Depiction of concrete structures with seismic separation under faraway fault earthquakes

  • One of the most suitable methods in structural design is seismic separator. Lead-Rubber Bearing (LRB) is one of the most well-known separation systems which can be used in different types of structures. This system mitigates the earthquake acceleration prior to transferring to the structure efficiently. However, the performance of this system in concrete structures with different heights have not been evaluated thoroughly yet. This paper aims to evaluate the performance of LRB separation system in concrete structures with different heights. For this purpose, three, 16, and 23 story concrete structures are equipped by LRB and exposed to a far-field earthquake. Next, a time history analysis is conducted on each of the structures. Finally, the performance of the concrete structures is compared with each other in the term of their response to the earthquakes and the formation of plastic hinges. The results of the paper show that the rate of change in acceleration response and the ratio of drift along the height of 8 and 23 stories concrete structures are more than those of the 16-stories, and the use of LRB reduces the formation of plastic joints.

  • This paper introduces a base isolation system-tuned mass damper inerter (BIS-TMDI) hybrid system to the AP1000 nuclear power plant (NPP), which reduces seismic damage potential of the NPP structure. The effects of fluid-structure interaction (FSI) caused by the passive containment cooling system water storage tank (PCCWST) on NPP's seismic performance are investigated. The FSI of water tank theoretical model is considered based on the Housner's model, and a series of time history analyses are performed to prove the rationality of the proposed model. Three single-objective optimization strategies are employed to minimize the relative displacement variance and absolute acceleration variance of the upper structure, as well as the filtered energy index (FEI). Furthermore, a multi-objective optimization strategy considering all these three indexes is proposed to obtain optimal parameters of vibration control. The influence of vibration control strategies on the relative deformation and acceleration of the upper structure is explored with various water level ratios. The analytical results indicate that the proposed BIS-TMDI strategy has significantly reduced the NPP structure's seismic response. The effectiveness of the vibration control strategy is influenced by the water level ratio, emphasizing the significance of designing an appropriate water level ratio to reduce NPP structure's seismic response.

  • Purpose: The evaluation of seismic performance of critical structures has been emerging a key issue in Korea, since a magnitude 5.8 earthquake, the worst in Koran history, struck Gyeongju, southern area in Korea on september 12th, 2016. In particular, the catastrophic failure of nonstructural components such as sprinkler piping systems can cause significant economic loss or loss of life during and after an earthquake. The nonstructural components can be more fragile than structural components in seismic behavior. Method: This study presents the seismic performance evaluation of fire protection piping system, using coupled building-piping system installed with Triple Friction Pendulum Bearings (TPBs). Kobe (Japan), Kocaeli (Turkey), and GyeongJu (Korea) were selected to consider the uncertainty of ground motions in this study. Result: In the simulation results, it was observed that the reduction of maximum displacements of the piping system with the TPBs' system was significant: Kobe, Kocaeli, and Gyeongju cases were 49%, 14.4% and 21.5%, respectively. Conclusion: Therefore, using seismically isolated system in a building-piping system can be more effective to reduce the seismic risk than a normally installed building-piping systems without TPBs in strong earthquakes.

The transposition pattern of the Ac element and its use for targeted transposition in Arabidopsis thaliana

  • In order to evlauate feasibility of the gene tagging by the maize transposable element Ac in heterologous plant systems, we have investigated physical distances and directions of transposition of the element in Arabidopsis thaliana and tobacco cultured cell line BY-2. We prepared a T-DNA construct that carried a non-autonomous derivative of Ac with a site for cleavage by endonuclease I-Scel (designated dAc-I-RS element). Another cleavage site was also introduced into the T-DNA region outside dAc-I-RS. A number of transgenic Arabidopsis plants were generated, each of which had a single copy of the T-DNA at a different chromosomal location. To examine the pattern of transposition, three out of these transgenic plants were crossed with the Arabidopsis plant that carried the gene for Ac transposase and progeny in which dAc-I-RS had been transposed were isolated. After digestion of the genomic DNA of these progeny with I-SceI, sizes of segment of DNA were determined byd pulse-field gel electrophoresis. We also performed linkage analysis for the transposed elements and sites of mutations near the elements. Our results with three transgenic lines showed that 50% of all transposition events had occurred within 1,700 kilo-base pairs (kb) on the same chromosome, with 35% within 200 kb, and that the elements transposed in both directions on the chromosome with roughly equal probability. The data thus indicate that the Ac-Ds system is most useful for tagging of genes that are present within 200 kb of the chromosomal site of Ac in Arabidopsis. In addition, determination of the precise localization of the transposed dAc-I-RS element should definitely assist in map-based cloning of genes around insertion sites. In the present paper, we report typical examples of such gene isolation studies.

Isolation and Characterization of Engineered Nucleoside Deoxyribosyltransferase with Enhanced Activity Toward 2'-Fluoro-2'-Deoxynucleoside

  • Nucleoside deoxyribosyltransferase (NDT) is an enzyme that replaces the purine or pyrimidine base of 2'-deoxyribonucleoside. This enzyme is generally used in the nucleotide salvage pathway in vivo and synthesizes many nucleoside analogs in vitro for various biotechnological purposes. Since NDT is known to exhibit relatively low reactivity toward nucleoside analogs such as 2'-fluoro-2'-deoxynucleoside, it is necessary to develop an enhanced NDT mutant enzyme suitable for nucleoside analogs. In this study, molecular evolution strategy via error-prone PCR was performed with ndt gene derived from Lactobacillus leichmannii as a template to obtain an engineered NDT with higher substrate specificity to 2FDU (2'-fluoro-2'-deoxyuridine). A mutant library of 214 ndt genes with different sequences was obtained and performed for the conversion of 2FDU to 2FDA (2'-fluoro-2'-deoxyadenosine). The E. coli containing a mutant NDT, named NDTL59Q, showed 1.7-fold (at 40℃) and 4.4-fold (at 50℃) higher 2FDU-to-2FDA conversions compared to the NDTWT, respectively. Subsequently, both NDTWT and NDTL59Q enzymes were over-expressed and purified using a His-tag system in E. coli. Characterization and enzyme kinetics revealed that the NDTL59Q mutant enzyme containing a single point mutation of leucine to glutamine at the 59th position exhibited superior thermal stability with enhanced substrate specificity to 2FDU.

  • Liquefied natural gas(LNG) is natural gas that has been converted temporarily to liquid form for ease of storage and transport it. Natural gas is the worlds cleanest burning fossil fuel and it has emerged as the environmentally preferred fuel of choice. In Korea, the demand of this has been increased since the first import from the Indonesia in 1986. LNG takes up about 1/600th the volume of natural gas in the gaseous state by cooling it to approximately $-162^{\circ}C(-260^{\circ}F)$. The reduction in volume therefore makes it much more cost efficient to transport and store it. Modern LNG storage tanks are typically the full containment type, which is a double-wall construction with reinforced concrete outer wall and a high-nickel steel inner tank, with extremely efficient insulation between the walls. The insulation will be installed to LNG outer tank for the isolation of cryogenic temperature. The insulation will be installed in the base slab, wall and at the roof. According to the insulation's arrangement, the different aspects of temperature transmission is shown around the outer tank. As the result of the thermal & stress analysis, by the installing cellular glass underneath the perlite concrete, the temperature difference is greatly reduced between the ambient temperature and inside of concrete wall, also reducing section force according to temperature load.

Hybrid Control of a Benchmark Cable-Stayed Bridge Considering Nonlinearity of a Lead Rubber Bearing (납고무받침의 비선형성을 고려한 벤치마크 사장교의 복합제어)

  • This paper presents a hybrid control strategy for seismic protection of a benchmark cable-stayed bridge, which is provided as a testbed structure for the development of strategies for the control of cable-stayed bridges. This benchmark problem considers the cable-stayed bridge that is scheduled for completion in Cape Girardeau, Missouri, USA in 2003. Seismic considerations were strongly considered in the design of this bridge due to the location of the bridge in the New Madrid seismic zone and its critical role as a principal crossing of the Mississippi river. Based on detailed drawings of this cable-stayed bridge, a three-dimensional linearlized evaluation model has been developed to represent the complex behavior of the bridge. A set of eighteen evaluation criteria has been developed to evaluate the capabilities of each control strategy. In this study, a hybrid control system is composed of a passive control system to reduce the earthquake-induced forces in the structure and an active control system to further reduce the bridge responses, especially deck displacements. Conventional base isolation devices such as lead rubber bearings are used for the passive control design and Bouc-Wen model is used to simulate the nonlinear behavior of these devices For the active control design, ideal hydraulic actuators are used and on $H_2$/LQG control algorithm is adopted. Numerical simulation results show that the performance of the proposed hybrid control strategy is quite effective compared to that of the passive control strategy and slightly better than that of the active control strategy. The hybrid control method is also more reliable than the fully active control method due to the passive control part. Therefore, the proposed hybrid control strategy can effectively be used to seismically excited cable-stayed bridges.

Isolation of $NH_4^+$-Tolerant Mutants of Actinobacillus succinogenes for Succinic Acid Production by Continuous Selection

  • Actinobacillus succinogenes, a representative succinicacid-producing microorganism, is seriously inhibited by ammonium ions, thereby hampering the industrial use of A. succinogenes with ammonium-ion-based materials as the pH controller. Therefore, this study isolated an ammonium-ion-tolerant mutant of A. succinogenes using a continuous-culture technique in which all the environmental factors, besides the stress (ammonium ions), were kept constant. Instead of operating the mutant-generating system as a nutrient-limited chemostat, it was used as a nutrient-unlimited system, allowing the cells to be continuously cultured at the maximum specific growth rate. The mutants were isolated on agar plates containing the acid-base indicator bromothymol blue and a high level of ammonium ions that would normally kill the parent strain by 100%. When cultured in anaerobic bottles with an ammonium ion concentration of 354 mmol/l, the mutant YZ0819 produced 40.21 g/l of succinic acid with a yield of 80.4%, whereas the parent strain NJ113 was unable to grow. When using $NH_4OH$ to buffer the culture pH in a 3.0 l stirredbioreactor, YZ0819 produced 35.15 g/l of succinic acid with a yield of 70.3%, which was 155% higher than that produced by NJ113. In addition, the morphology of YZ0819 changed in the fermentation broth, as the cells were aggregated from the beginning to the end of the fermentation. Therefore, these results indicate that YZ0819 can efficiently produce succinic acid when using $NH_4OH$ as the pH controller, and the formation of aggregates can be useful for transferring the cells from a cultivation medium for various industrial applications.

Design of Dual-Polarization Antenna with High Cross-Polarization Discrimination (높은 교차편파 분리도를 가지는 이중편파 안테나 설계)

  • In a small cell base station used in densely populated areas, a dual polarized multiple antenna(MIMO) is mainly used to increase the cell capacity. This paper demonstrates a dual-polarization antenna with high cross-polarization discrimination(XPD) that can improve the capacity of a small cell using a dual polarization multiple antenna (MIMO). By using the symmetric structure and differential feeding, high XPD in all directions is achieved. In addition, a very similar radiation pattern is observed between each polarization. Because of high XPD and similar radiation pattern in all directions, proposed antenna is well adopted for small-cell multiple-input multiple-output(MIMO) system. Experimental results shows that the proposed antenna has a bandwidth of 180 MHz (2.51~2.7 GHz), a maximum gain of 4.5 dBi (3.5~4.5 dBi), and a half-power beam width of 85 degrees. In addition, average XPD of 26.4 dB in all directions, more than 13.8 dB increase than previous dual-polarization antennas which use single emitter by using different feeding or selectively use polarization through switching.
