세균 분리주 KTB61의 담배 모자이크 바이러스(TMV) 감염 억제 효과 (Inhibitory Activity of Bacterial Isolate Pseudomonas sp. KTB61 against Tobacco Mosaic Virus(TMV) Infection to Tobacco Plants)

  • During the screening or antiviral substances having inhibitory effect on tobacco mosaic virus(TMV) infection to tobacco plants, we found that a bacterial isolate, KTB61, which was identified as a Pseudomonas sp., strongly inhibited the formation of TMV local lesions. When the culture filtrate from KTB61 was applied on the upper surface of leaves of N. tabaccum Xanthi-nc tobacco at the same time of or 24 hours before TMV inoculation, almost complete inhibition was achieved. Incidence of systemic TMV infection to the susceptible tobacco cultivar, NC82, was reduced by 95% when TMV was inoculated onto the upper surface of leaves 24 hours after spraying the culture filtrate. Also 75∼80% of inhibitory effect was obtained by the inoculation of TMV onto the under surface of the leaves treated with culture filtrate 24 hours beforehand. In field trials, the TMV infection was reduced by 96.5% when the tobacco seedlings, N. tabaccum cv. NC82, were soaked with culture filtrate before transplanting.

  • During the screening of antiviral substances having inhibitory effects on tobacco mosaic virus (TMV) infection to tobacco plants, we found a bacterial isolate KTGBP10, which was identified as a Stenotrophomonas sp., strongly inhibited the infection of TMV. When the culture filtrate from KTGBP10 was applied on the upper surface of leaves of Xanthi-nc tobacco plants at the same time or 24 hours before TMV inoculation, almost complete inhibition of TMV infection was achieved. And $40\%$ inhibition was shown with application of the culture filtrate to the under surface of leaves. In field trials, transmission of TMV from diseased seedlings to the healthy ones during transplanting work was reduced by $87.1\~92.6\%$ when the culture filtrate or cell suspension was sprayed onto the tobacco seedlings, cv. NC82, 24 hours before transplanting. No toxic effect was observed on the tobacco plants. When the broth filtrate of KTGBP10 was supplied by soaking through the cut-leaves before and/or after virus inoculation, the TMV infection was also inhibited by $50.4\~65.3\%$.

  • The typical dark brown stripe symptom of bacterial wilt disease was observed in the cuttings of tobacco stem treated with the culture filtrate of virulent Pseudomonas solanaceamm. And the tobacco callus create.4 with that culture filtrate showed deterioration of the callus 2 days after the treatment. On the contrary, the cuttings and the callus treated with the culture filtrate of the avirulent bacteria expressed no typical symptom and vigorous growth respectively. Therefore it was suggested that certain toxin which might be produced by the virulent bacteria could break down tobacco cells.

  • Three antagonistic bacterial strains against Valsa ceratosperma, one of the apple tree pathogens, were isolated from the nature and investigated. Out of the about 3,000 species of microorganisms which was isolated from the nature, the 3 strains designated as CH219, CH220 and CH245 were selected through the test of their antagonistic activity. The antagonists showed over 50% of antifungal activity against the growth of Valsa ceratosperma on PDA plates and, by the treatment of the culture broth and the heat-treated culture filtrate of it, showed over 95% of antifungal activity. When we tested on the medium which contained their culture filtrate or heat-treated culture filtrate, the antagonists strongly inhibited Valsa ceratosperma. In bioassay on the apple trees, the antagonists also showed their antifungal activity.

A Bacterial Endophyte, Pseudomonas brassicacearum YC5480, Isolated from the Root of Artemisia sp. Producing Antifungal and Phytotoxic Compounds

  • An endophytic bacterial strain YC5480 producing antifungal and phytotoxic compounds simultaneously was isolated from the surface sterilized root of Artemisia sp. collected at Jinju area, Korea. The bacterial strain was identified as a species of Pseudomonas brassicacearum based on its 16S rRNA gene sequence analysis and physiological and biochemical characteristics. The seed germination and growth of monocot and dicot plants were inhibited by culture filtrate (1/10-strength Tryptic Soy Broth) of the strain. The germination rate of radish seeds in the culture filtrate differed in various culture media. Only 20% of radish seeds germinated in the culture media of 1/2 TSB for 5 days incubation. Mycelial growth of fungal pathogens, Colletotrichum gloeosporioides, Fusarium oxysporum and Phytophthora capsici was also inhibited by the culture filtrate of the strain YC5480. An antifungal compound, KS-1 with slight inhibitory activity of radish seed germination at 1,000 ppm and a seed germination inhibitory compound, KS-2 without suppression of fungal growth were produced simultaneously in TSB. The compounds KS-1 and KS-2 were identified to be 2,4-diacetylphloroglucinol (DAPG) and 2,4,6-trihydroxyacetophenone (THA), respectively.

Inhibitory Effects of Acinetobacter sp. KTB3 on Infection of Tobacco mosaic virus in Tobacco Plants

  • During the screening of antiviral substances having inhibitory effects on Tobacco mosaic virus (TMV) infection on tobacco plants, we found a bacterial isolate KTB3, and identified it as Acinetobacter sp. which strongly inhibited the infection of TMV When the culture filtrate from KTB3 was applied on the upper surface of the Xanthi-nc tobacco leaves at the same time, or 24 hours before TMV inoculation, almost complete inhibition was achieved. Likewise, 86% inhibition was achieved, when the culture filtrate was applied on the underside of the leaves. In field trials, transmission of TMV from diseased seedlings to healthy ones during transplanting work was reduced by 92%, when the culture filtrate was sprayed onto the tobacco seedlings, cv. NC82, 24 hours before transplanting. No toxic effect was observed on the tobacco plants. Antiviral substance from the culture filtrate was purified by ethanol precipitation, dialysis, DEAE-cellulose, and Sephadex G75 gel column chromatography. The partially purified active material which showed positive color reaction to sugar and protein inhibited TMV infection by 60% at 1 ${\mu}$g/ml.

Inhibitory Effects of a Korean Strain Gpf01 Identified as Pseudomonas fluorescens on Cucumber mosaic virus

  • An antiviral producing bacterial strain was isolated from a ginseng rhizosphere in Kangwon province of Republic of Korea. In order to identify the bacterial strain, microbiological, physiological and biochemical tests were performed, along with RAPD, 16S rRNA, 16S-23S rRNA ITS (intergenic spacer region) and DNA-DNA hybridization analyses. The bacterium was found to be a strain of Pseudomonas fluorescens, which was designated as Gpf01. The strain was grown in Muller-Hinton (MH) broth, and the culture supernatant obtained was filtered through a $0.45{\mu}l$ filter. It was further boiled at $100^{\circ}C$ and tested in two experiments for its ability to control a yellow strain of Cucumber mosaic virus (CMV-Y). In the first experiment, boiled culture filtrate (RCF) was treated on one half of the leaves of Chenopodium amaranticolor followed by CMV- Y inoculation on both halves. In the second experiment, BCF was treated on the lower leaves of Nicotiana tobacum var. Xanthi-nc, with the CMV-Y mechanically inoculated onto the upper untreated leaves. In the first experiment, BCF treatment was able to considerably reduce the number of viral lesion, and in the second experiment, plants treated with BCF showed no visible viral symptoms compared to the Muller-Hinton (MH) media treated controls 15 days post inoculation (dpi), and remained symptomless throughout the study period. Thus, Gpf01, identified as P. fluorescence, was able to produce an antiviral component in the culture filtrate, which was found to be heat stable, non-phytotoxic and effective in local as well as systemic hosts of CMV.

사과 겹무늬썩음병균에 대한 Bacillus sp. SS279의 항진균활성과 생물학적 방제

  • From apple skin, we isolated a bacterial strain which strongly inhibited the growth of apple white rot fungus, Botryosphaeria dothidea. The isolated strain, designated as SS279, was identified to be the genus Bacillus. The antifungal activity of Bacillus sp. SS279 was found in the culture filtrate. The production of antifungal substances occurred during logarithmic phase and was the highest when cultures reached the stationary growth phase. The optimum ranges of temperature and pH for its production were 25-30$\circ$C and 4.5-9.0, respectively. The culture filtrate of Bacillus sp. SS279 exhibited a strong inhibitory effect on the spore germination and germ tube elongation of B. dothidea. Autoclaved culture filtrate of Bacillus sp. SS279 showed only a slight decrease in antifungal activity, indicating that the Bacillus sp. SS279 produce heat-stable antifungal substances. In in vivo bioassay, Bacillus sp. SS279 also showed antagonistic activity against apple white rot caused by B. dothidea.

적조생물 살조세균 탐색 II. 적조생물 Prorocentrum micans 살조세균 Pseudomonas sp. LG-2의 분리와 살조특성 (Isolation of Marine Bacteria Killing Red Tide Microalgae II. Isolation and Algicidal Properties of Pseudomonas sp. LG-2 Possessing Killing Activity for Dinoflagellate, Prorocentrum micans)

  • 1996년 7월 마산만의 해수를 0.8 $\mu$m filter에 여과한 여과액과 f/2배지에서 배양한 P. micans와의 혼합배양액에서 P. micans를 사멸시키는 해양세균을 분리하였다. 분리균의 형태학적 및 생화학적 검사와 균체지방산을 분석하여 동정한 결과 Pseudomonas 속으로 동정되어 Pseudomonas sp. LG-2로 명명하였으며, 그 살조특성을 조사한 결과는 아래와 같다. Pseudomonas sp. LG-2의 개체수와 배양여과액의 농도가 높을수록 P. micans의 사멸효과가 높게 나타났다. $1.3\times10^5,\;1.3\times10^6$ cells/ml의 농도로 접종한 시험관의 P. micans는 급격히 감소하여 배양 7일 후에는 $10^2$ cells/ml 이하로 사멸되었다. 또한, 배양석과액의 최종농도가 $30\%$일 경우에는 급격히 감소하여 배양 3일 후 거의 사멸하였다. Pseudomonas sp. LG-2의 성장시기에 따른 배양여과액의 사멸효과는 잠복기의 경우 P. micans의 성장에 큰 영향을 미치지 않았으나, 대수증식기 중기의 배양여과액은 접종 5일 후 P. micans의 개체수를 1/2로 감소시켰으며, 정상기의 배양여과액은 접종 후 급격히 감소시켜 배양 3일 후 대부분 사멸시켰다. Alexandrium tamarense, Prorocentrum micans, Scrippsiella trochoidea, Gymnodinium sanguineum, Cochlodinium polykrikoides의 5종의 적조원인 편모조류에 대한 Pseudomonas sp. LG-2의 살조특이성은 P. micans와 S. trochoidea에 살조효과를 나타내었다.

Biological Control of Gom-chwi (Ligularia fischeri) Phytophthora Root Rot with Enterobacter asburiae ObRS-5 to Suppress Zoosporangia Formation and Zoospores Germination

  • Gom-chwi (Ligularia fischeri) is severely infected with Phytophthora drechsleri, the causal organism of Phytophthora root rot, an economically important crop disease that needs management throughout the cultivation period. In the present study, Phytophthora root rot was controlled by using bacterial isolates from rhizosphere soils collected from various plants and screened for antagonistic activity against P. drechsleri. A total of 172 bacterial strains were isolated, of which, 49 strains showed antagonistic activities by dual culture assay. In the seedling assay, six out of the 49 strains showed a predominant effect on suppressing P. drechsleri. Among the six strains, the ObRS-5 strain showed remarkable against P. drechsleri when treated with seed dipping or soil drenching. The ObRS-5 strain was identified as Enterobacter asburiae based on 16S ribosomal RNA gene sequences analysis. The bacterial cells of E. asburiae ObRS-5 significantly suppressed sporangium formation and zoospore germination in P. drechsleri by 87.4% and 66.7%, respectively. In addition, culture filtrate of E. asburiae ObRS-5 also significantly inhibited sporangium formation and zoospore germination by 97.0% and 67.6%, respectively. Soil drenched bacterial cells, filtrate, and culture solution of E. asburiae ObRS-5 effectively suppressed Phytophthora root rot by 63.2%, 57.9%, and 81.1%, respectively. Thus, E. asburiae ObRS-5 could be used as a potential agent for the biological control of Phytophthora root rot infecting gom-chwi.
