모기 살충성 세균 B. thuringiensis subsp. israelensis의 효과적인 분리 방법 (The Effective Isolation of a Mosquitocidal Bacteria, Bacillus thuringiensis Subsp. israelensis)

  • For more convenient and rapidly isolation of Bacillus thuringiensis subsp. israelensis(Bti), 1) heat treatment spore forming bacteria, 2) growth in enrichment culture media for Bacillus sp. and 3) selection of bacteria producing a lecithinase for Bacillus thuringiensis subsp. israelensis, were performed. Spore forming bacteria were counted 4.8 $\times $ 10$^{8}$cells/g soil on NAPGCY media, 9.2 $\times $ 10$^{7}$cells/g soil on NA media, and 3.6 $\times $ 10$^{8}$cells/g soil on NAAC media, respectively. Bacteria producing only a lecithinase were reached at 25.2% on medium contained egg york, bacteria only producing a delta-endotoxin were reached at 23.2% by phase contrast microscope, and bacteria producing a lecithinase & a delta-endotoxin simultaneously were reached at 13.7%. Bacillus thuringiensis which producing a lecithinase and a delta-endotoxin simultaneously among bacteria producing a lecithinase, were reached at 56.5%; A half of Bacillus thuringiensis was produced a delta-endotoxin, but not produced a lecithinase. Among 8 isolates of Bacillus thuringiensis, two strain of Bti which has a mosquito-cidal toxin, were detected by PCR using a specific primer of $\delta $-endotoxin gene from Bti.

  • Bdellovibrio sp. is an ectoparasitic bacteria which is predatory and parasitic upon other bacteria. This study was carried out the isolation of Bdellovibrio sp. from several soil smaples and observation of this organisms by means of electron microscope. The results are as follows ; The primary isolated Bdellovibrio sp. from soil is an obligate intracellular rod form parasite and possess a monoflagella.

  • Anaerobic bacteria are the major residents of human skin and mucous membrane. The importance, as opportunistic pathogens, of anaerobic bacteria are well recognized because of more population. with decreased defense against bacterial invasion due to chemotherapy, rediation therapy, extensive surgical operation etc. are dealt at hospitals. An analysis of the anaerobe isolation results at Yonsei University Medical Center during the January 1974-May 1975 period was made and the following results were obtained. 1) From 118 patients 146 strains of anaerobes were isolated. Among these 81.3% were nonsporforming anaerobes. Most frequently isolated anaerobes were Pc. asaccharolyticus, Ps. anaerobius, Ps. intermedius, B. fragilis and Cl. perfringens. 2) Anaerobes were frequently isolated from wound, female genital, intraabdominal, and pleuropulmonary specimens. Fewer anaerobes were isolated from blood, spinal fluid and liver specimens. 3) The ratio of anaerobe isolation to total bacteria isolation were; liver 66.7%, intraabdominal 33.3%, pleuropulmonary 28.9%, spinal fluid 5.0% and blood 4.2%. 4) Among the 118 anaerobe isolated patients, 48.3% yielded anaerobes only and rest of them yielded anaerobes together with aerobes. 5) Most of the gram-positive anaerobes were susceptible to the antibiotics tested. Exception was to tetracycline to which appreciable number showed resistance. It was noteworthy that only 48% of B. fragilis was susceptible to tetracycline.

  • The objective of this study was isolation of halotolerant lactic acid bacteria for fermentation of food wastes. 5 strains of lactic acid bacteria were isolated from fermented foods. Among isolated strains, the strain 5-2 was selected according to the growth characteristics in food wastes containing medium. The selected strain 5-2 was identified as Pediococcus acidilactici based on its biochemical characteristics.

  • Bacterial grain rot by Burkholderia gluae cause severe damage in seedling and grain of rice after heading season. This seed-borne pathogen play a role as first infection agent that could be cause disease following cropping season. Until now the direct isolation of the bacteria has some trouble by interference of other bacteria existed inside seed. This study established convenient identification method as simple isolation with KB medium from seed showing symptom and using PCR identification. By this isolation method, B. glumae was isolated from 40 to 50% in brown rice and inner hull, however, there were saprophytic bacteria and fungi outer hull. In PCR identification with Ogf4 and Ogr3 primer to these 25 isolates, the amplified products were presented in all of the collections but not in 10 saprophytic germs. The isolation rate was constant to 3 months stored seeds. This result provide a rapid and convenient isolation and identification of B. glumae.

  • Blood is one of the most important clinical specimens for the isolation of bacteria. A rapid isolation and a high isolation rate of bacteria are very important in blood culture because bacteremic patients are mostly in grave condition. Various blood culture media which support growth of most fastidious bacteria are available commercially. However, growth of bacteria are frequently delayed because of antibacterial activity of blood. Sodium polyanethol sulfonate(Liquoid) has been reported to inactivate the antibacterial substance and disrupt phagocytic cells. The beneficial effect of SPS is well recognized in the isolation of gram-positive bacteria. However, the effect does not seem to be prominent for gram-negative bacilli isolation mainly due to the rapidity of their growth. It has been experienced with Sal. typhi that the growth is much slower than that of other gram-negative bacilli. For the rapid growth of the organism, use of bile broth has been recommended. Although Sal. typhi is the most frequently isolated organism at present, about one half of total isolates are other organisms and, in case bile broth is used, other media which support growth of these organisms should be used together. Fluid thioglycollate medium(FTM) which is always used in blood culture to isolate anaerobes is inferior to brain heart infusion(BHI) for the isolation of aerobes. This study was done to determine the effect of SPS on the isolation of Sal. typhi from blood. During the Sep. 1973 to Sep. 1974 study period, 2460 blood cultures were made from the Severance hospital patients: BHI and FTM sets 1431 specimens, BHI with SPS(0.05%) and FTM sets 396 specimens, BHI and FTM with SPS sets 359 specimens, BHI and BHI with SPS sets 274 specimens. Mean incubation time required for the macroscopic detection of growth of Sal. typhi were 3.5 days on BHI and 2.7 days on BHI with SPS. The 0.8 day difference was statistically significant. On FTM the mean incubation time was 3.8 days while it was 2.9 days on FTM with SPS. The 0.9 day difference was statistically significant. The result on BHI with and without SPS sets showed faster growth on BRI with SPS in 7 specimens and slower growth in one specimen and the remaining 12 showed growth at the same time. These specimens had mean incubation time of 3.2 days on BHI and 2.3 days on BHI with SPS. The 0.9 day difference was statistically significant. This study indicates beneficial effect of SPS for the rapid isolation of Sal. typhi from clinical blood specimens.

Influence of Isolation Temperature on Isolating Diverse Lactic Acid Bacteria from Kimchi and Cultural Characteristics of Psychrotrophs

  • Kimchi is a traditional Korean fermented vegetable that is stored and fermented at low temperatures. However, kimchi lactic acid bacteria (LAB) are typically isolated under mesophilic conditions, which may be inappropriate for isolating the diverse LAB. Therefore, this study investigated the suitable conditions for isolating various LAB from kimchi. Here, LAB were isolated from four kimchi samples using MRS, PES, and LBS media and varying isolation temperatures (30, 20, 10, and 5℃). Then, MRS was selected as the suitable medium for LAB isolation. A comparison of culture-dependent and culture-independent approaches indicated that 5℃ was not a suitable isolation temperature. Thus, the number and diversity of LAB were determined at 30, 20, and 10℃ using 12 additional kimchi samples to elucidate the effect of isolation temperature. With the exception of two samples, most samples did not substantially differ in LAB number. However, Leuconostoc gelidum, Leuconostoc gasicomitatum, Leuconostoc inhae, Dellaglioa algida, Companilactobacillus kimchiensis, Leuconostoc miyukkimchii, Leuconostoc holzapfelii, and Leuconostoc carnosum were isolated only at 10 and 20℃. The growth curves of these isolates, except Leu. holzapfelii and Leu. carnosum, showed poor growth at 30℃. This confirmed their psychrotrophic characteristics. In Weissella koreensis, which was isolated at all isolation temperatures, there was a difference in the fatty acid composition of membranes between strains that could grow well at 30℃ and those that could not. These findings can contribute to the isolation of more diverse psychrotrophic strains that were not well isolated under mesophilic temperatures.

Sensitive and Enzyme-Free Pseudomonas aeruginosa Detection and Isolation via DNAzyme Cascade Triggered DNA Tweezer

  • Effective isolation and sensitive detection of Pseudomonas aeruginosa (P. aeruginosa) is crucial for the early diagnosis and prognosis of various diseases, such as urinary tract infections. However, efficient isolation and simultaneous detection of P. aeruginosa remains a huge challenge. Herein, we depict a novel fluorescence assay for sensitive, enzyme-free detection of P. aeruginosa by integrating DNAzyme cascade-induced DNA tweezers and magnetic nanoparticles (MNPs)-based separation. The capture probe@MNPs is capable of accurately identifying target bacteria and transporting the bacteria signal to nucleic acid signals. Based on the DNAzyme cascade-induced DNA tweezers, the nucleic acid signals are extensively amplified, endowing the method with a high sensitivity and a low detection limit of 1 cfu/mL. In addition, the method also exhibits a wide detection of six orders of magnitudes. The proposed method could be extended to other bacteria detection by simply changing the aptamer sequence. Taking the merit of the high sensitivity, greatly minimized detection time (less than 1.5 h), enzyme-free characteristics, and stability, the proposed method could be potentially applied to diagnosing and preventing diseases caused by pathogenic bacteria.

담수식물 뿌리로부터 세균의 분리 (Isolation of Bacteria from Freshwater Plant Roots)

  • The isolation and identification of bacteria from freshwater plant root system were performed. Twenty four different strains were identified by microbiological identification system. Gram negative bacteria were isolated three times more than Gram positive ones. The ratio of rod and coccus was 11:1. The similarity above 0.5 was 37.5% from the tested samples in identification. Among these isolated bacteria, three dominant strains Pseudomonas cepacia JH10, Xanthomonas maltophilia JH12, Aeromonas salmonicida JH13 were selected to investigate biochemical properties and optimal growth conditions. These strains reached maximum growth after 24-36 hr of incubation in peptone broth, and their optimal temperature was 28$^{\circ}C$ and that of pH was between 7 and 8.
