In vivo evaluation of preventive effect of Lactobacillus reuteri on porcine epidemic diarrhea in suckling piglets

  • Lactic acid bacteria have been reported their beneficial roles on host including reduction of infectious diarrhea problems. In this study, preventive effect of Lactobacillus (L.) reuteri HY25101 and L. johnsonii HY25103 on porcine epidemic diarrhea virus (PEDV) was investigated in suckling piglets. Two groups of one day old PEDV naïve piglets were orally administered L. reuteri HY25101 and L. johnsonii HY25103 for three days respectively before challenge with lethal dose of PEDV. In second experiment, passive immunized one day old piglets using colostrums containing PEDV specific IgA were used. The survival rates of the L. reuteri HY25101 administered group were significantly higher than that of L. johnsonii HY25103 administered group and viral shedding was rapidly diminished in L. reuteri HY25101 administered group. Interestingly piglets born from the sow immunized with attenuated PEDV vaccine were not completely protected from PEDV challenge, however coadministeration of L. reuteri HY25101 and colostrums containing PEDV specific IgA were more effectively prevent PEDV infection. These results suggested that dietary treatment using L. reuteri HY25101 could reduce diarrheal problem and mortality rate caused by PEDV in suckling pigs. In addition, L. reuteri HY25101 could be used as one of effective compensation treatment with attenuated live vaccine for PED.

신종 인플루엔자 A (H1N1) 감염으로 입원한 성인 폐렴 환자의 임상양상 (Clinical Features of Hospitalized Adult Patients with Pneumonia in Novel Influenza A (H1N1) Infection)

  • Background: A novel 2009 influenza A (H1N1) virus emerged and disseminated to all over the world. There are few reports on the clinical characteristics of patients with complications. We describe the clinical features of pneumonia in adult patients hospitalized, who have novel influenza infection. Methods: There were 43 adult patients enrolled into the study with pneumonia of 528 hospitalized patients confirmed influenza A (H1N1) virus infection by real-time reverse transcriptase polymerase chain reaction testing, between 24 August 2009 and 31 January 2010. The clinical data of patients with pneumonia were collected retrospectively. Results: There were 22 of 43 (51.2%) influenza patients with pneumonia that had higher risk factors for complications. Compared to 28 patients with influenza A (H1N1) viral pneumonia and 15 patients, who had isolated bacteria from cultures, those with mixed viral and bacterial pneumonia were significantly more likely to have unilobar consolidations on chest radiographs (53.3 vs. 10.7%, p<0.01) and higher scores of pneumonia severity index (PSI; 90 [66~100] vs. 53 [28~90], p=0.04). Six patients required mechanical ventilation support in an Intensive Care Unit and were more likely to have dyspnea (83.3 vs. 29.3%, p=0.02) and low levels of $PaO_2$ (48.3 [37.0~70.5] vs 64.0 [60.0~74.5] mm Hg, p=0.02) and high levels of pneumonia severity index (PSI) score (108.0 [74.5~142.8] vs. 56.0 [40.5~91.0], p=0.03). Conclusion: The majority of pneumonia patients infected with novel influenza improved. Chest radiographic findings of unilobar consolidations suggest that mixed pneumonia is more likely. Initial dyspnea, hypoxemia, and high levels of PSI score are associated with undergoing mechanical ventilation support.

사료 내 마늘 추출액 첨가가 넙치(Paralichthys olivaceus)의 비특이적 면역반응 및 질병저항성에 미치는 영향 (Effects of Dietary Supplementation with Garlic Extract on Immune Responses and Diseases Resistance of Olive Flounder, Paralichthys olivaceus)

  • 본 연구에서는 1% 마늘 추출액을 사료에 첨가하여 항균력 시험, 혈액학적 분석 및 어류질병 원인체들 (Viral Hemorrhagic Septicemia Virus, Vibrio anguillarum, Streptococcus iniae, Edwardsiella tarda)에 대한 비특이적 면역반응을 조사하였다. 사료에 1% 마늘 추출액을 첨가하여 4주간 넙치에 섭이시킨 후 V. anguillarum, S. iniae, E. tarda을 인위 감염시켜 2주간 누적 폐사율을 관찰하였다. 공격 실험 결과 E. tarda를 제외한 모든 시험구에서 대조구에 비해 낮은 누적 폐사율이 관찰되었다. Hematocrit, glucose, total protein, lysozyme activity 및 macrophage activity에서는 대조구에 비해 유의적으로 높은 수치를 관찰하였다. 3가지 어병세균 (V. anguillarum, S. iniae, E. tarda)을 이용하여 마늘 항균력 및 MIC를 측정한 결과, 이전에 보고된 마늘 분말보다 1% 마늘 추출물에서 더 효과적인 결과를 관찰하였다. 이러한 결과는 마늘 추출물이 넙치의 V. anguillarum, S. iniae, E. tarda에 대한 비특이적 면역반응 및 질병저항력을 증가시키는 것으로 사료된다.

C 형 간염 바이러스의 외피당단백질 E1 및 E2의 융합단백질 $GST-E1_{192-283}$$-E2_{384-649}$의 대장균에서의 과량발현 및 면역원성 연구 (Overexpression of the $E1_{192-283}$ and $E2_{384-649}$ Proteins of Hepatitis C Virus in GST Fusion Forms in E. coli and Their Immunogenicity)

  • C 형 간염 바이러스 (Hepatitis C Virus, HCV)는 두종류의 외피당단백질 $E1_{192-383}$$E2_{384-746}$를 갖고 있다. $E1_{192-383}$$E2_{384-740}$ 단백질은 glutathione S-transferae (GST) 융합단백질의 형태로 대장균에서 발현되지 않았으나, 이 단백질들의 C말단에 존재하는 소수성영역을 제거하였을 때 $GST-E1_{192-283}$ 융합단백질은 과량으로 가용성 형태로 발현되었고, $GST-E2_{384-649}$ 융합단백질은 비 가용성 형태로 발현되었다. 이 융합단백질들 각각은 HCV 양성환자의 혈청과 특이적으로 반응하였다. Thrombin으로 처리하여 얻은 정제된 $E1_{192-283}$ 단백질 및 융합형태의 $GST-E2_{384-649}$ 단백질 각각을 생쥐에 접종하였을 때 E1 및 E2 특이적인 항체가 생성되었다. 이 결과들은 $E1_{192-383}$$E2_{384-649}$ 융합단백질 C 말단에 존재하는 소수성영역이 이 단백질들의 발현량 및 가용성에 영향을 주며 $E1_{192-283}$ 단백질 영역내에 HCV 양성환자의 혈청과 특이적으로 반응하는 epitope (s)이 존재한다는 것을 제시해 주고 있다.

수인성 전염병의 현황 및 효율적 관리 (Present status and effective control measure of water-borne infectious diseases in Korea)

  • Water- borne infectious diseases can be acquired by contact with contaminated water or by ingestion of contaminated water. There are many water- borne infectious agents such as bacteria, virus, and parasite. Among many of water- borne infectious diseases, health authorities of Korean government has particularly intensified to prevent and control typhoid fever(class I ), shigellosis(class I ), cholera(class I ), paratyphoid fever(class I), amebiasis(class II ) and leptospirosis(euivalent to class II ) under the communicable disease control law. Water- borne disease Prevention and control guideline itself has been also well provided by the health authorities. However, in practical public health point of view, there are still many problems remained to be solved out; no prospective investigation project to survey water borne infectious diseases under the national disease prevention and control programmes, incredible statistic data of annual notifiable disease report frequent appearance and varieties of drug resistance water- borne infectious agents, little cooperation and information- exchange system in between the related government authorities( the health authorities, the environment sanitation authorities and the food hygiene authorities) which should be closely collaborated, lack of health consciousness of the people, necessity of evaluation and Hndification on to the outcomes of performed health activities and programmes, neglect activities for water quality investigation, shortage of expertise and human resources in the related field, and poor investment of the government budget to develope and improve public health and sanitation field. In order to prevent and control water- borne infectious diseases effectively, it is emphasized that all the above indicated should be considered and performed to improve under the national health and sanitation development programmes.

Versatile Roles of Microbes and Small RNAs in Rice and Planthopper Interactions

  • Planthopper infestation in rice causes direct and indirect damage through feeding and viral transmission. Host microbes and small RNAs (sRNAs) play essential roles in regulating biological processes, such as metabolism, development, immunity, and stress responses in eukaryotic organisms, including plants and insects. Recently, advanced metagenomic approaches have facilitated investigations on microbial diversity and its function in insects and plants, highlighting the significance of microbiota in sustaining host life and regulating their interactions with the environment. Recent research has also suggested significant roles for sRNA-regulated genes during rice-planthopper interactions. The response and behavior of the rice plant to planthopper feeding are determined by changes in the host transcriptome, which might be regulated by sRNAs. In addition, the roles of microbial symbionts and sRNAs in the host response to viral infection are complex and involve defense-related changes in the host transcriptomic profile. This review reviews the structure and potential functions of microbes and sRNAs in rice and the associated planthopper species. In addition, the involvement of the microbiota and sRNAs in the rice-planthopper-virus interactions during planthopper infestation and viral infection are discussed.

재발성 감염 질환의 접근 방법 (Approach to the Children with Recurrent Infections)

  • The major function of immune system is to protect infections. The immune systems are composed of innate and adaptive immunity. In adaptive immunity, the cellular and humoral components interact each other. Neonates and infants are infected frequently, because immune systems are naive and easy to expose to infectious agents. The complete history and physical examination is essential to evaluate the child with recurrent infections. The environmental risk factors of recurrent infections are day care center, cigarette smoke, and air pollution. The underlying diseases such as immunodeficiency, autoimmune diseases, allergy, and disorders of anatomy or physiology increase the susceptibility to infections. In immunodeficiency, infections are characterized by severe, chronic, recurrent, and unusual microbial agents infection. The defects of antibody production are susceptible to sinopulmonary bacterial infections. T cells defects are vulerable to numerous organisms such as virus, fungi, bacteria and etc. The screening tests for immune functions are the quantitative and qualitative measurements of each immune components. A complete blood count with white blood cell, differential, and platelet provide quantitative informations of immune components. Total complement and immunoglobulin levels represent the humoral component. Antibody levels of previously injected vaccines also provide informations of the antigen specific antibody immune responses. T cell and subsets count is quantitative measurement of cell mediated immunity. Delayed hypersensitivity skin test is a crude measurement of T cell function. The long term outcome of children with recurrent infections is completely dependent on the underlying diseases, the initial time of diagnosis and therapy, continued management, and genetic counscelling.

Ginseng alleviates microbial infections of the respiratory tract: a review

  • The detrimental impact of air pollution as a result of frequent exposure to fine particles posed a global public health risk mainly to the pulmonary disorders in pediatric and geriatric population. Here, we reviewed the current literature regarding the role of ginseng and/or its components as antimicrobials, especially against pathogens that cause respiratory infections in animal and in vitro models. Some of the possible mechanisms for ginseng-mediated viral inhibition suggested are improvements in systemic and mucosa-specific antibody responses, serum hemagglutinin inhibition, lymphocyte proliferation, cell survival rate, and viral clearance in the lungs. In addition, ginseng reduces the expression levels of proinflammatory cytokines (IFN-γ, TNF-α, IL-2, IL-4, IL-5, IL-6, IL-8) and chemokines produced by airway epithelial cells and macrophages, thus preventing weight loss. In case of bacterial infections, ginseng acts by alleviating inflammatory cytokine production, increasing survival rates, and activating phagocytes and natural killer cells. In addition, ginseng inhibits biofilm formation and induces the dispersion and dissolution of mature biofilms. Most clinical trials revealed that ginseng, at various dosages, is a safe and effective method of seasonal prophylaxis, relieving the symptoms and reducing the risk and duration of colds and flu. Taken together, these findings support the efficacy of ginseng as a therapeutic and prophylactic agent for respiratory infections.

엘크의 구포자충 감염사례 (A case of elk (Cervus elaphus) coccidiosis in Korea)

  • A thirty-five week old elk was referred for Chronic Wasting Disease (CWD) confirmation from Chungnam province in Korea. Necropsy revealed much bloody contents in the lumen of small and large intestines, and reddening of its mucosa. Microscopically, few coccidia showing various developmental stages were infiltrated in lamina propria of small intestine with granulomatous inflammation and congestion. They were identified as Eimeria spp. according to their location and morphological characteristics. Because fecal examination and oocyst culture were not available, the species of the Eimeria could not be confirmed. There were no detection of pathogenic bacteria such as clostridia and virus in intestinal contents. CWD was shown to be negative in immunohistochemistry test. In conclusion, it was the first Eimeria case detected in small intestine of Korean domestic elk as far as we know although Eimeria spp. were not the main cause of death.

발열과 발진 (Fever and rash)

  • Patients with febrile illness and skin rashes need full and immediate attention. In general, these diseases show mild manifestations and good prognosis. However, causalities of some diseases with fever and rash may be life threatening or trivial. So, the differential diagnosis for those patients is extensive. A through history, a careful physical examination and close observation of clinical progress are very helpful and essential to confirming the diagnosis. Histories of recent travel, drug or specific food ingestion, exposure to human or an animal source of infection may be useful to discover the cause. Although laboratory tests can be useful in making the diagnosis, laboratory results usually are not available immediately. Knowledge and experiences of such diseases may be helpful to reduce the differential diagnosis to a few major possibilities. Rashes can be categorized as petechial, maculopapular, vesicular, urticarial and erythematous. Potential causes include infectious pathogens such as virus, bacteria, rickettsiae, spirohetes, connective tissue diseases, allergic diseases and heamto-oncologic diseases. Because the severity of these diseases can vary mild to life threatening, physicians must perform prompt management decisions regarding empirical therapies. In this article, the differential etiological diagnosis of each type rash is reviewed and discussed, and with emphasis on intensive care of life threatening febrile diseases with rashes that are seen in our country.
