해양균류의 Tyrosinase 저해활성 검색 (Screening of Tyrosinase Inhibiting Activity from the Marine-Derived Fungus)

  • In order to screen new tyrosinase inhibiting principle which is expected to be a new biofunctional skin whitening cosmetics, we have isolated 600 strains of the marine- derived fungi and investigated tyrosinase inhibiting activity for their acetone extracts. The significant activities (>70% Inhibition) were observed in the extract of 10 strains of fungi (MFA7, MFA27, MFA58, MFA317, MFA318, MFA345, MFA412, MFA552, MFA562, MFA581). These active strains were cultured in SWS medium with 1 L scale and the resulting broth and mycelium were extracted to afford mycelium extract (000M) and broth extract (000B), respectively. Tyrosinase inhibiting activity for all extracts has been tested. As the results, the broth extracts of 4 strains (27B, 58B, 552B and 581B) exhibited relatively high levels of activity of $IC_{50}$ values of $3.0-19.0\;{\mu}g/mL$. The active component of 581B was purified by assay-guided isolation to yield the known kojic acid (1), and its structure was determined by physicochemical evidence. Kojic acid showed the significant tyrosinase inhibitory activity with $IC_{50}$ values of $12.0\;{\mu}M$.

Biofunctional Properties of Enzymatic Squid Meat Hydrolysate

  • Squid is one of the most important commercial fishes in the world and is mainly utilized or consumed as sliced raw fish or as processed products. The biofunctional activities of enzymatic squid meat hydrolysate were determined to develop value-added products. Enzymatic squid hydrolysate manufactured by Alcalase effectively quenched 1,1-diphenyl-2-picrylhydrazyl radical, hydroxyl radical, and hydrogen peroxide radical with $IC_{50}$ values of 311, 3,410, and $111.5{\mu}g/mL$, respectively. Angiotensin I-converting enzyme inhibitory activity of squid hydrolysate was strong with an $IC_{50}$ value of $145.1{\mu}g/mL$, while tyrosinase inhibitory activity with an $IC_{50}$ value of 4.72 mg/mL was moderately low. Overall, squid meat hydrolysate can be used in food or cosmetic industries as a bioactive ingredient and possibly be used in the manufacture of seasoning, bread, noodle, or cosmetics.

빅벨리 해마(Hippocampus abdominalis) 유래 펩신 가수분해물의 In vitro와 In vivo에서의 항산화 효능 (Antioxidant Activity of Pepsin Hydrolysate Derived from Edible Hippocampus abdominalis in vitro and in Zebrafish Models)

  • Seahorse Hippocampus abdominalis a marine teleost fish, has long been used as one of the essential materials in traditional Chinese medicine. However, the uses of seahorse have been limited due to its high cost, despite its beneficial biological activities. Seahorse has not been widely explored for its biofunctional properties and active components. In the present study, the enzymatic hydrolysates of seahorse were prepared by using two digestive enzymes (trypsin and pepsin) and five food grade enzymes (neutrase, protamex, alcalase, kojizyme, and flavourzyme). The enzymatic hydrolysates indicated higher hydrolysis yields than its water extract. Among them, the distilled water-pepsin hydrolysate (DP) which was obtained by distilled water extraction followed by pepsin hydrolysis, showed the highest yield and protein content as well as the highest alkyl radical scavenging activity. Also, it provided protective effects against oxidative stress induced by AAPH in vero cell and zebrafish. Further fractionation based on the molecular weight was carried out to identify it’s active components, and < 5 kDa (less than 5 kDa) molecular weight fraction was confirmed to have the highest antioxidant activity. In conclusion, this study suggests that DP of seahorse has antioxidant properties, and might be a novel and useful material from the marine origin for healthy functional foods and cosmetics.

분리된 Simiduia sp. SH-4가 생산하는 β-agarase의 특성조사 (Characterization of β-agarase from Isolated Simiduia sp. SH-4)

  • 경상남도 남해군 미조면 연안으로부터 채취한 해수를 Marine agar 2216 배지에 도말하여 한천분해활성을 보이는 SH-4 균주를 분리하였다. 선택된 SH-4 균주는 16S rDNA 염기서열분석을 통해 Simiduia sp. SH-4로 명명하였다. Simiduia sp. SH-4 균주의 배양액으로부터 한천분해효소를 획득하여 한천분해활성을 측정하였다. 한천분해활성의 강도에 있어서 Simiduia sp. SH-4 유래 한천분해효소의 최고활성은 120.4 U/l로 나타났다. 한천분해활성은 30℃에서 최고치를 나타내었으며, 30℃에서의 활성을 100%로 하였을 때 20℃에서 30%, 40℃에서 75%의 상대활성을 나타내었다. 최적 pH는 pH 6.0으로 pH 6.0의 활성을 100%로 하였을 때 pH 5.0과 pH 7.0에서 각각 91%와 59%의 상대활성을 나타내었다. TLC 분석 결과, Simiduia sp. SH-4는 neoagarotetraose 및 neoagarobiose 등의 neoagarooligosaccharides를 생성하는 것으로 보아 β-agarase를 생산하는 균주로 확인되었다. 따라서 Simiduia sp. SH-4균주와 이 균주가 생산하는 β-agarase는 식품, 화장품, 의약품 산업에서 기능성소재 생산자로서 유용하게 활용할 수 있을 것으로 기대된다.
