The objectives of this study are to investigate the effect of media on the removal efficiency of nitrate-nitrogen and the biofilm thickness in the fluidized bed biofilm reactor(FBBR) used for the high rate denitrification of nitric acid wastewater. Granular activated carbon(GAC) of 1.274 mm diameter and sand of 0.455 mm diameter were used as the media in the FBBR of 0.05 m diameter and 1.5 m height. As the nitrate-nitrogen concentration of the influent was increased stepwise from 600 to 4800 mg/l, the nitrate- and nitrite-nitrogen concentration of the effluent, biofilm thickness and biofilm dry density were measured to study the effects of media on the denitrification efficiency. The biofilm thickness increased with the substrate loading rate, and the biofilm dry density decreased with the increase of the biofilm thickness. At the influent nitrate-nitrogen concentration of 2400 mg/l, the removal efficiency in the FBBR with GAC was 88%, while that in the FBBR with sand was 99.6%. The biofilm in the FBBR with GAC was so thick, 754.9 $\mu$m, as to increase the mass transfer resistance, compared to that, 143.7 $\mu$m, in the FBBR with sand. The maximum specific denitrification rate in the FBBR with GAC was 15.0 kg-N/m$^3\cdot$ day, while that in the FBBR with sand was 18.0 kg-N/m$^3\cdot$ day. The biomass concentration in the FBBR with sand exhibited the high value 37 kg/m$^3$.
A detachment of biofilm was investigated in an inverse fluidized bed biofilm reactor(IFRBR). The biofilm thickness, 5 and the bioparticle density, Pm were decreased by the increase of Reynolds number, Re and the decrease of biomass concentration, h. The correlations were expressed as $\delta$=6l.6+16.33$b_c$-0.004Re and Ppd=0.3+0.027$b_c$- 2.93x$l0^{-5}$ no by multiple linear regression analysis method. Specific substrate removal rate, q was derived by F/M ratio and biofilm thickness as q=0.44.+0.82F/M-5.Ix10$-4^{$\delta$}$. Specific biofilm detachment rate, bds was influenced by FIM ratio and Reynolds number as $b_{ds}$=-0.26+0.26F/M+ 2.17$\times$$10^{-4}$Re. Specific biofilm deachment rate in an IFBBR was higher than that in a FBRR(fluidized bed biofilm reactor) because of the friction between air bubble and the bioparticles.
The study was concerned with the biological denitrification of wastewater using the fluidization of biofilm-coated carbon particles. And the effect of Glucose and biofilm thickness on denitrification was mainly investigated. Experimental results showed that biofilm thickness increased with the growth of bacteria and 14 days after the beginning of operation, the thickness approached 300-310${\mu}{\textrm}{m}$. It was found that biofilm thickness was directly concerned with the removal efficiency of NO$_3$$^{[-10]}$ -N. As the results of experiments to find out the influence of Glucose on denitrification, Glucose, 60% excess from stoichiometric quantity was adequate and sufficient to react the bacteria with NO$_3$$^{[-10]}$ -N.
Using a rotating biological contactor modified with a sequencing bath reactor system (SBRBC) designed and operated to remove phosphate and nitrogen [58], the microbial community structure of the biofilm from the SBRBC system was characterized based on the extracellular polymeric substance (EPS) constituents, electron microscopy, and molecular techniques. Protein and carbohydrate were identified as the major EPS constituents at three different biofilm thicknesses, where the amount of EPS and bacterial cell number were highest in the initial thickness of 0-100${\mu}m$. However, the percent of carbohydrate in the total amount of EPS decreased by about 11.23%, whereas the percent of protein increased by about 11.15% as the biofilm grew. Thus, an abundant quantity of EPS and cell mass, as well as a specific quality of EPS were apparently needed to attach to the substratum in the first step of the biofilm growth. A FISH analysis revealed that the dominant phylogenetic group was $\beta$- and $\gamma$-Proteobacteria, where a significant subclass of Proteobacteria for removing phosphate and/or nitrate was found within a biofilm thickness of 0-250${\mu}m$. In addition, 16S rDNA clone libraries revealed that Klebsiella sp. and Citrobacter sp. were most dominant within the initial biofilm thickness of 0-250${\mu}m$, whereas sulfur-oxidizing bacteria, such as Beggiatoa sp. and Thiothrix sp., were detected in a biofilm thickness over 250${\mu}m$. The results of the bacterial community structure analysis using molecular techniques agreed with the results of the morphological structure based on scanning electron microscopy. Therefore, the overall results indicated that coliform bacteria participated in the nitrate and phosphorus removal when using the SBRBC system. Moreover, the structure of the biofilm was also found to be related to the EPS constituents, as well as the nitrogen and phosphate removal efficiency. Consequently, since this is the first identification of the bacterial community and structure of the biofilm from an RBC simultaneously removing nitrogen and phosphate from domestic wastewater, and it is hoped that the present results may provide a foundation for understanding nitrate and phosphate removal by an RBC system.
Activated carbon particles of 1.274 mm diameter and sand particles of 0.455 mm diameter were employed as the support of the biofilm formed in fluidized bed biofilm reactors(FBBRs) for the wastewater denitrification. Ethanol was used as the electron donor in the anoxic respiration. The steady-state biofilm thickness increased as the nitrate loading rate increased, and the activated carbon particles induced thicker biofilm than the sand particles. The FBBRs with sand support showed higher efficiency and rate of the nitrate removal than those with activated carbon support, and exhibited the biomass concentration of 37 kg/㎥ and the nitrate removal rate of 21 kg N/㎥d.
An inverse fluidized-bed biofilm reactor (IFBBR) was used for the treatment of highly-emulsified oily wastewater. When the concentration of biomass which was cultivated in the synthetic wastewater reached to 6000 mg/1, the oily wastewater was employed to the reactor with a input COD concentration range of 50 mg/1 to 1900 mg/l. Virtually the IFBBR showed a high stability during the long operation period although soma fluctuation was observed. The COD removal efficiency was maintained over 9% under the condition that organic loading rate should be controlled under the value of 1.5 kgCOD/$m^3$/day, and F/M ratio is 1.0 kgCOD/kgVSS/day at $22{\circ}C$ and HRT of 12 hrs. As increasing organic loading rates, the biomass concentration was decreased steadily with decreasing of biofilm dry density rather than biofilm thickness. Based on the experimental jesuits, it was suggested that the decrease in biofilm dry density was caused by a loss of biomass inside the biofilm.
Objectives: The purpose of this study was to investigate changes in the composition of artificial cariogenic biofilms using a Streptococcus mutans biofilm model over a period of time. Methods: We analyzed the dry weight, colony forming unit (CFU) number, extracellular polysaccharide (EPS) biovolume, and acid production rate of S. mutans biofilms formed on saliva-coated hydroxyapatite discs after 26 h, 50 h, 74 h, 98 h, 171 h, and 195 h. In addition, we performed a laser scanning confocal fluorescence microscopy to determine the bacterial volume, EPS biovolume, and biofilm thickness. We calculated the biofilm density using dry weight and EPS biovolume. Results: Over a period of time, there was no change in the CFU number and acid production rate of S. mutans biofilms, but there was an increase in the dry weight and EPS biovolume of S. mutans biofilms. The bacterial volume, EPS biovolume, and biofilm thickness only increased in the 50-h-old biofilm; however, no change was observed in 50-195-h-old biofilms. In addition, an increase in the biofilm density was observed over time. Conclusions: These results suggest that the acid production ability of cariogenic biofilms does not change, but the biofilm density increases over time. However, due to scientific information, further research needs to be conducted in the field of dentistry to get further insights on the progression of cariogenic biofilms over time.
Stability of reactor and effect on biofilm characteristics were investigated by varying the hydraulic residence time in an inverse fluidized bed biofilm reactor(IFBBR). The SCOD removal efficiency was maintained above 90 % in the HRT range of 12hr to 2hr, but the TCOD removal efficiency was dropped down to 50% because of biomass detachment from overgrown bioparticles. The reactor was stably operated up to the conditions of HRT of 2hr and F/M ratio of 4.5kgCOD/$m^3$/day, but above the range there was an abrupt increase of filamentous microorganisms. The optimum biofilm thickness and the biofilm dry density in this experiment were shown as $200\mu\textrm{m}$ and $0.08 g/cm^3$, respectively. The substrate removal rate of this system was found as 1st order because the biofilm was maintained slightly thin by the increased hydraulic loading rate.
Using rotating biological contactor(RBC) with artificial endogenous stage and aerobic fixed biofilm reactor(AFBR), organic material removal and biological nitrification of piggery wastewater has been studied at a pilot plant. RBC was operated in the endogenous phase at a interval of every 25 days. The concentration of COD, BOD and TKN in influent wastewater were from 2,940 to 3,800 mg/L, from 1,190 to 1,850 mg/L and from 486 to 754 mg/L respectively. The maximum active biomass content represented as VSS per unit aera was $2.0mg/cm$^{2}$ and biofilm dry density of $17mg/cm^{3}$ was observed at biofilm thickness of $900{\;}{\mu}m$. It was observed that the pilot scale RBC/AFBR process exhibited 72 percentage to 93 percentage of BOD removal, In order to obtain more than 90 percentage of BOD removal, the organic loading rate to the RBC/AFBR process should be maintained less than $0.09{\;}m^{3}/m^{2}{\cdot}day(125.9g{;\}BOD/m^{3}{\cdot}d$. The TKN removal efficiencies was from 45.5 to 90.9 percentage according to vary influent loading rate, It was estimated that the RBC/AFBR process consumed approximately 6.2 mg/L(as $CaCO_{3}$) of alkalinity per 1 mg/L of $NH_{3}$-N oxidized as the nitrification took piace.
Effect of the liquid circulation velocity on the biofilm development was investigated in an inverse fluidized bed biofilm reactor(IFBBR). To observe the effect of the influent COD concentration on biofilm simultaneously, the influent COD value was adjusted to 1000mg/1 f for 1st reactor, and 2500mg/l for 2nd reactor. The liquid circulation velocity was adjusted by controlling the initial liquid height. As the liquid circulation velocity was decreased, the settling amount of biomass was increased and the amount of effluent biomass was decreased. Since the friction of liquid was decreased by the decrease of liquid circulation velocity, the biofilm thickness was increased and the biofilm dry density was decreased. In the 1st reactor the SCOD removal efficiency was constant regardless of the variation of the liquid circulation velocity, but it was increased by the decrease of the liquid circulation velocity because of more biomass population in 2nd reactor.
