Improvement of Fungal Cellulase Production by Mutation and Optimization of Solid State Fermentation

  • Spores of Aspergillus sp. SU14 were treated repeatedly and sequentially with $Co^{60}$ ${\gamma}$-rays, ultraviolet irradiation, and N-methyl-N'-nitro-N-nitrosoguanidine. One selected mutant strain, Aspergillus sp. SU14-M15, produced cellulase in a yield 2.2-fold exceeding that of the wild type. Optimal conditions for the production of cellulase by the mutant fungal strain using solid-state fermentation were examined. The medium consisted of wheat-bran supplemented with 1% (w/w) urea or $NH_4Cl$, 1% (w/w) rice starch, 2.5 mM $MgCl_2$, and 0.05% (v/w) Tween 80. Optimal moisture content and initial pH was 50% (v/w) and 3.5, respectively, and optimal aeration area was 3/100 (inoculated wheat bran/container). The medium was inoculated with 25% 48 hr seeding culture and fermented at $35^{\circ}C$ for 3 days. The resulting cellulase yield was 8.5-fold more than that of the wild type strain grown on the basal wheat bran medium.

Phylogeography and Population Genetic Structure of Amur Grayling Thymallus grubii in the Amur Basin

  • Amur grayling, Thymallus grubii, is an important economic cold freshwater fish originally found in the Amur basin. Currently, suffering from loss of habitat and shrinking population size, T. grubii is restricted to the mountain river branches of the Amur basin. In order to assess the genetic diversity, population genetic structure and infer the evolutionary history within the species, we analysised the whole mitochondrial DNA control region (CR) of 95 individuals from 10 rivers in China, as well as 12 individuals from Ingoda/Onon and Bureya River throughout its distribution area. A total of 64 variable sites were observed and 45 haplotypes were identified excluding sites with gaps/missing data. Phylogenetic analysis was able to confidently predict two subclade topologies well supported by maximum-parsimony and Bayesian methods. However, basal branching patterns cannot be unambiguously estimated. Haplotypes from the mitochondrial clades displayed local homogeneity, implying a strong population structure within T. grubii. Analysis of molecular variance detected significant differences among the different geographical rivers, suggesting that T. grubii in each river should be managed and conserved separately.

Microsurgical Fenestration of Middle Cranial Fossa Arachnoid Cyst

  • Objective : The optimal surgical treatment for symptomatic middle cranial fossa arachnoid cysts is controversial. Therapeutic options include endoscopic fenestration, excision, cyst shunting, and craniotomy for fenestration of basal cistern. We reviewed the results of surgically treated middle cranial fossa arachnoid cysts. Methods : We performed a retrospective study in 18 cases of middle cranial fossa arachnoid cysts who had been treated with microsurgical fenestration between 1995 to 2003. The analysis was based on the results of the patients' age, sex distribution, developed area, clinical symptoms, treatment method, and complications. Results : Eighteen surgical treated middle cranial fossa arachnoid cysts patients were evaluated. The age range of cyst development was between 2 years and 44 years with the average of 16.4 years. The follow-up periods averaged 31.48 months. There were 15 male and 3 female patients, with significantly more cyst development in males than females. The most common clinical symptom was headache, followed by seizure. In the entire series, 77.8% of patients demonstrated a decrease in cyst size In serial imaging studies. Of them, 67.3% demonstrated a complete cyst effacement. Overall, 100% of patients with Grade I cysts, 81.8% of patients with Grade II cysts, 60% of patients with Grade III cysts exhibited evidence of decrease in cyst size during long-term monitoring. Complications included headache, meningitis, and hydrocephalus. Conclusion : Patients who were treated with microsurgical fenestration showed good outcome with acceptable complications. We concluded that microsurgical fenestration is a safe and effective surgical method for middle cranial fossa arachnoid cysts.

Status of the Red-Listed Plant Species, Smilax wightii A. DC. in Nilgiri Biosphere Reserve, the Western Ghats, India

  • The rare endemic plant, Smilax wightii, is generally distributed in shola forests at high altitudes in Nilgiri Biosphere Reserve, the Western Ghats, southern India. To determine the ecological status of the species, we surveyed 11 major shola forests in that region. S. wightii has a limited distribution in all sholas (frequency value of < 13%). The density of the species is generally around $15/100\;m^2$ in the sholas except in Thiashola where it occurred at a density of 40 individuals/$100\;m^2$. The density of S. wightii was generally greater in the margins of the shoals, while the interior forests contained fewer individuals per unit area. The basal cover occupied by this species ranged between 2246 and $3144\;mm^2/100\;m^2$. The importance value index for S. wightii was >2 in all shoals, which indicates that the species occupies an important position in the lower stratum of shola forests of Nilgiri Biosphere Reserve.

  • In the estimation of diameter distribution in a stand using Weibull distribution function, the calculation method of experimental distribution was presented in previous paper. This study was to estimate the diameter distribution of Korean pine stands by Weibull distribution which represents Gamma function, with mean diameter and mean basal-area diameter of the random sample trees. The results obtained fitted the diameter distribution in experimental stands. Thus, this method appears to be used for the estimation of diameter distribution in a stand as well as for the analysis and prediction of stand construction for the future.

Study on Antihyperlipidemic Effect of Kamiondamtang (가미온담탕(加味溫膽湯)의 항고지혈 작용에 대한 연구)

  • Kamiondamtang(KODT) has been used in oriental medicine as stress by many medical practitioner for the study of KODT, we had fed mice divided to three groups(basal diet, hyperlipidemic diet, hyperlipidemic diet + KODT), and observed the change of weight, total cholesterol, triglyceride, SGOT, SGPT, HDL-cholesterol, LDL-cholesterol on the serum per every 7days for 6weeks. To help comparison with the results above, we had tested endothelial cells and liver. The results of this Study were obtained as fallows ; 1. Total cholesterol, triglyceride and LDL-cholesterol were decreased significantly by KODT. 2. HDL-cholesterol was increasd significantly by KODT. 3. SGOT was intended to decrease by KODT, 4. %W was decreased significantly by KODT. 5. In case of supplying KODT, WBC attatched on endothelial cells and vacuoles in muscular layer were not observed. 6. In case of supplying KODT, fatty degeneration was not observed in liver portal area.

Effect of Cheunggansoyosan-gamibang on Hyperlipidemic mice (청간소요산가미방(淸肝逍遙散加味方)이 생쥐의 고지혈증에 미치는 영향)

  • Hyperlipidemia is one of dangerous factors causing the arteriosclerosis. For the study of Cheunggansoyosan-gamibang( CGSYS) on antihyperlipidemic effect, we had fed mice divided to three groups (basal diet, hyperlipidemic diet, hyperlipidemic diet + CGSYS), and observed the change of weight, total cholesterol. triglyceride, HDL-cholesterol. LDL-cholesterol. sGOT, sGPT on the serum per every 7days for 6weeks. Also to compensate for the results above, we had tested rat liver. The results of this Study were obtained as fallows ; 1. Cholesterol change on blood was decreased by CGSYS. 2. Total cholesterol. triglyceride and LDL-cholesterol were significantly decreased by CGSYS. 3. HDL-cholesterol was significantly increasd by CGSYS. 4. sGPT was significantly decreased by CGSYS. 5. sGOT was not changed by CGSYS. 6. In case of supplying CGSYS. fatty degeneration was not observed in liver portal area. This study suggests that Cheunggansoyosan-gamibang was significant effect with hyperlipidemia.

Phytophthora-Induced Diseases on Citrus in Jeju Island

  • Phytophthora-induced diseases on citrus in Jeju island have been considered of minor importance because of the use as root stock of trifoliate orange, which is immune to Phytophthora. However, brown rot on fruit, which severely occurred in 1998 and 1999, has become a great threat to citrus production in the island. About one-half of the surveyed orchards were infected in 1998 and 4 out of 19 infected fields showed over 20% fruit infection rate. The disease was less severe in 1999, with an estimated infected area and total fruit reduction of 3,155 ha and 15,300 tons, respectively. Typical gummosis was also occasionally observed on cv. Shiranugi, which is mostly cultivated under plastic film houses. Two types of Phytophthora were consistently isolated from various plant parts, identified as P. citrophthora and P. nicotianae. The former was isolated from the aerial parts of the fruit, young leaf, and shoot in the fields. Meanwhile, the latter was only isolated from the basal stem showing gummosis in plastic film houses.

Transjugular occlusion of patent ductus arteriosus using an Amplatz canine ductal occluder in a Cocker spaniel dog

  • A 5-year-old female Cocker spaniel dog (body weight 7.0 kg) was presented with primary complaints of exercise intolerance and loud precordial thrill which was noticed since she was a puppy. Physical examination revealed a grade V/VI continuous murmur over the maximal point of the left basal area, bounding femoral pulse, but no differential cyanosis. Tall R waves were detected in electrocardiogram, suggesting left ventricular enlargement. Diagnostic imaging studies showed enlarged left ventricle, bulged descending aorta (dAo), markedly dilated right pulmonary artery, and continuous shunt flow between the dAo and main pulmonary artery. Based on these findings, the dog was diagnosed as left to right shunted patent ductus arteriosus (PDA). The patent ductus arteriosus was treated by lodging a PDA duct occluder via the transvenous approach. Clinical signs were markedly improved after the ductal occlusion, the shunt flow was mildly persistent. The case presented is the first case of PDA occluded by the PDA duct occluder via the transvenous approach in a small breed of dog. Although the residual shunt flow was mildly persisted, the dog was clinically normal without detectable murmurs.

Canalicular Adenoma of the Parotid Gland (이하선에 발생한 소관 선종)

  • Parotid canalicular adenoma is a benign neoplasm that is predominantly composed of branching and interconnecting cords of single or double rows of columnar epithelium in a very loose stroma. There has been considerable confusion in the literature concerning the terminology of canalicular adenoma. However, thesedays it has been newly-recognized as a discrete entity of the monomorphic adenoma group. Canalicular adenoma has a remarkable predilection for occurrence in the minor salivary glands such as the upper lip, in contrast with basal cell adenoma that occurs predominantly in major salivary glands such as the parotid gland. We have experienced a case of canalicular adenoma of the parotid gland in a 65-year-old woman. The patient had a palpable mass on the preauricular area for the last 15 years and recently noticed a mild pain and discomfort on the mass. Neck ultrasonography showed a low echogenic mass of 1.0cm in diameter in the right parotid gland and a neck CT scan showed a well-enhanced rectangular-shaped mass. A superficial parotidectomy was performed for the lesion and the final pathologic diagnosis turned out to be 'multifocal canalicular adenoma'.
