LC-ESI-MS, 1D-NMR, and UV spectroscopy. ECG inhibited the melanogenesis of B16F10 cells in a dose-dependent manner. Particularly, it decreased the melanin content by 27.4% at 200 µM concentration, compared with the control, in B16F10 cells, without causing cytotoxicity. It is noteworthy that the expression of three key proteins, including tyrosinase, tyrosinase-related protein-1 (TRP-1), TRP-2, and microphthalmia-associated transcription factor (MITF), involved in melanogenesis, is significantly inhibited by ECG. The ECG isolated in this study caused the inhibition of body pigmentation and tyrosinase activity in vivo in the zebrafish model. These results suggest that the ECG isolated from P. amphibium L. is an effective anti-melanogenesis agent.

Morphological and Molecular Characterization of the Newly Reported Penicillium pimiteouiense from Field Soil in Korea

  • Penicillium pimiteouiense was discovered in South Korea during an investigation of fungal communities in soil collected from the Gyeongsangbuk-do province. In this study, we performed molecular analysis of this fungal isolate using internal transcribed spacer rDNA, β-tubulin, and Calmodulin gene sequences. We also performed morphological analysis using five agar media, potato dextrose, oatmeal, malt extract, czapek yeast extract, and yeast extract sucrose. In this study, the molecular and morphological analyses of P. pimiteouiense with detailed descriptions and figures has been carried out.

Five New Wood Decay Fungi (Polyporales and Hymenochaetales) in Korea

  • The wood decay fungi are a diverse taxonomic group that plays a pivotal role in forest carbon cycling. Wood decay fungi use various enzymatic pathways to digest dead or living wood in order to obtain carbon and other nutrients and these enzymatic systems have been exploited for both industrial and medical applications. Over 600 wood decay fungi species have been described in Korea; however, the recent application of molecular markers has dramatically altered the taxonomy of many of these wood decay fungi at both the genus and species levels. By combining molecular methods, specifically sequences of the internal transcribed spacer region, with traditional morphological characters, this study identified five new species records for Korea in five genera: Aurantiporus, Favolus, Neofavolus, Loweomyces, and Hymenochaetopsis. Three of these genera (Aurantiporus, Favolus, and Loweomyces) were previously unknown in Korea. The relatively simple morphology of the wood decay fungi often leads to ambiguous taxonomic assignment. Therefore, molecular markers are a necessary component of any taxonomic or evolutionary study of wood decay fungi. Our study highlights the need for a more robust and multifaceted approach in investigating new wood decay fungi in Korea.

Freshwater Invertebrates of Jindo Island in Korea

  • We surveyed the freshwater invertebrates of Jindo Island twice on July and September 2016, as part of a joint faunal survey conducted to commemorate the 30th anniversary of the Korean Society of Systematic Zoology. Aquatic insects were not included in this study. We found 38 freshwater species from seven phyla: Porifera, Platyhelminthes, Nematomorpha, Mollusca, Annelida, Arthropoda, and Bryozoa. Twenty-one of the 38 species (55.3%) represented the first record of their respective species on Jindo Island. Among these species, a freshwater shrimp-parasitic isopod, Tachaea n. sp., was identified as a new species.

Interrelationship of Biological Yield and Harvest Index in Mulberry and Its Association with Shoot Rearing in Silkworm (Bombyx mori L.)

  • Studies on the interrelationship between biological yield and harvest index, and their association with shoot rearing were carried out in 9 promising mulberry genotypes, which indicated significant variations among genotypes. Biological yield had close association with leaf yield components indicating that they are inter-related for higher productivity in mulberry, Biological yield revealed highly significant positive correlation with important leaf yield components. However height of the longest branch showed negative correlation with biological yield. Genotypes with higher biological yield associated with low ratio of stem weight to leaf weight and high Harvest index on dry weight basis are to be selected for shoot rearing. Biological yield recorded on fresh weight basis had less impact on the present study in comparison to dry weight basis.

Improvement of Biological Control against Bacterial Wilt by the Combination of Biocontrol Agents with Different Mechanisms of Action

  • Despite the increased interests in biological control of soilborne diesease for environmental protection, biological control of bacterial wilt caused by Ralstonia solanacearum have not provided consistent or satisfying results. To enhance the control efficacy and reducing the inconsistency and variability, combinations of specific strains of microorganisms, each having a specific mechanism of control, were applied in this study. More than 30 microorganisms able to reduce the activity of pathogen by specific mechanism of action were identified and tested for their disease suppressive effects. After in vitro compatibility examinations, 21 individual strains and 15 combinations were tested in the greenhouse. Results indicated three-way combinations of different mode of control, TS3-7+A253-16+SKU78 and TS1-5+A100-1+SKU78, enhanced disease suppression by 70%, as compared to 30-50% reduction for their individual treatments. This work suggests that combining multiple traits antagonizing the pathogen improve efficacy of the biocontrol agents against Ralstonia solanacearum.
