A process for the production of mannitol from fructose (5% to 25%) using Leuconostoc mesenteroides NRRL B-1149 was investigated. Fermentations were carried out In batch or fed-batch fermentations without aeration at 28$\^{C}$, pH 5.0. When 5% fructose was used In batch culture fermentation, the yield of mannitol was 78% of that expected theoretically. When the frurtose concentration was Increased to 10%, the yield dropped to 59.6% of the theoretical value. However, In the fed-batch culture, using 10% fructose, the yield was 81.9% of the theoretical value. In a 15% fruttose fed-batch culture, with 5% fructose being added initially and the other 10% fructose being added as a continuous supply the final yield was 83.7% of the theoretical yield. When 20% fructose was used In the same manner, the yield was 89.5% of theoretical yield.

Multivariate analysis and batch monitoring on a pilot-scale sequencing batch reactor (SBR) are described for integrated wastewater treatment management system, where a batchwise multiway independent component analysis method (MICA) are used to extract meaningful hidden information from non-Gaussian wastewater treatment data. Three-way batch data of SBR are unfolded batch-wisely, and then a non-Gaussian multivariate monitoring method is used to capture the non-Gaussian characteristics of normal batches in biological wastewater treatment plant. It is successfully applied to an 80L SBR for biological wastewater treatment, which is characterized by a variety of error sources with non-Gaussian characteristics. The batchwise multivariate monitoring results of a pilot-scale SBR for integrated wastewater treatment management system showed more powerful monitoring performance on a WWTP application than the conventional method since it can extract non-Gaussian source signals which are independent and cross-correlation of variables.
A comparative study on response of a toxic compound thiocyanate ($SCN^-$) was carried out in continuous and fed batch moving bed reactor systems. Both systems had three sequential anaerobic, anoxic and aerobic reactors and operated at same hydraulic retention time. Feed $SCN^-$ was first increased from 600 mg/L to 1,000 mg/L for 3 days (shock 1) and then from 600 to 1,200 mg/L for 3 days (shock 2). In anaerobic continuous reactor, increase of effluent COD (chemical oxygen demand) due to shock load was only 2%, whereas in fed batch reactor it was 14%. In anoxic fed batch reactor recovery was partial in terms of $SCN^-$, phenol, COD and $NO{_3}{^-}$-N and $NO{_2}{^-}$-N removals and in continuous reactor complete recovery was possible. In both systems, inhibition was more significant on aerobic reactors than anaerobic and anoxic reactors. In aerobic reactors ammonia removal efficiency deteriorated and damage was irreversible. Present study showed that fed batch reactors showed higher substrate removal efficiency than continuous reactors during regular operation, but are more susceptible to toxic feed shock load and in nitrifying reactor damage was irreversible.
Zooloea ramigera 115를 사용하여 생물응집제로서 사용되는 생물고분자생산 실험을 하였다. 생물고분자 생산을 높이기 위하여 회분식, 유가배양, 연속배양방법을 사용하였다. 탄소원으로는 포도당, 유당, 당밀, 유청을 사용하였다. 기질이 포도당의 경우에는 C/N배 98일 때 생물고분자 생산 효과가 좋았으며, 유당의 경우에는 C/N비 30, 당밀과 유청의 경우에는 C/N비 60일 때 생물고분자 생산이 가장 좋았다. 유가배양 방법이 회분식 배양방법 보다 최종 생물고분자 생산이 우수하였다. C/N비를 달리한 2단계 연속 배양방법으로 생산성을 향상시켰다. 당밀의 경우 0.048$hr^{-1}$의 희석속도에서, 유청의 경우 0.096$hr^{-1}$에서 생산성이 가장 좋았다.
산화물 형태의 사용후핵연료를 용융염에서 금속 형태로 전환하여, 발열량, 부피 및 방사능을 1/4로 감소시킬 수 있는 전기화학적 금속전환 공정을 개발하고, 5kg $U_3O_8$/Batch 규모의 mock-up 실험을 수행하였다. 본 연구에서는 전해 셀의 운전변수를 해석하였으며, 아울러 hot test를 위한 장치개발 연구도 병행하였다. 전기화학적 금속전환 공정을 이용하여 $U_3O_8$ 형태의 천연우라늄 분말을 99% 이상 금속전환할 수 있었으며, 또한 20kg $U_3O_8$/batch 규모 장치의 설계자료를 산출할 수 있었다.
The biodegradability of organics has become essential for the design and modeling of a biological nutrient removal process. Respirometry for the batch test just with wastewater has been conducted to estimate active biomass and readily biodegradable organics in municipal wastewater simultaneously. Municipal wastewater contains significant active biomass, which is estimated about 17% of COD. Compared to the batch test seeded with sludge, the batch test just with wastewater represents a little higher readily biodegradable organics. This might be due to the different environment of the logarithmic growth of active biomass. The nitrate uptake rate test has been also performed for the estimation of the readily biodegradable organics. The nitrate uptake rate test results in a little higher readily biodegradable organics compared to the batch test seeded with sludge and similar organics compared to the batch test just with wastewater. This might be caused by the different sludge of a sequencing batch reactor process. Taking the result of the previous research into account, the readily biodegradable, slowly biodegradable, active biomass, soluble inert, and particulate inert organics are estimated about 11%, 49%, 17%, 11%, and 12% of COD, respectively.
Acarbose, a pseudo-oligosaccharide, is widely used clinically in therapies for non-insulin-dependent diabetes. In the present study, S-adenosylmethionine (SAM) was added to selected media in order to investigate its effect on acarbose fermentation by Actinoplanes utahensis ZJB-08196. Acarbose titer was seen to increase markedly when concentrations of SAM were added over a period of time. The effects of glucose and maltose on the production of acarbose were investigated in both batch and fed-batch fermentation. Optimal acarbose production was observed at relatively low glucose levels and high maltose levels. Based on these results, a further fed-batch experiment was designed so as to enhance the production of acarbose. Fed-batch fermentation was carried out at an initial glucose level of 10 g/l and an initial maltose level of 60 g/l. Then, 12 h post inoculation, 100 ${\mu}mol/l$ SAM was added. In addition, 8 g/l of glucose was added every 24 h, and 20 g/l of maltose was added at 96 h. By way of this novel feeding strategy, the maximum titer of acarbose achieved was 6,113 mg/l at 192 h. To our knowledge, the production level of acarbose achieved in this study is the highest ever reported.
Laboratory scale experiments were conducted to study the applicability, and to compare the performance of two types of sequencing batch reactor (SBR)systems, a conventional SBR and sequencing batch biofilm reactor (SBBR) on the biological nitrogen and phosphorus removal. The nitrification rate in SaR was higher than that in SBBR both in high influent TOC concentration. The denitrification was completed at the first non-aeration period in SBR, however, the additional non-aeration period should be installed or the first aeration period should be extended more in order to complete the nitrogen removal in SBBR. The time at the first aeration period was more needed as about 4-5 h in order to uptake all the released $PO_4^{3-}\;-P$ at the first non-aeration period. SBBR needed more operation time, especially the first aeration time, than SBR at the high influent TOC concentration in order to complete nitrogen and phosphorus removal.

Recombinant Bacillus subtilis LKS88[pASA240] containing the amylase gene from Streptomyces albus KSM-35 was exploited in fed-batch cultivation for mass production of maltotetraose-producing amylase. The effects of dissolved oxygen, additional organic nutrients (peptone and yeast extract) and mixed carbon sources (glucose plus soluble starch) on amylase production were examined in fed-batch operations in an effort to determine the optimum conditions for a maximum amylase productivity. Under the optimum conditions, maximum amylase activity was about 4.2 times higher than that obtained in batch cultivations, indicating that mass production of maltotetraose-producing amylase could be accomplished in fed-batch cultivation of the recombinant B. subtilis strain.
