Characterization of promotor sequences for strong expression of groEx IN Escherichia coli.

  • The cloned X-bacterial gene (groEx) which is analogous to groE of E. Coli strongly expressed in E. coli when grown at the temperature 27.deg. C or higher without having to add any inducers. By S1-nuclease mapping, primer extension analysis and site directed mutagenesis, we found 4 promoters in the gene. Among them two promoters located at 5'-extended region of the gene are homologous to the promoters found in groE family of heat-shock genes ; they are , .sigma.$^{32}$ factor-dependent P1 promotor and .delta$^{70}$factor-dependet P2 promoter. The other two promoters found within the coding region of groESx were P3, 5'-TTGGCG-(18 bases)-AATACT-3' and P4, 5'-TTGGCA-(19 bases)-TAAGT which overlapped within 49 bases. These unique intragenic .delta.$^{70}$-dependent promoters are the first to be cloned and characterized in groE analogous heat-shock genes so far. These P3 and P4 promoters appeared to be responsible for the strong expression of GroElx in X-bacteria in vivo.

Factors Determining Brand Equity for Retail Stores: A Market Segmentation Approach

  • The present study aims to analyze factors of brand equity in the retail stores in Vietnam; to recognize sub-dimensions connected with the forms of brand equity and thereby study the extent of the relationship among bases of market segmentation and brand equity. The present research made a collection of data by conducting a preliminary study. Responses were collected from the customers through a structured questionnaire to buy goods and services from the retail store in Vietnam. The sample size selected for the study was 196. The study applied various statistical tools, namely, Cronbach's Alpha for reliability checking, correlation and regression statistics with the support of SPSS Software. The brand association is treated as a vital determinant of brand equity that significantly influences the bases of market segmentation. Further, the study also recognized the fact that the majority of consumers who prefer to buy from retail stores are young and in the age-group of 18-24 years. The present study made an initial attempt to study the relationship among bases of market segmentation with the factors related to brand equity; thereby to offer a novel conceptual framework designed for retail stores in Vietnam for brand positioning.

Biodegradibility Tests of Biodiesel-derived Pentaerythritol Lubricant Oil Bases (바이오디젤 유래 펜타에리쓰리톨계 윤활유 베이스의 생분해성 테스트)

  • Biodegradability test for various synthetic lubricant oil bases derived from biodiesel was carried out. The biodegradability was estimated under aerobic aqueous condition, according to the method by OECD 301 B, which is based on CO$_2$ evolution test. The ultimate biodegradability of pentaerythritol methyl esters were estimated as 61.1∼80.3%, at 28 day with which the test compounds were indicated as ultimately biodegradable. Among the tested samples, biodiesel showed the highest biodegradability (83.5%). The validation with several criteria, regarding relative errors of test results, toxicity control and procedure control, was performed through the biodegradability test. The test procedure was validated for all the tested lubricant oil bases and biodiesel, except for petroleum diesel.

Novel and Efficient Palladium Complexes with β-Ketoiminate Ligands for the Polymerization of Norbornene

  • A series of the noble palladium complexes containing $\beta$-ketoiminate ligands with internal bases, [Pd(${\eta}^3$-allyl)($\beta$- ketoiminate)], [Pd(Me)($PPh_3$)($\beta$-ketoiminate)] and [Pd(Me)($\beta$-ketoiminate)], have been successfully prepared. Crystallographically determined structures showed that these complexes are distorted square planar and pendant bases of the $\beta$-ketoiminate ligands fail to coordinate to the metal in the first two classes of complexes while bases do coordinate in the 3rd class complexes. These complexes are active towards norbornene polymerization on activation with $H(OEt_2)_2BAr^'_4$ (Ar' = 3,5-bistrifluoromethylphenyl) and modified methylalumioxane (MMAO). MMAO is more efficient for the activation for polymerization. Generally, the polymerization activity increases with the following order; [Pd(allyl)($\beta$-ketoiminate)] < [Pd(Me)($PPh_3$)($\beta$-ketoiminate)] < [Pd(Me)($\beta$-ketoiminate)].

Investigation of rotational characteristics of column 'PINNED' bases of steel portal frames

  • Most of the portal frames are designed these days by the application of plastic analysis, with the normal assumption being made that the column bases are pinned. However, the couple produced by the compression action of the inner column flange and the tension in the holding down bolts will inevitably generate some moment resistance and rotational stiffness. Full-scale portal frame tests conducted during a previous research program had suggested that this moment can be as much as 20% of the moment of resistance of the column. The size of this moment of resistance is particularly important for the design of the tensile capacity of the holding down bolts and also the bearing resistance of the foundation. The present research program is aiming at defining this moment of resistance in simple design terms so that it could be included in the design of the frame. The investigation also included the study of the semi-rigid behaviour of the column base/foundation, which, to a certain extent, affects the overall loading capacity and stiffness of the portal frames. A series of column bases with various details were tested and were used to calibrate a finite element model which is able to simulate the action of the holding down bolts, the effect of the concrete foundation and the deformation of the base plate.

  • Three decades after its establishment in 1946, Seoul National University was transferred to its current location in Gwan-ak District in accordance with the Seoul National University Integration Plan. In the 40 years since Seoul National University transferred to the Gwan-ak area, much quantitative and qualitative growth has taken place, and the university has grown into a world-class research institution. In this process, the campus was expanded externally and internally, and many changes have been made to the campus life bases and facilities which were planned at the beginning of the campus' construction. This study examines the expansion of the facilities at Seoul National University's Gwan-ak campus, as well as the expansion of the campus' boundaries. In addition, through quantitative analysis (Space Syntax), we predicted the location of campus life bases of the present multi-centralized campus and suggested a future developmental direction for the campus.

  • World practice is evidence of that computer systems of an intellectual support of medical activities bound up with examination of patients, their diagnosis, therapy and so on are the most effective means for attainment of a high level of physician\`s qualification. Such systems must contain large knowledge bases consistent with the modern level of science and practice. To from large knowledge bases for such systems it is necessary to have a medical ontology model reflecting contemporary notions of medicine. This paper presents a description of an observation ontology, knowledge base for the physician of general tipe, architecture, functions and implementation of problem independent shell of the system for intellectual supporting patient examination and mathematical model of the dialog. The system can be used by the following specialist: therapeutist, surgeon, gynecologist, urologist, otolaryngologist, ophthalmologist, endocrinologist, neuropathologist and immunologist. The system supports a high level of examination of patients, delivers doctors from routine work upon filling in case records and also automatically forms a computer archives of case records. The archives can be used for any statistical data processing, for producing accounts and also for debugging of knowledge bases of expert systems. Besides that, the system can be used for rise of medical education level of students, doctors in internship, staff physicians and postgraduate students.

Development of Standardized Korean Plant Ontology for International Harmonization of Environmental and Ecological Knowledge Bases (환경·생태 지식베이스의 국제적 조화를 위한 한국형 표준 식물 온톨로지 개발)

  • Background: To describe domain knowledge consistently and precisely, the establishment of a controlled vocabulary, a so-called ontology, is essential. Internationally, the plant ontology (PO) in the ecology field has been developed for the anatomy and developmental stages of plants in English, Spanish, and Japanese, but there is no Korean version of the PO due to a lack of knowledge on standardization for Korean plants. Objectives: We aimed to establish a Korean plant ontology with core PO architectures. Methods: The latest ontology web language (OWL)-formatted raw version of the PO was collected from the PO consortium site. A formal workflow process and OWL file-handing tools for efficient Korean content development were conducted and executed. Results: The macro- and micro-perspective frameworks of the PO were presented by analyzing the upper model and the internal OWL-leveled physical structure, respectively. We developed and validated Korean knowledge content for a total of 1,957 classes included in the PO and transplanted them into an ontology modeling system. Conclusions: A Korean plant ontology was established for international harmonization through improved compatibility and data exchangeability with multilingual environmental and ecological knowledge bases.

Site-Specific Mutagenesis on the 32-T and 39-T of E. coli $tRNA^{phe}$ Gene (E. coli $tRNA^{phe}$ 유전자의 32-T와 39-T 염기의 부의 특이적 돌연변이)

  • There are three pseudourdine ($Psi$)bases in the E. coli $tRNA^{phe}$ In order to study the function of the pseudouridine bases in the $tRNA^{phe}$, changes of bases $tRNA^{phe}$ gene to other bases were undertaken by the site-specific mutagenesis. Site-specific mutagenesis of T in the pheW gene, a $tRNA^{phe}$ gene of E. coli, corresponding to the baseat the No.32 position to C and also T corresponding to the base at the No.39 position to C were performed using Kunkel's uracil-containing template method. Identification of mutants were undertaken by the KNA sequencing techniques of the mutated pheW genes and activities of the mutated pheW genes complementing to E. coli NP37 mutant($pheS^{-ts}$) using the recombinant plasmid containing the mutated genes. Neither NP37 harboring pheW gene mutated at No.32 position nor NP37 harboring pheW gene mutated at No.39 position can be grown at non-permissive temperature. The result means that both mutated pheW genes can not complement to E. coli NP37, and that the pseudouridine bases are essential to the activity of the E. coli $tRNA^{phe}$ in vivo.
