재배방법이 다른 미나리의 항세균 활성 (Antibacterial Activities in Watercress(Oenanthe javanica D.C.) Cultivated with Different Culture Methods)

  • Antibacterial activities in each part of watercress(Oenanthe javanica D.C.) grown under different culture conditions were measured to determine the possibility to use watercress as a resource to develop the antibacterial substance. The leaves of watercress were extracted with methanol and the methanol extract was further fractionated with various organic solvents. Antibacterial activities against Shigella dysenteriae ATCC 9361 in all fractions were determined according to the agar diffusion method using paper disc. Methanol extract of watercress leaves was more effectively inhibited the growth of the tested bacteria than the extracts of roots or stems at the concentration of 0.5 g eq./disc, and the extract of watercress from Hwasoon was the most effective one as compared to others. Phenolic and neutral fractions fractionated from methanol extract of watercress had a considerable inhibiting activity on the growth of the bacteria, but acidic and basic fractions did not show any inhibitory effect. Minimum inhibitory concentrations of phenolic and neutral fractions against Shigella dysenteriae ATCC 9361 were $400\;{\mu}g/disc$ and $550\;{\mu}g/disc$, respectively.

광릉쥐오줌풀의 정유향 특성 (Odor Characteristics of Essential Oil of Valeriana fauriei var. dasycarpa HARA)

  • 광릉쥐오줌풀 뿌리의 정유성분 조성 및 정유의 특성을 더욱 정밀하게 조사하기 위하여 수증기 증류에 의해 얻어진 정유성분을 산성, 페놀성, 염기성 및 중성성분 분획으로 분리하여 분획별 향의 관능평가 및 분획물의 조성을 조사한 결과는 다음과 같다. 1. 광릉쥐오줌풀 뿌리의 정유성분분획물 각각의 조성은 중성성분 분획물 92%, 산성성분 분획물 6%, 염기성성분 및 페놀분획물은 각각 1% 미만이며 각 분획물의 향특성은 중성성분 분획은 sweet-balsamic, woody, musky, medicinal 향이며 산성성분 분획은 sweat-socks, valeric-like, cheese-like 향이었다. 2. 중성성분분획물을 용매의 극성을 달리하여 분획한 결과 용출 용매별 분획 비율은 ethyl ether분획물이 44%, pentane : ethyl ether분획물 34%, pentane과 methanol분획물은 각각 11%였다. 분획별 향 특성은 ethyl ether 분획물은 woody, medicinal 향이며 pentane :. ethyl ether 분획물은 sweet-balsamic, woody, herb-like향 특성을 나타내었다. 극성을 달리한 용출용매별 분획물에서 확인된 성분은 ethyl ether 분획물에서 valeranone 등 12종, pentane : ethyl ether(9 : 1) 분획물에서는 bornyl acetate등 26종, pentane 분획물에서 camphene등 43종을 확인하였다. 3. 산성성분분획에서는 n-butanoic acid 외 44종을 확인하였고 그 중 양적으로 많은 성분은 3-methyl butanoic acid 42.1%, dimethoxy-2-propanoic acid 11.5%, 5-ethyldihydro-2H-furanone 5.7% 등 이었다.

무화과잎 페놀성 분획중의 항미생물 활성물질의 정제 및 동정 (Purification and Identification of Antimicrobial Substances in Phenolic Fraction of Fig Leaves)

  • 무화과잎의 메탄올추출물을 용매분획하여 얻은 분획중 산성 분획과 페놀성 분획이 항미생물 활성이 강한 것으로 나타났고 페놀성 분획이 산성 분획보다 항미생물 활성이 더 강하여 페놀성 분획중의 항미생물 활성물질을 동정하기 위하여 먼저 silica gel column chromatography, RP-8 column chromatography 및 preparative liquid chromatography를 행하여 정제하였고 정제된 항미생물 활성물질을 IR, $^{1}H-NMR$ 및 MS에 의한 기기분석을 통하여 동정한 결과 각각 psoralen, bergapten, umbelliferone 및 ${\beta}$-sitosterol로 구명되었다.

수도권 상수중 유기오염물질의 돌연변이원성 (Mutagenic Activity of Organic Pollutans in Drinking Water in Seoul)

  • To measure the mutagenic activity of micro-organic pollutants in drinking water, mutagenicity test was conducted using Salmnella typhimurium TA 98 strain on the water sample taken from three water supply stations and six tap water in Seoul in July and November 1987. The results were as follows : 1. The average amounts of organic matters in raw, treated, and tap water sampled in July were 0.38mg/l, 0.28mg/l, and 0.45mg/l. respectively, and sampled in November were 0.34mg/l, 0.24mg/l, and 0.22mg/l, respectively. The amount of organic matters of tap water sampled in November did not increase while that of tap water sampled in July increased compare to those of raw or treated water. 2. The amount of organic matters is the highest in neutral fraction compare to acidic and basic fractions. 3. In the five out of six tap water and raw water of Paldang and Kuui station sampled in July, the mutagenicity ratios were greater then two (both direct and indirect mutagenicity). 4. In the three out of six tap water and raw and treated water of Kuui station sampled in November, the mutagenicity ratios were greater than two. 5. While mutagenic activities were low in acidic and basic fraction, they were high in neutral fraction. The samples which had high mutagenic acitivity in the total amount also showed high mutagenic activity in neutral fraction. 6. While mutagenic acitivity was decreased after the treatement of water, it was increased in tap water as the distance from the water supply station increases.

대황으로부터 세균성 질염 치료를 위한 항균성 물질의 분리 및 특성 (Isolation and Characterization of an Antibacterial Substance from Rheum palmatum for Treatment of Bacterial Vaginosis)

  • Rheum palmatum has traditionally been used as a preventive agent and medication against fever and infection. The aim of this study was to isolate and characterize an antibacterial substance from R. palmatum that is effective against bacterial vaginosis. A methanol extract from R. palmatum showed antibacterial activity against Lactobacillus vaginalis KC TC 3515, Chryseobacterium gleum KCTC 2904, and Sphingomonas paucimobilis KCTC 2834, which cause bacterial vaginosis. After extraction and pH control of the methanol extract from R. palmatum, we found that acidic and alkaline extracts did not show antibacterial activity. A neutral extract (50 mg/mL) displayed an inhibitory zone of 18 mm on a nutrient agar plate with C. gleum KCTC 2904. Fractions No. 11 and 12 among 41 fractions obtained by silica gel column chromatography produced inhibitory zones of 10 mm on nutrient agar plates with C. gleum KCTC 2904. $R_f0.15$ and $R_f0.17$ spots produced by TLC of fraction No. 11 showed antibacterial activity against C. gleum KCTC 2904. Isolation and purification of the peak at a retention time (Rt) of 9.427 min was achieved by HPLC of $R_f0.29spots$. The peak at Rt 9.427 min showed antibacterial activity against C. gleum KCTC 2904.

부숙식물유체(腐熟植物遺體)에서 추출(抽出)한 Fulv 산(酸) 가수분해(加水分解) 용액중(溶液中)의 Amino 산함량(酸含量) (Amino Acid Contents in the Hydrolysates of Fulvic Acids Extracted from Decomposing Plant Residues)

  • 볏짚, 보릿짚, 밀짚, 호밀짚의 4가지 작물유체(作物遺體)와 산야초(山野草), 활엽수(闊葉樹)와 침엽수(針葉樹) 낙엽등(落葉等) 7가지 식물유체(植物遺體)를 부열(腐熱)시키면서 경시적(經時的)으로 시료(試料)를 채취(採取)하여 추출(抽出)한 fulv산(酸)을 가수분해(加水分解)시킨후, amino산(酸)을 정량분석(定量分析)한 결과(結果) aspartic acid, glutamic acid, arginine, histidine, lysine, glycine, alanine, valine, leucine, isoleucine, phenylalanine, tyrosine, serine, threonine, proline, methionine 등(等) 16종(種)의 amino산(酸)이 검출(檢出)되었다. 1. 부숙(腐熟) 90일(日)후에 유출(抽出)한 fulvic acid중에는 0.15% 내지 0.53%의 amino산(酸)이 함유되어 있었고, 산야초(山野草)에 가장 많이 그리고 밀짚에 가장 적게 함유되어 있었다. Humic acid의 함량에 비해 1/5내지 1/31의 정도로 함랑이 적었다. 2. Neutral amino acids, acidic amino acids, basic amino acids의 순(順)으로 많이 함유되어 있었다. 3. 부숙화(腐熱化)가 잘된 시료(試料)에서는 arginine이 검출되지 않았다. 4. Aromatic amino acid인 phenylalanine과 tyrosine은 일반적으로 검출되지 않았다. 5. Glycine, glutamic acid와 aspartic acid가 주요 amino acid였으며 glutamic acid와 aspartic acid를 제외하고 비교해 본 결과, 각 amino산중(酸中)에서 분자량(分子量)이 낮을수록 함유된 농도가 높은 경향이었다.

목초액의 향균 및 DPPH 라디칼 소거 활성에 관한 연구 (Study of Antimicrobial and DPPH Radical Scavenger Activity of Wood Vinegar)

  • The antimicrobial and antioxidative activities were investigated to confirm the utility of wood vinegar. Antimicrobial activity was performed by paper disc method and liquid culture. The growth inhibition was observed in all microbial species at a dose of as low as 25 $\mu$m of wood vinegar by paper disc method. Also, in liquid culture, S. cerevisiae and P. aeruginosa were more inhibited the growth than others in the concentration of $2\%$ (v/v). For measuring of antioxidative activity, wood vinegar was fractionated with acidic, phenolic, basic and neutral fraction; and their antioxidant activities were measured by the radical scavenging effect on DPPH radical. In four fractions, phenolic fraction showed high antioxidative activity.

Character Impact Compounds in Flavors of Korean Soy Sauce Manufactured with the Traditional and the Improved Meju

  • We characterized the character impact compounds of flavors of the fermented Korean soy sauce manufactured with both the traditional and the improved Meju made with different strains. The whole flavor samples were obtained by extracting each volatile flavor phase from both the traditional and the improved soy sauce. To get more detailed information, each whole volatile flavor was further fractionated into the basic, acidic, phenolic and neutral fractions. Each separated peak from the whole and fractionated flavor samples on gas chromatogram was identified by GC/MS and Kovat s retention index, and likewise the aroma of each peak was investigated by a sniffing test with the exercised panel. We were able to identify 15 groups of ingredients with the characteristic soy sauce aroma from the soy sauce made with the traditional Meju and 6 groups from the soy sauce manufactured with the improved Meju made with Aspergillus oryzae. The character impact compounds the two soy sauces were different from each other.

  • Chemical studies of nitrogen compounds of Panax ginseng seem relatively rare, Probably due to the isolation difficulties, subsequently the investigations of biological activities are little. The experimental conditions were established for highly complete extraction of peptides (basic, acidic and neutral) from Panax ginseng. This task was achieved by applying the follow isolation procedure: 1 , the sequential extraction with water, 0.1% TFA in 20% acetonitril and buffer pH 6.5 (water-pyridine-acetic acid 100:3:900) : 2, fractionation by ultrafiltration : 3, n-butanol extraction 4, cation- and anion-exchange chromatography : 5, chromato-electrophoresis. The comparison of red ginseng (xylem Sl pith part) and red ginseng inside white (xylem Sc pith part) was also provided. To analyze the peptide mixture the chromato-electrophoresis method of separation was applied. Optimal conditions for peptides mapping of sample were explored. Our experiments revealed the quantitative difference of peptide between xylem & pith and phloem & cortex part. We have also found the qualitative difference in the composition of polypeptides between normal red ginseng (xylem Sc pith part) and red ginseng inside-white (xylem St pith part)

수삼의 휘발성 향기 성분 (VOLATILE FLAVOR COMPONENTS OF FRESH GINSENG)

  • 수삼(Panax ginseng C.A.Meyer)의 휘발성 향기성분을 SE-54 fused silica capillary를 이용한 개스크로와 질량분석방법을 이용해서 연구했다. 수삼을 수증기 종류해서 얻어진 증류물을 diethyl ether로 추출해서 농축하고 이 농축물을 중성, 산성, 폐놀성 및 염기성 분획으로 분리했다. 가장 방향이 풍부하고 원 수삼취와 비슷한 중성분획을 분석한 결과 인삼의 주요한 향기성분은 200종 이상 많은 성분중 monoterpene과 sesquiterpene계 성분이었다. Sesquiterpene 화합물은 질량 204를 갖는 azulene, menthene, napthalene계 화합물이었다.
