THE EFFECT OF PRP AND FIBRIN SEALANT WITH THE DEPROTENIZED BOVINE BONE IN THE RABBIT CRANIUM (가토의 두개골 골결손부에 탈단백 우골 이식시 혈소판 농축 혈장과 피브린 실란트의 효과)

  • Purpose The purpose of this study is to evaluation of effect on bone formation of PRP and fibrin sealant with deproteinized bovine bone(Bio-Oss) grafts on rabbit cranial defect. Material and Methods Twelve rabbits were used as experimental animal Two equal 9mm diameter cranial bone defects were created in each rabbit and immediately grafted with Bio-Oss only, Bio-Oss and PRP, and Bio-Oss and Fibrin sealant. Rabbits were sacrificed at 4 and 8 week. The defects were evaluated by histomorphometric analysis. Results Kruskal-Wallis tests were performed comparing new bone formation via histomorphometric analysis. No statistically significant difference of new bone formation was found between Bio-Oss only, Bio-Oss and PRP, and Bio-Oss and fibrin sealant at 4 and 8 weeks (P>0.05). Conclusion This study fails to find a stimulatory effect of PRP and Fibrin sealant on New bone formation of Bio-Oss grafts by histomorphometric analyses.

A chemical conjugate between HER2-targeting antibody fragment and Pseudomonas exotoxin A fragment demonstrates cytotoxic effects on HER2-expressing breast cancer cells

  • Conventionally, immunotoxins have been produced as a single polypeptide from fused genes of an antibody fragment and a toxin. In this study, we adopted a unique approach of chemical conjugation of a toxin protein and an antibody fragment. The two genes were separately expressed in Escherichia coli and purified to high levels of purity. The two purified proteins were conjugated using a chemical linker. The advantage of this approach is its ability to overcome the problem of low recombinant immunotoxin production observed in some immunotoxins. Another advantage is that various combinations of immunotoxins can be prepared with fewer efforts, because the chemical conjugation of components is relatively simpler than the processes involved in cloning, expression, and purification of multiple immunotoxins. As a proof of concept, the scFv of trastuzumab and the PE24 fragment of Pseudomonas exotoxin A were separately produced using E. coli and then chemically crosslinked. The new immunotoxin was tested on four breast cancer cell lines variably expressing HER2. The chemically crosslinked immunotoxin exhibited cytotoxicity in proportion to the expression level of HER2. In conclusion, the present study revealed an alternative method of generating an immunotoxin that could effectively reduce the viability of HER2-expressing breast cancer cells. These results suggest the effectiveness of this method of immunotoxin crosslinking as a suitable alternative for producing immunotoxins.

A Study on the Regional Distribution Characteristics and Innovation Activity Performance of Bio-Industry in Korea: Focusing on Metropolitan and Non-metropolitan Areas (국내 바이오산업의 지역별 분포특성과 혁신 활동 성과에 관한 연구: 수도권과 비수도권 지역을 중심으로)

  • The study empirically analyzed the differences in industry distribution and innovation activity performance in the metropolitan and non-metropolitan areas of Korea's bio companies, which are highlighted as future growth engines. The main innovation activities of the bio industry, which are focused on science and technology and expressed with high uncertainty, were analyzed, centering on human resources, technology cooperation, and investment promotion. As a result of the analysis, the biomedical industry in the metropolitan area was found to have a high proportion, and bio foods, bio-based chemicals, and energy industries in the non-metropolitan area, respectively. Moreover, the innovation activity performances differed between the two regions. In particular, the notable characteristics included human resources, investment promotion, and technical cooperation with medical institutions in the metropolitan area with a high proportion of biomedical industries, and technology personnel exchange and cooperation with private research institutions in the non-metropolitan area, which has a high proportion of bio foods, bio-based chemicals, and energy industries. This study is significant in that it is the first study to compare and analyze the performance of innovative activities based on the distribution of industries in the bio-industry, focusing on human resources, technology cooperation, and investment promotion. In addition, after investigating the distribution status and competitiveness of the domestic bio-industry by region, it will analyze the status and characteristics of the domestic bio-industry and present policy implications to implement relevant promotion policy more efficiently.

Design and Implementation of Medical Information System using QR Code (QR 코드를 이용한 의료정보 시스템 설계 및 구현)

  • The new medical device technologies for bio-signal information and medical information which developed in various forms have been increasing. Information gathering techniques and the increasing of the bio-signal information device are being used as the main information of the medical service in everyday life. Hence, there is increasing in utilization of the various bio-signals, but it has a problem that does not account for security reasons. Furthermore, the medical image information and bio-signal of the patient in medical field is generated by the individual device, that make the situation cannot be managed and integrated. In order to solve that problem, in this paper we integrated the QR code signal associated with the medial image information including the finding of the doctor and the bio-signal information. bio-signal. System implementation environment for medical imaging devices and bio-signal acquisition was configured through bio-signal measurement, smart device and PC. For the ROI extraction of bio-signal and the receiving of image information that transfer from the medical equipment or bio-signal measurement, .NET Framework was used to operate the QR server module on Window Server 2008 operating system. The main function of the QR server module is to parse the DICOM file generated from the medical imaging device and extract the identified ROI information to store and manage in the database. Additionally, EMR, patient health information such as OCS, extracted ROI information needed for basic information and emergency situation is managed by QR code. QR code and ROI management and the bio-signal information file also store and manage depending on the size of receiving the bio-singnal information case with a PID (patient identification) to be used by the bio-signal device. If the receiving of information is not less than the maximum size to be converted into a QR code, the QR code and the URL information can access the bio-signal information through the server. Likewise, .Net Framework is installed to provide the information in the form of the QR code, so the client can check and find the relevant information through PC and android-based smart device. Finally, the existing medical imaging information, bio-signal information and the health information of the patient are integrated over the result of executing the application service in order to provide a medical information service which is suitable in medical field.

Fabrication and Characterization of PCL/TiO2 Nanoparticle 3D Scaffold (PCL/TiO2 Nanoparticle 3차원 지지체 제조 및 특성 평가)

  • Polycaprolactone (PCL) is a synthetic biodegradable polymer with excellent mechanical properties. $TiO_2$ (titanium dioxide) has a hydrophilic, high density and excellent biocompatibility. In this work, we produced three-dimensional porous scaffolds with PCL and $TiO_2$ nanoparticles using a salt-leaching method. Physical properties of the scaffolds were analyzed by FE-SEM, FTIR, TGA and compressive strength. Interestingly, the addition of $TiO_2$ nanoparticles decreased the water absorption and swelling ratio of the porous scaffolds. However, the compressive strength was increased by $TiO_2$. CCK-8 assay, which is generally used for the analysis of cell growth, shows that $TiO_2$ nanoparticles have no cytotoxicity. Taken together, we suggest that the PLC/$TiO_2$-scaffold can be used for biomedical applications.

Fabrication and Characterization of Silk/PVA Hydrogels by Sonication and Freezing-Thawing Technique (초음파와 동결/융해에 의한 실크/PVA 하이드로젤의 제조 및 특성 평가)

  • Biomaterials like silk fibroin (SF) and poly(vinyl alcohol) (PVA) have received increasing attention in biomedical applications because of their attractive properties such as hydrophobicity and biocompatibility. In this study, efficient systems consisting of interpenetrating SF/PVA hydrogels were prepared as potential candidate for wound dressing applications. A simple approach consisting of sonication and a freezing-thawing technique was adopted to fabricate the hydrogels. Different blend ratios consisting of SF (100, 75, 50, 25 and 0%) with respect to the weight of PVA were prepared. The produced hydrogels were characterized for physico-chemical investigations using various states of techniques like; FE-SEM, TGA, FTIR and tensile strength. The addition of PVA to SF was proved to be beneficial in terms of reducing the pore size and swelling ratio of hydrogels. The mechanical property of SF had been increased by addition of PVA. These results show that SF/PVA hydrogels may serve as potential candidates for wound dressing application.

Protein Tyrosine Phosphatase, Receptor Type B (PTPRB) Inhibits Brown Adipocyte Differentiation through Regulation of VEGFR2 Phosphorylation

  • Brown adipocytes have an important role in the regulation of energy balance through uncoupling protein-1 (UCP-1)-mediated nonshivering thermogenesis. Although brown adipocytes have been highlighted as a new therapeutic target for the treatment of metabolic diseases, such as obesity and type II diabetes in adult humans, the molecular mechanism underlying brown adipogenesis is not fully understood. We recently found that protein tyrosine phosphatase receptor type B (PTPRB) expression dramatically decreased during brown adipogenic differentiation. In this study, we investigated the functional roles of PTPRB and its regulatory mechanism during brown adipocyte differentiation. Ectopic expression of PTPRB led to a reduced brown adipocyte differentiation by suppressing the tyrosine phosphorylation of VEGFR2, whereas a catalytic inactive PTPRB mutant showed no effects on differentiation and phosphorylation. Consistently, the expression of brown adipocyte-related genes, such as UCP-1, $PGC-1{\alpha}$, PRDM16, $PPAR-{\gamma}$, and CIDEA, were significantly inhibited by PTPRB overexpression. Overall, these results suggest that PTPRB functions as a negative regulator of brown adipocyte differentiation through its phosphatase activity-dependent mechanism and may be used as a target protein for the regulation of obesity and type II diabetes.
