Growth of Red-leaf Lettuce and Changes in Soil Solution Chemical Properties of Coir-dust Containing Root Media Influenced by Application Rates of Pre-planting Fused-Superphosphate (코이어 더스트 혼합상토에 용과린의 시비수준에 의한 적축면 상추의 생장과 근권부 화학성 변화)

  • This research was conducted to investigate the influence of various levels of fused superphosphate as pre-planting fertilizer on the growth of red-leaf lettuce and changes in the chemical properties of the soil solution in three root media, namely coir-dust plus expanded rice hull (8:2, v/v; CD+ERH), carbonized rice hull (6:4; CD+CRH), or ground and aged pine bark (8:2; CD+GAPB). The amounts of fused superphosphate (FSP) incorporated into the three root media during formulation were controlled from 0 to $6.0g{\cdot}L^{-1}$ in $1.5g{\cdot}L^{-1}$ increments. The root media containing fertilizers were packed into 300 mL plastic pots and seedlings of red-leaf lettuce at the 3rd leaf stage were transplanted. After transplanting, the crops were fed with a solution of neutral fertilizer ($100mg{\cdot}L^{-1}$). The growth of red-leaf lettuce was investigated 5 weeks after transplanting and soil solutions were extracted and analyzed every week for pH, EC, and concentrations of macro-nutrients. The elevation of application rates of FSP in the three root media resulted in better growth, and the crops grown in CD+ERH and CD+GRPB had greater fresh and dry weights than those in CD+CRH when compared among the treatments of equal amounts of FSP. The pH and $PO_4{^{-3}}$ concentrations in the soil solution of CD+CRH at 3 weeks after transplant were in the ranges of 4.0 to 4.8 and 20 to $100mg{\cdot}L^{-1}$, respectively. These were lower pH and higher $PO_4{^{-3}}$ concentrations than those in CD+ERH and CD+GAPB. The $K^+$ concentrations were higher in CD+CRH than those in the other two root media, and the elevation of FSP application rates resulted in higher $Ca^{+2}$, $Mg^{+2}$ and $SO_4{^{-2}}$ concentrations in soil solution of the three root media. The $NO_3$-N concentrations in soil solution rose continuously during crop cultivation, implying that the leaching percentage was elevated. The soil solution EC varied, showing the same tendencies as the $NO_3$-N concentrations. The above results indicated that the CD+ERH and CD+GRPB media performed better than CD+CRH, and optimum application rates of FSP in the three root media were 4.5 to $6.0g{\cdot}L^{-1}$ for pot cultivation of red-leaf lettuce.

Studies on Methanol-assimilating Yeasts (메탄올 자화효모에 관한 연구)

  • The distribution of methanol-assimilating yeasts on three different sources (elm bark, soil and fresh-water mud) and the growth conditions of a new strain of Candidaboidinii (SIO) wereexamines. From 150 samples, 91 methanol yeasts were isolated through enrichment culture ; they were identified as 77 strains of Candida boidinii including four new strains, 5 isolates of Torulopsis pinus, 3 strains of Hansenula polymorpha and one sstrain of Pichia pastoris respectively. The comparison of these yeasts with three sources indicated that decaying bark of elm tree other two, and that Gandida boidinii was most frequently distributed in all three sources. Four new strains of Candida boidinii were freshly isolated and their taxonomical properties were discussed. Of them, SIO strain was selected and characterized for its growth on methanol. This yeast could grow well on less than 1%(v/v) methanol. However, its growth was inhibited at 10% methanol. The cell yield was 3.1g (dry weight) per 1000ml of mineral mediurr, containing 1%(v/v) methanol as well as 01.% yeast extract as additive. The concentration of 0.1% yeast extract appears to be effective for the biomass production. Optimum conditions for growth on methanol was found to be : $28^{\circ}C,\;NH_4^+$ as nitrogen sources, thiamine as vitamin, and pH 4.5 to 6.0. The cell composition was as follows : crude protein and nucleic acids were 54% and 7% respectively. The amino acids were also described.

  • Morinda citrifolia, commercially known as Noni, is a tree that grows widely throughout the Pacific, and is recognized as one of the most significant sources of traditional medicines among Pacific Island societies. All parts of the plant have traditional and/or modern uses, including roots and bark(dyes, medicine), trunks(firewood, tools), and leaves and fruits(food, medicine). The bark and the roots of the tree contain red and yellow pigments respectively, which are both used in the manufacture of dyes. Dyes from Noni are being used traditionally, to color clothing and fabrics. In this study, mordanting effects on the dyeing properties of Noni root extract on silk fabrics were investigated.

Anxiolytic-like effects of extracts from Albizzia julibrissin bark in the elevated plus- maze in rats

  • The purpose of the this study was to characterize the putative anxiolytic-like effects of the aqueous extract of Albizzia julibrissin stem bark using the elevated plus maze (EPM) in rats. The water extract of Albizzia julibrissin was orally administered at 10, 50, 100 or 200 mg/kg to adult male SD rats, 1 h before behavioral evaluation in an EPM, respectively. Control rats were treated with an equal volume of saline, and positive control rats buspirone (1 mg/kg). Single or repeated treatment (for 7 days) of the water extract of Albizzia julibrissin (at 100 or 200 mg/kg) significantly increased time-spent and arm entries into the open arms of the EPM, and decreased time-spent and arm entries in the closed arms of the EPM versus saline controls (P < 0.05).   • The dried bark of Platycarya strobilacea were ground, extracted with 95% EtOH, concentrated, and one of EtOH extracts was fractionated with a series of n-hexane, dichloromethane and another was fractionated with a series of petroleumether, $Et_2O$, ethyl acetate on a separatory funnel. A portion of dichloromethane soluble was chromatographed on a Sephadex LH-20 column ($72.0{\times}5.0cm$) using EtOH-$CHCl_3$ (7:3, v/v) as eluent and A portion of $Et_2O$ soluble was chromatographed on a silica gel column ($42.0{\times}3.5cm$) using $CHCl_3$-MeOH (9:3, v/v) as eluent. The isolated compounds were identified by TLC, $^1H$-, $^{13}C$-NMR, HMBC and EI-MS. Two flavonoids and three flavonoid glycosides were isolated from the bark of P strobilacea. The structures were determined to quercetin (compound 1), myricetin (compound 2) as flavonol compounds and afzelin (compound 3), quercitrin (compound 4), myricitrin (compound 5) as flavonol glycosides, respectively, on the basis of spectrosopic data.

Antioxidative Activities and Nitrite Scavenging Abilities of Extracts from Ulmus devidiana (느릅나무 추출물의 항산화 효과 및 아질산염 소거능)

  • To measure antioxidative activities, the various extracts from Ulmus devidiana were examined in oil emulsion. Water, ethanol, methanol and butanol were used as extract solutions. The activity oxygen species ($H_2O_2,-OH,\;KO_2$) bound the extracts for antioxidative activities were excellent. The extracts bound $Fe^{2+}$ ion and $Cu^{2+}$ ion showed effective antioxidative activities and strong chelating effects. The concentration of $Fe^{2+}$ ion and total ion in ethanol and methanol extracts from Ulmus devidiana root parts (Chinese) were higher than those of the other products. The highest superoxide dismutase-like activities showed butanol extracts from Ulmus devidiana root parts (Chinese) and water extracts from Ulmus devidiana bark parts (Korean). Electron donating abilities and nitric scavenging abilities of ethanol, methanol and butanol extracts were higher than those of water extracts. The nitrite scavenging abilities also reached the maximum at pH 1.2 and the minium at pH 6.0.

  • This study was performed to investigate antioxidant activities and glutathione S-transferase (GST) activity according to parts of the Acer mono and A. okamotoanum. Most extracts showed high scavenging activities on DPPH. Especially, the bark of A. okamotoanum showed higher activity as 98.4% than the control, BHA as 96.5%. A. mono and A. okamotoanum showed high ability on nitrite scavenging, but decreasing tendency according to decreasing of pH. On SOD-like test, the wood of A. okamotoanum showed highest activity as 35.4% at 1.0mg/ml concentration. Also, the extracts obtained high activity on GST test. Therefore, the water extracts from the bark of A. mono and A. okamotoanum have relatively good antioxidant activity and GST activity. Especially, the bark of A. okamotoanum showed the highest activity on all of extracts, could be the use of functional foods and biomaterials.

  • The study of the scars formed on the bark of pine trees damaged by forest fire was carried out in the burned area of Samchuk and Donghae in 2000. Fire scars were formed on the bark of trees when fire passed by the tree. Fire scarring is usually found on the windward and leeward sides of the tree. Fire spread was analyzed topographically using the fire scars formed at the tree stem. Fire spread was closely related to the shape, slope and direction of the forestland. Four fire direction types are classified according to the shapes of the forestland. The height of the fire scar was higher on the leeward side than on the windward side of the trees studied. The burnt area of the total bark of the tree was over 30% in dead trees and below 12% in living trees. The regression model of the burnt stem area using the height of the fire scar, the diameter of breast height and the height of tree was Y=-2.484${\times}$Height+0.04199${\times}$D.B.H-1.686${\times}$Windward+11.172${\times}$Leeward+23.432(r=0.936, F=409.968, P>0.0001).

The Physiological Activities of Bark Extract of Albizia julibrissin (자귀나무(A. julibrissin) 수피 추출물의 생리활성)

  • Three bark extracts of Albizia julibrissin were prepared using water (AW), 70% (v/v) ethanol (AE), and hot water (AHW). Organic solvent fractions were analyzed for total flavonoids and polyphenols, antioxidant activities, and inhibitory activities against xanthine oxidase. Total flavonoid and polyphenol contents of the AHW extract were 8.57 mg/g and 108.67 mg/g, respectively. The SOD-like activities of all extracts, assayed at 1.0 mg/mL, were 10.46-16.73%. The nitrite-scavenging ability of the AHW extract, assayed at pH 1.2, was 60.82%, and the $IC_{50}$ value was $770.18\;{\mu}g$/mL. The electron-donating ability of the AHW extract, at 0.3 mg/mL, was 92.30%; the $IC_{50}$ values of the AW and AHW extracts were $31.31\;{\mu}g$/mL and $36.22\;{\mu}g$/mL, respectively; thus higher than that of ascorbic acid ($39.06\;{\mu}g$/mL). Xanthine oxidase inhibition by the AHW extract, at 1.0 mg/mL, was 94.05%. These results indicate that the AHW of A. julibrissin has potential as a natural antioxidant, for addition to foods and nutraceuticals.

Effects of Silkworm Powder, Mulberry Leaves and Mulberry Root Bark Adminstered to Rat on Gastrointestinal Function (누에분말, 뽕잎 및 상백피 투여가 흰쥐의 장관기능에 미치는 영향)

  • This experiment was carried out to investigate the effect of silkworm powder, mulberry leaves and mulberry root bark administered to S.D. rat on gastrointestinal function. Plot containing ten percent sericultural products(silkworm powder, mulberry leaves and mulberry root bark) was supplied with S.D. rat. The food efficiency ratio(FER) of S.D. rat fed with sericultural products were 0.1486~0.1573. The transit time also were 512.5~574.0 minutes, and transit speed were 25.05~29.11 mm/min. The pH of S.D. rat's feces fed with sericultural products were 5.95~6.72. The daily amount of S.D. rat's feces were 4.12~5.42g. As above results, sericultural products was evaluated to improve the function of S.D. rat's gastrointestine.
