Dyeing properties of rosemary colorants on cotton fabrics were investigated. Effect of dyeing conditions on dye uptake and effect of mordanting and cationizing on dye uptake, color change and colorfastness were explored. Also, antimicrobial activity of rosemary colorants was ascertained and further effect of cationizing on antimicrobial activity was investigated. Affinity of rosemary colorants to cotton fiber was considerably low, and its isotherm adsorption curve was Freundlich type, indicating that hydrogen bonding was involved in the adsorption of rosemary colorants to cotton fiber. The cotton fabrics showed generally high colorfastness except fastness to washing and light. The cationized cotton with Cationon UK(quarterly ammonium salt) showed higher dye uptake and shorter dyeing time, compared with the untreated cotton. The cationized cotton showed good colorfastness to washing, perspiration and rubbing. Antimicrobial activity of rosemary colorfastness was confirmed. The cationized cotton itself showed high bacterial reduction rate. For cationized and dyed samples, as dye uptake increased, bacterial reduction rate was decreased slightly.
Gamma irradiation was evaluated for its in vitro and in vivo antibacterial activity against a postharvest bacterial pathogen, Erwinia carotovora subsp. carotovora (Ecc). Gamma irradiation in a bacteria cell suspension resulted in a dramatic reduction of the viable counts as well as an increase in the amounts of DNA and protein released from the cells. Gamma irradiation showed complete inactivation of Ecc, especially at a dose of 0.6 kGy. In addition, scanning electron microscopy of irradiated cells revealed severe damage on the surface of most bacterial cells. Along with the morphological changes of cells by gamma irradiation, it also affected the membrane integrity in a dose-dependent manner. The mechanisms by which the gamma irradiation decreased the bacterial soft rot can be directly associated with the disruption of the cell membrane of the bacterial pathogen, along with DNA fragmentation, results in dose-dependent cell inactivation. These findings suggest that gamma irradiation has potential as an antibacterial approach to reduce the severity of the soft rot of paprika.
BACKGROUND: Biological iron redox transformation alters iron minerals, which may act as effective adsorbents for arsenate [As(V)] in the environments. In the viewpoint of alleviating arsenate, microbial Fe(III) reduction was sought under high concentration of As(V). In this study, Fe(III)-reducing bacteria were isolated from the wild plant rhizosphere soils collected at abandoned mine areas, which showed tolerance to high concentration of As(V), in pursuit of potential agents for As(V) bioremediation. METHODS AND RESULTS: Bacterial isolation was performed by a series of enrichment, transfer, and dilutions. Among the isolated strains, two strains (JSAR-1 and JSAR-3) with abilities of tolerance to 10 mM As(V) and Fe(III) reduction were selected. Phylogenetic analysis using 16S rRNA genesequences indicated the closest members of Pseudomonas stutzeri DSM 5190 and Paenibacillus selenii W126, respectively for JSAR-1 and JSAR-3. Ferric and ferrous iron concentrations were measured by ferrozine assay, and arsenic concentration was analyzed by ICP-AES, suggesting inability of As(V) reduction whereas ability of Fe(III) reduction. CONCLUSION: Fe(III)-reducing bacteria isolated from the enrichments with arsenate and ferric iron were found to be resistant to a high concentration of As(III) at 10 mM. We suppose that those kinds of microorganisms may suggest good application potentials for As(V) bioremediation, since the bacteria can transform Fe while surviving under As-contaminated environments. The isolated Fe(III)-reducing bacterial strains could contribute to transformations of iron minerals which may act as effective adsorbents for arsenate, and therefore contribute to As(V) immobilization
The objective of this study was to investigate the bacterial adhesion to iron (hydr)oxide-coated sand (IHCS) and aluminum oxidecoated sand (AOCS) in the presence of Tween 20 (nonionic surfactant) and lipopeptide biosurfactant (anionic surfactant) through column experiments. Results show that in the presence of Tween 20, bacterial adhesion to the coated sands was slightly decreased compared to the condition of deionized water; the mass recovery (Mr) increased from 0.491 to 0.550 in IHCS and from 0.279 to 0.380 in AOCS. The bacterial adhesion to the coated sands was greatly reduced in lipopeptide biosurfactant; Mr increased to 0.980 in IHCS and to 0.797 in AOCS. Results indicate that the impact of lipopeptide biosurfactant on bacterial adhesion to metal oxide-coated sands was significantly greater than that of Tween 20. Our results differed from those of the previous report, showing that Tween 20 was the most effective while the biosurfactant was the least effective in the reduction of bacterial adhesion to porous media. This discrepancy could be ascribed to the different surface charges of porous media used in the experiments. This study indicates that lipopeptide biosurfactant can play an important role in enhancing the bacterial transport in geochemically heterogeneous porous media.
This study investigated the efficacy of ultraviolet-C (UV-C) irradiation in enhancing the quality of raw bovine milk by targeting microbial populations and lipid peroxidation, both of which are key factors in milk spoilage. We categorized the raw milk samples into three groups based on initial bacterial load: low (<3 Log 10 CFU/mL), medium (3-4 Log 10 CFU/mL), and high (>4 Log 10 CFU/mL). Using a 144 W thin-film UV-C reactor, we treated the milk with a flow rate of 3 L/min. We measured the bacterial count including standard plate count, coliform count, coagulase-negative staphylococci count, and lactic acid bacteria count and lipid peroxidation (via thiobarbituric acid reactive substances assay) pre- and post-treatment. Our results show that UV-C treatment significantly reduced bacterial counts, with the most notable reductions observed in high and medium initial load samples (>4 and 3-4 Log 10 CFU/mL, respectively). The treatment was particularly effective against coliforms, showing higher reduction efficiency compared to coagulase-negative staphylococci and lactic acid bacteria. Notably, lipid peroxidation in UV-C treated milk was significantly lower than in pasteurized or untreated milk, even after 72 hours. These findings demonstrate the potential of UV-C irradiation as a pre-treatment method for raw milk, offering substantial reduction in microbial content and prevention of lipid peroxidation, thereby enhancing milk quality.
Background: Some studies confirm the reduction of the number of Streptococcus mutans in saliva and dental plaque by Lactobacillus, however, these effects are not always confirmed in in vitro and clinical studies, and only the risk of dental caries has been reported. Our in vitro study aimed to reveal microbial and biochemical changes in the single cultures of S. mutans, Lactobacillus casei and Aggregatibactor actinomycetemcomitans and co-cultures of S. mutans and L. casei or A. actinomycetemcomitans according to sucrose concentration. We also aimed to confirm the anti-oral bacterial and anti-biofilm activities of L. casei and A. actinomycetemcomitans against S. mutans according to sucrose concentration. Methods: S. mutans (KCCM 40105), L. casei (KCCM 12452), and A. actinomycetemcomitans (KCTC 2581) diluted to 5×106 CFU/ml were single cultured, and L. casei or A. actinomycetemcomitans applied at concentrations of 10%, 20%, 30% and 40% to S. mutans were co-cultured with selective medium containing 0%, 1% and 5% sucrose at 36.5℃ for 24 hours. Measurements of bacterial growth value and acid production, disk diffusion and biofilm formation assays were performed. Results: In the medium containing sucrose, the bacterial growth and biofilm formation by S. mutans, L. casei, and A. actinomycetemcomitans were increased. In contrast, 30% and 40% of L. casei in the medium containing 0% sucrose showed both anti-oral bacterial and anti-biofilm activities. This implies that L. casei can be used as probiotic therapy to reduce S. mutans in a 0% sucrose environment. Conclusion: The concentration of sucrose in the oral environment is important for the control of pathogenic bacteria that cause dental caries and periodontitis. To apply probiotic therapy using L. casei for S. mutans reduction, the concentration of sucrose must be considered.
Bacterial abundance, production, and environmental parameters were investigated three times to study distribution of bacterial variables and to examine how estuarine mixing would influence the distribution of bacterial variables in the euphotic zone of the estuarine system of the Mankyung river and Dongjin river during a period of October, 1990-August, 1991. Although a limited number of investigations were made, bacterial abundance and production showed large variations from 0.4 to 5.8${\times}$10/SUP 9/ 1/SUP -1/ and from 0.1 to 22.2 ug C 1/SUP -1/ d/SUP -1/, respectively. The wide ranges of bacterial variables indicated very dynamic changes in conditions of bacterial growth in the estuary. Interestingly, bacterial abundance substantially increased with depth in most stations f shallow depth. but bacterial production remarkably decreased with depth. We propose that the observed distribution of bacterial abundance and production would be explained by estuarine mixing in the estuary. Analyses of mixing diagrams showed that estuarine mixing would mix conservatively bacteria and bacterial production. Further, estuarine mixing often seemed to cause an increase in bacterial abundance and reduction of bacterial production presumably due to resuspension of sediment. This suggests that roles of estuarine mixing would be significant in the distribution of bacterial abundance and production, and thus in biogeochemical cycles in the estuary.
Indigenous bacterial mediation of As in contaminated sediment after biostimulation with a variety of carbon sources was investigated under aerobic and anaerobic conditions. Under the aerobic condition with lactate supply, indigenous bacteria increased the amount of total As extracted from the sediment and most dissolved As existed as As(V). Under the anaerobic, glucose-supplied condition, dissolved As diminished with time likely due to production of As sulfide(s) and subsequent precipitation, which resulted from bacterial reduction of ${SO_4}^{2-}$. The results implied that bacterial natural attenuation of As in subsurface has a potential to be practically applied.
Despite the increased interests in biological control of soilborne diesease for environmental protection, biological control of bacterial wilt caused by Ralstonia solanacearum have not provided consistent or satisfying results. To enhance the control efficacy and reducing the inconsistency and variability, combinations of specific strains of microorganisms, each having a specific mechanism of control, were applied in this study. More than 30 microorganisms able to reduce the activity of pathogen by specific mechanism of action were identified and tested for their disease suppressive effects. After in vitro compatibility examinations, 21 individual strains and 15 combinations were tested in the greenhouse. Results indicated three-way combinations of different mode of control, TS3-7+A253-16+SKU78 and TS1-5+A100-1+SKU78, enhanced disease suppression by 70%, as compared to 30-50% reduction for their individual treatments. This work suggests that combining multiple traits antagonizing the pathogen improve efficacy of the biocontrol agents against Ralstonia solanacearum.
Bacillus subtilis JW-1 was isolated from rhizosphere soil as a potential biocontrol agent of bacterial wilt caused by Ralstonia solanacearum. Seed treatment followed by a soil drench application with this strain resulted in >80% reduction in bacterial wilt disease compared with that in the untreated control under greenhouse conditions. The antibacterial compound produced by strain JW-1 was purified by bioactivity-guided fractionation. Based on mass spectroscopy and nuclear magnetic resonance spectral data ($^1H$, $^{13}C$, $^1H-^1H$ correlation spectroscopies, rotating frame nuclear Overhauser effect spectroscopy, and heteronuclear multiple-bond correlation spectroscopy), the structure of this compound was elucidated as a cyclic lipopeptide composed of a heptapeptide (Gln-Leu-Leu-Val-Asp-Leu-Leu) bonded to a ${\beta}$-hydroxy-iso-hexadecanoic acid arranged in a lactone ring system.
