The study was designed to observe the effect of conjugated linoleic acid (CLA) on tumor incidence, eicosanoid formation and antioxidant enzyme activities in colonic mucosa and the fecal excretion of deoxycholic acid and lithocholic acid in 1,2-dimethylhydrazine (DMH)-treated rats. One hundred twenty male Sprague Dawley rats were divided into 2 groups, BT (beef tallow diet) group and FO (fish oil diet) group, and each group was again subdivided into 2 groups depending on CLA supplementation, i.e.4 groups of BT, BTC, FO, FOC. All rats were fed experimental diet for 30 weeks, which contained 12% (wt/wt) total dietary fat including 1% (wt/wt) CLA, and were intramuscularly injected with DMH for 6 weeks to give total dose of 180 mg/kg body. CLA-supplemented to BT and FO diet reduced tumor incidence, eicosanoid (PGE$_2$ and TXA$_2$) level in colonic mucosa. N-3 fatty acids (mainly DHA) of fish oil diet (FO, FOC group) also reduced tumor incidence and significantly reduced eicosanoid (PGE$_2$ and TXA$_2$) level in colonic mucosa. CLA supplementation and n-3 fatty acid significantly increased colonic mucosal level of superoxide dismutase and glutathione peroxidase activities but reduced secondary bile acids (deoxycholic acid and lithocholic acid) excretion in the feces. In conclusion, CLA supplementation and n-3 fatty acid could reduce tumor incidence by reducing eicosanoids and increasing antioxidant enzyme activities in colon and decreasing the excretion of deoxycholic acid and lithocholic acid in the feces. The data might suggest that CLA supplementation and n-3 DHA rich fish oil may modulate colon carcinogenesis.termediate level of endurance exercise training for 6 weeks did not influence concentrations of most of free amino acid in soleus muscle of rats collected at an overnight fasted and rested state. In contrast, isolucine and leucine concentrations in extensor digitorum longus muscle of exercise-trained rats were significantly lower than those for control animals. These results indicate that aerobic energy metabolism had not been efficiently conducted, and thereby the utilization of BCAA for energy substrate was enhanced in fast twitch oxidative glycolytic fibers of extensor digitorum longus muscle of rats followed exercise-training protocol for 6 weeks.
Lipid-lowering effects of the inulin have been demonstrated in animal, yet attempts to reproduce similar effects in humans have generated conflicting results. In this study, the lipid-lowering potential of inulin and especially its effect on bile acid and neutral sterol excretion were investigated in Korean postmenopausal women. Nineteen postmenopausal women were randomly divided into two groups in a double-blind parallel design and consumed one of two supplements for 12 weeks; placebo of 8g maltodextrins/sucrose mixture (placebo group) or 8g inulin (inulin group). There were no significant changes in body weight during the supplementation period in either inulin or placebo group. Dietary consumption of animal fat in both group tended to decrease after 12 weeks of experiment. Intake of cholesterol was lower in placebo group, whereas the decrease of cholesterol intake in inulin group did not reach statistical significance after 12 weeks. The levels of serum total cholesterol (TC) and LDL-cholesterol (LDL-C) were significantly decreased in both placebo (p<0.05) and inulin group (p<0.01) after supplementation for 12 weeks compared with the baseline. The levels of serum triglyceride (TG) and HDL-cholesterol (HDL-C) were not significantly affected by inulin supplements, but atherogenic index (AI) and LDL-C/HDL-C ratio (LHR) as a predictor for coronary heart disease were improved (p<0.01) significantly after inulin supplementation. Therefore, inulin supplement may decrease the risk of cardiovascular disease via improving blood cholesterol level. Fecal weight and pH were not changed after 12 weeks of supplementation. There were no statistically significant changes for the fecal short-chain fatty acids (SCFAs). In inulin group, fecal deoxycholic acid (DCA) was significantly lowered compared with the baseline (p<0.05) whereas other bile acids were not changed. During the 12 weeks of intervention, no differences were found in fecal excretion of neutral sterol in the two groups. In summary, dietary inulin decreases serum TC, LDL-C, AI, LHR and lowers excretion of fecal DCA in the Korean postmenopausal women. These results support the use of inulin for reducing risk factors for hyperlipidemic postmenopausal women. However, the exact mechanism (s) responsible for the blood lipid lowering action of inulin including altered fecal bile acid remain to be elucidated.
Biliary atresia (BA) is an infantile cholestatic disease of progressive obliterative cholangiopathy with varying degrees of damage to both extra and intrahepatic bile ducts due to unknown causes. The diagnostic studies should be done to diagnose or exclude BA without unnecessary delay. Kasai portoenterostomy is the first choice of treatment for bile drainage from microscopic bile ductules present in the portal fibrous mass. The medical management after Kasai portoenterostomy should be done carefully to maintain bile excretion and prevent and treat complications Including cholangitis, hepatic fibrosis, portal hypertension and nutritional problem. The reported five years-survival rates after Kasai portoenterostomy range from 30 to 60 %. About 20 % of all patients undergoing Kasai portoenterostomy during infancy survive into adulthood with their native liver. Even if Kasai portoenterostomy remains as the first line of treatment In BA, liver transplantation serves as a good salvage treatment when portoenterostomy fails or liver function gradually deteriorates after initially successful establishment of bile flow, Overall 5-year survival rate in BA is about 90 % in recent series.
This study was performed to investigate the effect of dietary chitin & chitosan on cadmium (Cd) toxicity and lipid metabolism in rats. Forty-two male rats of Sprague-Dawley strain weighing 137$\pm$2g were blocked into 6 groups according to body weight, and were raised for 4 weeks. Cadmium chloride was given at the level of 0 or 400ppm in diet and chitin and chitosan were given at the level of 0 or 4%(w/w) of diet. The results are summarized as follow. Chitosan decreased the toxicity of Cd on liver, kidney and femur and increase the Cd content of fecal excretion. Chitosan increased the lipdi & cholesterol content of fecal excretion by combining with lipid and bile acid. Chitosan decreased lipid, cholesterol and TG content in serum and liver by combining with lipid and bile acid. Chitin was less effective than chitosan in decreasing of Cd toxicity and lipid content of rat.
This study was carried out to explore the hypocholesterolemic effect of dehyulled defatty soy flour and its possible mechanisms including endocrine status, cholesterol biosynthesis, and fecal excretion in rats. Animals fed casein were used as control and each phospholipid compared with casein feeding. Cholesterol concentrations in all lipoprotein fraction were significantly lower in defatted soy flour group compared with casein-fed control. Defatted soy flour feeding also significantly lowered hepatic total lipid, cholesteol and TG, and increaed fecal bile acid excretion by 270% compared with casein feeding. Defattd soy flour feeding had no significant effect on plasma thyroid hormone levels and hepatic 3-hydroxy-3-methyl glutary coenzyme A(HMG-CoA) reductase activity. However, plasma T4 concentration was slightly elevated and HMG-CoA reductase activity was suppressed in defatted soy flour group. These metabolic alterations partially explain the reduced plasma and hepatic cholesterol levels of rats fed defatted soy flour.

In order to observe the effect of tannin on plasma lipid status in human, tannin was supplemented with increasing amounts from 0 to 0.5, 1.0, 2.5g/d early week until 4th week. The results obtained from this research are a follows. Concentration of total lipid in plasma was significantly lower compared to the control group when the level of tannin supplementation was 0.5g/day while the level of tannin 1.0g/day and 2.5g/day did not affect to change the total lipid level. There seems to be no obvious improvement of the plasm lipid status respect to different tannin levels. Despite the result, the excretion of total bile acids in feces was increased and total lipid in plasma was reduced significantly during first time supplementation compared the control.
This study was conducted to investigate the effects of corn peptide(CP) on lipid metabolism. Sprague-Dawley(S.D) male rats were assigned to three dietary groups {control diet(CD), high fat diet (HFD) & high fat com peptide diet(FCD)} and fed 4 weeks to examine the effects of CP. There were no significantly different in cholesterol concentrations in the liver among the groups. However, triglyceride(TG) concentrations of the FCD & CD significantly lower than the HFD. Fecal excretion of neutral steroids & bile acids of the FCD significantly higher than the CD & HFD. Serum total cholesterol TG & LDL-cholesterol concentrations of the FCD & CD significantly lower than the HFD. These results suggest the improvement of lipid composition in serum by CP might be inhibit of lipid absorption in intestine & increment of neutral steroids & bile acids excretion in feces.

The effect of sulfur-containing amino acids on lipid metabolism was studied in rats fed casein as a protein source. Plasma cholesterol, HDL-cholesterol and atherosclerotic index decreased in the cysteine group compared to the methionine group. Plasma triglyceride and phospholipid level were not affected by the supplementation of the sulfur-containing amino acids. The levels of cholesterol and triglyceride in liver decreased by both methionine and cysteine. Cysteine increased the fecal excretion of coprostanol, total neutral steroid and bile acid. The results suggest that plasma cholesterol level is affected by dietary ratio of cysteine/methionine and that the hypocholesterolemic effects of cysteine is, at least in part, through reducing cholesterol absorption from small intestine and through enhancing fecal excretion of bile acids.
The effects of indigestible dextrin on lipids in serum, tissue, and feces were investigated in two experiments carried out with 64 male Sprague-Dawley rats for eight weeks. Experiment I diet(normal fat diet) containing 5% corn oil and experiment II diet(high fat diet) containing 18% fat(13% beef tallow and 5% corn oil) and 1% cholesterol were divided into four groups with 0.5% cellulose(0.5CL : control), 10% cellulose(10CL), 10% Indigestible dextrin I(ID-I) and 10% indigestible dextrin II(ID-II), respectively. ID-I and ID-II diets produced a decrease in serum total lipid and cholesterol concentrations in both normal and high fat diets but more significantly in the groups fed high fat diet. ID-I, ID-II, and 10CL of high fat diet decreased serum glucose concentration. ID-I and ID-II diets also decreased liver cholesterol and triglyceride concentrations in rats fed normal and high fit diets. The fecal excretion of cholesterol in ID-II/high fat diet was significantly higher than that of 0.5CL diet. Bile acid excretion was increased in the rats fed 10CL and ID-II diets of normal and high fat diets. These observations indicat the hypolipidemic and hypocholesterolemic effects of indigestible dextrin on the rats. These effects are mediated by the increase of facal cholesterol and bile acid excretion. (Korean J Nutrition31(6) : 981-990, 1998)
The metabolic fate of ginsenosides including gastrointestinat absorption, organ distribution, excretion and metabolism in liver was investigated by tracer studies using the radio-labeled ginsenosides. 3H-ginsenosides were shown to be absorbed from the mouse digestive tract and then to be excreted rapidly into urine and/or bile. Bile juice was concluded to play a significant role in absorption of ginsenosides. The total concentration of radioactivity persisted in tissues 24 hrs after oral administration was less than 1.3% of the administered dose and Rbl showed the highest value. The concentrations of radioactivity were relatively high in the liver and kidney. After administration of Rbl radioactivity was detected in the brain. After oral administration of 8H-ginsenosides, major component excreted into urine was found to be the intact ginsenosides and decomposed and/or metabolized products were found in GIT in the case of Rbl. 3H-ginsenoside Rbl was shown to be metabolized in the liver and the metabolite was suggested to be an acylated compound of Rbl by a certain organic acid.
