에탄올 발효 배지로서 홍삼박의 활용 (Potential of Red Ginseng Marc for Ethanol Production as a Fermentation Medium)

  • 홍삼박을 에탄올 발효의 원료로 활용하기 위하여 홍삼박을 탄수화물 분해효소와 단백질분해효소로 처리한 결과 환원당의 함량이 7.2 g/L에서 101.1 g/L로 증가하였다. 홍삼박 효소분해물에 영양성분을 추가하지 않고 시판 효모를 접종하여 $25^{\circ}C$에서 7일간 발효한 결과 에탄올은 29.3 g/L까지 생성되었으며 총당대비수율은 0.274 g/g로 계산되었다. 이는 대조군보다 에탄올 생산량은 1,000% 이상, 수율은 50% 이상 향상된 결과이다.

Potential Health-Promoting Benefits of Paraprobiotics, Inactivated Probiotic Cells

  • Viability plays an important role in the beneficial microbes (probiotics) to produce health benefits. However, this idea has been changed after the invention of the term "paraprobiotics," indicating that non-viable microbes could produce health benefits similar to those produced by live probiotics. Occasionally, it might be dangerous to administer live probiotics to people with weak immunity. In such cases, ingestion of paraprobiotics could be a potential alternative. The definition of paraprobiotics refers to the use of inactivated (non-viable) microbial cells or cell fractions to provide health benefits to the consumer. Paraprobiotics have attracted much attention because of their long shelf life, safety, and beneficial effects, such as modulation of immunity, modification of biological responses, reduction of cholesterol, anti-inflammatory, and antiproliferative properties. These features indicate that paraprobiotics may play a vital role in improving the health of the consumer by enhancing particular physiological functions, even though the exact underlying mechanisms have not yet been completely elucidated. In this mini-review, we briefly discuss the historical backgrounds of paraprobiotics and evidence of their health-promoting effects, prophylactic, and therapeutic properties.

Effect of Soil Water Contents on Urea Hydrolysis and Nitrification in a Newly Reclaimed Tidal Soils

  • The effect of soil water content on the transformation potential of N compounds derived from hydrolysis of urea applied in a reclaimed tidal soils which was saline-sodic was observed to evaluate nitrification rates of urea. Soil samples were collected from Moonpo series at the newly reclaimed area in Saemanguem. For the transformation potential of N compounds from urea (46% N), newly reclaimed tidal soils (RS) were amended with urea at the rates of 0, 10, and 20 kg $10a^{-1}$. With leachate obtained from the incubated RS in a leaching tube at $25^{\circ}C$, urea hydrolysis and nitrification were measured for a total of 30days. The cumulative amounts of $NO_3{^-}$-N in each of the four soils treated with urea was linear with time of incubation. Results showed that increase in pH occurred with increasing application rate of urea and volumetric water content due to hydrolysis of urea. The total N in the RS was decreased with incubation time, indicating that rates of urea hydrolysis was influenced by soil moisture conditions. Also, the cumulative amount of nitrate in RS gradually increased with increase in time of incubation.

Wnt5a attenuates the pathogenic effects of the Wnt/β-catenin pathway in human retinal pigment epithelial cells via down-regulating β-catenin and Snail

  • Activation of the Wnt/β-catenin pathway plays a pathogenic role in age-related macular degeneration (AMD) and is thus a potential target for the development of therapeutics for this disease. Here, we demonstrated that Wnt5a antagonized β-catenin response transcription (CRT) induced with Wnt3a by promoting β-catenin phosphorylation at Ser33/Ser37/Thr41 and its subsequent degradation in human retinal pigment epithelial (RPE) cells. Wnt5a decreased the levels of vascular endothelial growth factor (VEGF), tumor necrosis factor-α(TNF-α), and nuclear factor-κB (NF-κB), which was up-regulated by Wnt3a. Furthermore, Wnt5a increased E-cadherin expression and decreased cell migration by down-regulating Snail expression, thereby abrogating the Wnt3a-induced epithelial-mesenchymal transition (EMT) in human RPE cells. Our findings suggest that Wnt5a suppresses the pathogenic effects of canonical Wnt signaling in human RPE cells by promoting β-catenin phosphorylation and degradation. Biological Control Potential of Bacillus amyloliquefaciens KB3 Isolated from the Feces of Allomyrina dichotoma Larvae

  • Most biocontrol agents for plant diseases have been isolated from sources such as soils and plants. As an alternative source, we examined the feces of tertiary larvae of the herbivorous rhino beetle, Allomyrina dichotoma for presence of biocontrol-active microbes. The initial screen was performed to detect antifungal activity against two common fungal plant pathogens. The strain with strongest antifungal activity was identified as Bacillus amyloliquefaciens KB3. The inhibitory activity of this strain correlated with lipopeptide productions, including iturin A and surfactin. Production of these surfactants in the KB3 isolate varied with the culture phase and growth medium used. In planta biocontrol activities of cell-free culture filtrates of KB3 were similar to those of the commercial biocontrol agent, B. subtilis QST-713. These results support the presence of microbes with the potential to inhibit fungal growth, such as plant pathogens, in diverse ecological niches.

Inhibitory Effect Against Akt by Cyclic Dipeptides Isolated from Bacillus sp.

  • Among thirteen strains of the genus Bacillus isolated from Shrimp-jeotkal in our laboratory, a strain BA34 showing good antifungal activity against Phytophthora infestans in a previous experiment was tested for the inhibitory effect against Akt, protein kinase B. Since Akt is known to play an important role in controlling apoptosis, its inhibitors can be used as potential apoptosis-inducing agents in the treatment of cancer. Two active compounds were isolated and their structures were determined. They have similar structures, despite showing different inhibitory effects. In order to elucidate the reasons for these different effects, three-dimensional studies were carried out.

N3S-ligated Copper(II) Complex Catalyzed Selective Oxidation of Benzylic Alcohols to Aldehydes under Mild Reaction Conditions

  • A Cu(II) complex with an three nitrogens and one sulfur coordination environment was synthesized and characterized. Its redox potential was observed at 0.483 V vs. NHE, very similar to that of a Cu-containing fungal enzyme, galactose oxidase, which catalyzes the oxidation of alcohols to corresponding aldehydes with the concomitant reduction of molecular oxygen to water. The Cu(II) complex selectively oxidizes the benzylic alcohols using TEMPO/$O_2$ under mild reaction conditions to corresponding aldehydes without forming any over-oxidation product. Moreover, the catalyst can be recovered and reused multiple times for further oxidation reactions, thus minimizing the waste generation.

  • Recently, the next-generation advanced flow visualization techniques such as holographic PIV, dynamic PIV, echo-PIV, micro/nano-PIV, and X-ray PIV have been introduced. These advanced measurement techniques have a big potential as the core technology for analyzing outmost thermo-fluid flows in future. These would be indispensable in solving complicated thermo-fluid flow problems not only in the industrial fields such as automotive, space, electronics, aero- and hydro-dynamics, steel, and information engineering, but also in the research fields of medical science, bio-medical engineering, environmental and energy engineering etc. Especially, NT (Nano Technology) and BT (Bio Technology) strongly demand these advanced measurement techniques, because it is impossible for conventional measurement methods to observe most complicated nano- and bio-fluidic phenomena. In this presentation, the basic principle of these high-tech flow visualization techniques and their practical applications which cannot be resolved by conventional methods, such as blood flows in a micro-tube, in vivo analysis of micro-circulation, and flow around a living body will be introduced as a blue ocean strategy.

자연치즈의 제조과정 중 숙성의 중요성 (Importance of Ripening during Natural Cheese Making)

  • The ripening of cheese allows for the development of characteristic taste and flavour, nutritional substances, bio-active components and texture, helping to improve quality. Many different microbiological, biochemical and nutritional changes occur during the process depending on the quality of raw milk, added cultures and enzymes, as well as specific processing and ripening conditions. During the ripening lactose is hydrolyzed to lactic, propionic and acetic acid, helping to reduce potential effects of the problem of lactose intolerance. Fat is hydrolyzed to butyric, propionic and conjugated linoleic acid, which function as bio-active substances. Protein is hydrolyzed to different peptides and amino acids which all show various bio-activities. However, errors of cheese ripening can happen and affect the quality of the product. To guarantee good quality cheese the process needs to be managed carefully with the right microbes used and ensuring cleanliness of processing facilities, staff, ventilation and hazard analysis and critical control points (HACCP). Research into and controlling of ripening technology is crucial for producing high quality cheeses.

  • Acupuncture effect principle on meridians is what physical stimulation does on general physiology so that it gets into new equilibrium from the fallen function state of human body. However, it is generally accepted that western medicine has regarded the action passageway of various kinds stimulation as nerve fluids tissue or nerve-endocrine-immune system. These regard any action for all meridians as a response to the stimulation. These are not treatments for transmission of the energy. This paper compares the change of electric potential when operator's bio-energy is passed on and when is not. As a result of it, the effect of acupuncture is different when a operator's bio-energy is passed on to meridians and it isn't. Therefore, it means that the general acupuncture is more effective than the insulation one. Moreover, we could certify that there is close relation between an operator's energy transmission and acupuncture effect.
