Transformation of a Filamentous Fungus Cryphonectria parasitica Using Agrobacterium tumefaciens

  • As Agrobacterium tumefaciens, which has long been used to transform plants, is known to transfer T-DNA to budding yeast, Saccharomyces cerevisiae, a variety of fungi were subjected to the A. tumefaciens-mediated transformation to improve their transformation frequency and feasibility. The A. tumefaciens-mediated transformation of chestnut blight fungus, Cryphonectria parasitica, is performed in this study as the first example of transformation of a hardwood fungal pathogen. The transfer of the binary vector pBIN9-Hg, containing the bacterial hygromycin B phosphotransferase gene under the control of the Aspergillus nidulans trpC promoter and terminator, as a selectable marker, led to the selection of more than 1,000 stable, hygromycin B-resistant transformants per 1${\times}$10$\^$6/ conidia of C. parasitica. The putative transformants appeared to be mitotically stable. The transformation efficiency appears to depend on the bacterial strain, age of the bacteria cell culture and ratio of fungal spores to bacterial cells. PCR and Southern blot analysis indicated that the marker gene was inserted at different chromosomal sites. Moreover, three transformants out of ten showed more than two hybridizing bands, suggesting more than two copies of the inserted marker gene are not uncommon.

  • We propose a BASK (Binary Amplitude Shift Keying) system for Giga-bit Modem in millimeter band. The proposed system consists of a high speed shutter of the transmitter and a repeater of the receiver. The shutter of the proposed system is introduced for pulse shaping to improve the intersymbol interference (ISI). The repeater consists of several stage converters. A converter is constructed with a low pass filter and a limiter. The repeater can improve the signal-to-noise ratio (SNR) and make the rectangular pulse train. The proposed system is a simple system that uses conversion method without IF (Intermediate Frequency) process.

  • The direction component uses mainly the sobel and FFT method. The sobel method is difficult to set representative direction when we wish to extract representative direction when we wish to extract representative direction component and the complicated processing process of sobel mask because same value appears in the low provision image or high provision image and we cannot accumulate tilt size in case of making accumulate after making unit vector to pixel. The method that uses FFT conversion for direction extraction is possible in case that ridge has correct direction specification and must use a special direction filter. After thinning the binary image to supplement above weak point in the paper, we extract direction component by pixel unit, and we extract the most direction components of pixel that exist in block of 8${\times}$8 pixels size as representative direction of ridge.

Production of transgenic potato exhibiting enhanced resistance to fungal infections and herbicide applications

  • Potato (Solanum tuberosum L.), one of the most important food crops, is susceptible to a number of devastating fungal pathogens in addition to bacterial and other pathogens. Producing disease-resistant cultivars has been an effective and useful strategy to combat the attack of pathogens. Potato was transformed with Agrobacterium tumefaciens strain EHA101 harboring chitinase, (ChiC) isolated from Streptomyces griseus strain HUT 6037 and bialaphos resistance (bar) genes in a binary plasmid vector, pEKH1. Polymerase chain reaction (PCR) analysis revealed that the ChiC and bar genes are integrated into the genome of transgenic plants. Different insertion sites of the transgenes (one to six sites for ChiC and three to seven for bar) were indicated by Southern blot analysis of genomic DNA from the transgenic plants. Expression of the ChiC gene at the messenger RNA (mRNA) level was confirmed by Northern blot analysis and that of the bar gene by herbicide resistance assay. The results obviously confirmed that the ChiC and bar genes are successfully integrated and expressed into the genome, resulting in the production of bialaphos-resistant transgenic plants. Disease-resistance assay of the in vitro and greenhouse-grown transgenic plants demonstrated enhanced resistance against the fungal pathogen Alternaria solani (causal agent of early blight).

Learning Directional LBP Features and Discriminative Feature Regions for Facial Expression Recognition (얼굴 표정 인식을 위한 방향성 LBP 특징과 분별 영역 학습)

  • In order to recognize the facial expressions, good features that can express the facial expressions are essential. It is also essential to find the characteristic areas where facial expressions appear discriminatively. In this study, we propose a directional LBP feature for facial expression recognition and a method of finding directional LBP operation and feature region for facial expression classification. The proposed directional LBP features to characterize facial fine micro-patterns are defined by LBP operation factors (direction and size of operation mask) and feature regions through AdaBoost learning. The facial expression classifier is implemented as a SVM classifier based on learned discriminant region and directional LBP operation factors. In order to verify the validity of the proposed method, facial expression recognition performance was measured in terms of accuracy, sensitivity, and specificity. Experimental results show that the proposed directional LBP and its learning method are useful for facial expression recognition.

Orthonormal Polynomial based Optimal EEG Feature Extraction for Motor Imagery Brain-Computer Interface

  • In this paper, we explored the new method for extracting feature from the electroencephalography (EEG) signal based on linear regression technique with the orthonormal polynomial bases. At first, EEG signals from electrodes around motor cortex were selected and were filtered in both spatial and temporal filter using band pass filter for alpha and beta rhymic band which considered related to the synchronization and desynchonization of firing neurons population during motor imagery task. Signal from epoch length 1s were fitted into linear regression with Legendre polynomials bases and extract the linear regression weight as final features. We compared our feature to the state of art feature, power band feature in binary classification using support vector machine (SVM) with 5-fold cross validations for comparing the classification accuracy. The result showed that our proposed method improved the classification accuracy 5.44% in average of all subject over power band features in individual subject study and 84.5% of classification accuracy with forward feature selection improvement.

Will You Buy It Now?: Predicting Passengers that Purchase Premium Promotions Using the PAX Model

  • Upselling is often a critical factor in revenue generation for businesses in the tourism and travel industry. Utilizing passenger data from a major international airline company, we develop the PAX (Passenger, Airline, eXternal) model to predict passengers that are most likely to accept an upgrade offer from economy to premium. Formulating the problem as an extremely unbalanced, cost-sensitive, supervised binary classification, we predict if a customer will take an upgrade offer. We use a feature vector created from the historical data of 3 million passenger records from 2017 to 2019, in which passengers received approximately 635,000 upgrade offers worth more than $422,000,000 U.S. dollars. The model has an F1-score of 0.75, outperforming the airline's current rule-based approach. Findings have several practical applications, including identifying promising customers for upselling and minimizing the number of indiscriminate emails sent to customers. Accurately identifying the few customers who will react positively to upgrade offers is of paramount importance given the airline 'industry's razor-thin margins. Research results have significant real-world impacts because there is the potential to improve targeted upselling to customers in the airline and related industries.

Stochastic Non-linear Hashing for Near-Duplicate Video Retrieval using Deep Feature applicable to Large-scale Datasets

  • With the development of video-related applications, media content has increased dramatically through applications. There is a substantial amount of near-duplicate videos (NDVs) among Internet videos, thus NDVR is important for eliminating near-duplicates from web video searches. This paper proposes a novel NDVR system that supports large-scale retrieval and contributes to the efficient and accurate retrieval performance. For this, we extracted keyframes from each video at regular intervals and then extracted both commonly used features (LBP and HSV) and new image features from each keyframe. A recent study introduced a new image feature that can provide more robust information than existing features even if there are geometric changes to and complex editing of images. We convert a vector set that consists of the extracted features to binary code through a set of hash functions so that the similarity comparison can be more efficient as similar videos are more likely to map into the same buckets. Lastly, we calculate similarity to search for NDVs; we examine the effectiveness of the NDVR system and compare this against previous NDVR systems using the public video collections CC_WEB_VIDEO. The proposed NDVR system's performance is very promising compared to previous NDVR systems.

Blind Quality Metric via Measurement of Contrast, Texture, and Colour in Night-Time Scenario

  • Night-time image quality evaluation is an urgent requirement in visual inspection. The lighting environment of night-time results in low brightness, low contrast, loss of detailed information, and colour dissonance of image, which remains a daunting task of delicately evaluating the image quality at night. A new blind quality assessment metric is presented for realistic night-time scenario through a comprehensive consideration of contrast, texture, and colour in this article. To be specific, image blocks' color-gray-difference (CGD) histogram that represents contrast features is computed at first. Next, texture features that are measured by the mean subtracted contrast normalized (MSCN)-weighted local binary pattern (LBP) histogram are calculated. Then statistical features in Lαβ colour space are detected. Finally, the quality prediction model is conducted by the support vector regression (SVR) based on extracted contrast, texture, and colour features. Experiments conducted on NNID, CCRIQ, LIVE-CH, and CID2013 databases indicate that the proposed metric is superior to the compared BIQA metrics.

Modulation Recognition of BPSK/QPSK Signals based on Features in the Graph Domain

  • The performance of existing recognition algorithms for binary phase shift keying (BPSK) and quadrature phase shift keying (QPSK) signals degrade under conditions of low signal-to-noise ratios (SNR). Hence, a novel recognition algorithm based on features in the graph domain is proposed in this study. First, the power spectrum of the squared candidate signal is truncated by a rectangular window. Thereafter, the graph representation of the truncated spectrum is obtained via normalization, quantization, and edge construction. Based on the analysis of the connectivity difference of the graphs under different hypotheses, the sum of degree (SD) of the graphs is utilized as a discriminate feature to classify BPSK and QPSK signals. Moreover, we prove that the SD is a Schur-concave function with respect to the probability vector of the vertices (PVV). Extensive simulations confirm the effectiveness of the proposed algorithm, and its superiority to the listed model-driven-based (MDB) algorithms in terms of recognition performance under low SNRs and computational complexity. As it is confirmed that the proposed method reduces the computational complexity of existing graph-based algorithms, it can be applied in modulation recognition of radar or communication signals in real-time processing, and does not require any prior knowledge about the training sets, channel coefficients, or noise power.
