Korea Global Atmosphere Watch Center (KGAWC), which is located in Anmyeondo and, belongs to the Korea Meteorological Administration (KMA), measures sulfur hexafluoride ($SF_6$) in every hour since 2007. In this study, $SF_6$ observed in 2007 are discussed. A gas chromatograph-electron capture detector (GC-ECD) with pre-cooled device is applied during the observation, and produced data are qualified by means of periodic calibration with $SF_6$ standard gas made by Korea Research Institute of Standard and Science (KRISS). $SF_6$ has been greatly paid attention since Kyoto protocol because of its high global warming potential(GWP) with 22,200 times of $CO_2$ in the period of 100 years. It is a man-made compound and has been usually used for gas insulation since 1970s and for etching process in the information technology-based industry since 1990. Average mixing ratio of $SF_6$ in 2007 was 6.65 pmol/mol at Anmyeondo. According to the GAW report published in 2008, average mixing ratio of $SF_6$ in the atmosphere is continuously growing. At present, the average mixing ratio of $SF_6$ in the atmosphere is known to be approximately 6.25 pmol/mol at global observatory. $SF_6$ value in Anmyeondo shows 0.40 pmol/mol greater than that of the Mauna Loa observatory in 2007.
In this study, we have fabricated a fiber-optic radiation sensor using an organic scintillator for high energy electron beam therapy. The intensities of scintillating light from a fiber-optic radiation sensor are measured with different field size, electron beam energy and monitor unit of a clinical linear accelerator. To obtain percent depth dose(PDD), the amount of scintillating light is measured at different depth of polymethylmethacrylate(PMMA) phantom. Also the intensity of Cerenkov light is measured and characterized as a function of incident angle of electron beam and a subtraction method is investigated using a background optical fiber to remove a Cerenkov light.
Background: Salvia miltiorrhiza Bunge has been used in traditional medicine. The type of light source has an effect on the growth properties and composition of functional compounds in plants. In this study, we analyzed the effects of different artificial light sources on the growth characteristics as well as antioxidant and antimicrobial activities of S. miltiorrhiza. Methods and Results: Seedlings of S. miltiorrhiza were grown under various artificial light sources, including fluorescent light (FL), light emitting diode (LED), and microwave electrodeless light (MEL), for 8 weeks. Growth characteristics were the best in plants treated with MEL. DPPH scavenging activity of the shoot was more pronounced with the FL treatments, while the roots were more active in plants grown under single wavelength lights (i.e., blue and red LEDs). Among the different light source treatments, the blue LED resulted in a higher total phenolic content in the plants. Furthermore, growing plants growth under the red LED enhanced their total flavonoid content. Notably, the antimicrobial properties of plants varied significantly between light source treatments in this study. Except for E. coli, all the tested microorganisms were susceptible to the plant extracts. Conclusions: The type of light source may be an important parameter for the enhancement of plant growth and functional compounds in S. miltiorrhiza.
Background: Breast cancer is one of the most frequent cancer types within female populations. Silibinin is a chemotherapeutic agent ative against cancer. Niosomes are biodegradable, biocompatible, safe and effective carriers for drug delivery. Objective:To prepare nanoniosomal silibinin and evaluate its cytotoxicity inthe T-47D breast cancer cell line. Materials and Methods: Niosomes were prepared by reverse phase evaporation of a mixture of span 20, silibinin, PEG-2000 and cholesterol in chloroform and methanol solvent (1:2 v/v). The solvent phase was evaporated using a rotary evaporator and the remaining gel phase was hydrated in phosphate buffer saline. Mean size, size distribution and zeta potential of niosomes were measured with a Zetasizer instrument and then nanoparticles underwent scanning electron microscopy. The drug releasing pattern was evaluated by dialysis and the cytotoxicity of nanoniosomes in T-47D cells was assessed by MTT assay. Results: Particle size, size variation and zeta potential of the niosomal nanoparticles were measured as $178.4{\pm}5.4nm$, $0.38{\pm}0.09$ and $-15.3{\pm}1.3mV$, respectively. The amount of encapsulated drug and the level of drug loading were determined $98.6{\pm}2.7%$ and $22.3{\pm}1.8%$, respectively; released drug was estimated about $18.6{\pm}2.5%$ after 37 hours. The cytotoxic effects of nanoniosome were significantly increased when compared with the free drug. Conclusions: This study finding suggests that silibinin nanoniosomes could serve as a new drug formulation for breast cancer therapy.
Background: Hepatitis B virus (HBV) infection is one of the worldwide public health problem affecting about 300 million people. The envelope protein of HBV consists of three components known as preS1, preS2, and S antigen. According to the recent study, anti-HBs Ab showed effective neutralization ability against HBV from chronic hepatitis B and liver transplant patients, suggesting the possible development of therapeutic antibody. Methods: Spleen cells immunized with S antigen of HBV were fused with myeloma cell line to obtain HBsAg specific monoclonal antibodies. High affinity antibodies against HBsAg (adr, ad and ay type) were selected by competitive ELISA method. Nucleotide sequence of the variable regions of monoclonal antibodies was analyzed by RT-PCR followed by conventional sequencing method. Results: We produced 14 murine monoclonal antibodies which recognize S antigen of HBV. Two of them, A9-11 and C6-9 showed the highest affinity. The sequence analysis of A9-11 revealed that variable regions of the heavy chain and light chains are members of mouse heavy chain I (B) and light chain lambda 1, respectively. Likewise, the sequence analysis of C6-9 revealed that variable regions of the heavy chain and light chains are members of mouse heavy chain II (B) and light chain kappa 1, respectively. Neutralization assay showed that A9-11 and C6-9 effectively neutralize the HBV infection. Conclusion: These results suggest that A9-11 and C6-9 mouse monoclonal antibodies can be used for the development of therapeutic antibody for HBV infection.

Background: In situ gamma spectrometry has been used to measure environmental radiation, assumptions are usually made about the depth distribution of the radionuclides of interest in the soil. The main limitation of in situ gamma spectrometry lies in determining the depth distribution of radionuclides. The objective of this study is to develop a method for subsurface characterization by in situ measurement. Materials and Methods: The peak to valley method based on the ratio of counting rate between the photoelectric peak and Compton region was applied to identify the depth distribution. The peak to valley method could be applied to establish the relation between the spectrally derived coefficients (Q) with relaxation mass per unit area (${\beta}$) for various depth distribution in soil. The in situ measurement results were verified by MCNP simulation and calculated correlation equation. In order to compare the depth distributions and contamination levels in decommissioning KRR site, in situ measurement and sampling results were compared. Results and Discussion: The in situ measurement results and MCNP simulation results show a good correlation for laboratory measurement. The simulation relationship between Q and source burial for the source layers have exponential relationship for a variety depth distributions. We applied the peak to valley method to contaminated decommissioning KRR site to determine a depth distribution and initial activity without sampling. The observed results has a good correlation, relative error between in situ measurement with sampling result is around 7% for depth distribution and 4% for initial activity. Conclusion: In this study, the vertical activity distribution and initial activity of $^{137}Cs$ could be identifying directly through in situ measurement. Therefore, the peak to valley method demonstrated good potential for assessment of the residual radioactivity for site remediation in decommissioning and contaminated site.
Background: Disialoganglioside GD2 is a tumor-associated antigen that is overexpressed on tumor cells of neuroectodermal origin, such as melanoma and neuroblastoma. Anti-idiotypic antibodies that mimic GD2 may induce more effective immune responses than GD2 antigen itself, because they are protein antigens and are known to be able to break immune tolerance. In this study, to explore the potential of anti-idiotypic antibodies as tumor vaccines, the ability of anti-idiotypic antibodies (Ab2) to induce anti-anti-idiotypic antibodies (Ab3) that bind to the original antigen GD2 was investigated. Methods: Six monoclonal anti-idiotypic antibodies (1A8, 1G5, 2B6, 3A4, 3D6, 3H9) to monoclonal antibody M2058, which is a monoclonal antibody to GD2, were produced in mice. Three (1A8, 3A4, 3H9) of them were selected based on their ability to inhibit the binding of Ab1 to D142.34 (murine melanoma cell expressing GD2). These 3 different Ab2 were injected into rabbits, and rabbit Ab3 induced by each of them were characterized. Results: Ab3-containing sera from two rabbits immunized with 1A8, 3A4, or 3H9 bound significantly (P<0.05) to D142.34 but not to B78.96 (GD2-negative cell), and bound significantly (P<0.05) to isolated GD2 but not to GD1a. Ab3-containing sera from two rabbits immunized with 3A4 or 3H9 inhibited significantly (P<0.05) the binding of Ab1 M2058 to D142.34, and inhibited significantly (P<0.05) the binding of Ab1 M2058 to the Ab2. Conclusion: These results suggest that anti-idiotypic antibodies 3A4 and 3H9 have a potential to be used as vaccines against tumors expressing GD2 by inducing GD2-specific antibodies (Ab3).
Background: Novel prognostic biomarkers or therapeutic molecular targets for laryngeal squamous cell carcinoma (LSCC) are an urgent priority. We here sought to identify multiple novel LSCC-associated genes. Methods: Using high-density microarray expression profiling, we identified multiple genes that were significantly altered between human LSCCs and paired normal tissues. Potential oncogenic functions of one such gene, DCUN1D5, were further characterized in vitro. Results: Our results demonstrated that DCUN1D5 was highly expressed in LSCCs. Overexpression of DCUN1D5 in vitro resulted in 2.7-fold increased cellular migration, 67.5% increased invasive capacity, and 2.6-fold increased proliferation. Endogenous DCUN1D5 expression was decreased in a time-dependent manner after genotoxic stress, and silencing of DCUN1D5 by siRNA decreased the number of cells in the S phase by 10.2% and increased apoptosis by 11.7%. Conclusion: Our data suggest that DCUN1D5 in vitro might have vital roles in DNA damage response, but further studies are warranted to assess its significance in vivo.
Background: To investigate the characteristics of allelic distribution of IGF2 and H19 gene polymorphisms in molar tissues compared to normal placentas. Materials and Methods: Forty-nine specimens of molar tissues as well as 100 control normal placental tissues, delivered on the same days, were collected. Polymerase chain reaction (PCR) with restriction fragment length polymorphism (RFLP) on 2% agarose gel electrophoresis was conducted to determine the allelic distribution. The ApaI polymorphism within exon 9 of IGF2 and the RsaI polymorphism within exon 5 of H19 were employed to identify the allelic distribution of the IGF2 and H19 genes, respectively. Then the data for these genes in the molar and normal placenta tissues were compared. Results: The allelic distribution of IGF2 genes found in molar tissue were 21 (42.9%) aa (undigested), 10 (20.4%) ab (heterozygous) and 18 (36.7%) bb (digested), while in normal placenta tissue the values were 22 (22%) aa, 51 (51%) ab, and 27 (27%) bb. The allelic distribution of H19 in molar tissues was 8 (16.2%) aa (undigested), 8 (16.3%) ab (heterozygous) and 33 (67.4%) bb (digested) and in normal placental tissue was 16 (16%) aa, 36 (36%) ab and 48 (48%) bb in normal placenta tissue. These results were significantly different with P values of 0.001 and 0.037 for the allelic distribution of IGF2 and H19, respectively. Conclusions: Molar tissues showed significant differences of allelic distribution of IGF2 and H19 from normal placenta tissues.

Background: Additive manufacturing (AM) is a rapidly expanding new technology involving challenges to occupational health. Here, metal exposure in an AM facility with large-scale metallic component production was investigated during two consecutive years with preventive actions in between. Methods: Gravimetric analyzes measured airborne particle concentrations, and filters were analyzed for metal content. In addition, concentrations of airborne particles <300 nm were investigated. Particles from recycled powder were characterized. Biomonitoring of urine and dermal contamination among AM operators, office personnel, and welders was performed. Results: Total and inhalable dust levels were almost all below occupational exposure limits, but inductively coupled plasma mass spectrometry showed that AM operators had a significant increase in cobalt exposure compared with welders. Airborne particle concentrations (<300 nm) showed transient peaks in the AM facility but were lower than those of the welding facility. Particle characterization of recycled powder showed fragmentation and condensates enriched in volatile metals. Biomonitoring showed a nonsignificant increase in the level of metals in urine in AM operators. Dermal cobalt and a trend for increasing urine metals during Workweek Year 1, but not in Year 2, indicated reduced exposure after preventive actions. Conclusion: Gravimetric analyses showed low total and inhalable dust exposure in AM operators. However, transient emission of smaller particles constitutes exposure risks. Preventive actions implemented by the company reduced the workers' metal exposure despite unchanged emissions of particles, indicating a need for careful design and regulation of the AM environments. It also emphasizes the need for relevant exposure markers and biomonitoring of health risks.
