  • To find antibacterial strains of the soil microorganisms in Korea, they were isolated from the soil samples of different locations and screened for antibacterial activity against several standard microorganisms. An isolate among them had antibacterial activities against gram-positive and gram-negative bacteria. The examination of its morphological, biochemical, cultural and physiological characteristics according to the International Streptomyces Project methods showed that it belongs to the genus Streptomyces. This strain appears to be a novel strain when it was compared with those species of the genus which have been so far reported. The antibiotic metabolite was produced in the submerged culture of the strain. This metabolite was extracted from the culture filtrate and purified by ion-exchange column chromatography. Physico-chemical properties of the antibacterial metabolite were characterized.

Isolation and Identification of Microorganism Producing Glutary 7-Aminodeacetoxycephalosporanis Acid Acylase (Glutary 7-Aminodeacetoxycephalosporanis Acid Acylase 생산균의 분리 및 동정)

  • Microorganism producing glutaryl 7-aminodeacetoxycephalosporanic acid (GL-7-ADCA) acylase was screened from soil. The microorganism was identified as Alcaligenes sp. J-421 by its morphology and biochemical properties. Cultural conditions of Alcaligenes sp. J-421 were investigated for the production of GL-7-ADCA acylase. Optimum medium composition was 1% glucose, 1% beef extract, 0.5% yeast extract, 0.2% monosodium L-glutamate, 0.1% glutaric acid, 0.2% NaCl, 0.5% $K_2$ $HPO_4$, and 0.05% $CuSO _4{\cdot}5H_2O$. Optimum cultivation conditions for the production of the enzyme in 5 l jar fermentor were $37^{\circ}C$, tip speed 300 rpm, aeration 1 vvm. Optimum reaction pH of the enzyme was 8.0 and the enzyme was stable at pH7.0-11.0.

Isolation and Identification of Lactic acid Producing Bacteria from Kimchi and Their Fermentation Properties of Soymilk (젖산 생성능이 우수한 김치 유래 젖산균의 분리 및 두유 발효 특성)

  • Lactic acid bacteria were selected on the basis of lactic acid producing ability from kimchi, a traditional Korean fermented food. Among the initial screening of over 150 strains selected from the sample, 27 strains were selected as lactic acid producing bacteria, and 4 strains were finally selected based on their ability to produce relatively high levels of lactic acid. The four strains were identified as Lactobacillus (L.) plantarum Gk04, Pediococcus pentosaceus Gk07, L. brevis Gk35 and L. curvatus Gk36 by the conventional morphological, cultural, physiological and biochemical characteristics, as well as by 16S rRNA sequence analysis. Among the identified lactic acid bacteria, L. curvatus Gk36 was used for soymilk fermentation. The viable cell counts and acidity values measured for the L. curvatus Gk36 were comparable to the commmercial L. acidopillus. Thus, the L. curvatus Gk36 is a potential probiotic strain to prepare fermented soy products, such as kephir, yogurt, tempeh and soy sauce.

Cultural Conditions of Lactobacillus sp. GM7311 for the Production of Bacteriocin (Lactobacillus sp. GM7311에 의한 박테리오신의 생산 조건)

  • A lactic acid bacteria which showed antimicrobial activity was isolated from dairy products and identified as Lactobacillus sp. according to the morphological, physiological and biochemical properties, which was named Lactobacillus sp. SH 7311. The bacteriocin of Lactobacillus sp. GM 7311 showed a broad range of inhibitory spectrum against some gram positive and negative bacteria. Especially, Proteus mirabilis was highly sensitive to bacteriocin and used as indicator strain for further investigation. The optimal condition for the production of bacteriocin was showed on MRS broth at $37^{\circ}C$ and pM 6.0. Bacteriocin production of this strain cultured under optimal condition was increased late logarithmic phase to early stationary phase. This bacteriocin was fully active at the pH range $2.0\~5.0$, also was stable at $100^{\circ}C$ for 60 min. at pH 5.0, But about $40\%$ of bacteriocin activity was diminished by the treatment of acetone, ethanol, iso-butanol and ethyl ether during 2 hours at $4^{\circ}C$.
