Although enumerating bacterial cells is a fundamental step in understanding microbial ecosystems in marine environments, substantial decrease in bacterial counts with increasing sample storage time hampers the accurate estimation of bacterial biomass. We compared the variations in bacterial cell numbers caused by freezing and thawing of sample bottles or slides. Bacterial counts of seawater samples frozen only once in a sampling bottle yielded approximately 95% of the original numbers after 90 days, whereas 80% of the original count was obtained for samples prepared on slides. Only 67% and 58% of the original counts were recovered in samples repeatedly frozen and thawed in bottles or on slides, respectively. The results indicated that freezing a seawater sample in a bottle increased the consistency of the epifluorescence microscopic enumeration of bacterial cells.
Purpose: The purpose of this study was to investigate the condition of feminine hygiene behaviors to identify factors predicting bacterial vaginosis infection. Methods: A self-reporting survey was conducted with 385 female university students in Korea through a descriptive survey. Data were collected on demographics, feminine hygiene behaviors, and bacterial vaginosis infection. Results: The bacterial vaginosis infection rate was 37.9%. Factors predicting bacterial vaginosis infection were grade, age of menarche, history of sexual intercourse, history of sexually transmitted infection, experience of using a bidet, internal douche during menses, over the counter drug anti-itch products, tampons, and tampons/pads combination. Conclusion: To prevent bacterial vaginosis infection among female university students, information and education should be provided to promote positive feminine hygiene behaviors at the university and at home. Results of this study are essential to improve education and practices that will prevent bacterial vaginosis in female university students.
The distribution of physicochemical environmental factors and microbiological factors was studied at 6 sampling sites in Kyeongge Bay of Yellow Sea from October 1989 to October 1990. The total bacterial number, saprophytic bacterial number, petroleum-degrading bacterial number, bacterial biomass, and bacterial secondary production were measured in the range of 0.09~1.24*10$^{7}$ cells/ml, 7~60000 CFUs/ml, 0~240 cells/ml, 14.16~301 .$\mu$g-C/l, and 0.13~11.82 mg-C/m$^{3}$/hr, respectively. The turnover times of $^{3}$H-glucose and $^{3}$H-acetate were in range of 6.5~6984 and 41~24897 hours, respectively. The spatial distribution of heterotrophic bacterial communities were hightly affected by influx of organic pollutants from the coastal area and the seawater exchange with offshore.
Objectives : This study was designed to evaluate the clinical effects of herb medicine and acupuncture therapy on chronic bacterial prostatitis. Method : A clinical observation was made on two patients with chronic bacterial prostatitis diagnosed with DNA analysis. They were treated with herb medicine and acupuncture therapy. Before and after treatment we measured National Institutes of Health Chronic Prostatitis Symptom Index(NIH-CPSI) and WBC counts on expressed prostatic secretion(EPS). Results : According to DNA analysis, Streptococcus mitis and Veillonella was proved to be the cause for chronic bacterial prostatitis for each patients. After treatment, NIH-CPSI score improved from severe to mild, and WBC counts on EPS decreased considerably from many to under 10. Conclusion : The present result suggest that oriental medical therapy has effect on chronic bacterial prostatitis. Further clinical study about chronic bacterial prostatitis, and research on herbal medicine sensitive to each bacterium causing chronic bacterial prostatitis is needed.
Accurate and rapid detection of bacterial plant pathogen is the first step toward disease management and prevention of pathogen spread. Bacterial plant pathogens Clavibacter michiganensis subsp. nebraskensis (Cmn), Pantoea stewartii subsp. stewartii (Pss), and Rathayibacter tritici (Rt) cause Goss's bacterial wilt and blight of maize, Stewart's wilt of maize and spike blight of wheat and barley, respectively. The bacterial diseases are not globally distributed and not present in Korea. This study adopted comparative genomics approach and aimed to develop specific primer pairs to detect these three bacterial pathogens. Genome comparison among target pathogens and their closely related bacterial species generated 15-20 candidate primer pairs per bacterial pathogen. The primer pairs were assessed by a conventional PCR for specificity against 33 species of Clavibacter, Pantoea, Rathayibacter, Pectobacterium, Curtobacterium. The investigation for specificity and sensitivity of the primer pairs allowed final selection of one or two primer pairs per bacterial pathogens. In our assay condition, a detection limit of Pss and Cmn was $2pg/{\mu}l$ of genomic DNA per PCR reaction, while the detection limit for Rt primers was higher. The selected primers could also detect bacterial cells up to $8.8{\times}10^3cfu$ to $7.84{\times}10^4cfu$ per gram of grain seeds artificially infected with corresponding bacterial pathogens. The primer pairs and PCR assay developed in this study provide an accurate and rapid detection method for three bacterial pathogens of grains, which can be used to investigate bacteria contamination in grain seeds and to ultimately prevent pathogen dissemination over countries.
Objectives : This experimental study was performed to investigate the continuous anti-bacterial potency of Sean-tang on cultivation of Staphylococcus species(S. aureus, S. epidermidis) that induce eye disease. Methods : Minimal inhibitory concentration(MIC) was measured by dropping to 50${\mu}$l diluted Sean-tang(100%, 10%,1%, 0.1%) on S. aureus, S. epidermidis that were cultivated from 2 to 6 days. 1. Anti-bacterial potency was measured by the size of inhibition zone with change of volume(20${\mu}$l, 30${\mu}$l, 40${\mu}$l, 50${\mu}$l). Results : 1. Anti-bacterial potency of Sean-tang on S. aureus was appeared in 100% and increased on 6 days. Anti-bacterial potency with change of volume was increased in propotion to increase volume. Anti-bacterial potency of Sean-tang on S. aureus was appeared continuous. 2. Anti-bacterial potency of S. epidermidis was appeared in 100%, 10%, 1% on 2 days and in100%, 10% on 6 days. In 100% Sean-tang, Anti-bacterial potency of 6 days was increased, in 10%, 1%, Anti-bacterial potency of 2 days was increased. Anti-bacterial potency with change of volume was increased inpropotion to increase volume and increased on 6 days, but bacteria was increased. Anti-bacterial potency Sean-tang on S. epidermidis wasn't appeared continuous. Conclusions : Anti-bacterial potency of Sean-tang on cultivation of S. aureus was showed continuous, but on cultivation of S. epidermidis was not showed continuous.
The total bacteria count is significantly important factor for hygienic quality in raw milk. BactoScan FMTM and BactoCount IBCTM are the automated instruments for the determination of the total bacterial count in raw milk. They have been used after calibration by standard plate count (SPC) method in South Korea since 2000. It is necessary to re-evaluate for total bacterial counter according to the improvement of milk quality and the change of milk quality grade. Therefore, this study was evaluated the conversion mode of the two machines by SPC method. We collected 921 bulk-tank milk samples throughout the concentration range of 1,000~1,000,000 CFU/mL from June 2020 to October 2021. As a result, when compared by the SPC value, there was a slight difference in total bacterial count in BactoScan below 10,000 CFU/mL and above 200,000 CFU/mL and in BactoCount above 100,000 CFU/mL, respectively. Therefore, the conversion factor for BactoScan and the conversion equation for BactoCount were newly adjusted based on SPC value, and then the correlation coefficients (R2) was 0.85 or higher. In addition, the correlation (R2) between BactoScan and BactoCount was 0.91, which means the results were high positive correlation. These results are expected to contribute to improving the accuracy of the automated instruments for determining of total bacterial count in raw milk.
Four major diseases of chili pepper including two fungal diseases, anthracnose (Colletotrichum acutatum) and Phytophthora blight (Phytophthora capsici), and two bacterial diseases, bacterial wilt (Ralstonia solanacearum) and bacterial spot (Xanthomonas campestris pv. vesicatoria), were investigated under future climate-change condition treatments in growth chambers. Treatments with elevated $CO_2$ and temperature were maintained at $720ppm{\pm}20ppm$$CO_2$ and $30^{\circ}C{\pm}0.5^{\circ}C$, whereas ambient conditions were maintained at $420ppm{\pm}20ppm$$CO_2$ and $25^{\circ}C{\pm}0.5^{\circ}C$. Pepper seedlings or fruits were infected with each pathogen, and then the disease progress was evaluated in the growth chambers. According to paired t-test analyses, bacterial wilt and spot diseases significantly increased by 24% (p=0.008) and 25% (p=0.016), respectively, with elevated $CO_2$ and temperature conditions. On the other hand, neither Phytophthora blight (p=0.906) nor anthracnose (p=0.125) was statistically significant. The elevated $CO_2$ and temperature accelerated the progress of bacterial wilt by two days and bacterial spot by one day compared to the ambient treatment. Temperature regime studies of the diseases without changes in $CO_2$ confirmed that the accelerated bacterial disease progress was mainly due to the increased temperature rather than the elevated $CO_2$ conditions.
Our study aimed to discover indigenous prokaryotic species in Korea. A total of 29 bacterial species in the phylum Proteobacteria were isolated from freshwater and sediment of rivers and brackish zones in Korea. From the high 16S rRNA gene sequence similarity (${\geq}98.8%$) and formation of a robust phylogenetic clade with the closest species, it was determined that each strain belonged to an independent and predefined bacterial species. To our knowledge, there is no official report or publication that has previously described these 29 species in Korea. Specifically, we identified 10, 12, and seven species of eight, 12, and seven genera that belong to classes Alphaproteobacteria, Betaproteobacteria, and Gammaproteobacteria, respectively; all are reported as previously unrecorded bacterial species in Korea. The Gram reaction, colony and cell morphology, basic biochemical characteristics, isolation source, and strain IDs for each are also described.
