Protective Antitumor Activity through Dendritic Cell Immunization is Mediated by NK Cell as Well as CTL Activation

  • Dendritic cells (DCs) are potent professional antigen-presenting cells (APC) capable of inducing the primary T cell response to antigen. Although tumor cells express target antigens, they are incapable of stimulating a tumor-specific immune response due to a defect in the costimulatory signal that is required for optimal activation of T cells. In this work, we describe a new approach using tumor-DC coculture to improve the antigen presenting capacity of tumor cells which does not require a source of tumor-associated antigen. Immunization of a weakly immunogenic and progressive tumor cocultured with none marrow-derived DCs generated an effective tumor vaccine. Immunization with the cocutured DCs was able to induce complete protectiv immunity against tumor challenges and was effective for the induction of tumor-specific CTL (cytotoxic T lymphocyte) activity. Furthermore, high NK cell activity was observed in mice in which tumors were rejected. In addition, immunization with tumor-pulsed DC s induced delayed tumor growth, but not tumor eradication in tumor-bearing mice. Our results demonstrate that coculture of DCs with tumors generated antitumor immunity due to the NK cell activation as well as tumor-specific T cell. This approach would be used for designing tumor vaccines using DCs when the information about tumor antigens is limited.

국민건강증진사업 인력 교육 프로그램 개발 (Development of Health Professional Training Program for National Health Promotion Services)

  • National health promotion services are any planned combination of comprehensive and specific services to help people to achieve and maintain health. The purpose of the service activities is to identify health needs, to obtain information and resources and to achieve change of individuals, families, groups, or entire communities. It also includes environmental support of social, political, economic, and organizational policy and regulatory arrangements bearing on behavior or more directly on health. To be most effective, the service must be planned and delivered by health professional. Therefore, the aim of this study has been to develop a health professional training program for national health promotion. The specific aims of this study were: 1) to clarify the roles, responsibilities and competencies of health promotion practitioners; 2) to help health promotion practitioners to get the knowledge, skills, and abilities for any heath promotion project or program that seeks to improve health; 3) to help health promotion practitioners to acquire the skills and abilities to encourage people to participate in the health promotion project, to access the health needs and available community resources, and develop community-wide health promotion program strategies. The health professional training program developed in this study included factors affecting education, the demands of training program and roles and responsibilities of health promotion practitioners. This study also developed the curriculum for health promotion practitioners. The curriculum had six topics: 1) government's health promotion policies and projects or programs; 2) health management and plan; 3) recent health issues and future perspectives; 4) data on various health status indices; 5) strategies to implement health promotion projects or programs; and 6) introduction of some effective and comprehensive health promotion projects or programs.

병솔꽃나무(Callistemon citrinus (Curt.) Skeels)의 개화결실 및 증식특성 (Characteristics of flowering, fruit setting and propagation of Callistemon citrinus (Curt.) Skeels)

  • 본 연구는 외래도입 병솔꽃나무(Callistemon citrinus)의 개화결실 습성과 효율적인 증식방법을 구명 하고자 수행하였다. 병솔꽃나무의 삭과는 개화후 4년간 가지에 존속하며, 종자의 수와 크기는 3년생 삭과와 2년생 삭과가 비슷한 반면 1년생 삭과는 크게 차이가 나는 것으로 나타났다. 삭과로부터 종자를 탈리시키기 위해 건조시킨 결과 삭과의 상대 수분함량이 약 75%일 때 삭과열개율이 100%에 달했다. 발아율은 3$0^{\circ}C$에서 건조시킨 종자가 가장 높았으며, 그 중 2년생 삭과에서 탈리된 종자의 발아율이 높았다. 반면 50, 7$0^{\circ}C$에서 건조되어 탈리된 종자의 발아율은 낮았다. 가을철 2차 개화를 유도하기 위하여 봄철 개화 후 삭과가 달린 가지를 전정한 개체에서는 모든 개체에서 가을에 개화하였으며 , 개화량은 평균 14송이였다. 반숙지를 IBA 100mgι$^{-1}$ 에 24시간 침적처리한 결과 86.4%가 발근되었으며, 평균 12.6개의 1차근을 가지며 생장이 가장 양호하였다.

  • 보안정책의 두 가지 구성요소는 내용과 형식이다. 지금까지 보안 정책 내용에 대한 연구는 폭 넓게 수행되어온 반면, 보안 정책의 형식에 대해서는 관련연구가 매우 부족한 실정이다. 형식이 보안정책의 성공을 결정하는 중요한 요인이기 때문에 어떻게 보안 정책의 형식을 작성할 것인가는 매우 중요하다. 따라서 본 연구의 목적은 보안 정책의 형식과 조직 구성원들의 보안 정책 준수간의 관계를 규명하는 것이다. 분석결과 보안정책의 형식은 보안준수태도, 주관적 규범, 인지된 행위 통제, 그리고 인지된 보안정책 준수비용에 유의한 영향을 미치는 것으로 나타났으며, 보안정책 준수태도와 주관적 규범은 지속적 보안준수 의도에 유의한 영향을 미치는 것으로 나타났다.

GIS 기반 Weight of Evidence 기법을 이용한 포천 지역의 지하수 산출특성 예측도 작성 (Feasibility Mapping of Groundwater Yield Characteristics using Weight of Evidence Technique based on GIS in the Pocheon Area)

  • 본 연구에서는 경기도 포천군 일대의 지하수 부존특성 가능지역을 공간적으로 예측하기 위하여 GIS 기반으로 Weight of Evidence(WofE) 기법을 적용하였다. 기본 자료로는 지하수 산출 특성분석과 관련된 수문지질학적 인자인 지표피복, 지형, 지질 등의 자료를 GIS 정보로 입력하였다. 그리고, 베이지안 확률 분석기법(Bayesian Method)에 기반하여 기본자료와 함께 선구조, 암상, 수계밀도, 식생, 토양, 토지이용현황등과 같이 산출성에 영향을 주는 인자들에 대하여 지하수 부존가능 지수와 사전/사후확률을 구하였다. 이들 자료에 대해 다시 WofE 기법을 적용하여, 각 인자들의 W+, W- 가중 값들을 계산하였다. 또한 이러한 결과의 차이 값으로 공간적인 상관관계를 구하여 지하수 산출특성의 예측가능도를 작성하고자 하였다. 본 연구에 적용한 방법은 잠재된 지하수 부존 지역과 주변 지역의 공간적 분포를 파악하는 데 유용한 것으로 생각된다.

Prestress evaluation in continuous PSC bridges by dynamic identification

  • In the last decades, research efforts have been spent to investigate the effect of prestressing on the dynamic behaviour of prestressed concrete (PSC) beams. Whereas no agreement has been reached among the achievements obtained by different Researchers and among the theoretical and the experimental results for simply supported beams, very few researches have addressed this problem in continuous PSC beams. This topic is, indeed, worthy of consideration bearing in mind that many relevant bridges and viaducts in the road and railway networks have been designed and constructed with this structural scheme. In this paper the attention is, thus, focused on the dynamic features of continuous PSC bridges taking into account the effect of prestressing. This latter, in fact, contributes to the modification of the distribution of the bending stress along the beam, also by means of the secondary moments, and influences the flexural stiffness of the beam itself. The dynamic properties of a continuous, two spans bridge connected by a nonlinear spring have been extracted by solving an eigenvalue problem in different linearized configurations corresponding to different values of the prestress force. The stiffness of the nonlinear spring has been calculated considering the mechanical behaviour of the PSC beam in the uncracked and in the cracked stage. The application of the proposed methodology to several case studies indicates that the shift from the uncracked to the cracked stage due to an excessive prestress loss is clearly detectable looking at the variation of the dynamic properties of the beam. In service conditions, this shift happens for low values of the prestress losses (up to 20%) for structure with a high value of the ratio between the permanent load and the total load, as happens for instance in long span, continuous box bridges. In such conditions, the detection of the dynamic properties can provide meaningful information regarding the structural state of the PSC beam.

Geotechnical engineering behavior of biopolymer-treated soft marine soil

  • Soft marine soil has high fine-grained soil content and in-situ water content. Thus, it has low shear strength and bearing capacity and is susceptible to a large settlement, which leads to difficulties with coastal infrastructure construction. Therefore, strength improvement and settlement control are essential considerations for construction on soft marine soil deposits. Biopolymers show their potential for improving soil stability, which can reduce the environmental drawbacks of conventional soil treatment. This study used two biopolymers, an anionic xanthan gum biopolymer and a cationic ${\varepsilon}-polylysine$ biopolymer, as representatives to enhance the geotechnical engineering properties of soft marine soil. Effects of the biopolymers on marine soil were analyzed through a series of experiments considering the Atterberg limits, shear strength at a constant water content, compressive strength in a dry condition, laboratory consolidation, and sedimentation. Xanthan gum treatment affects the Atterberg limits, shear strength, and compressive strength by interparticle bonding and the formation of a viscous hydrogel. However, xanthan gum delays the consolidation procedure and increases the compressibility of soils. While ${\varepsilon}-polylysine$ treatment does not affect compressive strength, it shows potential for coagulating soil particles in a suspension state. ${\varepsilon}-Polylysine$ forms bridges between soil particles, showing an increase in settling velocity and final sediment density. The results of this study show various potential applications of biopolymers. Xanthan gum biopolymer was identified as a soil strengthening material, while ${\varepsilon}-polylysine$ biopolymer can be applied as a soil-coagulating material.

가변피치 프로펠러의 축기전력 특성에 관한 연구 - 세계로호를 중심으로 - (A Study on the Characteristics of Shaft Electric Motive Force of Controllable Pitch Propeller - Focusing on the M/V Segero -)

  • 대부분의 기계는 여러 종류의 금속으로 구성된다. 특히 선박의 축계는 프로펠러 날개의 황동과 스테인리스로 된 축으로 이루어져 있다. 이 이종금속이 바닷물의 전해액에 들어가면 볼타 전지를 이루고, 기전력이 발생된다. 이 기전력은 축계를 받치고 있는 베어링과 축을 전기부식 시키는 원인이 된다. 선박에서는 이 부식을 막기 위해 선박에서는 축 접지 시스템을 설치하여 운용하고 있다. 본 연구는 가변피치 프로펠러의 축기전력을 측정하기 위하여 추진축의 전압과 주기관의 회전수를 동시에 측정하였다. 측정장치는 내셔널인스트루먼트사의 24bit A/D컨버터를 사용하여 측정하였고, 프로그램은 LabVIEW를 사용하였다. 주기관의 회전수와 축기전력의 발생, 블레이드 각도에 따른 기전력과, 배의 항해 방향에 따른 축기전력을 측정하고 분석하였다.

Modeling of composite MRFs with CFT columns and WF beams

  • A vast amount of experimental and analytical research has been conducted related to the seismic behavior and performance of concrete filled steel tubular (CFT) columns. This research has resulted in a wealth of information on the component behavior. However, analytical and experimental data for structural systems with CFT columns is limited, and the well-known behavior of steel or concrete structures is assumed valid for designing these systems. This paper presents the development of an analytical model for nonlinear analysis of composite moment resisting frame (CFT-MRF) systems with CFT columns and steel wide-flange (WF) beams under seismic loading. The model integrates component models for steel WF beams, CFT columns, connections between CFT columns and WF beams, and CFT panel zones. These component models account for nonlinear behavior due to steel yielding and local buckling in the beams and columns, concrete cracking and crushing in the columns, and yielding of panel zones and connections. Component tests were used to validate the component models. The model for a CFT-MRF considers second order geometric effects from the gravity load bearing system using a lean-on column. The experimental results from the testing of a four-story CFT-MRF test structure are used as a benchmark to validate the modeling procedure. An analytical model of the test structure was created using the modeling procedure and imposed-displacement analyses were used to reproduce the tests with the analytical model of the test structure. Good agreement was found at the global and local level. The model reproduced reasonably well the story shear-story drift response as well as the column, beam and connection moment-rotation response, but overpredicted the inelastic deformation of the panel zone.

Amphidinium stirisquamtum sp. nov. (Dinophyceae), a new marine sand-dwelling dinoflagellate with a novel type of body scale

  • Amphidinium species are amongst the most abundant benthic dinoflagellates in marine intertidal sandy ecosystems. Some of them produce a variety of bioactive compounds that have both harmful effects and pharmaceutical potential. In this study, Amphidinium cells were isolated from intertidal sand collected from the East China Sea. The two strains established were subjected to detailed examination by light, and scanning and transmission electron microscopy. The vegetative cells had a minute, irregular, and triangular-shaped epicone deflected to the left, thus fitting the description of Amphidinium sensu stricto. These strains are distinguished from other Amphidinium species by combination characteristics: (1) longitudinal flagellum inserted in the lower third of the cell; (2) icicle-shaped scales, 276 ± 17 nm in length, on the cell body surface; (3) asymmetrical hypocone with the left side longer than the right; and (4) presence of immotile cells. Therefore, they are described here as Amphidinium stirisquamtum sp. nov. The molecular tree inferred from small subunit rRNA, large subunit rRNA, and internal transcribed spacer-5.8S sequences revealed that A. stirisquamtum is grouped together with the type species of Amphidinium, A. operculatum, in a fully supported clade, but is distantly related to other Amphidinium species bearing body scale. Live A.stirisquamtum cells greatly affected the survival of rotifers and brine shrimp, their primary grazers, making them more susceptible to predation by the higher tropic level consumers in the food web. This will increase the risk of introducing toxicity, and consequently, the bioaccumulation of toxins through marine food webs.
