들현호색으로부터 Berberine과 Coptisine의 분리 및 함량분석 (Isolation and Quantitative Determination of Berberine and Coptisine from Tubers of Corydalis ternata)

  • Corydalis Tuber has been used in traditional medicine for an analgesic, antispasmodic and gastric ulcers. For the quality control on this drug, isolation and quantitative determination of berberine and coptisine from Corydalis ternata Nakai (Papaveraceae) has been conducted by using HPLC method. Berberine and coptisine in quarternary alkaloidal fraction from the crude drug were separated on silicagel column using a $CHCl_3:MeOH\;(85:15)$ and $CHCl_3:MeOH:H_2O\;(70:30:4)$ as an eluent, and the average contents were about 0.93 and 0.36%.

흡광도측정법에 의한 황백과 제제 중 프로토베르베린 알칼로이드의 정량 (Determination of Protoberberine Alkaloids in Phellodendri Cortex and Preparation by Spectrophotometric Method)

  • The Phellodendri Cortex of Phellodendron amurense (Rutaceae) is known to contain a number of isoquinoline alkaloid, and berberine, palmatine, jateorrhizine, phellodendrine and magnoflorine are the major constituents of protoberberine alkaloids. For the determination of protoberberine alkaloids from Phellodendri Cortex and berberine chloride from the preparation, the new spectrophotometric method was developed with a simple and selective sample clean-up using thiocyanatocobaltate[II] complex ion. Samples were extracted with 0.1 mM hydrochloric acid, potassium biphthalate reagent, thiocyanatocobaltate reagent and 1.2-dichloroethane for 60 min. The absorbance of protoberberine alkaloid complexes in 1.2-dichloroethane solution was measured at 625 nm. Calibration curve for berberine was linear over the concentration range of 0.05~0.30 mg/ml 1.2-dichloroethane. The method proved to be rapid, simple and reliable for the determination of protoberberine alkaloids from Phellodendri Cortex and berberine chloride from the preparation.

황련과 감초의 수용성 혼합물로부터 얻어진 반응침전물의 세포독성 (Cytotoxicity of Reaction-Precipitate from Coptidis Rhizoma and Glycyrrhizae Radix Aqueous Mixture)

  • The purpose of this research was to investigate the effect of reaction-precipitate from Coptidis Rhizoma and Glycyrrhizae Radix aqueous mixture(CGP) on the cytotoxicity. The effects of CGP on the growth of tumor cells, Balb/c 3T3 cell, mouse spleen cell and human lymphocyte were compared with those of berberine, glycyrrhizin and berberine glycyrrhizinate(BG), which were estimated by MTT colorimetric assay or cell counting. CGP, berberine and BG inhibited the growth of several tumor cells, such as Hep G2, A549, Raji, MCF-7, HeLa and KHOS-NP. Whereas, glycyrrhizin inhibited the growth of Raji and MCF-7, CGP did not affect on Balb/C 3T3 cells, mouse spleen cells and human lymphocyte at $10^{-6}{\sim}10^{-5}g/ml$. CGP increased the number of leukocyte in mice. This results indicate that CGP have the inhibitory action of the growth of human tumor cells, and the side effect of CGP is less than berberine and BG.

Calcium Channel Blocking and $\alpha$-Adrenoceptor Blocking Action of Coptidis Rhizoma Extracts and their Alkaloid Components in Rat Aorta

  • Vascular relaxation action of crude extracts of two kinds of Coptidis rhizoma (Coptis chinensis and Coptis japonica, Ranunculaceae) was investigated and compared with that of berberine and palmatine, active alkaloid components of these plants. The results show that total extracts, berberine, and palmatine induced a concentration-dependent vasodilatation of rat thoracic aorta contracted with phenylephrine (PE). Palmatine, unlike to berberine, did not inhibit contraction induced by KCI. In calcium-free media, not only berberine but also crude extracts inhibited calcium-induced contraction. Furthermore, pretreatment of crude extracts inhibited contraction induced by PE noncompetitively. In PE-induced contraction, berberine was 2.5 times more potent than Coptis chinensis in the relaxation of rat aorta in terms of $IC_{50}$ values. Analysis of the effects of crude extracts on the Emax and $IC_{50}(PE)IC_{50}(KCI)$ ratios provides information on selectivity and indicates that extracts exhibit greater inhibition of the contrac tile response induced by PE than by KCI. We concluded that crude extracts have .alpha.-adrenoceptor blocking action and possesses inhibitory effect on calcium influx, which may be at least in part responsible for the antihypertensive action.

섬유아세포에서의 MMP-2 및 MMP-9 활성 억제에 의한 베르베린의 항주름 효과 (Anti-wrinkle effect of berberine by inhibition of MMP-2 and MMP-9 activity in fibroblasts)

  • 우리는 황련에서 분리된 베르베린의 화장품소재로의 활성을 평가하기 위해 항산화와 항주름 효능을 분석하였다. 시료의 독성을 평가하기 위해 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay를 수행하였으며 항산화와 항주름 활성평가를 위해 ROS 생성능, hyaluronic acid 생성능, MMP-2, -9, TIMP-1,-2, $TNF-{\alpha}$의 발현을 각각 측정하였다. 베르베린에 대한 자외선의 세포 독성은 CCD-986sk 섬유아세포를 이용하여 MTT 분석으로 측정되었고, 세포 독성이 $25{\mu}g/mL$의 농도 미만에서 관찰되지 않았다. 또한 베르베린이 ROS 생산을 농도 의존적으로 감소시키고 히알루론산의 합성을 촉진한다는 것을 발견했다. 베르베린은 주름 형성과 관련된 MMP-2 및 MMP-9의 protein 단계 및 mRNA 발현을 감소시키고 TIMP-1 및 TIMP-2의 발현을 증가시켰다. 프로 pro-inflammatory cytokine 으로 알려진 $TNF-{\alpha}$에 대한 베르베린의 억제효능을 실험한 결과, 농도의존적으로 $TNF-{\alpha}$ 유전자를 억제하는 효과를 나타내었다. 이러한 결과를 통해 베르베린은 활성산소종의 억제와 TIMP-1과 -2의 발현 조절에 의한 MMP-2,-9의 감소로 항산화 및 항주름에 효과가 있는 화장품 소재로서의 가치를 가진다고 사료된다.

Hep3B 세포에서 베르베린은 DNA methyltransferase3b 억제를 통해 p53을 발현시켜 세포사멸을 유도 (Berberine Induces p53-Dependent Apoptosis through Inhibition of DNA Methyltransferase3b in Hep3B Cells)

  • 종양 억제 유전자 p53은 인간 간암세포 Hep3B에서는 불활성화되어 있다. 베르베린(berberine)은 암세포의 증식을 억제하는 것으로 보고되어 있다. 우리는 베르베린을 처리한 Hep3B 세포에서 세포사멸이 유도되는지를 조사하였고 이 세포사멸이 p53과 DNA methyltransferase의 발현과 연관되어 있는지를 관찰하였다. MTT 분석을 통하여 세포 생존력을 측정하였다. 세포사멸은 각각 Annexin V flow 세포 분석을 사용하여 측정하였다. 베르베린이 처리된 세포에서 DNMT 효소 활성, mRNA 발현, 단백질 발현 정도가 검사되었으며, p53 단백질 발현은 웨스턴 블롯 분석에 의해 검사되었다. 베르베린 처리는 시간 및 용량 의존적으로 Hep3B세포의 세포사멸을 증가시켰다. 베르베린 처리 시 DNMT3b의 활성 정도, mRNA 발현 그리고 단백질 발현 정도가 모두 감소되었다. 이와는 대조적으로, Hep3B에서는 비활성인 p53 단백질의 발현은 DNMT3b의 감소와 동시에 증가했다. ERK의 발현은 변화가 없었으나, P-ERK의 발현은 농도 의존적으로 감소하는 것으로 나타났다. 이러한 결과는 Hep3B 세포에 베르베린의 처리는 DNMT3b의 발현을 감소시켜서 종양 억제 유전자인 p53의 증가를 유도할 수 있고, 이를 통해서 세포사멸을 증가 시킬 수 있다는 것을 나타낸다. 이는 베르베린이 간암 세포의 증식 억제에 효과적으로 작용할 수 있음을 보여준다.

Berberine Chloride Inhibits Receptor Activator of $NF-{\kappa}B$ Ligand-induced Osteoclastogenesis via Preventing ERK Activation

  • An imbalance in bone remodeling that is caused by increased bone resorption over bone formation leads to most adult skeletal diseases including osteoporosis. Since the development of anti-resorptive agents from natural substances has recently gained more interest in the treatment of osteoporosis, we evaluated the effects of 222 natural compounds on receptor activator of $NF-{\kappa}B$ ligand (RANKL)-induced of tartrate-resistance acid phosphatase (TRAP) activity in RAW264.7 murine macrophage cell, and found that berberine chloride is one of compounds inhibiting RANKL-induced TRAP activity. Berberine chloride significantly inhibited the RANKL-induced TRAP activity and the formation of multinucleated osteoclasts in a dose-dependent manner. In addition, berberine chloride prevented the RANKL-induced mRNA expression of TRAP, matrix metalloproteinase 9 and c-Src, which have been known to be highly expressed in the process of osteoclastogenesis. Interestingly, berberine chloride prevented the RANKL-induced activation of extracellular signal-regulated kinase (ERK) which is one of mitogen-activated protein (MAP) kinases. In conclusion, berberine chloride could inhibit the osteoclastogenesis via preventing the activation of ERK/MAP kinase signaling pathway.

Enhancement of Arsenic Trioxide ($As_2O_3$)-Mediated Apoptosis Using Berberine in Human Neuroblastoma SH-SY5Y Cells

  • Objective : Arsenic trioxide ($As_2O_3$) has been used as an anticancer agent in traditional Chinese medicine for thousand years and berberine is an isoquinoline alkaloid present that has indicated significant antimicrobial activity. We have examined the combined anticancer effects of $As_2O_3$ and berberine against the human neuroblastoma (HNB) SH-SY5Y cells in vitro, and to elucidate underlying molecular mechanism. Methods : HNB SH-SY5Y cells were treated with $2\;{\mu}M\;As_2O_3$ and $75\;{\mu}g/ml$ berberine, and their survival, cell death mechanism as well as synergistic cytotoxic effects were estimated by using MTT assay, DAPI staining, agarose gel electrophoresis, flow cytometric analysis, and western blot analysis. Results : The combined treatment of two drugs also markedly decreased cell viability. The cytotoxic effects of two drugs were revealed as apoptosis characterized by chromatin condensation, DNA fragmentation, and the loss of mitochondrial membrane potential. The apoptotic cytotoxicity was accompanied by activation of caspase-3 protease as well as decreased the expression of Bcl-2, Bid, and Bcl-x/L. In addition, the cells treated with combination of two drugs also showed significantly increased intracellular reactive oxygen species levels and lipid peroxidation compared to cells $As_2O_3$or berberine only. Conclusion : Combined treatment of $As_2O_3$ with berberine induced activation of apoptotic signaling pathways in HNB SH-SY5Y cells. These results suggest that the possibility of the combined treatment of two chemotherapeutic agents with low concentration improving cytotoxic effect for cancer cells with minimal side effects.

Combination of berberine and ciprofloxacin reduces multi-resistant Salmonella strain biofilm formation by depressing mRNA expressions of luxS, rpoE, and ompR

  • Bacterial biofilms have been demonstrated to be closely related to clinical infections and contribute to drug resistance. Berberine, which is the main component of Coptis chinensis, has been reported to have efficient antibacterial activity. This study aimed to investigate the potential effect of a combination of berberine with ciprofloxacin (CIP) to inhibit Salmonella biofilm formation and its effect on expressions of related genes (rpoE, luxS, and ompR). The fractional inhibitory concentration (FIC) index of the combination of berberine with CIP is 0.75 showing a synergistic antibacterial effect. The biofilm's adhesion rate and growth curve showed that the multi-resistant Salmonella strain had the potential to form a biofilm relative to that of strain CVCC528, and the antibiofilm effects were in a dose-dependent manner. Biofilm microstructures were rarely observed at $1/2{\times}MIC/FIC$ concentrations (MIC, minimal inhibition concentration), and the combination had a stronger antibiofilm effect than each of the antimicrobial agents used alone at $1/4{\times}FIC$ concentration. LuxS, rpoE, and ompR mRNA expressions were significantly repressed (p< 0.01) at $1/2{\times}MIC/FIC$ concentrations, and the berberine and CIP combination repressed mRNA expressions more strongly at the $1/4{\times}FIC$ concentration. The results indicate that the combination of berberine and CIP has a synergistic effect and is effective in inhibiting Salmonella biofilm formation via repression of luxS, rpoE, and ompR mRNA expressions.

향류분배 크로마토그라피법에 의한 황련($Coptis$ $chinensis$) 뿌리로 부터 Berberine의 분리 (Isolation of Berberine from the Rhizome of $Coptis$ $chinensis$ by Centrifugal Partition Chromatography)

  • 황련은 염증, 항균, 고혈압 및 항암 작용을 지닌 중요한 전통 한약재이다. 황련 뿌리 메탄올 추출성분은 CPC와 HPLC 방법으로 정제하였다. CPC의 최적 용매 조건은 n-butanol:acetic acid:water(4:1:5)이었다. Berberine(16.8 mg)은 CPC와 HPLC 방법에 의하여 효과적으로 분리하였다. 이 물질의 화학적 구조는 $^1H$, $^{13}C$-NMR과 ESI-MS 데이터 분석에 의하여 확인되었다.
